Anal fissure nanocarrier of lercanidipine for enhanced transdermal delivery: formulation optimization, ex vivo and in vivo assessment.

OBJECTIVE: Pathogenesis of anal fissure (AF) is associated with raised resting anal pressures (RAP) involving contraction of smooth muscle. Therefore, the drug delivery strategy should be customized to reduce this raised RAP. In this investigation, in order to achieve this task, a transdermal nanoemulsion (NE) gel of lercanidipine (LER) was developed and optimized to evaluate its permeation ability and in vivo performance. Further, the same formulation was explored for droplet size analysis, zeta potential measurement and stability studies. METHODS: Pseudo-ternary phase diagram was constructe

Longitudinal changes in hand dominance and manual dexterity in normal aging and mild cognitive impairment (MCI): a kinematic study.

INTRODUCTION: Decrements on hand dominance in older populations, including neurodegenerative conditions, have been reported cross-sectionally. At present, no longitudinal research on this issue is available. METHODS: To address this research gap, we conducted a detailed kinematic analysis of unimanual movements of the dominant and non-dominant hands in 16 patients with mild cognitive impairment (MCI) (M age = 69.3 yrs) and 20 healthy controls (HC) (M age = 69.9 yrs) at two points in time with an averaged time interval of 4.5 years between evaluations (3.5 yrs. for HC; 5.35 yrs. for MCI). Functional mapping of the somatotopic organization of the supplementary motor area using navigated repetitive transcranial magnetic stimulation and computer vision-based analysis.

BACKGROUND: The supplementary motor area (SMA) is a cortical region involved in motor and language functions. Motor representations within the SMA follow a somatotopic organization: Anterior regions are linked to orofacial movements, middle regions to upper limb movements, and posterior regions to lower limb movements. SMA lesions may produce impairments that correspond to this somatotopy; therefore, preoperative assessment may aid diagnosis. OBJECTIVE: This study aimed to revise and extend a protocol for assessing the SMA using navigated repetitive transcranial magnetic stimulation (nrTMS),

Oscillatory Neural Signatures of Visual Perception Across Developmental Stages in Individuals With 22q11.2 Deletion Syndrome.

BACKGROUND: Numerous behavioral studies have highlighted the contribution of visual perceptual deficits to the nonverbal cognitive profile of individuals with 22q11.2 deletion syndrome. However, the neurobiological processes underlying these widespread behavioral alterations are yet to be fully understood. Thus, in this paper, we investigated the role of neural oscillations toward visuoperceptual deficits to elucidate the neurobiology of sensory impairments in deletion carriers. METHODS: We acquired 125 high-density electroencephalography recordings during a visual grating task in a group of

A computer-aided method based on geometrical texture features for a precocious detection of fetal Hydrocephalus in ultrasound images.

BACKGROUD: Hydrocephalus is the most common anomaly of the fetal head characterized by an excessive accumulation of fluid in the brain processing. The diagnostic process of fetal heads using traditional evaluation techniques are generally time consuming and error prone. Usually, fetal head size is computed using an ultrasound (US) image around 20-22 weeks, which is the gestational age (GA). Biometrical measurements are extracted and compared with ground truth charts to identify normal or abnormal growth. METHODS: In this paper, an attempt has been made to enhance the Hydrocephalus characteriz

Altered prefrontal cortex activity during working memory task in Bipolar Disorder: A functional Magnetic Resonance Imaging study in euthymic bipolar I and II patients.

BACKGROUND: Working memory (WM) deficits are among the most frequently impaired cognitive domains in patients with Bipolar Disorder (BD), being considered promising cognitive endophenotype of the disorder. However, the related neurobiological correlates still deserve further investigation. The present study was aimed to explore whether dorsolateral prefrontal cortex (DLPFC) activity during WM processing was abnormal in euthymic bipolar patients and may represent a potential trait-related phenotype associated with the disorder. METHODS: Using 3 Tesla functional Magnetic Resonance Imaging (3T f

High-rate x-ray spectroscopy in mammography with a CdTe detector: a digital pulse processing approach.

PURPOSE: Direct measurement of mammographic x-ray spectra under clinical conditions is a difficult task due to the high fluence rate of the x-ray beams as well as the limits in the development of high resolution detection systems in a high counting rate environment. In this work we present a detection system, based on a CdTe detector and an innovative digital pulse processing (DPP) system, for high-rate x-ray spectroscopy in mammography. METHODS: The DPP system performs a digital pile-up inspection and a digital pulse height analysis of the detector signals, digitized through a 14-bit, 100 MH

Precisely targeted brain stimulation for social anxiety: Proactive cognitive control and attentional biases as pathways to symptom reduction.

BACKGROUND: In individuals with high social anxiety (HSAs), threat-related attentional bias is robust. However, traditional attentional bias modification shows small and unstable effects, suggesting that this bias may be more of a symptom than a cause. Additionally, existing transcranial direct current stimulation (tDCS) trials have primarily focused on symptoms or single behavioral outcomes without clarifying the cognitive processes being modulated. OBJECTIVE: To examine whether high-definition tDCS (HD-tDCS) targeting the left dorsolateral prefrontal cortex (DLPFC) enhances proactive cognit

Effects of variable resistance training with different load ratios on lower-limb strength and power in male college students.

OBJECTIVE: The optimum relationship combination of variable to constant resistance within variable resistance training (VRT) for enhancing lower-limb strength and power remains unclear. This study aimed to investigate how different ratios of variable to constant resistance in VRT affect lower-limb strength and power. METHODS: Forty-five male college students (age: 21.65 ± 2.20 years; height: 179.65 ± 4.98 cm; body mass: 77.21 ± 6.77 kg; resistance training experience: 3.00 ± 0.96 years) were randomly assigned to three groups: 20 % VRT (n = 15), 35 % VRT (n = 15), and constant resistance train

Functional modulation of contralateral bias in early and object-selective areas after stroke of the occipital ventral cortices.

Object agnosia is a rare symptom, occurring mainly after bilateral damage of the ventral visual cortex. Most patients suffering from unilateral ventral lesions are clinically non-agnosic. Here, we studied the effect of unilateral occipito-temporal lesions on object categorization and its underlying neural correlates in visual areas. Thirteen non-agnosic stroke patients and twelve control subjects performed an event-related rapid object categorization task in the fMRI scanner where images were presented either to the left or to the right of a fixed point. Eight patients had intact central visua

Neural correlates of response inhibition in pediatric bipolar disorder.

OBJECTIVES: Pediatric bipolar disorder is characterized by core deficits in mood and executive function and commonly co-occurs with attention-deficit/hyperactivity disorder (ADHD). We aimed to examine response inhibition in this population, as an element of executive function, which, if aberrant, may interfere with learning and information processing. METHODS: Children (9-18 years) with bipolar I or II disorder (BD, n = 26) and age, gender, and intelligence quotient (IQ) comparable healthy children (HC, n = 22) without any psychopathology were given a standardized Go/NoGo computerized task me

The role of dorsomedial striatum and parvalbumin interneurons in schizophrenia-related cognitive deficits: insights from disrupted strategic decision-making in Akt1 heterozygous mice.

Schizophrenia, a debilitating disorder with genetic and neurobiological underpinnings, often manifests cognitive deficits, including impaired decision-making. Utilizing Akt1 heterozygous mutant (HET) mice as a model, which mimic schizophrenia due to AKT1's implication as a susceptibility gene, we investigated the involvement of Akt1 and its neural mechanisms influencing strategic decision-making to identify potential therapeutic targets for schizophrenia-associated cognitive impairments. In six experiments, we first revealed that lesions targeting the dorsomedial striatum (DMS) significantly i

Notch3-Dependent Effects on Adult Neurogenesis and Hippocampus-Dependent Learning in a Modified Transgenic Model of CADASIL.

We and others have reported that Notch3 is a regulator of adult hippocampal neurogenesis. Cerebral Autosomal Dominant Arteriopathy with Subcortical Infarcts and Leukoencephalopathy (CADASIL), the most common genetic form of vascular dementia, is caused by mutations in Notch3. The present study intended to investigate whether there is a correlation between altered adult hippocampal neurogenesis and spatial memory performance in CADASIL transgenic mice. To overcome visual disabilities that hampered behavioral testing of the original mice (on an FVB background) we back-crossed the existing TgN3 R

Matching strategies in cognitive research with individuals with high-functioning autism: current practices, instrument biases, and recommendations.

A meta-analysis was performed on the 133 cognitive and behavioral papers in autism using comparison groups in the 1999-2002 period. High-functioning (average IQ: 84.7), adolescents (average, 14.4 years) are largely dominant. IQ is the most frequent matching variable in use (51.2%). The instruments that are most frequently used to determine IQ or general level are Wechsler scales (46.9%), British Picture Vocabulary Scale (BPVS; 22.3%), and Raven Progressive Matrices (RPM; 16.9%). In order to determine if these instruments were equivalent when applied to individuals with pervasive developmental

Neuromechanical adaptations to EMG-guided SSC training in elite badminton players: a predictive multivariate approach.

BACKGROUND: The stretch-shortening cycle (SSC) is essential for explosive lower-limb actions in court-based sports like badminton. Traditional jump assessments may miss subtle neuromechanical changes. Recent developments in real-time electromyography (EMG) and multivariate analysis-such as synergy-based models-enable more precise, individualized diagnostics in sport-specific contexts. OBJECTIVES: This study examined the neuromechanical effects of a 4-week EMG-guided SSC training program in elite badminton players and developed predictive models to identify early training responders. METHODS:
