A dynamic microcosm biofilm model for root carious-like lesion development: analysis of demineralization and microbiological characterization.

This study investigated the multifunctional oral cavity simulator (MOCS) in terms of microbial composition, functional profile, and dentin root demineralization. Microcosm biofilms were grown on dentin using human saliva for 4, 7, and 14 days, with exposure to sucrose and a mucin-enriched medium. Biofilms were analyzed for microbial viability and composition through CFU count and 16S-rRNA gene sequencing. Demineralization was quantified by percentage surface hardness change (%SHC), mineral loss (ML), and lesion depth (LD). The results showed microbial viability at all time points. After 7 days

The influence of surface materials on microbial biofilm formation in aviation fuel systems.

The ability of different microbes to form biofilms on materials found in aviation fuel systems was assessed using both individual isolates and complex microbial communities. Biofilm formation by the Gram-negative bacterium, Pseudomonas putida, the fungus Amorphotheca resinae and the yeast, Candida tropicalis, was influenced by material surface properties although this differed between isolates. Biofilm formation was greatest at the fuel-water interface. The Gram-positive bacterium Rhodococcus erythropolis, in contrast, was able to grow on most surfaces. When a subset of materials was exposed t

Quantifying biofilm matrix components: effects of chlorhexidine and orthophthalaldehyde on Candida parapsilosis and Staphylococcus aureus.

Candida and Staphylococcus species are responsible for hospital-acquired infections, forming resilient biofilms. This study evaluated the impact of biocides on the biofilm matrix components of Candida parapsilosis and Staphylococcus aureus in monospecies and mixed biofilms. Proteins, carbohydrates, and extracellular DNA (eDNA) were quantified using the Bradford reagent, phenol-sulfuric acid, and silica column extraction with spectrophotometric readings at 260 and 280 nm. Biofilms were treated with 0.5% chlorhexidine (CLX) and 0.55% ortho-phthalaldehyde (OPA) for 3 and 10 min, respectively. Res

Exploring quinazoline-derived copper(I) complex coated intravaginal ring against vulvovaginal candidiasis causing Candida species.

Vulvovaginal candidiasis (VVC) is especially prevalent among intrauterine device (IUD) and intravaginal ring (IVR) users. Candida albicans is the leading causative agent of VVC followed by Candida glabrata. Ascribed to the increased drug resistance by Candida spp. to the currently available drugs, this study has focused on the novel quinazoline-derived copper(I) complexes as anti-candida agents. As a novel approach, a vaginal ring was coated with the best quinazoline-derived copper(I) complex, and biofilm disruption ability was evaluated. The coated vaginal ring eradicated 70% of preformed bio

Nano-boosted thymoquinone: moving beyond antibiotics to inhibit Staphylococcus aureus biofilms.

Staphylococcus aureus is a high-priority pathogen requiring novel antimicrobial strategies. Thymoquinone (TQ), a bioactive compound from Nigella sativa, exhibits antimicrobial and antibiofilm properties, but its precise mechanisms remain unclear. This study evaluated free and nano-encapsulated TQ against multidrug-resistant S. aureus isolates. Nano-encapsulation enhanced biofilm penetration, with TQ significantly reducing extracellular DNA (eDNA) at sub-MIC levels without affecting initial adhesion, exopolysaccharides, or enzymatic virulence. Comparative analysis with vancomycin, ciprofloxacin

Butenolide synergises with vancomycin to eradicate pre-formed biofilm of Staphylococcus aureus by interfering with energy-associated metabolism.

Bacterial biofilms significantly contribute to persistent infections and the emergence of drug resistance of Staphylococcus aureus. Integrating conventional antibiotics with antibiofilm agents represents a promising strategy for combating biofilm-associated infections. This study systematically investigated the antibiofilm activity and underlying mechanisms of butenolide (BU) against methicillin-resistant S. aureus (MRSA), with a focus on the synergistic effects between BU and vancomycin (VAN). BU exhibited dual antibiofilm activities by efficiently preventing biofilm formation and eradicating

Glutaraldehyde fixation is more advantageous for Malassezia furfur biofilm measurements.

The fixative most commonly used in biofilm measurement studies of pathogenic yeasts is ethanol. However, due to lipid dissolution in ethanol, this method may not be the optimal choice for certain yeasts which have a high lipid content in their cell walls, such as human pathogen Malassezia furfur. We conducted a study to compare the measurement values of 26 clinical strains of Malassezia furfur using glutaraldehyde and paraformaldehyde instead of ethanol. After the fixation step, standard staining methods were applied for biomass and extracellular polymers. Imaging was performed using scanning

Changes of microbial communities in loose sediments with accumulation time in chlorinated drinking water distribution systems.

'Yellow water' issues frequently occur in drinking water supply systems on university campuses, primarily caused by the resuspension of loose sediments within pipelines, which may pose high potential microbiological risks. In order to investigate the accumulation of sediments accompanying microbiological risks in municipal pipelines, 'water + sediment' samples were collected periodically over two semesters in a university campus exhibiting divergent temperature patterns. Results indicated that sediment quantity increased rapidly within a short period; however, microbial biomass did not increas

Baicalein of Scutellaria baicalensis inhibits Mycobacterium abscessus biofilm formation to restore the antibiotics susceptibility.

Mycobacterium abscessus is resistant to traditional anti-TB medications and most currently available antibiotics, its strong biofilm formation may contribute to the resistance and make the situation even worse. In this study, Scutellaria baicalensis extract significantly inhibited the planktonic growth as well as the biofilm formation of M. abscessus. Baicalein, the principal active element in S. baicalensis extract, inhibited the biofilm formation instead of the planktonic growth and reduced extracellular matrix lipids in M. abscessus biofilm. The synergistic effects of baicalein in combinati

Improvement of a low-cost protocol for a simultaneous comparative evaluation of hydrolytic activity between sessile and planktonic cells: Candida albicans as a study model.

Candida albicans is often implicated in nosocomial infections with fatal consequences. Its virulence is contributed to hydrolytic enzymes and biofilm formation. Previous research focused on studying these virulence factors individually. Therefore, this study aimed to investigate the impact of biofilm formation on the hydrolytic activity using an adapted low-cost method. Eleven strains of C. albicans were used. The biofilms were formed on pre-treated silicone discs using 24-well plates and then deposited on the appropriate agar to test each enzyme, while the planktonic cells were conventionally
