Waardenburg Syndrome type 1: A case report.

Waardenburg Syndrome (WS) is a rare hereditary disorder that is characterized by the clinical manifestations of oculocutaneous anomalies of pigmentation, congenital deafness, dystopia canthorum, and broad nasal root. It demonstrates both genetically and clinically heterogenous characteristics. In this article, we report an 11-month-old boy with WS1, one of four clinicat types of WS. He exhibited white forelock, hypopigmented macules and patches, heterochromia irides, and dystopia canthorum.

A method to simultaneously monitor hepatitis C virus NS3 helicase and protease activities.

The hepatitis C virus NS3 protein contains an N-terminal serine protease and a C-terminal helicase that unwinds RNA or DNA duplexes. The HCV NS3 protein is the target for several antiviral drugs in clinical trials, which inhibit the protease function. A method is reported to simultaneously monitor the helicase and protease function of the NS3 protein in a single reaction using fluorescence spectroscopy and a single chain recombinant protein where NS3 is fused to its protease activator NS4A. The method monitors both activities together in real time and is amenable to high-throughput screening.

Serum ferritin correlates with activity of anti-MDA5 antibody-associated acute interstitial lung disease as a complication of dermatomyositis.

Dermatomyositis (DM) is occasionally complicated by interstitial lung disease. Acute/subacute interstitial pneumonia (A/SIP) with DM is intractable and life threatening. Clinically amyopathic dermatomyositis (C-ADM) is also reported to be complicated with A/SIP, especially in those patients with anti-melanoma differentiation-associated gene 5 (MDA5) antibody. In the present cases, we indicate that serum ferritin level correlated with activity of A/SIP with DM. Two patients, a 65-year-old woman and a 30-year-old woman, were diagnosed with anti-MDA5 antibody-associated A/SIP with DM. Serum ferri

PAX6 immunoreactivity in the diencephalon and midbrain of alligator during early development.

PAX6 expression was examined during early development of the diencephalon and midbrain of Alligator using an immunocytochemical methodology. These observations focused on the basal plate to determine whether diencephalic prosomere organization in this region followed a pattern previously identified for alar plate areas. PAX6 expression was also described in alar diencephalic regions and the adjacent midbrain. PAX6 (+) cells in the basal plate were first seen in prosomere 1 at stage 7, in the midbrain at stage 10, and lastly in prosomeres 2 and 3 at stage 11. By stage 12, a nearly continuous co

Tbx2b is required for the development of the parapineal organ.

Structural differences between the left and right sides of the brain exist throughout the vertebrate lineage. By studying the zebrafish pineal complex, which exhibits notable asymmetries, both the genes and the cell movements that result in left-right differences can be characterized. The pineal complex consists of the midline pineal organ and the left-sided parapineal organ. The parapineal is responsible for instructing the asymmetric architecture of the bilateral habenulae, the brain nuclei that flank the pineal complex. Using in vivo time-lapse confocal microscopy, we find that the cells th

Tbx3 is required for outflow tract development.

Conotruncal and ventricular septal congenital heart anomalies result from defects in formation and division of the embryonic outflow tract. Cardiac remodeling during outflow tract and ventricular septation converts the tubular embryonic heart into a parallel circulatory system with an independent left ventricular outlet and right ventricular inlet. Tbx3 encodes a T-box-containing transcription factor expressed in the developing conduction system of the heart. Mutations in TBX3 cause ulnar-mammary syndrome. Here we show that mice lacking Tbx3 develop severe outflow tract defects, including conn

Spinocerebellar ataxia 17 (SCA17) and Huntington's disease-like 4 (HDL4).

Spinocerebellar ataxia 17 (SCA17) or Huntington's disease-like-4 is a neurodegenerative disease caused by the expansion above 44 units of a CAG/CAA repeat in the coding region of the TATA box binding protein (TBP) gene leading to an abnormal expansion of a polyglutamine stretch in the corresponding protein. Alleles with 43 and 44 repeats have been identified in sporadic cases and their pathogenicity remains uncertain. Furthermore, incomplete penetrance of pathological alleles with up to 49 repeats has been suggested. The imperfect nature of the repeat makes intergenerational instability extrem

Maturation of ribbon synapses in hair cells is driven by thyroid hormone.

Ribbon synapses of inner hair cells (IHCs) undergo developmental maturation until after the onset of hearing. Here, we studied whether IHC synaptogenesis is regulated by thyroid hormone (TH). We performed perforated patch-clamp recordings of Ca2+ currents and exocytic membrane capacitance changes in IHCs of athyroid and TH-substituted Pax8-/- mice during postnatal development. Ca2+ currents remained elevated in athyroid IHCs at the end of the second postnatal week, when it had developmentally declined in wild-type and TH-rescued mutant IHCs. The efficiency of Ca2+ influx in triggering exocytos

Precraniate origin of cranial motoneurons.

The craniate head is innervated by cranial sensory and motor neurons. Cranial sensory neurons stem from the neurogenic placodes and neural crest and are seen as evolutionary innovations crucial in fulfilling the feeding and respiratory needs of the craniate "new head." In contrast, cranial motoneurons that are located in the hindbrain and motorize the head have an unclear phylogenetic status. Here we show that these motoneurons are in fact homologous to the motoneurons of the sessile postmetamorphic form of ascidians. The motoneurons of adult Ciona intestinalis, located in the cerebral ganglio

22q11 deletion syndrome: a review of some developmental biology aspects of the cardiovascular system.

The morphology and molecular genetics of the 22q11 deletion syndrome cardiovascular anomalies are reviewed. Special emphasis is given to TBX1, recently identified and considered to be the potential key gene for this clinical syndrome. The TBX1 downstream molecular pathways modulating the normal development of the pharyngeal apparatus are also discussed, and emphasis is given to the possible, equally fundamental role of downstream molecular pathway disruption in causing the clinical 22q11 deletion phenotype features.

Characterization of the uridine diphosphate-glucuronosyltransferase-catalyzing thyroid hormone glucuronidation in man.

Increased thyroid hormone glucuronidation in rats caused by exposure to xenobiotics has stimulated a search for the individual uridine diphosphate-glucuronosyltransferases (UGTs) catalyzing this reaction in rats and man. Microsomal preparations from Crigler-Najjar liver, normal human liver, and kidney have been used to try to identify the UGT isoforms responsible for glucuronidation of the thyroid hormones. The predominant thyroid hormone released from the thyroid gland, T4, and the inactive rT3 are glucuronidated by cloned expressed bilirubin UGT1A1 and also phenol UGT1A9. Results from Crigle

At the crossroads of immunity and hijacking: dual roles of DDX proteins during host-virus interactions.

The DEAD-box (DDX) protein family, constituting a critical subgroup of the Superfamily 2 (SF2) helicase family, plays extensive roles in nucleic acid metabolism, gene expression regulation and innate immune signaling, and exerts a key regulatory role during viral infection. During viral infection, DDX proteins function as both RNA helicases and host restriction factors, exerting antiviral effects by recognizing viral nucleic acids, regulating signaling pathways and maintaining the host immune response. Conversely, through complex and diverse mechanisms, viruses can hijack or antagonize DDX pro

Effect of Glycosylphosphatidylinositol-specific phospholipase D1 knockout on the expression of antioxidant enzymes and sirtuin.

BACKGROUND: Glycosylphosphatidylinositol-specific phospholipase D1 (Gpld1) is a membrane-associated enzyme that modulates diverse cellular processes through the cleavage of glycosylphosphatidylinositol (GPI)-anchored proteins. Although recent studies have linked circulating Gpld1 to exercise-induced rejuvenation, its cell-autonomous role in coordinating redox homeostasis, melanogenesis, and cellular aging has not been fully elucidated. METHODS AND RESULTS: To address this, a stable Gpld1 knockout (KO) was generated in B16F1 melanoma cells using CRISPR/Cas9-mediated genome editing to investiga

Bufalin reverses oxaliplatin resistance in gastric cancer by targeting YBX1 to suppress glycolysis.

Oxaliplatin resistance is a key challenge in gastric cancer therapy. This study explored how bufalin, an active component of Chansu, reverses this resistance. We found that bufalin targets the transcription factor YBX1, which is upregulated in resistant cells. YBX1 activates STC1 expression, leading to PI3K/Akt pathway activation, increased HK2 levels, and enhanced glycolysis-all contributing to resistance. Bufalin inhibits this YBX1/STC1/glycolysis axis. Using in vitro assays (proliferation, apoptosis, glycolysis measurement) and in vivo models, we demonstrated that bufalin suppresses glycoly

FOXO transcription factors in Alzheimer's Disease: balancing neuroprotection and neuronal degeneration.

Alzheimer's disease (AD) is a progressive neurodegenerative condition marked by the accumulation of amyloid-β, hyperphosphorylation of tau, oxidative stress, synaptic dysfunction, and neuroinflammation. Recent research underscores the Forkhead box O (FOXO) family of transcription factors (FOXO1, FOXO3, FOXO4, FOXO6) as crucial regulators of these pathogenic processes. FOXOs regulate antioxidant defenses, autophagy, mitochondrial quality control, and apoptosis by functioning downstream of insulin/PI3K-Akt and stress-responsive pathways. This context-dependent activity enables FOXOs to act as du

Accelerating bone healing in femoral defect model using FBXO6-modified bone marrow-derived mesenchymal stem cells on a collagen scaffold.

Bone repair following large defects remains a significant clinical challenge due to limited osteogenic capacity. F-box only protein 6 (FBXO6), an E3 ubiquitin ligase known to promote degradation of downstream proteins, was highly expressed after osteogenic differentiation. Herein, the role of FBXO6 in osteoblastic differentiation and bone remodeling was explored. Rat bone marrow mesenchymal stem cells (BMSCs) were first isolated and characterized. Following 14 days of osteogenic induction, FBXO6 was significantly upregulated (approximately 4-fold increase). BMSCs overexpressing FBXO6 demonstra

SAA/FPR2 Signaling Between Pericentral Hepatocytes and Macrophages Exacerbates Zonated Liver Transplant Injury.

Liver transplantation (LT) grafts exhibit selective pericentral hepatic ischemia-reperfusion injury (HIRI), but the zonal mechanism is unclear. To address this, we integrated single-cell and spatial transcriptomics with chromatin accessibility profiling, proteomics, and bulk RNA-seq, then validated key findings in clinical LT specimens, mouse HIRI models, and in vitro assays. Across datasets, HIRI disproportionately aggravated pericentral hepatocyte injury and markedly coincided with heightened macrophage crosstalk. Mechanistically, we identified FOXO1-dominated SAA secretion from pericentral

Molecular insights into DDX3X-androgen receptor mRNA regulation via non-canonical G-quadruplex in castration-resistant prostate cancer.

Prostate cancer (PC) is one of the most common malignancies in men, and the emergence of androgen receptor-low/negative castration-resistant PC (ARL/- CRPC) following androgen receptor signaling inhibitor (ARSI) therapy remains a critical clinical challenge. The RNA-binding protein DEAD-box helicase 3 X-linked (DDX3X) has been implicated in the translational regulation of androgen receptor (AR) mRNA; however, the underlying binding mechanisms are not well defined. Here, we show that DDX3X colocalizes with AR mRNA in ARL/- CRPC cells and selectively recognizes non-canonical RNA G-quadruplex (rG

Age-Associated YBX1 Phosphorylation Regulates the Keratinocyte Senescence-Associated Secretory Phenotype Through Translational Control.

Skin aging is characterized by epidermal atrophy, reduced keratinocyte proliferation, and the accumulation of senescent cells that sustain a chronic, low-grade inflammatory secretome known as the senescence-associated secretory phenotype (SASP). We previously showed that Y-box binding protein 1 (YBX1) limits keratinocyte senescence in human epidermis by acting as a translational repressor of SASP chemokines, including CXCL1 and IL8. How this brake is regulated during skin aging, however, remains undefined. Here we demonstrate that, although total YBX1 protein is reduced in keratinocytes from a

Case Reviews for Two Families With Unique Variants in TBX22 Causing Abruzzo-Erickson Syndrome.

The purpose of this study is to explore the phenotypic spectrum observed in individuals and between families with confirmed variants in the T-Box Transcription Factor 22 gene (TBX22). Pathogenic variants in TBX22 have been identified in individuals with classic X-linked cleft palate (CPX) and also in Abruzzo-Erickson Syndrome (ABERS). We compare the phenotypic features of a newly suspected family with ABERS to those of the original family with ABERS to help determine if family-specific pathogenic variants in TBX22 are the cause of ABERS. Furthermore, we discuss possible mechanisms of action of
