MicroRNAs as Regulators of Reproductive Function: From Gametogenesis to Pregnancy.

The regulation of transcriptional activity constitutes a primary layer of control over cellular function across diverse tissues and cell types. This regulatory transcriptional landscape is largely governed by epigenetic mechanisms, including DNA and histone modifications, the dynamic activity of epigenetic "writers" and "erasers," and the influence of small regulatory molecules such as microRNAs (miRNAs). miRNAs are small non-coding RNA molecules that play critical roles in transcriptional and post-transcriptional gene regulation. By modulating the expression of transcription factors, kinases,

MEOX2: a homeobox transcription factor with multifaceted biological functions and broad disease associations.

The mesenchymal homeobox transcription factor MEOX2 is recognized for its roles in development and homeostasis, but the full landscape of its complex and often contradictory functions in human diseases, along with its potential unifying regulatory logic, remains to be systematically elucidated. This Review synthesizes current evidence to provide a comprehensive overview of MEOX2's widespread roles in both neoplastic and non-neoplastic diseases. MEOX2 is remarkably context-dependent: it acts as an oncogene in glioblastoma and lung cancer, but as a tumour suppressor in breast cancer, hepatocellu

Integrative multi-omics profiling and machine learning reveal enhancer RNA signatures for early detection and prognostic stratification in breast cancer.

Current breast cancer biomarkers rely predominantly on protein-coding transcriptomes, leaving the regulatory information encoded by enhancer RNAs (eRNAs) largely unexploited. Here, we profiled eRNA expression across 1073 The Cancer Genome Atlas Breast Cancer (TCGA-BRCA) samples integrated with The Cancer eRNA Atlas annotations, using GTEx healthy breast tissue (n = 179) as a curated normal reference. For prognostic stratification, least absolute shrinkage and selection operator (LASSO)-Cox regression identified a 10-eRNA signature that stratified patients into high- and low-risk groups in both

XIST expression and hypermethylation of the X chromosome in males with systemic lupus erythematosus.

INTRODUCTION: Systemic Lupus Erythematosus (SLE) exhibits a pronounced sex bias, affecting females approximately nine times more frequently than males; however, males tend to experience a more severe clinical course yet the molecular basis for these differences remains unclear. METHODS: Leveraging epigenomic, transcriptomic, and proteomic data from the whole blood of 720 SLE patients (679 females, 41males) and 84 healthy controls (77 females, 7 males), we conducted comprehensive multi-omic analyses to identify sex-specific molecular features of this disease. Specifically, differential express

Comparative insights into miRNA-mediated repair across nervous system injuries: Implications for histology and regenerative medicine.

Neural regeneration in the adult mammalian central nervous system (CNS) is severely restricted by both intrinsic neuronal limitations and an inhibitory post-injury microenvironment. In contrast, the peripheral nervous system (PNS) retains a greater capacity for spontaneous repair. MicroRNAs (miRNAs), a class of small non-coding RNAs, have emerged as key epigenetic regulators of neural regeneration by coordinately modulating multiple genes and signaling pathways involved in axonal growth, neuroinflammation, glial responses, and tissue remodeling. This review summarizes current advances in miRNA

The DANCR/miR-145-5p/CD133 Axis Drives Osteosarcoma Stemness and Progression: Implications for Tumor Biology and Therapeutic Innovation.

Characterized by its highly aggressive behavior and propensity for metastasis, osteosarcoma remains a formidable clinical challenge with restricted treatment modalities. Cancer stem-like cells (CSCs) are widely recognized as central orchestrators of oncogenic progression and therapeutic intractability; however, the precise epigenetic regulations governing these processes are yet to be fully elucidated. Here, we investigated the role of the long non-coding RNA DANCR in regulating osteosarcoma stemness. DANCR expression was significantly upregulated in human osteosarcoma tissues and positively c

Lumbar Spinal Endoscopy: Mature, Durable, and Established-Not Investigational.

BACKGROUND: Lumbar spinal endoscopy has long been mischaracterized by payers as investigational. Such claims conflict with decades of refinement, randomized trials, and policy precedent. METHODS: We reviewed the historical evolution, prospective trials, meta-analyses, and policy milestones of transforaminal endoscopic lumbar discectomy (TELD), with emphasis on coding, reimbursement, and comparative effectiveness. RESULTS: Evidence demonstrates that TELD, performed with direct visualization, provides durable outcomes with complication, reoperation, and conversion rates equal or superior to op

Emerging roles of PIWI-interacting RNAs in cancer molecular diagnostics and therapeutics: a molecular biosciences perspective.

Cancer claims nearly 10 million lives yearly, demanding innovative diagnostics and therapies beyond surgery and chemotherapy's limitations, such as resistance and toxicity. Next-generation sequencing has unveiled PIWI-interacting RNAs (piRNAs), 26-31 nucleotide small non-coding RNAs, as pivotal regulators in cancer pathogenesis, offering fresh biomarkers and targets from a molecular biosciences lens. Once deemed germline-exclusive for transposon silencing via PIWI proteins (PIWIL1-4), piRNAs exhibit somatic dysregulation across malignancies, driving hallmarks like proliferation, metastasis, an

Oscillatory shear stress-induced endothelial extracellular vesicles exacerbate aortic valve calcification.

AIMS: Calcific aortic valve disease (CAVD) lacks effective pharmacotherapies. Although small extracellular vesicles (sEVs) are established mediators of cellular communication, how they translate oscillatory shear stress (OSS) into pro-calcific signals through endothelial-interstitial crosstalk remains unknown. This study aimed to delineate a complete mechanosensitive pathway by which sEVs drive aortic valve calcification (AVC). METHODS AND RESULTS: Using circRNA microarray sequencing, we identified circILRUN as the most markedly upregulated circRNA in sEVs from OSS-stimulated human valvular e

AI in the Prediction of Hepatic Fibrosis Progression Using Non-Coding RNAs.

Hepatic fibrosis is a dynamic and progressive condition that can lead to cirrhosis and hepatocellular carcinoma (HCC) if left untreated. Appropriate assessment of the disease progression of fibrosis is critical for early intervention and individualized treatment regimens. Traditional biopsy techniques are invasive and prone to sampling errors, highlighting the need for less invasive predictive techniques. Non-coding RNAs (ncRNAs), including microRNAs (miRNAs), long ncRNAs (lncRNAs), and circular RNAs (circRNAs), have emerged as key regulators of hepatic fibrogenesis and as a possible biomarker

Molecular characterization of Cdh12-SCON conditional knockout mice reveals unexpected splicing changes.

Functional validation of candidate genes in congenital anomalies of the kidneys and urinary tract (CAKUT) and other disorders is essential for translating genetic discoveries into clinical applications. Conditional knockout mouse models are indispensable for studying gene function in complex organ systems. The Short Conditional intrON (SCON) system accelerates the generation of such models by inserting the artificial SCON into a coding exon. SCON is designed to be spliced out after transcription, without affecting gene expression. Upon Cre activity, SCON is converted into the ΔSCON allele whic

Biomolecular condensates as dynamic regulators of musculoskeletal homeostasis, disease, and therapeutic challenges.

Biomolecular condensates are membraneless assemblies that concentrate proteins, nucleic acids, and other biomolecules into dynamic cellular compartments. Liquid-liquid phase separation (LLPS) is one important route by which such condensates form, particularly when multivalent interactions generate liquid-like, reversible assemblies. However, not every condensate or disease-associated assembly should be explained solely by LLPS. In the musculoskeletal system, condensates have been linked to transcription, signal transduction, RNA metabolism, stress responses, tissue development, homeostasis, an

FNIP1 variants are associated with favourable metabolism in 1 million humans.

Altered energy metabolism is a shared driver across cardiometabolic diseases-the leading cause of death globally1. Energy metabolism varies between individuals and is partly heritable2-9. Here, to investigate the genetic basis of energy metabolism, we perform an exome-sequencing analysis of 1,032,116 people from America, Europe and Asia, and estimate associations between rare protein-coding variants and the ratio of triglyceride to high-density-lipoprotein cholesterol (TG:HDL)-an energy-state biomarker that we associate with diverse cardiometabolic risk factors and diseases. We identify 59 ind

Exosomal miRNA in cerebrospinal fluid as biomarkers for neurodegenerative disease.

Cerebrospinal fluid protein biomarkers, such as the Aβ42/Aβ40 ratio, phosphorylated tau, and neurofilament light chain, have significantly advanced the diagnostic process for Alzheimer's disease. Nonetheless, these biomarkers face challenges in effectively distinguishing Alzheimer's disease from frontotemporal dementia or Parkinson's disease from dementia with Lewy bodies. This limitation arises from overlapping protein profiles and the variability inherent in immunoassay techniques. A complementary class of analytes is exosomal microRNAs in cerebrospinal fluid, where these non-coding RNAs are

Irisin in airway remodeling in COPD: Regulatory mechanisms from epithelial barrier to smooth muscle.

This review synthesizes the emerging evidence positioning irisin, a myokine released during physical activity, as a critical molecular link in chronic obstructive pulmonary disease (COPD) airway remodeling. Clinically, irisin deficiency is consistently observed in COPD and correlates with key features including reduced physical activity, respiratory muscle weakness, sarcopenia, emphysema severity, and exacerbation risk, supporting a hypothesis of a "muscle-lung crosstalk" axis. At the cellular level, irisin exerts direct protective effects on airway structural cells by preserving epithelial ba

Blood-based circular RNAs for early diagnosis of Alzheimer's disease.

Detection of Alzheimer's disease (AD) before the development of clinical symptoms is critical for enabling the use of new treatments. Circular RNAs (circRNAs) are highly stable non-coding RNAs enriched in the brain that can cross the blood-brain barrier. Here, analyzing blood data from 1,221 individuals with AD and healthy individuals, we identified 34 circRNAs associated with AD status. A predictive model including these 34 circRNAs was comparable to plasma phosphorylated Tau-217 (pTau217) in classifying AD based on biomarker-confirmed (amyloid-β and Tau) status and replicated in independent

Cardiovascular toxicity induced by immune checkpoint inhibitors: mechanisms linking immune dysregulation and myocardial fibrosis.

Immune checkpoint inhibitors (ICIs) play a key role in cancer immunotherapy, enhancing outcomes for patients with multiple malignancies by blocking the programmed cell death protein 1 (PD-1)/programmed death-ligand 1 (PD-L1) and cytotoxic T-lymphocyte-associated protein 4 (CTLA-4) pathways. Nevertheless, their application is limited due to immune-related adverse events (irAEs), which comprise various manifestations, among which cardiovascular toxicities - primarily myocarditis, arrhythmias, pericarditis, and heart failure - are a major clinical challenge. While relatively rare, myocarditis has
