Metabolomic analysis of pressure-overloaded and infarcted mouse hearts.

BACKGROUND: Cardiac hypertrophy and heart failure are associated with metabolic dysregulation and a state of chronic energy deficiency. Although several disparate changes in individual metabolic pathways have been described, there has been no global assessment of metabolomic changes in hypertrophic and failing hearts in vivo. Hence, we investigated the impact of pressure overload and infarction on myocardial metabolism. METHODS AND RESULTS: Male C57BL/6J mice were subjected to transverse aortic constriction or permanent coronary occlusion (myocardial infarction [MI]). A combination of LC/MS/M

Effects of ultraviolet light on biogenic amines and other quality indicators of chicken meat during refrigerated storage.

Radiation from UV-C has been demonstrated as a potential surface decontamination method in addition to several advantages over regular sanitation methods. However, UV-C radiation possibly affects the physicochemical properties of meat products. To determine the optimum exposure time for bacterial reduction, 39 chicken breasts, inoculated with a pool of Salmonella spp., were submitted to 3 levels of UV-C intensities (0.62, 1.13, and 1.95 mW/cm²) for up to 120 s. After the optimum exposure time of 90 s was determined, changes in the biogenic amines, total aerobic mesophilic bacteria, Enterobacte

Prevention of a plant disease by specific inhibition of fungal polyamine biosynthesis.

DL-alpha-Difluoromethylornithine (DFMO), an inhibitor of the polyamine biosynthetic enzyme ornithine decarboxylase (EC 4.1.1.17), strongly retards the growth of several species of phytopathogenic fungi in vitro. Such inhibition can be completely reversed by putrescine or spermidine, confirming the essentiality of polyamines for growth of fungal hyphae. We now show that DFMO can protect bean plants (Phaseolus vulgaris Linnaeus cv. Pinto) against infection by uredospores of the bean rust fungus, Uromyces phaseoli Linnaeus, race O. Unifoliolate leaves of 10-day-old greenhouse-grown seedlings were

Lead stress triggers the synthesis of protecting salicylic acid through polyamine accumulation in the Pb-resistant species Piptatherum miliaceum (L.) Cosson.

To assess the impact of polyamines and salicylic acid on the response of Piptatherum miliaceum to lead toxicity, plants were cultivated in nutrient solution containing 500 μM Pb(NO3)2 in the presence or absence of polyamine synthesis inhibitors (d-Arginine 5 mM, cyclohexylammonium 5 mM, methylglyoxal-bis-guanyl-hydrazone 1 mM), ethylene synthesis inhibitor (2-aminoethoxyvinyl-glycine 2 μM) and/or exogenous supplementation of 100 μM putrescine, spermidine, spermine or 10 μM salicylic acid. Although all plants remained alive until the end of the treatment, Pb toxicity induced a decrease in growt

Characterising the Response of Human Breast Cancer Cells to Polyamine Modulation.

Breast cancer is a complex heterogeneous disease with multiple underlying causes. The polyamines putrescine, spermidine, and spermine are polycationic molecules essential for cell proliferation. Their biosynthesis is upregulated in breast cancer and they contribute to disease progression. While elevated polyamines are linked to breast cancer cell proliferation, there is little evidence to suggest breast cancer cells of different hormone receptor status are equally dependent on polyamines. In this study, we characterized the responses of two breast cancer cells, ER+ (oestrogen receptor positive

The Development of Microbiota and Metabolome in Small Intestine of Sika Deer (Cervus nippon) from Birth to Weaning.

The dense and diverse community of microorganisms inhabiting the gastrointestinal tract of ruminant animals plays critical roles in the metabolism and absorption of nutrients, and gut associated immune function. Understanding microbial colonization in the small intestine of new born ruminants is a vital first step toward manipulating gut function through interventions during early life to produce long-term positive effects on host productivity and health. Yet the knowledge of microbiota colonization and its induced metabolites of small intestine during early life is still limited. In the prese

New insights of polyamine metabolism in testicular physiology: A role of ornithine decarboxylase antizyme inhibitor 2 (AZIN2) in the modulation of testosterone levels and sperm motility.

The specific role of polyamines in the testis physiology is not fully understood. Antizymes (OAZs) and antizyme inhibitors (AZINs) are modulators of ornithine decarboxylase (ODC), a key enzyme in polyamine biosynthesis and polyamine uptake. Although the three known OAZs are expressed in the testis, only OAZ3 is testis specific and has been proven to have an essential role in male fertility. Regarding the two existing AZINs, AZIN2 is the most abundantly expressed member in this gonad. Whereas previous studies suggested that AZIN2 might participate in mouse spermatogenesis, immunohistological an

Optimization of GC/TOF MS analysis conditions for assessing host-gut microbiota metabolic interactions: Chinese rhubarb alters fecal aromatic amino acids and phenol metabolism.

In this paper, an optimized method based on gas chromatography/time-of-flight mass spectrometry (GC-TOFMS) platform has been developed for the analysis of gut microbial-host related co-metabolites in fecal samples. The optimization was performed with proportion of chloroform (C), methanol (M) and water (W) for the extraction of specific metabolic pathways of interest. Loading Bi-plots from the PLS regression model revealed that high concentration of chloroform emphasized the extraction of short chain fatty acids and TCA intermediates, while the higher concentration of methanol emphasized indol

Evaluation of difluoromethylornithine for the chemoprevention of Barrett's esophagus and mucosal dysplasia.

Patients with Barrett's esophagus (BE) and dysplasia are candidates for chemopreventive strategies to reduce cancer risk. We determined the effects of difluoromethylornithine (DMFO) on mucosal polyamines, gene expression, and histopathology in BE. Ten patients with BE and low-grade dysplasia participated in a single-arm study of DFMO (0.5 g/m(2)/d) given continuously for 6 months. Esophagoscopy with biopsies was conducted at baseline, 3, 6, and 12 months. Dysplasia was graded by a gastrointestinal pathologist. Audiology was assessed (at baseline and at 6 months). Mucosal polyamines were measur

Functional coupling of rat myometrial alpha 1-adrenergic receptors to Gh alpha/tissue transglutaminase 2 during pregnancy.

Gh alpha protein, which exhibits both transglutaminase and GTPase activities, represents a new class of GTP-binding proteins. In the present study, we characterized Gh alpha in rat uterine smooth muscle (myometrium) and followed its expression during pregnancy by reverse transcription-PCR and Western blot. We also measured transglutaminase and GTP binding functions and used a smooth muscle cell line to evaluate the role of Gh alpha in cell proliferation. The results show that pregnancy is associated with an up-regulation of Gh alpha expression at both the mRNA and protein level. Gh alpha induc

Use of trifluoroperazine isolates a [(3)H]Ifenprodil binding site in rat brain membranes with the pharmacology of the voltage-independent ifenprodil site on N-methyl-D-aspartate receptors containing NR2B subunits.

The use of trifluoroperazine in a well washed rat brain membrane preparation revealed [(3)H]ifenprodil binding to a single high affinity state with the pharmacology of N-methyl-D-aspartate (NMDA) receptors containing NR2B subunits. Inhibition of [(3)H]ifenprodil binding in the presence of trifluoroperazine by 10 NR1a/NR2B selective agents was highly correlated with their inhibition at rat NR1a/NR2B receptors expressed in Xenopus ooctyes and [(3)H]TCP binding to rat brain NR2B subunit containing NMDA receptors but not with their inhibition of [(3)H]DTG binding. Copper, biogenic amines, and amine oxidases.

Amine oxidases have been classified in the past on the basis of either (a) the structural requirements in the substrate or (b) the tissue (or species) of origin, or both. As knowledge about the chemistry of these enzymes grows, their classification on the basis of chemical structure is becoming possible. Currently, many amine oxidases can be categorized according to whether they contain riboflavin (e.g. the monoamine oxidases -- EC 1.4.3.4) or copper (e.g. the amine oxidases of plasma and the diamine oxidases EC 1.4.3.6 -- found prominently in pig kidney cortex, placenta, and pea seedlings). Multiple active site conformations revealed by distant site mutation in ornithine decarboxylase.

Ornithine decarboxylase (ODC) is an obligate homodimer that catalyzes the pyridoxal 5'-phosphate-dependent decarboxylation of l-ornithine to putrescine, a vital step in polyamine biosynthesis. A previous mutagenic analysis of the ODC dimer interface identified several residues that were distant from the active site yet had a greater impact on catalytic activity than on dimer stability [Myers, D. P., et al. (2001) Biochemistry 40, 13230-13236]. To better understand the basis of this phenomenon, the structure of the Trypanosoma brucei ODC mutant K294A was determined to 2.15 A resolution in compl

Bergamot (Citrus bergamia) preserves cardiac structure and function and modulates myocardial redox and metabolic profiles in diet-induced obesity.

Obesity promotes cardiac remodeling and dysfunction through oxidative stress, inflammation, and metabolic disturbances. Citrus bergamia (bergamot) has shown beneficial effects on obesity-related cardiometabolic risk factors, but its cardiac effects remain unclear. We evaluated bergamot supplementation in a rat model of diet-induced obesity using integrated assessments of cardiac structure and function, redox and inflammatory status, biogenic amines, fatty acids, and metabolites. Wistar rats (n = 54) were randomized to Control, Obese, or Obese + Bergamot groups for 20 weeks; bergamot was admini

Multiple polyamine transport pathways in cultured pulmonary artery smooth muscle cells: regulation by hypoxia.

The polyamines putrescine (PUT), spermidine (SPD), and spermine (SPM) are a family of low molecular weight organic cations that play essential intracellular regulatory roles in cell growth and differentiation. Consistent with this important function, increases in cellular polyamine contents are necessary for a variety of physiologic and pathologic events in the lung, including development of hypertensive pulmonary vascular disease secondary to chronic alveolar hypoxia. In intact rat lungs, hypoxia depresses ornithine decarboxylase activity, the initial rate-limiting enzyme in de novo polyamine

Decreased plasma postheparin diamine oxidase levels in celiac disease.

The highest diamine oxidase activity is contained in small-bowel mucosa and, after heparin administration, the enzyme is released by the intestine into the plasma. Previous experimental studies showed that measurement of plasma postheparin diamine oxidase activity is a sensitive test for quantitating the length and severity of small-bowel mucosal injury. On this basis, we measured plasma diamine oxidase activity in celiac disease, a condition characterized by a loss of mature enterocyte mass. Twenty-five untreated celiac patients, 21 celiac patients on a gluten-free diet, 16 patients with smal
