An antifungal effector from a plant-parasitic nematode modulates host fungal community composition and supports ecological fitness.

Host-associated microbiomes are increasingly recognized as key determinants of plant health, disease development, and ecosystem functioning. Plant pathogens, especially fungal pathogens, have been reported to secrete antimicrobial effectors to modulate the host microbiota and promote colonization. Plant-parasitic nematodes (PPNs) could also modulate host microbial communities, but the processes involved remain to be clarified. Here, we identify a secreted antifungal effector, BxylTLP6, from Bursaphelenchus xylophilus, the causal agent of pine wilt disease. BxylTLP6 degrades fungal cell walls a

Temperature-Dependent Oviposition Models for Monochamus saltuarius (Coleoptera: Cerambycidae).

Monochamus saltuarius Gebler is a serious insect pest in Europe and East Asia regions, including Portugal, Spain, China, Japan, and Korea. It transfers the pine wood nematode Bursaphelenchus xylophilus to conifer trees, resulting in pine wilt disease (PWD). As temperature is a key factor influencing insect population dynamics, temperature-dependent models describing M. saltuarius oviposition could estimate population growth potential and evaluate outbreak risks. In this study, the longevity and fecundity of M. saltuarius females were measured under constant temperature conditions ranging from

Biosynthetic pathway and mating-regulated synthesis of 2-(undecyloxy)-ethanol in Monochamus saltuarius: Implications for pheromone-mediated communication and pest management.

Pine wilt disease (PWD), caused by the pine wood nematode (PWN, Bursaphelenchus xylophilus), poses a severe threat to pine forests worldwide. In northeastern China, Monochamus saltuarius (Coleoptera: Cerambycidae) is a primary vector of this disease. The male-produced aggregation pheromone 2-undecyloxy-1-ethanol plays a critical role in mating and reproductive behavior across several Monochamus species. Understanding its biosynthesis can reveal key targets for pheromone disruption, providing a foundation for pest management. In this study, we investigated the site, temporal dynamics, and regul

The diffusible factor synthase XanB2 is a bifunctional chorismatase that links the shikimate pathway to ubiquinone and xanthomonadins biosynthetic pathways.

The diffusible factor synthase XanB2, originally identified in Xanthomonas campestris pv. campestris (Xcc), is highly conserved across a wide range of bacterial species, but its substrate and catalytic mechanism have not yet been investigated. Here, we show that XanB2 is a unique bifunctional chorismatase that hydrolyses chorismate, the end-product of the shikimate pathway, to produce 3-hydroxybenzoic acid (3-HBA) and 4-HBA. 3-HBA and 4-HBA are respectively associated with the yellow pigment xanthomonadin biosynthesis and antioxidant activity in Xcc. We further demonstrate that XanB2 is a stru

Design, synthesis, nematicidal activity, and preliminary mechanism of thiazole derivatives through the scaffold hopping strategy.

BACKGROUND: Plant-parasitic nematodes (PPNs) pose a serious threat to global agricultural productivity. Most of the currently available nematicides have limitations in terms of efficacy and environmental safety. Therefore, there is an urgent need to discover new, highly effective and environmentally friendly nematicides. RESULTS: A series of thiazole derivatives were designed and synthesized through a scaffold-hopping strategy. Some of these compounds exhibited excellent nematicidal activity. Compound A1 displayed remarkable toxicity against Bursaphelenchus xylophilus, Aphelenchoides besseyi,

Chemical Signals of Vector Beetle Facilitate the Prevalence of a Native Fungus and the Invasive Pinewood Nematode.

In China, the invasive Bursaphelenchus xylophilus, the vector Monochamus alternatus beetle, and associated fungi exhibit a symbiotic relationship causing serious losses to pine forests. Although this complex system has been intensively investigated, the role of vector beetles on the development of associated fungi and their indirect contribution to the prevalence of pinewood nematode (PWN) is yet unknown. Here, three of the highly prevalent fungal species, viz., Sporothrix sp. 1, Ophiostoma ips, and Sporothrix sp. 2 were isolated from beetle chambers in diseased trees in Guangdong province, so

Pre-Visual Detection of Pine Wilt Disease Using an Optimized PSRI Derived from Hyperspectral Drone Imagery.

Pine wilt disease (PWD) is a devastating infectious disease of pine trees caused by the invasion of Bursaphelenchus xylophilus. Achieving rapid and accurate identification of pine trees in the early stages of infection is critical for preventing and controlling its spread, particularly for early warning and intervention before the plants exhibit obvious discoloration symptoms. The Plant Senescence Reflectance Index (PSRI) has shown significant potential for the early detection of PWD. However, existing studies often use its default band combinations for calculation, which may fail to fully exp

Ultrastructural Description of Amphid Neurons in the Pine Wood Nematode Indicates Concurrent Evolution of the Stylet and Specific Neurons.

Understanding how animals perceive environmental stimuli is essential for reconstructing the evolution of their sensory systems. Nematodes provide a useful model for studying sensory adaptation due to their relatively simple nervous systems and broad ecological diversity. The amphid, the primary sensory organ in nematodes, has been well characterized in Caenorhabditis elegans and other bacterivorous species. However, comparatively little is known about amphid structures in nematodes with different ecological niches. In this study, we performed serial section transmission electron microscopy a

Reproductive system development in Monochamus alternatus hope subjected to maturation feeding.

Monochamus alternatus Hope (Coleoptera: Cerambycidae), a vector of the invasive pathogenic nematode Bursaphelenchus xylophilus (Steiner & Buhrer) Nickle, is a notorious forest pest in Asia. After emergence, adult M. alternatus must supplement nutrition to further develop and reach sexual maturity before they can engage in courtship and mating. However, there is a lack of information on the precopulation duration of the Chinese population of M. alternatus and the reproductive system development of both sexes subjected to maturation feeding. In this study, we investigated the precopulation durat

PmWRKY-10 Enhances Pine Wilt Disease Resistance by Promoting Pinosylvin Monomethyl Ether Biosynthesis Through Transcriptional Activation of PmPMT2-1.

Pine wilt disease (PWD), caused by the pine wood nematode (PWN, Bursaphelenchus xylophilus), poses a severe threat to global pine forests. Plants can enhance their resistance to external stresses by eliciting secondary metabolites; however, the transcriptional regulatory mechanisms governing this defence response in pines remain largely unexplored. Here, we report a novel defence regulatory module in Pinus massoniana. PmWRKY-10 may participate in PWN resistance by regulating the biosynthesis of the nematicidal metabolite pinosylvin monomethyl ether (PME). We first demonstrated that PME accumul

Amino Acid-Optimized Medium Improves Long-Term Growth, Conidiation, Stress Tolerance, and Biocontrol Performance of Esteya vermicola Against Pinewood Nematode.

Esteya vermicola is a nematophagous fungus with strong parasitic ability against the pinewood nematode (Bursaphelenchus xylophilus) and shows great potential for the biological control of pine wilt disease. However, this fungus is highly sensitive to environmental stress factors and often exhibits early necrosis when cultured on conventional nutrient-rich media, limiting its large-scale application. In this study, we optimized the long-term cultivation and conidiation conditions of E. vermicola CBS115803 by supplementing minimal medium (MM) with amino acids, and evaluated its stress tolerance

Effect of trap color, pheromone and kairomone release rates, and α-pinene and limonene enantiomers on the capture of Monochamus saltuarius Gebler (Coleoptera: Cerambycidae).

Monochamus saltuarius (Gebler) (Coleoptera: Cerambycidae) is a significant forest pest that spreads pine wood nematodes [Bursaphelenchus xylophilus (Steiner & Bührer) Nickle) (Aphelehncida: Parasitaphelenchidae)] across northern Eurasia, including South Korea. The Korea Forest Service recommends the use of pheromone traps to monitor the spread of this insect vector. However, to improve the accuracy of occurrence monitoring and enhance control through mass trapping, it is necessary to increase the capture efficiency of pheromone traps. In this study, we evaluated various pheromone trap conditio

Immune tolerance of vector beetle to its partner plant parasitic nematode modulated by its insect parasitic nematode.

Immune response of insect vectors to transmitted pathogens or insect hosts against parasites are well studied, whereas the mechanism of tripartite interactions remains elusive. In this study, we investigated the immune interactions of the vector beetle Monochamus alternatus ( Ma) to the devastating plant parasitic nematode Bursaphelenchus xylophilus ( Bx) and the insect parasitic nematode Howardula phyllotretae ( Hp). We report the unique immune mechanism by which the vector beetle tolerates many devastating Bx in its trachea, yet that immune tolerance is compromised by the parasitic nematode

Design, synthesis, nematicidal activity, and preliminary mechanism of pyrimidine derivatives through the scaffold hopping strategy.

BACKGROUND: Plant-parasitic nematodes pose a severe threat to crops, causing over $175 billion in economic losses to global agriculture. RESULTS: To discover novel nematicides, a database of more than 2000 molecules was constructed, and several active scaffold structures were obtained through rapid in vitro nematicidal activity screening. Eight pyrimidine derivatives containing 1,2,4-oxadiazole moieties were designed and synthesized using scaffold hopping strategies that grafted tioxazafen onto the active pyrimidine structure. Among the 38 pyrimidine compounds screened or synthesized, the maj

Plant pathogenic nematode exosomes remodel vector tracheae to enhance pathogen transmission.

Pathogens frequently employ vector-manipulation strategies to enhance their transmission efficiency. Exosomes are increasingly recognized as mediators of interspecific communication between pathogens and their vectors. However, the mechanisms by which plant pathogenic nematode exosomes mediate cross-kingdom manipulation of vector development remain largely unexplored. Here, we demonstrate that the plant pathogenic nematode (Bursaphelenchus xylophilus), transmitted by the vector beetle (Monochamus alternatus), secretes exosomes containing microRNAs (miRNAs) that remodel the tracheal development

Integrating niches of the pathogen, multiple host and vector species highlights abiotic constraints and host-vector interactions shaping pine wilt disease risk in China.

BACKGROUND: Pine wilt disease (PWD), caused by Bursaphelenchus xylophilus (PWN), is a major threat to forest health and pest management in China. Because PWD results from interactions among the pathogen, vector insects, and host plants, single-component analyses may misrepresent disease risk and overlook species-specific contributions. We integrated the environmental niches of the pathogen, hosts, and vectors to characterize abiotic constraints and biotic influences on PWD risk, quantify species-specific contributions and interactions, and inform surveillance priorities. RESULTS: Ensemble eco

Residual Dynamics of Fluopyram and Its Compound Formulations in Pinus massoniana and Their Efficacy in Preventing Pine Wilt Disease.

Injecting chemical agents into tree trunks is a key method for preventing pine wilt disease (PWD). However, the long-term use of conventional trunk injection agents such as emamectin benzoate (EB) and avermectin (AVM) may lead to nematode resistance. Therefore, it is crucial to evaluate the potential of new-generation nematicides, including fluopyram (FLU) and its compound formulations, as alternatives to EB and AVM in PWD prevention. In this study, four trunk injection agents, i.e., 5% FLU microemulsion (ME), 2% AVM + 6% FLU ME, 5% EB ME, and 5% AVM emulsifiable concentrate (EC), were injecte

CO2 drives the pine wood nematode off its insect vector.

Insects have developed special organs, spiracles and the trachea, for oxygen-carbon dioxide exchange to adapt to terrestrial life. The plant-parasitic nematode Bursaphelenchus xylophilus, also known as pine wood nematode (PWN), is vectored by pine sawyer beetles (Monochamus spp.) and causes destructive pine wilt disease, threatening the safety and stability of pine forest ecosystems. Unlike the free-living nematode model species Caenorhabditis elegans, PWN have two distinct life stages (dispersive and propagative), each requiring a unique host relationship ranging from symbiotic/commensal to p

The nematicide emamectin benzoate increases ROS accumulation in Pinus massoniana and poison Monochamus alternatus.

Pine wilt disease (PWD) is caused by the pine wood nematode (PWN, Bursaphelenchus xylophilus) and transmitted by a vector insect, the Monochamus alternatus. The PWN has caused much extensive damage to pine-dominated forest ecosystems. Trunk injection of emamectin benzoate (EB) has been found to be the most useful protective measure against the PWN, due to its low effective dose and long residence time in the field. However, the interactions between EB and the host or the environment remain largely unknown, which limits the efficacy and stability of EB in practical field settings. In this study
