Cell membrane-camouflaged double emulsion-based SNEDDS for targeted melanoma therapy using Inula viscosa.

Metastatic melanoma is an aggressive skin cancer with limited treatment options. Inula viscosa (IV), a medicinal plant rich in anticancer compounds such as chlorogenic acid shows therapeutic potential; however, low water solubility and rapid clearance of chlorogenic acid limit its clinical application. This study developed double emulsions-based self-nanoemulsifying drug delivery systems (SNEDDS) loaded with Inula viscosa extract (IVE-G), further coated with membranes from SK-MEL28 metastatic melanoma cells to enhance solubility, stability, and tumor-specific targeting. The optimized double em

Posterior capsular Opacification: Pathogenesis, challenges, and innovative therapeutic strategies.

Posterior capsular opacification (PCO), a clouding of the lens capsule, can occur as a major complication after cataract surgery. The standard cataract surgical procedure involves the removal of the lens nucleus and cortex, while retaining a portion of the anterior capsule and the complete posterior capsule. Lens epithelial cells (LECs) are difficult to eliminate entirely, and the residual LECs are prone to proliferate and migrate, resulting in light scattering within the visual axis. Despite advancements in surgical interventions and intraocular lens (IOL) designs, PCO remains a widespread is

Isolation and Characterization of Compounds from Glycyrrhiza uralensis as Therapeutic Agents for the Muscle Disorders.

Skeletal muscle is the most abundant tissue and constitutes about 40% of total body mass. Herein, we report that crude water extract (CWE) of G. uralensis enhanced myoblast proliferation and differentiation. Pretreatment of mice with the CWE of G. uralensis prior to cardiotoxin-induced muscle injury was found to enhance muscle regeneration by inducing myogenic gene expression and downregulating myostatin expression. Furthermore, this extract reduced nitrotyrosine protein levels and atrophy-related gene expression. Of the five different fractions of the CWE of G. uralensis obtained, the ethyl a

Comparison of immunoregulatory effects of polysaccharides from three natural herbs and cellular uptake in dendritic cells.

Polysaccharides from different types of natural herbs have not been compared with each other to determine their differential potencies on innate immune response, such as maturation of dendritic cells (DC). In addition, the role of endocytosis of polysaccharides in DC maturation has not been explored previously. Polysaccharides isolated from Astragalus membranaceus (APS), Ganoderma lucidum (GLP) and Radix ophiopogonis (OGP) were characterized and applied in bone marrow derived DC. Compared to immature DC, three polysaccharides with immunoactivities showed elongated dendrites, decreased phagocyt

Impaired nucleotide excision repair upon macrophage differentiation is corrected by E1 ubiquitin-activating enzyme.

Global nucleotide excision repair is greatly attenuated in terminally differentiated mammalian cells. We observed this phenomenon in human neurons and in macrophages, noting that the transcription-coupled repair pathway remains functional and that there is no significant reduction in levels of excision repair enzymes. We have discovered that ubiquitin-activating enzyme E1 complements the repair deficiency in macrophage extracts, and although there is no reduction in the concentration of E1 upon differentiation, our results indicate a reduction in phosphorylation of E1. In preliminary studies,

In vitro steroid secretion by staged spermatocysts (Sertoli/germ cell units) of dogfish (Squalus acanthias) testis.

Using the dogfish shark (Squalus acanthias) testis model in which different germ cell stages are topographically separated, we previously observed that steroidogenic enzyme activities vary qualitatively and quantitatively during spermatogenesis. To determine whether these data, obtained by radiolabeled tracer analysis of testicular microsomes, accurately predict steroid secretion by intact cultured spermatocysts (germ cell/Sertoli cell units), steroids were radioimmunoassayed directly in spent medium. Steroid output in basal medium was low, but after addition of 25-hydroxycholesterol (25-OH ch
