Antimicrobial resistance (AMR) represents a major global health challenge and motivates the development of rapid analytical approaches for characterizing clinical bacterial isolates. Surface-enhanced Raman spectroscopy (SERS), combined with machine learning (ML), has emerged as a promising approach for label-free spectral classification of bacterial isolates. However, the field lacks methodological consensus on how hierarchical spectral datasets should be structured, standardized, and partitioned for reliable model evaluation. This gap has led to widespread data leakage and inflated performanc

Infected wounds represent a formidable clinical challenge, as persistent bacterial colonization and disrupted bio-signaling collectively hinder effective tissue repair. Conventional passive dressings lack the capacity to actively modulate this complex microenvironment. Here, we report a wireless, dual-responsive hydrogel (PVA/P(VDF-TrFE)/BiOClBr@PDA, PPBP) based on porous poly(vinyl alcohol) (PVA), piezoelectric poly(vinylidene fluoride-trifluoroethylene) (P(VDF-TrFE)), and polydopamine-modified bismuth oxybromide chloride (BiOClBr@PDA). The constructed hydrogel orchestrates piezoelectric stim

BACKGROUND: Polyphenols' interaction with gut microbiota is crucial for their bioactivity and intestinal health benefits, yet this effect varies individually due to differences in microbiota structure. Using fecal samples from healthy individuals (n = 3), ulcerative colitis (UC) patients (n = 3), and Crohn's disease (CD) patients (n = 3), we employed an in vitro digestion and fermentation model combined with 16S rRNA sequencing to compare polyphenol-microbiota interactions among these groups.
RESULTS: Polyphenol intervention consistently increased Bifidobacterium abundance while decreasing Fi

Cross-linking mass spectrometry is a powerful method for structural analysis, but choosing between cleavable and non-cleavable cross-linkers remains challenging. We rigorously compared non-cleavable DSS with cleavable DSSO and found that DSS consistently yields more cross-link identifications from isolated protein complexes to bacterial lysates. The advantage of DSS diminishes as sample complexity increases. At the highest complexity tested-human cell lysate-the trend reverses, with DSSO outperforming DSS. The superior performance of DSS in less complex samples is likely explained by its longe

Microplastics create unique microbial habitats known as plastispheres, functioning as embedded "micro-enclaves" in soil ecosystems. However, their assembly mechanisms and ecological implications remain poorly understood. Here, we sampled microplastics and adjacent soils at 27 urban wetland sites in Changsha, China, to sequence bacterial communities and assess their assembly processes and environmental drivers. Plastispheres selectively recruit soil bacteria, with bacterial richness representing only 51.6-69.3% of that in adjacent soils despite positive correlations with source communities. Imp

BACKGROUND: Antibiotic resistance among multidrug-resistant pathogens necessitates alternative antimicrobial strategies. Photodynamic therapy (PDT) and sonodynamic therapy (SDT), particularly with methylene blue (MB), show promise but remain underexplored in clinical contexts.
METHODS: We conducted an in vitro evaluation of MB-PDT and SDT against Staphylococcus aureus, a major cause of skin and soft tissue infections. Assays included growth curve analysis, scanning electron microscopy (SEM) to assess bacterial ultrastructure. A pilot clinical study in 20 healthy volunteers assessed in vivo an

Antibiotic-resistant bacteria (ARB) and their genes (ARGs) pose a significant risk to public health and the ecosystem. Conventional water disinfection efficiently eliminates pathogens, however, it requires intensive chemical consumption but inefficiently removes ARGs. Herein, we propose an on-demand electrochemical platform that simultaneously eliminate both ARB and ARGs with in-situ generated hydroxyl radicals (·OH) without additional chemical inputs. Single-atom copper catalysts with CuN4 coordination structure (SA-CuN4/NC) are precisely tuned to directly convert oxygen into controllable ·OH

Wound infection remains a significant clinical challenge, exacerbated by the growing prevalence of bacterial resistance due to the overuse of conventional antibiotics. Photodynamic therapy (PDT) offers a promising approach for wound sterilization that circumvents the issue of antibiotic resistance. However, conventional photosensitizers are prone to inactivation and exhibit poor retention at the wound site, limiting their clinical efficacy. To overcome these limitations, we developed a biomimetic mineralization approach to encapsulate the small-molecule photosensitizer chlorin e6 (Ce6) within

Plant-derived antimicrobial proteins represent a promising but underexplored source of new anti-infective molecules. In this study, we purified and identified AtQQS (Qua-Quine Starch, NP_189695), a 59-amino-acid orphan protein previously known as a metabolic regulator in Arabidopsis thaliana, and demonstrated direct broad-spectrum antimicrobial activity against bacterial and fungal pathogens. An antimicrobial polypeptide, AtQQS, was purified by C18 reversed-phase high-performance liquid chromatography (RP-HPLC) from A. thaliana plants and confirmed by SDS-PAGE. Recombinant AtQQS protein inhibi

Ash fertilization is a forest management practice used to improve forest growth in boreal peatland forests. It impacts soil chemistry, vegetation, and soil organisms, yet little is known about its long-term impacts on soil microbial communities. Here, we focus on the long-term impacts of ash fertilization on the soil microbiome of drained boreal peatland forests in Finland. We hypothesized that the ash-induced changes in soil chemistry have led to long-term changes in soil microbial communities. To study this, we analyzed soil bacterial and archaeal communities from seven drained peatland fore

Traditional fermented cereal products are important for food security and nutrition. In Zambia and neighbouring countries, Munkoyo is a traditional beverage produced by spontaneous fermentation of cooked grains inoculated with roots from specific wild plant species ("Munkoyo" plants). These plants host distinct root endosphere microbial communities that also vary across geographic locations. However, how plant species identity, geographic origin, and cereal type shape the microbial composition and functional properties of Munkoyo remains unclear. We conducted fermentation experiments to assess

Nine novel dihydro-α-pyrone derivatives, talaromypyrones A-I (1-9), were isolated from the soil-derived fungus Talaromyces sp. G23. These compounds share a conserved 3'R-configuration within the 4-hydroxy-5,6-dihydro-2-pyrone core but display notable variations in their flanking polyketide chains. Talaromypyrones A-H (1-8) demonstrate potent antibacterial activity against Gram-positive bacteria, including drug-resistant strains, while exhibiting excellent safety profile by showing no cytotoxicity toward normal human LO2 cells. However, they exhibit no activity against Gram-negative bacteria (l

It is imperative to provide timely and effective treatment for bacterially infected wounds to prevent progression to more severe pathological conditions. Herein, we report a near-infrared (NIR)-activated nanoassembly (BC@P) integrating dual functions of bactericidal activity and tissue repair. The nanoassembly is facilely fabricated via the self-assembly of organic photothermal molecules B(TIBC)2, natural antioxidant molecules curcumin, and biocompatible eight-arm polyethylene glycol (8-arm-PEG). B(TIBC)2 is rationally designed with an acceptor-donor-acceptor (A-D-A) structure and tailored pho

OBJECTIVE: Microbiome being a potential biomarker holds the future hope for insight into GD pathogenesis, diagnosis and cure. Many factors such as diet, lifestyle, environment, host genetics shape the diversity and composition of human microbiome. Although recent studies have indicated that gut microbiome dysbiosis was significantly associated with the onset of gestational diabetes mellitus (GDM). Information on the alteration of gut microbiota composition in Pakistani women is limited. Therefore, present study was designed to elucidate gut microbiota taxonomic composition and relative abundan

Host stress is associated with elevated catecholamine neurohormones that influence gut physiology and host-microbe interactions, yet how bacterial systems detect and interpret these signals remains incompletely understood. Enteric pathogens exploit inter-kingdom adrenergic signaling to sense host-derived norepinephrine and epinephrine, but whether such pathways can be rationally rewired to produce predictable, programmable outputs has not been systematically explored. Here, we reconstitute adrenergic signaling in Escherichia coli Nissle 1917 by repurposing the enterohemorrhagic E. coli QseBC t

Pathogen infections, exacerbated by emerging drug resistance, remain among the most challenging health issues, for which multitargeting approaches may offer effective solutions. In this context, medicinal plants, including essential oils, provide complex mixtures of diverse molecules that can exert therapeutic effects, either alone or synergistically with established antibiotics. Although several databases comprehensively collect information on the antibacterial properties of medicinal plants, including chemical composition, bioactivity data, and ethnobotanical uses, there is a notable lack of

As early-spring pioneer ephemeroids in arid and semi-arid regions, Ferula species exert irreplaceable ecological functions through distinctive life-history strategies. However, their impacts on soil microhabitats and soil quality remain unclear. Taking Ferula species (Ferula feruloides, Ferula lehmannii, Ferula krylovii, and Ferula sinkiangensis) as the research subject, this study conducted a comprehensive analysis of the soil abiotic (physicochemical properties) and biological (microbial (bacterial/fungal) community diversity, composition, structure, and co-occurrence networks) factors as we

Dry and wet heat treatments are widely used physical methods for microbial inactivation, and their effectiveness in destroying bacterial spores has been extensively demonstrated. This review systematically discusses the bactericidal effects and underlying mechanisms of dry heat, wet heat, and steam-based treatments on bacterial spores. Wet heat inactivates spores through heat-induced protein denaturation and dipicolinic acid release, resulting in multi-level structural damage, including spore coat collapse, cortical peptidoglycan hydrolysis, and core hydration. On the other hand, dry heat trea

Diabetic osteomyelitis, exacerbated by a hyperglycemic microenvironment, leads to increased bacterial infections and bone tissue destruction, raising the risk of amputation. Current antibiotic therapies are limited in effectiveness due to rising antibiotic resistance and biofilm barrier. In response, we developed a microgel-based hydrogel system that delivers H2Se gas combined with mild photothermal therapy (MPTT) to achieve integrated treatment for infection control, anti-inflammation, and osteogenesis. This system utilizes Fe3O4 nanoparticles as photothermal-responsive carriers to encapsulat

Urbanization reshapes soil ecosystems, yet its impacts on the composition and interactions of soil microbial communities remain insufficiently understood. Here, we use high-throughput sequencing to profile bacteria, fungi, and protists across four representative habitat types of different urbanization level-campuses, roadside greenbelts, suburban unmanaged lands, and concrete pavements in four Chinese cities spanning a broad latitudes (Changchun, Beijing, Hangzhou, Haikou). Vegetated habitats consistently structure microbial communities, whereas concrete pavements form a distinct disturbed hab
