Empathy and suicide-related stigma among emerging adults: A structural equation modeling study.

Suicide-related stigma may hinder help-seeking and timely mental health support among emerging adults aged 18-29 years. Although empathy may be protective, evidence on its association with suicide-related attitudes remains limited. This cross-sectional study examined associations between empathy and the three subscales of the Stigma of Suicide Scale-Short Form among 1,003 Peruvian university students (Mage = 20.47, SD = 1.97; 51.4% men). Participants completed the Cognitive and Affective Empathy Test and the Stigma of Suicide Scale-Short Form. Structural equation modeling with WLSMV showed tha

Non-Neutralising Antibodies and T Cell Responses to SARS-CoV-2 Detected in Unvaccinated SARS-CoV-2-Naive Spanish Adolescents.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection in children and adolescents usually follows a mild or asymptomatic course. In this brief report, we investigated the cellular and humoral responses to SARS-CoV-2 in six healthy Spanish female adolescents, aged 14 to 16, using blood samples collected early in 2021, well before COVID-19 vaccinations. Using IFNγ-ELISPOT assays, we detected T cell recall responses in four of these adolescents to a peptide pool encompassing the SARS-CoV-2 Spike protein. The same subjects also exhibited IgM antibodies against the Nucleocapsid and

Targeted blood-brain barrier penetration for traumatic brain injury therapy via RXR/PPAR-driven microglial M2 polarization.

As a leading global cause of death and disability, traumatic brain injury (TBI) triggers a complex secondary injury process leading to long-term neurological impairment. Current therapeutic efficacy is limited by the blood-brain barrier (BBB) and the absence of integrated approaches to address neuroinflammation and neuroregeneration. We designed a biomimetic nanomedicine (Ali-NPs@Mexo) by encapsulating alitretinoin(Ali)-loaded albumin nanospheres within mesenchymal stromal cell (MSC)-derived exosomal membranes. This platform exploits the natural BBB-crossing and inflammation-targeting capabili

Embryonic sheep placenta alleviates muscle-fat wasting symptoms and improves gut microbiome dysbiosis in cachexia mice.

Cachexia is a multifactorial wasting syndrome characterized by muscle-fat depletion, accompanied by significant gut microbiome disturbances. Embryonic sheep placenta (ESP), a functional food known to modulate immunity and gut microbiome, represents a potential intervention substance. By in vitro and in vivo experiments combined with multi-omics analysis, this study provides experimental evidence for the effect of ESP on cachexia. These studies have found that ESP and its extract alleviate cachexia by increasing 91.80% (P < 0.0001) muscle tube diameter, 19.56% (P < 0.01) muscle cross-sectional

BBB-permeable carbon dots ameliorate Alzheimer's-like phenotypes in mice by suppressing oxidative stress, neuroinflammation, and amyloid-β aggregation.

Alzheimer's disease (AD) is a common neurodegenerative disorder wherein reactive oxygen species (ROS) and Amyloid-β-protein (Aβ) play critical roles. Inspired by traditional Chinese charcoal drug and the anti-inflammatory properties of some carbon dots, we developed Radix Isatidis derived carbon dots (RI-CDs) via a hydrothermal method. The RI-CDs can cross the blood-brain barrier (BBB) and were thus evaluated for AD therapy. In vitro, RI-CDs scavenged ROS, inhibited Aβ42 aggregation, protected SH-SY5Y cells, and regulated inflammatory factors. In AD mice, the Morris water maze test and nesting

Observed frequency of increased iron indices and associated factors in welders.

Increased iron indices among welders are underrecognized. This study investigated their frequency and associated factors in an observational cross-sectional study including 55 male welders. Increased iron indices were defined as serum ferritin >400 µg/L and/or TS ≥45%. Standard statistical and regression analyses were used. Increased iron indices were identified in 25.5% of welders. Compared to those with normal indices, affected welders had longer employment duration. Iron parameters were significantly higher in the increased group (serum iron 139.6 vs. 93.3 µg/dL, ferritin 498.9 vs. 142.3 µg

Experimental hepatitis E virus genotype 1 infection in three types of wild rodents.

Hepatitis E virus genotype 1 (HEV-1) has long been considered human-specific, with no known natural animal hosts. Here, we demonstrate that three wild rodent species-Apodemus peninsulae, Clethrionomys rufocanus, and Lasiopodomys brandtii-are susceptible to HEV-1 infection. Among them, A. peninsulae infected with HEV-1 exhibited the highest susceptibility, characterized by robust fecal viral shedding, systemic viral replication, seroconversion, and mild liver pathology mimicking human acute HEV-1 infection. Intrahepatic transcriptomic analysis of infected animals revealed activation of inflamma

Immunogenicity of poxvirus A16/G9 entry-fusion subcomplex and its restriction by A56/K2 protein informs vaccine design.

Poxviruses rely on a conserved multiprotein entry-fusion complex, providing numerous potential antibody targets, although the immunogenicities of only a few have been analysed. Here we tested the ectodomains of ten orthopoxvirus entry-fusion complex proteins in rabbit and mouse immunization experiments and determined that six induce neutralizing antibodies. Focusing on the apical A16/G9 heterodimer, we show that it induces antibodies that prevent vaccinia virus (VACV) entry, cross-neutralize cowpox virus (CPXV) and monkeypox virus (MPXV), and protect mice against lethal challenge infections wi

Mechanisms of Paclitaxel Resistance: Recent Advances and Future Perspectives.

Paclitaxel (PTX) is a core first-line chemotherapeutic agent used to treat a broad spectrum of cancers, which remain leading causes of death worldwide. However, the emergence of intrinsic and acquired resistance critically contributes to chemotherapy failure, thereby promoting tumor recurrence and metastasis and resulting in a poor prognosis. PTX resistance is a complex biological process driven by multiple factors and cross-regulated signaling pathways. It encompasses a wide variety of mechanisms, including aberrations in microtubules and related proteins; changes in mitotic systems and chrom

Resurgence of human metapneumovirus in the post-COVID-19 era: pathogenesis, epidemiological shifts, clinical impact, and future challenges.

Human metapneumovirus (hMPV), a respiratory pathogen identified in 2001, is a substantial cause of community-acquired respiratory infections across all age groups. This Review explores the impact of hMPV after the COVID-19 pandemic, emphasising its resurgence as a public health concern. Epidemiological shifts, as well as unusual seasonal patterns, increased co-infection rates, and altered age distributions, have been observed globally. Phylogenetic analysis has shown the variation across three distinct periods, especially before and after the COVID-19 pandemic, in terms of genotypic distributi

Vancomycin-Resistant E. faecium: Addressing Global and Clinical Challenges.

Antimicrobial resistance (AMR) poses a profound threat to modern healthcare, with vancomycin-resistant Enterococcus faecium (VREfm) emerging as a particularly resilient and clinically significant pathogen. This mini-review examines the biological mechanisms underpinning VREfm resistance, including biofilm formation, stress tolerance, and the acquisition of resistance genes such as vanA and vanB. It also explores the behavioural, social, and healthcare system factors that facilitate VREfm transmission, highlighting disparities in burden across vulnerable populations and low-resource settings. The Longitudinal Relationship Between Co-Rumination and Emotional Problems in Early Chinese Adolescents: The Moderating Role of Self-Compassion.

The longitudinal relationship between co-rumination and emotional problems has been understudied, particularly regarding the role of protective factors in moderating the relationship. This study employed a cross-lagged analysis to examine the bi-directional relationship between co-rumination and emotional problems, and the moderating role of self-compassion in this dynamic. The participants comprised 814 Chinese junior school students (Mage = 13.98 ± 0.83), who were surveyed over two tracking periods spaced four months apart. The results revealed that (1) baseline levels of co-rumination posit

Perceived Social Isolation and Fentanyl Use Among Adults Who Inject Drugs in Rural Kentucky.

Despite evidence linking perceived social isolation (PSI) with substance use disorders (SUDs), limited research has examined its effect on fentanyl use among people who inject drugs (PWID). Cross-sectional data of 350 PWID in Appalachian Kentucky were utilized to examine the association between PSI and lifetime fentanyl use. Study findings highlighted a significant association between PSI and lifetime fentanyl use. Additionally, PWID with higher scores on the PSI scale tend to use fentanyl less frequently, suggesting that social isolation may serve as a protective factor against fentanyl use w

Discovery of non-retinoid compounds that suppress the pathogenic effects of misfolded rhodopsin in a mouse model of retinitis pigmentosa.

Pathogenic mutations that cause rhodopsin misfolding lead to a spectrum of currently untreatable blinding diseases collectively termed retinitis pigmentosa. Small molecules to correct rhodopsin misfolding are therefore urgently needed. In this study, we utilized virtual screening to search for drug-like molecules that bind to the orthosteric site of rod opsin and improve its folding and trafficking. We identified and validated the biological effects of 2 non-retinoid compounds with favorable pharmacological properties that cross the blood-retina barrier. These compounds reversibly bind to unli

In-Situ Cross-Linked Polymers for Enhanced Thermal Cycling Stability in Flexible Perovskite Solar Cells.

Flexible perovskite solar cells (FPSCs) are a promising emerging photovoltaic technology, with certified power conversion efficiencies reaching 24.9 %. However, the frequent occurrence of grain fractures and interface delamination raises concerns about their ability to endure the mechanical stresses caused by temperature fluctuations. In this study, we employ an in situ polymerization molecule with extended functional end groups to preserve mechanical integrity during thermal cycling. The AMPS-DEA molecule chemically anchors to grain boundaries and cross-links neighboring grains, protecting th

APOE4 to APOE2 allelic switching in mice improves Alzheimer's disease-related metabolic signatures, neuropathology and cognition.

Compared to individuals carrying two copies of the ε4 allele of apolipoprotein E (APOE), ε2 homozygotes have an approximate 99% reduction in late-onset Alzheimer's disease (AD) risk. Here we develop a knock-in model that allows for an inducible 'switch' between risk and protective alleles (APOE4s2). Gene expression and proteomic analyses confirm that APOE4s2 mice synthesize E4 at baseline and E2 after tamoxifen administration. A whole-body allelic switch results in a metabolic profile resembling E2/E2 humans and drives AD-relevant alterations in the lipidome and single-cell transcriptome, part

Contribution of the different Neisseria gonorrhoeae lipooligosaccharide structural variants to functional responses elicited by GMMA outer membrane vesicles.

Despite decades of research, an effective vaccine against Neisseria gonorrhoeae remains elusive due to the pathogen's antigenic variability and immune evasion capabilities. Retrospective studies of OMV-based meningococcal vaccines have shown a partial effectiveness against gonorrhea, reigniting hopes for a feasible vaccine. Our study focused on the role of gonococcal lipooligosaccharides (LOS), the most abundant antigens on the surface, in stimulating functional immune responses. By employing detoxified OMV from Neisseria gonorrhoeae isogenic strains expressing different LOS glycoforms, we obs
