Pigment epithelium-derived factor 34-mer peptide prevents liver fibrosis and hepatic stellate cell activation through down-regulation of the PDGF receptor.

Pigment epithelium-derived factor (PEDF) has been shown previously to prevent liver fibrosis and hepatic stellate cell (HSC) activation. By investigating the functional domains in PEDF, we identified a 34-mer peptide (residues Asp44-Asn77) that harbors the same function as the full-length PEDF protein. Not only did the 34-mer suppress the development of fibrosis in carbon tetrachloride (CCl4)-treated mouse liver but it also upregulated peroxisome proliferator-activated receptor-gamma (PPARγ) expression in HSCs in vivo. Platelet-derived growth factor (PDGF) plays a crucial role on the process o

Cornelia de Lange Syndrome: NIPBL haploinsufficiency downregulates canonical Wnt pathway in zebrafish embryos and patients fibroblasts.

Cornelia de Lange Syndrome is a severe genetic disorder characterized by malformations affecting multiple systems, with a common feature of severe mental retardation. Genetic variants within four genes (NIPBL (Nipped-B-like), SMC1A, SMC3, and HDAC8) are believed to be responsible for the majority of cases; all these genes encode proteins that are part of the 'cohesin complex'. Cohesins exhibit two temporally separated major roles in cells: one controlling the cell cycle and the other involved in regulating the gene expression. The present study focuses on the role of the zebrafish nipblb paral

NG2-Glia in Central Nervous System Following Injury: From Pathological Involvement to Repair Potential.

BACKGROUND: Central nervous system (CNS) injury remains a severe neurological condition with limited regenerative capacity. NG2-glia are abundant and functionally versatile glial cells that have attracted increasing attention in CNS repair research. However, their roles are complex and context-dependent, ranging from early pathological involvement to later reparative potential, and a systematic synthesis of this temporal dichotomy is lacking. METHOD: We systematically searched PubMed and Web of Science from inception to 2026 using combinations of keywords including "NG2-glia," "NG2 cells," "o

Longitudinal Changes of Circulating miRNAs During Bisphosphonate and Teriparatide Treatment in an Animal Model of Postmenopausal Osteoporosis.

MicroRNAs regulate bone homeostasis, and circulating microRNAs have been proposed as novel bone biomarkers. The effect of anti-osteoporotic treatment on circulating microRNAs has not been described in detail. Therefore, we performed a comprehensive analysis of microRNA serum levels in ovariectomized (OVX) and sham-operated (SHAM) rats over 12 weeks of antiresorptive or osteoanabolic treatment. Forty-two Sprague Dawley rats underwent SHAM surgery (n = 10) or ovariectomy (n = 32). After 8 weeks, OVX rats were randomized to antiresorptive treatment with zoledronate (n = 11), osteoanabolic treatme

Guben Qingyuan Herbal Medicine delays enzalutamide resistance in prostate cancer by alleviating tumor hypoxia and reprogramming energy metabolism in cancer stem cells.

ETHNOPHARMACOLOGICAL RELEVANCE: Enzalutamide resistance is a major clinical bottleneck limiting the therapeutic efficacy of castration-resistant prostate cancer (CRPC). Enzalutamide treatment can exacerbate tumor hypoxia, which is a critical contributor to the development of its resistance. The traditional Chinese formula Guben Qingyuan Herbal Medicine (GBQY), coupled with enzalutamide, has demonstrated potential in delaying enzalutamide resistance. Nevertheless, its interaction with the tumor microenvironment (TME) remains insufficiently investigated. AIM OF THE STUDY: This study aims to exp

Genetic Influences and Targeted Treatments in Osteoporosis: A Systematic Review.

Osteoporosis is a chronic skeletal disorder marked by reduced bone mineral density (BMD) and increased fracture risk, posing a substantial global health burden. Traditionally considered multifactorial, growing evidence highlights a significant genetic contribution across both early-onset monogenic and adult-onset polygenic forms. Understanding the molecular and genetic architecture of osteoporosis is crucial for guiding targeted diagnostics and developing personalised therapeutic strategies. This review aimed to: (1) identify and summarise genetic mutations and polymorphisms associated with os

Immunohistochemical Assessment of Maspin, β-Catenin, and MMP-14 in Oral Potentially Malignant Lesions and Oral Squamous Cell Carcinoma: A Retrospective Observational Study.

Background and Objectives: Oral cancer remains a critical global health burden. Oral potentially malignant disorders (OMPDs) such as leukoplakia and oral lichen planus can precede oral squamous cell carcinoma (OSCC). Inflammation, tissue remodeling, and dysregulated signaling pathways are central to malignant transformation. This observational study aimed to evaluate the expression patterns of Maspin, β-catenin, and MMP-14 by immunohistochemistry (IHC) in oral leukoplakia, oral lichen planus, OSCC, and normal mucosa, exploring associations with lesion type, with no prognostic inferences drawn

Factors associated with posttraumatic growth: gratitude, PTSD and distress; one year into the COVID-19 pandemic in Lebanon.

DNA-PK is an enzyme that is required for proper DNA-repair and is thought to confer radio-resistance in cancer cells. As a consequence, it is a high-profile validated target for new pharmaceutical development. However, no FDA-approved DNA-PK inhibitors have emerged, despite many years of drug discovery and lead optimization. This is largely because existing DNA-PK inhibitors suffer from poor pharmacokinetics. They are not well absorbed and/or are unstable, with a short plasma half-life. Here, we identified the first FDA-approved DNA-PK inhibitor by "chemical proteomics". In an effort to unders

Vibration and β-hydroxy-β-methylbutyrate treatment suppresses intramuscular fat infiltration and adipogenic differentiation in sarcopenic mice.

BACKGROUND: Sarcopenia is an aging-induced deterioration of skeletal muscle mass and function. Low-magnitude high-frequency vibration (LMHFV) was shown to improve muscle functions and β-hydroxy-β-methylbutyrate (HMB) to increase muscle mass and strength. Muscle-derived stem cells (MDSCs) are progenitor cells important for muscle regeneration. We hypothesized that LMHFV and HMB could retard sarcopenia by reducing fat infiltration through inhibiting adipogenesis in MDSCs. METHODS: Senescence-accelerated mouse P8 male mice were randomized into control (CTL), HMB, LMHFV (VIB), and combined (COM)

BBOX1-AS1 promotes gastric cardia adenocarcinoma progression via interaction with CtBP2 to facilitate the epithelial-mesenchymal transition process.

It is recognized that lncRNA BBOX1-AS1 exerts a crucial oncogenic property in several cancer types. However, the functions and underlying mechanisms of BBOX1-AS1 in the epithelial-mesenchymal transition (EMT) process of gastric cardia adenocarcinoma (GCA) have remained unclarified. The findings of this study demonstrated that GCA tissues had elevated BBOX1-AS1 expression levels, which was associated with a worse prognosis in GCA patients. BBOX1-AS1 dramatically enhanced cell proliferation, invasion, and TGF-β1-induced the EMT process in vitro. Further mechanism analysis revealed that BBOX1-AS1

Integrated genomic analysis reveals mutated ELF3 as a potential gallbladder cancer vaccine candidate.

Gallbladder cancer (GBC) is an aggressive gastrointestinal malignancy with no approved targeted therapy. Here, we analyze exomes (n = 160), transcriptomes (n = 115), and low pass whole genomes (n = 146) from 167 gallbladder cancers (GBCs) from patients in Korea, India and Chile. In addition, we also sequence samples from 39 GBC high-risk patients and detect evidence of early cancer-related genomic lesions. Among the several significantly mutated genes not previously linked to GBC are ETS domain genes ELF3 and EHF, CTNNB1, APC, NSD1, KAT8, STK11 and NFE2L2. A majority of ELF3 alterations are fr

An Organoid-Based Model of Cortical Development Identifies Non-Cell-Autonomous Defects in Wnt Signaling Contributing to Miller-Dieker Syndrome.

Miller-Dieker syndrome (MDS) is caused by a heterozygous deletion of chromosome 17p13.3 involving the genes LIS1 and YWHAE (coding for 14.3.3ε) and leads to malformations during cortical development. Here, we used patient-specific forebrain-type organoids to investigate pathological changes associated with MDS. Patient-derived organoids are significantly reduced in size, a change accompanied by a switch from symmetric to asymmetric cell division of ventricular zone radial glia cells (vRGCs). Alterations in microtubule network organization in vRGCs and a disruption of cortical niche architectur

The transcription factor SOX6 contributes to the developmental origins of obesity by promoting adipogenesis.

An association between impaired fetal growth and the postnatal development of obesity has been established. Here, by comparing adipocytes differentiated from mesenchymal stem cells (MSCs) taken from the umbilical cord and derived from normal and growth-restricted neonates, we identified the transcription factor SOX6 as highly expressed only in growth-restricted individuals. We found that SOX6 regulates adipogenesis in vertebrate species by activating adipogenic regulators including PPARγ, C/EBPα and MEST. We further show that SOX6 interacts with β-catenin in adipocytes, suggesting an inhibitio

Loss of Axin2 results in impaired heart valve maturation and subsequent myxomatous valve disease.

AIMS: Myxomatous valve disease (MVD) is the most common aetiology of primary mitral regurgitation. Recent studies suggest that defects in heart valve development can lead to heart valve disease in adults. Wnt/β-catenin signalling is active during heart valve development and has been reported in human MVD. The consequences of increased Wnt/β-catenin signalling due to Axin2 deficiency in postnatal valve remodelling and pathogenesis of MVD were determined. METHODS AND RESULTS: To investigate the role of Wnt/β-catenin signalling, we analysed heart valves from mice deficient in Axin2 (KO), a negat

Defective postnatal endochondral bone development by chondrocyte-specific targeted expression of parathyroid hormone type 2 receptor.

The human parathyroid hormone type 2 receptor (PTH2R) is activated by PTH and by tuberoinfundibular peptide of 39 residues (TIP39), the latter likely acting as its natural ligand. Although the receptor is expressed at highest levels in the nervous system, we have observed that both PTH2R and TIP39 are expressed in the newborn mouse growth plate, with the receptor localizing in the resting zone and the ligand TIP39 localizing exclusively in prehypertrophic and hypertrophic chondrocytes. To address the role of PTH2R in postnatal skeletal growth and development, Col2a1-hPTH2R (PTH2R-Tg) transgeni

A Novel CAF-Related Signature for Precise Prediction of Clinical Outcomes and Immunotherapy Response for Breast Cancer Patients: Based on Multiomics Analyses and Experimental Validation.

OBJECTIVE: Breast cancer (BC) remains a leading cause of cancer-related mortality worldwide. Cancer-associated fibroblasts (CAFs) is a central stromal component of the tumor microenvironment (TME), critically influence BC progression and therapeutic resistance. However, the association between CAF heterogeneity and patient prognosis or response to immunotherapy remains poorly characterized. Here, we aimed to develop a CAF-associated gene signature by integrating single-cell RNA sequencing (scRNA-seq) and bulk RNA-seq data to predict clinical outcomes and immunotherapeutic response in BC. Fine mapping of PTSD GWAS reveals a role for amygdala Foxp2 in regulation of fear and threat responses.

Post-traumatic Stress Disorder (PTSD) is a debilitating psychiatric condition caused by severe trauma exposure and characterized by ongoing dysregulation of fear processing, hyperarousal, and amygdala activation, but with limited effective treatments. Recent large-scale genome-wide association studies (GWAS) of PTSD have identified the transcription factor FOXP2 as a highly-significant, top putative risk gene. Both fine-mapping of the PTSD GWAS with amygdala-specific expression quantitative trait loci (eQTL) data, and transcriptome-wide association analyses, show that altered expression of FOX

Maternal Endocrine and Vascular Perturbations Reprogram the Transcriptomic and Secretory Profile of Perinatal Stem Cells: A Comparative Study of Umbilical Cord-Derived Stem Cell UC-MSCs and Human Milk-Derived Stem Cell MM-MSCs.

Perinatal mesenchymal stem cells (MSCs) are characterized by high phenotypic plasticity, allowing them to dynamically adapt to microenvironmental stimuli. However, the extent to which maternal pathophysiology and clinical history in vivo permanently alter their biological properties remains poorly understood. The aim of this study is to determine the influence of the maternal in vivo environment-including vascular stress (gestational hypertension), metabolic stress (hypothyroidism), and obstetric history (miscarriages, stillbirths)-on the transcriptomic, proteomic, and epigenetic profiles of g
