Effective Motor Skill Learning Induces Inverted-U Load-Dependent Activation in Contralateral Pre-Motor and Supplementary Motor Area.

Motor learning involves complex interactions between the cognitive and sensorimotor systems, which are susceptible to different levels of task load. While the mechanism underlying load-dependent regulations in cognitive functions has been extensively investigated, their influence on downstream execution in motor skill learning remains less understood. The current study extends the understanding of whether and how learning alters the load-dependent activation pattern by a longitudinal functional near-infrared spectroscopy (fNIRS) study in which 30 healthy participants (15 females) engaged in ex

Thalamocortical structural connectivity with sleep oscillatory coupling in insomnia disorder.

BACKGROUND: Thalamocortical interactions play a critical role in sleep oscillatory activities. However, in individuals with insomnia disorder (ID), the structural organization of these circuits, their relationship with sleep oscillatory coupling, and their potential modulation by repetitive transcranial magnetic stimulation (rTMS) remain unclear. METHODS: In Study 1, we recruited 62 ID patients and 62 healthy controls and assessed white matter tract strength between thalamic subregions and cortical areas using probabilistic tractography. In Study 2, we administered 20 sessions of 1 Hz rTMS (a

Electroclinical features of lateral and medial orbitofrontal epilepsy: a case series.

To better understand the electroclinical features and epileptic network of lateral and medial orbitofrontal epilepsy (OFE). We evaluated four patients who had undergone epilepsy surgery. Epileptic foci in two patients originated from the lateral orbitofrontal cortex, and those in the other two originated from the medial orbitofrontal cortex, which was confirmed by stereoelectroencephalography (SEEG). Time-frequency spectrograms were also provided for assistance, and the change in high-frequency energy was superimposed on the 3D reconstructed brain with a colour code in order to more intuitivel

Brain Structural Correlates of Metacognition in First-Episode Psychosis.

Metacognition is impaired in schizophrenia and is an important predictor of functional outcome, but the underlying neuropathology is not clear. Studies have implicated frontal regions and there is also some evidence that the hippocampus might play a pivotal role, but findings are inconsistent. We set out to more comprehensively investigate the neural underpinnings of insight in first-episode psychosis (FEP) using 2 metacognitive measures (the Beck Cognitive Insight Scale [BCIS]) and a perceptual metacognitive accuracy task alongside structural magnetic resonance imaging (MRI). We measured cort

Acute kick-boxing exercise alters effective connectivity in the brain of females with methamphetamine dependencies.

OBJECTIVE: Methamphetamine (METH) dependence, especially among women, is a serious global health problem. Kick-boxing exercise can be used to reduce cue-induced craving and develop a healthy lifestyle for female with METH dependencies. This study aimed to assess acute kick-boxing related changes in effective connectivity (EC) in the brain of females with METH dependencies by using functional near-infrared spectroscopy (fNIRS) signals. METHODS: The fNIRS signals were continuously recorded from the left and right prefrontal cortices (LPFC/RPFC) and left and right motor cortices (LMC/RMC) of 30

Perceptual load in decision making: The role of anterior insula and visual areas. An ERP study.

The present study aimed at describing the effects of perceptual load on neurocognitive processes of decision-making. To this aim, we used a visual-motor discriminative task in which pairs of stimuli were assigned to either target or non-target categories. For each category, stimulus configuration was defined as simple or complex according to orientation and arrangement of the constituent segments. Analyses of prefrontal ERPs revealed that the pP1 component (at 180 ms) was larger for complex stimuli than simple for both categories, and the same result was found for the pP2 component (at 320 ms)

Structural and cognitive correlates of fatigue in progressive multiple sclerosis.

BACKGROUND: Fatigue in multiple sclerosis (MS) is a debilitating symptom and experienced by most patients. In recent studies investigating this phenomenon, the majority of patients had a relapsing-remitting disease course. METHODS: Patients with progressive MS participating in one of three treatment trials during a period from 2010 to 2014 were included. Fatigue was assessed with the Fatigue Scale for Motor and Cognitive Functions (FSMC) and patients were further examined with a cognitive test battery, including Symbol Digit Modalities Test (SDMT), and 3 T MRI with subsequent quantitative ana

An investigation of potential neural correlates of intrusive retrieval of distressing memories.

BACKGROUND AND OBJECTIVES: Despite the prevalence of intrusive memories across psychological disorders, little is known about the neural networks that underpin this form of memory. This study used functional magnetic resonance imaging (fMRI) to identify neural circuits associated with the retrieval of intrusive memories. METHODS: Participants with moderate levels of anxiety (N = 30) underwent a cold pressor task to induce a physiological stress response, after which they viewed 10 neutral and 10 negative film clips. In a method designed to induce intrusive memories, participants then complete

Roles of default-mode network and supplementary motor area in human vigilance performance: evidence from real-time fMRI.

We used real-time functional magnetic resonance imaging (fMRI) to determine which regions of the human brain have a role in vigilance as measured by reaction time (RT) to variably timed stimuli. We first identified brain regions where activation before stimulus presentation predicted RT. Slower RT was preceded by greater activation in the default-mode network, including lateral parietal, precuneus, and medial prefrontal cortices; faster RT was preceded by greater activation in the supplementary motor area (SMA). We examined the roles of these brain regions in vigilance by triggering trials bas

Epileptic negative myoclonus: An EEG-single-photon emission CT study indicating involvement of premotor cortex.

We report a combined EEG-single-photon emission CT (SPECT) study on a patient with epileptic negative myoclonus (ENM). Clinically, the ENM was characterized by brief repetitive lapses in postural tone of the right upper extremity when the arms were held outstretched, whereas no movement effect was observed during rest. Ictal EEG showed repetitive left frontal spikes with a maximum at electrodes EC1 and F1. EMG silent periods lasting from 100 to 200 ms followed the onset of the EEG transients by a latency of 20 to 40 ms. The N20 component of median nerve somatosensory evoked potentials-represen

Sumoylation profiles and mitochondrial dynamics in in vivo and in vitro models of Parkinson's disease.

Parkinson's disease (PD) is characterized by progressive dopaminergic neuron loss and motor and non-motor impairments. Mitochondrial dysfunction and altered post-translational modifications, including SUMOylation, have emerged as key mechanisms in PD pathogenesis. Here, we investigated global SUMOylation changes in in vivo and in vitro PD models induced by MPTP and MPP+, respectively. Intranasal MPTP administration in rats induced region- and time-dependent changes in Ubc9 and SENP3 across the olfactory bulb, prefrontal cortex, striatum, and hippocampus. In primary cortical neurons, MPP+ reduc

A prefrontal peptidergic circuit for psychogenic itch.

Psychological stress can evoke itch without a peripheral stimulus, yet the underlying mechanism remains undefined. Here, we demonstrate that a peptidergic circuit within the medial prefrontal cortex (mPFC) mediates this phenomenon. In mice under chronic restraint stress, stress-elicited scratching coincided with elevated gastrin-releasing peptide (GRP) release and heightened activity of GRP receptor (GRPR)-expressing neurons in the mPFC. Microinjection of GRP into the mPFC reproduced stress-induced itch, whereas ablation of GRPR neurons abolished it. Optogenetic activation of GRPR neurons or t

SpaDiff denoises sequence-based spatial transcriptomics via diffusion process.

Spot-swapping in sequence-based spatial transcriptomics redistributes RNA molecules across neighboring spots, distorting spatial gene expression and biasing downstream analysis. We introduce SpaDiff, a mass-conserving denoising framework that corrects this artifact through score-guided reverse diffusion. Across simulated and real spatial transcriptomics datasets, SpaDiff consistently improves the recovery of spatial gene-expression distributions. In particle-level simulations with known ground truth, SpaDiff robustly restores gene-level spatial patterns across diverse contamination regimes. In

Behavioral Decoding Reveals Cortical Endocannabinoid Potentiation during Δ9-THC Impairment.

How Δ9-tetrahydrocannabinol (THC) impairs natural behaviors in mice remains unknown. We developed a video-monitored behavioral platform with machine learning classifiers to unravel discrete changes in natural mouse behaviors. THC infusion into the medial prefrontal cortex (mPFC) disrupted walking kinematic features characteristic of impairment responses. THC predominantly increased mPFC GABAergic activity preceding walk initiation shifting the mPFC excitatory/inhibitory (E/I) balance. Pose-defined closed loop photo-stimulation of mPFC GABAergic neurons demonstrated that THC exacerbates selecte

Gestational diabetes mellitus, not obesity, triggers postpartum brain inflammation and premature aging in Sprague-Dawley rats.

Previous studies showed that women with gestational diabetes mellitus (GDM) are susceptible to cognitive dysfunction. We investigated the effects of GDM on brain pathologies and premature brain aging in rats. Seven-week-old female Sprague-Dawley rats were fed a normal diet (ND) or a high-fat diet (HFD) after two weeks of acclimatization. On pregnancy day 0, HFD-treated rats received streptozotocin (GDM group) or vehicle (Obese mothers). ND-treated rats received vehicle (ND-control mothers). On postpartum day 21, brains and blood were collected. The GDM group showed increased inflammatory and p

Isolation of psychedelic-responsive neurons underlying anxiolytic behavioral states.

Psychedelics hold promise as alternate treatments for neuropsychiatric disorders. However, the neural mechanisms by which they drive adaptive behavioral effects remain unclear. We isolated the specific neurons modulated by a psychedelic to determine their role in driving behavior. Using a light- and calcium-dependent activity integrator, we genetically tagged psychedelic-responsive neurons in the medial prefrontal cortex (mPFC) of mice. Single-nucleus RNA sequencing revealed that the psychedelic drove network-level activation of multiple cell types beyond just those expressing 5-hydroxytryptam

Cognitive impairments in type 1 diabetes mellitus model mice are associated with synaptic protein disorders.

An association between type 1 diabetes mellitus (T1DM) and cognitive impairment was recently reported. However, the mechanisms by which T1DM induces cognitive impairment are still unknown. Here, we confirmed that T1DM mice induced by streptozotocin (STZ) injection had impaired working memory and spatial memory. We observed long-term potentiation (LTP) induction defects and synaptic loss in mice 20 weeks after STZ injection. We also found decreased levels of synaptic proteins, including the N-methyl-D-aspartic acid receptor (NMDAR) subunit NR2A, synaptophysin (SYP), and postsynaptic density 95

What part of the brain is involved in graphic design thinking in landscape architecture?

Graphic design thinking is a key skill for landscape architects, but little is known about the links between the design process and brain activity. Based on Goel's frontal lobe lateralization hypothesis (FLLH), we used functional magnetic resonance imaging (fMRI) to scan the brain activity of 24 designers engaging in four design processes-viewing, copy drawing, preliminary ideas, and refinement-during graphic design thinking. The captured scans produced evidence of dramatic differences between brain activity when copying an existing graphic and when engaging in graphic design thinking. The res
