Personalized Medicine in Pediatric Urology: From Diagnosis to Individualized Risk Assessment.

BACKGROUND: Pediatric urology has traditionally relied on anatomical classifications, standardized diagnostic pathways, and disease-specific treatment algorithms. However, children with the same anatomical diagnosis may have substantially different risks of disease progression, renal injury, complications, and need for intervention. Personalized and precision medicine aim to integrate clinical, imaging, functional, biological, genomic, and longitudinal information to support individualized risk assessment and decision-making. METHODS: This narrative review was based on a targeted literature s

SHAP-explained machine-learning model for high-risk gastric cancer identification.

INTRODUCTION: Gastric cancer (GC) remains a major public health concern in Asia. Risk prediction tailored to regional biological features such as Helicobacter pylori (H. pylori) status and high-risk mucosal findings such as atrophic gastritis (AG) and intestinal metaplasia (IM) may help improve the screening workflow. METHODS: Using a large, real-world, nationwide screening cohort with available endoscopic AG/IM codes, we developed 2-year GC risk prediction models that integrate AG/IM with regional demographic and lifestyle factors. We compared a conventional Cox proportional hazards model (C

Single-cell and metabolomic profiling reveals brain cellular and metabolic disruptions in pregnant mice exposed to micro- and nanoplastics.

Micro- and nanoplastics (MNPs) are emerging environmental contaminants that can cross biological barriers, but their direct effects on the developing brain remain poorly defined. We hypothesized that prenatal MNPs exposure impairs neurodevelopment by disrupting cerebral homeostasis. To test this hypothesis, we established a prenatal MNP exposure model in mice and integrated single-cell RNA sequencing (scRNA-seq) with untargeted metabolomics. Our results demonstrate that MNPs can cross both the placental and blood-brain barriers, accumulating in fetal brain tissue, particularly in grey matter.

3D printing in ophthalmology: From basic research to clinical transformation.

3D printing is reshaping ophthalmic biomaterials, tissue models, implants, biosensors, and drug-delivery systems, but its clinical value depends on matching each printing strategy to ocular-specific requirements rather than on printing capability alone. This review classifies ophthalmic 3D printing into extrusion-based, inkjet-based, electric field-assisted, and light-assisted approaches, while clarifying the boundary between acellular material inks and cell-laden bioinks. We summarize how ink formulation, spatial resolution, optical transparency, mechanical stiffness, degradation, sterilizati

New Insights into Acinetobacter baumannii Pathogenesis and Therapeutic Implications.

Acinetobacter baumannii is a leading cause of healthcare-associated infections and is classified among the highest-priority antimicrobial-resistant pathogens. Its clinical success reflects the convergence of antimicrobial resistance (AMR) and biological traits that promote environmental persistence and transmission. Acinetobacter baumannii has undergone a remarkable transformation over the past few decades, evolving from a relatively obscure environmental bacterium into a globally recognized multidrug-resistant pathogen. Its prevalence in healthcare settings, particularly intensive care units,

Biological sex influences immune response to influenza infection in juvenile outbred mice.

Children are highly susceptible to influenza virus infection and lung inflammation. The majority of studies of influenza pneumonia in mice are in adult, inbred, same-sex mice with limited variation in disease outcomes, unlike what is observed in children. In this study, juvenile outbred mice were challenged with influenza A/California/07/2009 H1N1 virus to examine the temporal effect of infection on lung inflammation in genetically dissimilar male and female mice. In this model, influenza infection induced variable disease severity characterized by viral load, airspace leak and inflammation, a

The Gut-Immune-Brain Axis in Aging: Integrating Immunosenescence, Inflammaging, and Neuroinflammation for Precision Medicine.

Background: Population aging is accompanied by progressive immune remodeling, chronic low-grade inflammation, and increased susceptibility to neurodegenerative diseases. Although the microbiota-gut-brain axis is increasingly recognized as a regulator of neuroimmune homeostasis, mechanisms linking age-associated dysbiosis with immunosenescence, barrier dysfunction, and brain aging remain incompletely understood. This review integrates current evidence into the proposed Gut-Immune-Brain Resilience Axis (GIBRA), a systems-level model describing how microbial signaling shapes neuroimmune resilienc

The Dual Role of Natural Killer Cells in the Septic Liver.

Sepsis is a life-threatening organ dysfunction caused by dysregulated host responses to infection. As an important immune organ, the liver plays a vital role in the pathophysiological process of sepsis. When sepsis occurs, the liver exhibits unique immunological characteristics, including abnormal activation of pattern recognition receptors, loss of control of the complement system, adaptive immune regulation, and hepatic Kupffer cell polarization, which together determine the outcome of septic liver. Natural killer (NK) cells are innate immune cells with different biological characteristics i

AI-integrated multi-omics platform to revolutionize anti-metastatic therapy development through circulating tumor cell profiling: SCRUM-MONSTAR-CTC.

BACKGROUND: Metastatic disease remains the leading cause of cancer-related death, yet most precision oncology strategies still emphasize profiling primary tumors and tracking cell-free tumor DNA (ctDNA). Although ctDNA has transformed genomic profiling, molecular residual disease monitoring, and early cancer detection, it cannot directly capture viable tumor cell states, phenotypic plasticity, or functional adaptations that drive metastatic spread. We propose that the next phase of precision oncology should integrate the cellular dimension of metastasis through systematic circulating tumor cel

Sepsis in end-stage liver disease and acute-on-chronic liver failure: pathophysiology, diagnostic challenges, and pharmacological management.

BACKGROUND: Sepsis represents a leading cause of acute decompensation, acute-on-chronic liver failure (ACLF), and short-term mortality in patients with end-stage liver disease (ESLD). Its clinical course is shaped by the coexistence of profound systemic inflammation, cirrhosis-associated immune dysfunction (CAID), and early multiorgan failure, which together complicate diagnosis, antimicrobial management, and supportive care. METHODS: This narrative review synthesizes current evidence on the epidemiology, immunopathophysiology, microbiology, diagnostic strategies, and therapeutic management o

Scenarios of Engineered Membranes in Cultural Heritage: How Do Membranes Act on Microclimates and Microenvironments?

Conservation is one of the critical issues for cultural heritage assets. A lot of materials are daily proposed to clean, repair, and restore monuments, but best practices are still in high demand to prevent artwork deterioration. Membrane science provides practical and sustainable solutions, including buffer environmental zones, to preserve cultural assets from physical, chemical, and biological agents. Protection can be obtained in an outdoor context in order to counteract the effects of climatic changes, variations of temperature, violent storms, bright lighting, and the wear of time. Remova

Antigen-directed p53 restoration in TP53-mutated myeloid neoplasms: a hypothesis and theory perspective.

TP53-mutated myeloid neoplasms, including acute myeloid leukemia (AML) and myelodysplastic syndromes (MDS), remain among the hardest-to-treat hematologic malignancies, with short survival despite hypomethylating agents +/- venetoclax, and allogeneic transplantation. Despite encouraging early-phase data, Phase 3 trials of mutant-p53 reactivators to date have not yet translated into clear, practice-changing survival benefits, highlighting a need to explore complementary strategies that aim to directly restore wild-type p53 function in the leukemic niche. Building on clinical experience with the

Application and characterisation of four genetically engineered animal models (mouse/zebrafish/rhesus monkey/Drosophila) in acute myeloid leukemia.

Acute myeloid leukemia (AML), a highly prevalent hematological malignancy, heavily relies on animal models that can mimic the genetic and biological characteristics of the disease for research advancement. Among various modeling approaches, genetically engineered animal models have emerged as critical tools for deciphering AML pathogenesis and developing novel therapies, leveraging their precise gene targeting, robust experimental reproducibility, and diverse model types. These models not only enable meticulous dissection of gene functions but also highly simulate human disease traits, fulfill

Generative AI for the Design of Molecules: Advances and Challenges.

The design of novel molecules underpins advances in both drug discovery and biomaterials engineering. Traditional approaches, from natural product isolation to high-throughput screening, have delivered important therapeutics but remain costly, inefficient, and limited in exploring the chemical and biomolecular space. While predictive machine learning models improved aspects of discovery, they cannot fully address the complexity of modern precision medicine. Generative artificial intelligence (AI) offers a paradigm shift by enabling de novo molecular creation guided by data-driven optimization.

New insights into the molecular biology of Alzheimer's-like cerebral amyloidosis achieved through multi-omics approaches.

BACKGROUND: One of the neuropathologic hallmarks of Alzheimer's disease (AD) is amyloid plaques composed of fibrillar amyloid beta (Aβ) that accumulate in the hippocampus and cerebral cortex. The identification of molecular changes and interactions associated with Aβ-dependent cerebral amyloidosis is a need in the field. We hypothesize that structured datasets linking proteins to differentially abundant metabolites may provide an indirect but effective means of elucidating the processes and functions in which these metabolites are involved. The goal of this study was to identify core network m

Generation of a Transgenic Plasmodium cynomolgi Parasite Expressing Plasmodium vivax Circumsporozoite Protein for Testing P. vivax CSP-Based Malaria Vaccines in Non-Human Primates.

Background/Objectives: Malaria, caused by infection with Plasmodium parasites, exacts a heavy toll worldwide. There are two licensed vaccines for malaria as well as two monoclonal antibodies that have shown promising efficacy in field trials. The vaccines and monoclonal antibodies target the major surface protein (circumsporozoite protein, CSP) of Plasmodium falciparum. Yet P. falciparum is only one of the four major species of Plasmodium that infect humans. Plasmodium vivax is the second leading cause of malaria, but the P. vivax vaccine and monoclonal development lags far behind that for P.
