A robust xanthylium-based near-infrared fluorescent probe for viscosity imaging in inflammatory and hepatic injury mice models.

As the principal organ for the metabolism and detoxification of both exogenous and endogenous substances, the liver demonstrates heightened vulnerability to the onset of hepatic pathologies. Aberrant viscosity, acting as a potential biomarker, exhibits a significant correlation with liver diseases such as fatty liver, hepatic fibrosis, and liver injury. Therefore, real-time monitoring of viscosity fluctuations in animal models of liver disease is essential for related pathological investigations. Here, we report a novel viscosity-sensitive fluorescent probe (DJXP) with NIR excitation at 736 nm

Preparation of an activatable benzothiadiazole-based nanoprobe for multispectral optoacoustic and NIR-II fluorescence dual-mode imaging of liver injury.

Light-excited dual-mode imaging that integrates multispectral optoacoustic tomography (MSOT) and near-infrared region II fluorescence (NIR-II FL) imaging allows complementary deep-tissue visualization with high anatomical resolution and molecular sensitivity, thereby enhancing the accuracy of biomedical diagnostics. Activatable probes further advance this approach by producing signals upon recognition of disease-related biomarkers, which reduces background interference and improves imaging specificity. BTPE-NO2@F127, a benzothiadiazole-based nanoprobe, exhibits selective activation of both opt

A NIR Molecular-level Framework Tracks Mercury Ions via Fast and Real-time Determination in Real Samples and Live Cells.

Bio-degradation of mercury ions and their abnormal bioaccumulation in the human body through the food chain can cause various diseases. Therefore, we have designed and synthesized a novel near-infrared (NIR) ratiometric fluorescent probe (E)-2-(3-(3-(1,3-dithiolan-2-yl)-4-hydroxystyryl)-5,5-dimethylcyclohex-2-en-1-ylidene)malononitrile (ICM-S-OH) for detecting Hg2+. Due to the Hg2+-promoted deprotection reaction of mercaptal, ICM-S-OH showed rapid response within 3 min, accompanied by a noticeable fluorescence change from yellow emission to red emission and colour change from yellow to pink. A Fluorescence-Enhanced Probe Enables Rapid Detection of BRAF(V600E) Mutant Activity in Living Cells and Tumor Tissues.

BRAF is the most frequently mutated protein kinase in human cancers, and in particular, the V600E mutation in the kinase domain confers a constitutively active conformation on the protein, thereby promoting oncogenesis. To date, several BRAF kinase inhibitors have been approved for treating cancer patients with the BRAF(V600E) mutation. However, the benefits of BRAF kinase inhibitors in clinical practice are limited, with an objective response rate of less than ∼50%. Therefore, novel diagnostic tools enabling the visualization of BRAF-mutant activity in an activity-based manner are quite desir

Renal-clearable fluorescent probe for early detection of pancreatic cancer in living murine models.

Pancreatic cancer (PC) is a highly aggressive and lethal malignancy with extremely poor patient survival rates. The lack of biomarkers and deep tissue detection barriers has presented long-standing and unsolved issues, severely hindering the early diagnosis of PC even in living rodent models. Here, we report a renal-clearable leucine aminopeptidase (LAP)-activatable fluorescence probe, overturning the probe's pharmacokinetics from the liver to kidney pathway (90% of the injection dosage). This strategy reduces interference in the liver and enables LAP as a biomarker for PC detection in murine

Orange Peel-Assisted GO/rGO Blended to Functionalize Ag Nanoparticles: A Sensing Probe for Organo-Hg2+ Ions in Factual Analytes.

Heavy metals such as Hg2+, Pb2+, and Cd2+ are among the most hazardous environmental pollutants due to their high toxicity and tendency to bioaccumulate in living organisms. Mercury, particularly in the form of alkylated mercury, poses a serious public health concern owing to its accumulation in fish and other aquatic life, which subsequently enters the human food chain. There are different fish species across Asia, such as Catla, Rui, Aar, Bhetki, and Tilapia, that have been reported to bioaccumulate these toxic metals in their tissues. In this study, we present a green-synthesized nanohybrid

Lanthanide Complexes-Based Probe for Dual-Modal Time-Gated Luminescence and Magnetic Resonance Imaging of Physiological pH Fluctuations.

Monitoring pH fluctuations with high sensitivity and spatiotemporal resolution is essential for understanding their roles in physiology and pathology; however, existing approaches are often limited by invasiveness, low sensitivity, or inadequate tissue penetration. Here we report the design and synthesis of pH-Eu/Gd, a small-molecule lanthanide complex that integrates time-gated luminescence (TGL) and magnetic resonance (MR) readouts for dual-modal pH sensing. Constructed by chelating quinolinesulfonamide-DO3A ligands with Eu3+ and Gd3+ ions, pH-Eu/Gd exhibits complementary pH-dependent lumine

A blood-brain barrier-penetrating NIR fluorescent probe enables cross-model optical imaging of H2O2 in three neuroinflammation-related experimental mice models of brain injury.

BACKGROUND: Hydrogen peroxide (H2O2) is a key mediator of oxidative stress in neuroinflammation-related brain injury, yet brain H2O2 imaging remains challenging because of the blood-brain barrier (BBB) and tissue autofluorescence. In addition, most reported probes have been evaluated in only one or a few disease models, limiting cross-model comparison. Here, we developed MH-1, an H2O2-responsive near-infrared (NIR) fluorescent probe for brain-section optical mapping after systemic administration. RESULTS: MH-1 exhibited an approximately 60-fold fluorescence turn-on at 667 nm, enabled H2O2 det

Unsaturated Fatty Acid-Conjugated Amonafide Fluorescent Prodrugs for Autophagy Disruption and Cancer Theranostics.

Despite the powerful antitumor activity of amonafide (ANF), its use in the clinic has been limited by dose-dependent toxicities. To address this challenge, we developed a series of unsaturated fatty acid-amonafide conjugates, leveraging the biocompatibility and inherent tumor-targeting capacity of unsaturated fatty acids. Through amidation reactions, five conjugates were synthesized and evaluated. ANF-DHA stood out as the most promising candidate, demonstrating not only targeted cytotoxicity against cancer cells but also the ability to overcome chemoresistance. Additionally, the design allows

A folate receptor-targeted NIR-II probe improves intraoperative imaging and surgery of metastatic ovarian cancer.

Intraoperative fluorescence imaging holds great promise for achieving complete cytoreduction in ovarian cancer, but its clinical utility is currently constrained by the limitations of existing imaging agents. In particular, the leading clinical probe, OTL38, operates in the near-infrared I (NIR-I; 700 to 1000 nanometers) window, which suffers from limited tissue penetration and suboptimal resolution. Here, a folate receptor α (FRα)-targeted NIR-II (1000 to 3000 nanometers) imaging probe was developed by incorporating a 4-(4-iodophenyl)butanoic acid moiety. This structural modification enables

Flexible disulfide-bridged hyperbranched poly(amido amine)s based nonconjugated carbonized polymer dots for iron (III) sensing and its application in meat by-product.

Herein, a novel flexible disulfide-bridged hyperbranched poly(amido amine)s based nonconjugated carbonized polymer dots (ssHPA-CPDs) was developed using citric acid and ssHPA with disulfide bonds as precursors. The structure and composition of ssHPA-CPDs were characterized by transmission electron microscopy, X-ray photoelectron spectroscopy, Fourier transform infrared spectroscopy, raman spectroscopy, and small-X-ray scattering. The results show that when the mass ratio of ssHPA-CPDs/citric acid is 0.5, graphite nitrogen and thiophenone/thiazole sulfur occupy a dominant position, with a highe

Indole-bipyridyl fluorophores as dual-functional probes for endoplasmic reticulum imaging and Zn2+ sensing in live cells.

The endoplasmic reticulum (ER) plays a central role in protein folding, lipid biosynthesis, and calcium regulation, and its dysfunction is closely linked to diseases such as cancer, neurodegeneration, and metabolic disorders. Here, we report a new class of indole-bipyridyl (IBP) fluorophores designed for high-contrast visualisation of ER architecture and dynamic ER-associated processes in living cells. Three derivatives (IBP-1, IBP-2, and IBP-3) were synthesised through a condensation-alkylation strategy and exhibited strong absorption around 420-430 nm, large Stokes shifts, and pronounced env

Imaging alkaline phosphatase activity in alcoholic liver disease via a rational-designed NIR fluorogenic probe.

Alkaline phosphatase is a zinc-based metalloenzyme present in various tissues, and its levels increase in alcoholic liver injury. Herein, we report an enzyme-activatable NIR probe, BDP-R-ALP, that enables highly sensitive and selective detection of alkaline phosphatase activity. At the cellular level, the probe realized real-time imaging of endogenous ALP in ALP-positive HepG2 cells and an alcoholic-liver-disease (ALD) cell model. Furthermore, it enabled tracking of endogenous ALP in a tumor-bearing mouse model. Additionally, two murine models of ALD, simulating hazardous drinking and excessiv

A near-infrared fluorescent probe activated by cysteine for imaging hyperglycemia-induced oxidative stress injury in the liver.

BACKGROUND: Chronic hyperglycemia can cause oxidative stress damage to the liver, which is one of the main complications of diabetes. Cysteine (Cys), acting as a key thiol involved in maintaining the redox balance of the liver, plays an important role in the pathogenesis of diabetic liver injury. There is currently a lack of means for real-time monitoring of the dynamic changes of Cys in the liver. Therefore, the development of related technologies is crucial for revealing the potential pathological mechanisms of diabetes. RESULTS: Here, we developed a near-infrared (NIR) fluorescent probe th

Ratiometric near-infrared fluorescent probe for imaging Peroxynitrite fluxes in rheumatoid arthritis and drug-induced liver injury models.

BACKGROUND: Peroxynitrite (ONOO-) plays a pivotal role in the pathogenesis of Rheumatoid arthritis (RA) and drug-induced liver injury (DILI), with its dynamic accumulation closely associated with disease progression. To effectively monitor the level of ONOO- in biological system, fluorescent probes offer distinct advantages for ONOO-, including high spatiotemporal resolution, non-invasive imaging capability, and exceptional sensitivity for detecting transient ONOO- fluctuations in living systems. These probes enable real-time visualization of ONOO- in inflamed joints or drug-stressed livers, f

Development of a Fluorescent Probe for Real-Time In Vivo Imaging of Cysteine Dynamics in a Mouse Model of Lipopolysaccharide-Induced Acute Lung Injury.

Acute lung injury (ALI) involves inflammatory dysregulation and oxidative stress, processes in which cysteine (Cys), an endogenous sulfur-containing biothiol, participates through thiol redox homeostasis. We designed HD-Cys, an activatable hemicyanine probe, to visualize Cys in real-time in an LPS-induced murine ALI model. The reaction with Cys releases the emissive HD form and produces a pronounced near-infrared turn-on response. HD-Cys showed high selectivity, a detection limit of 160 nM, and a linear concentration-dependent response. Its performance was examined in BEAS-2B cells, serum, and

An activatable naphthalimide-based fluorescent probe for hydrogen peroxide detection: visualizing microcystin-LR induced oxidative stress in zebrafish and hepatocyte injury mechanism.

Microcystin-LR (MC-LR), a toxic cyanotoxin, accumulates in fish through uptake, posing severe threats to fisheries' safety and human health. The interaction between MC-LR and aquatic fish induces oxidative stress, which triggers excessive production of reactive oxygen species (ROS), notably hydrogen peroxide (H2O2). Abnormal ROS fluctuations are closely linked to the onset of various diseases, making the development of in vivo H2O2 detection and regulation methods critically important. Herein, we designed and synthesized an activatable fluorescent probe using naphthalimide as the fluorophore.

Intracellular Localization of DNA-Wrapped Single-Walled Carbon Nanotube Sensors for Nitric Oxide Detection in Triple-Negative Breast Cancer Cells.

Spatiotemporal sensors capable of monitoring analyte activity in subcellular compartments of cells are critical to advancing our understanding of complex disease progression. Nitric oxide (NO) plays a key role in tumor progression, yet its precise function remains unclear due to its bimodal effect, which is influenced by its subcellular distribution. Factors such as membrane permeability, NO reactivity, and redox states of intracellular compartments may determine whether NO promotes or suppresses tumor growth. Current sensors are limited in providing spatial and temporal information about NO i

A chemodosimetric chemosensor for the ratiometric detection of nerve agent-mimic DCP in solution and vapor phases.

Nerve agents are among the most deadly and lethal chemical warfare agents (CWAs). Rapid identification is crucial for specialized individuals to take action against dangerous drugs. This paper describes the synthesis and characterisation of a probe (MNFZ) based on the methoxy naphthalene-furoic hydrazide group. The probe rapidly (100 s) detects and quantifies the nerve-agent simulant diethyl chlorophosphate (DCP) in both solution and vapor phases. This sensor uses a new recognition center, furoic hydrazide, where the nitrogen atom of the imine group (CN) attacks the electrophilic core phospho
