Availability and accessibility of SARS-CoV-2 vaccines among rural populations in sub-Saharan Africa: a systematic review and meta-analysis protocol.

INTRODUCTION: The COVID-19 pandemic has posed unprecedented global public health challenges since its emergence in late 2019. Vaccination remains the most effective intervention for preventing severe disease and reducing transmission. However, disparities in vaccine distribution and access have been widely reported, particularly in low-income and middle-income countries and rural communities. Rural populations in sub-Saharan Africa (SSA) often face structural barriers, including weak health systems, limited infrastructure and an inadequate healthcare workforce, which may affect the availabilit

Immunity gaps for several vaccine preventable diseases in Norway.

Childhood immunisation programs (CIP) have led to a substantial reduction in vaccine preventable diseases. However, simply monitoring a decline in disease incidence does not accurately reflect an individual's immunity to each disease. To identify the immune status in the Norwegian population, we measured the IgG levels against antigens of 7 vaccine preventable diseases (diphtheria, tetanus, pertussis (DTP), measles, mumps, rubella and Hemophilus influenzae type b infection) in 3051 residual sera representative of the Norwegian population aged 2-95 years in 2012. We found that 39.8% of women an

First-Line PD-1 Inhibitor-Based Therapy for Advanced or Metastatic Esophageal Squamous Cell Carcinoma: A Revised Systematic Review and Meta-Analysis of Phase III Trials.

BACKGROUND AND PURPOSE: Immune checkpoint blockade has changed the first-line treatment landscape for advanced esophageal squamous cell carcinoma (ESCC), but the magnitude and consistency of benefit across phase III trials require careful synthesis. METHODS: We conducted a PRISMA 2020-guided systematic review of PubMed, Embase, CENTRAL, and Web of Science (January 1, 2015 to April 30, 2025) for phase III randomized trials comparing first-line PD-1 inhibitor-based therapy with chemotherapy in advanced/metastatic ESCC. The primary endpoint was overall survival (OS). Secondary endpoints were saf

Microbial-derived immunostimulatory small molecule augments anti-PD-1 therapy in lung cancer.

We previously showed that enrichment of the Bacteroides genus is associated with improved anti-PD-1-mediated tumor therapy. Here, we isolate 183 Bacteroides isolates from the feces of humanized anti-PD-1 responder mice. Supernatants from 6 of 183 isolates stimulate IFNγ production from primary CD8+ T cells. These six isolates (6-consort) enhance anti-PD-1-induced anti-tumor efficacy in syngeneic and orthotopic lung cancer models compared to non-responder feces-colonized mice, an effect dependent on the production of IFNγ. Bioassay-guided fractionation and comparative metabolomics lead to the d

Preclinical models of immune checkpoint inhibitors-related interstitial pneumonia for anti-PD1 tumor immunotherapy.

Immune-related adverse reactions (irAEs) are common adverse reactions after immune checkpoint inhibitor treatment, impacting the universality and continued use of immunotherapy. Currently, preclinical models to investigate the mechanisms underlying these adverse effects are inadequate. This study aims to develop both in vitro and in vivo models of irAEs to advance basic research on these adverse reactions. For vitro models, we designed two co-culture systems: "Lung epithelial cells-PBMC" conditional co-culture model and "organoid-PBMCs" co-culture model. These involve culturing spheroids, pati

Long-term overall survival with dual CTLA-4 and PD-L1 or PD-1 blockade and biomarker-based subgroup analyses in patients with advanced non-small-cell lung cancer: a systematic review and reconstructed individual patient data meta-analysis.

BACKGROUND: Immune checkpoint inhibitors targeting PD-L1 or PD-1 as monotherapy or combined with CTLA-4 inhibitors or chemotherapy (or both) are the standard of care for patients with advanced non-small-cell lung cancer (NSCLC). However, it remains unclear which patients benefit from the addition of CTLA-4 inhibitors. We aimed to evaluate whether dual checkpoint blockade with CTLA-4 and PD-L1 or PD-1 inhibitors provides similar efficacy to PD-L1 or PD-1 inhibitor monotherapy, or whether these strategies produce distinct outcomes across NSCLC subpopulations. METHODS: We conducted a search of P

Nonredundant Immune Checkpoints Direct Therapeutic Resistance to Chemoimmunotherapy in Pancreatic Ductal Adenocarcinoma.

Pancreatic ductal adenocarcinoma (PDA) is characterized by a myeloid-enriched microenvironment and has shown remarkable resistance to immune checkpoint blockade (e.g., anti-PD-1 and anti-CTLA-4). In this study, we sought to define the role of myeloid immunosuppression in immune resistance in PDA. We report that although depletion of CSF1R+ myeloid cells in combination with anti-PD-1 and chemotherapy triggers T-cell infiltration into PDA, it also causes compensatory remodeling of the myeloid compartment with limited tumor control. Combination therapy against multiple myeloid targets, including

Preclinical models of melanoma leptomeningeal disease to assess intrathecal checkpoint blockade.

Leptomeningeal disease (LMD) is a subtype of central nervous system metastatic disease that is associated with poor patient outcomes and limited treatment options. There is an unmet need to develop preclinical models of LMD to expedite and improve the development of new therapeutics. Here, we describe the development of multiple orthotopic immunocompetent murine models of melanoma LMD, including their use to assess the efficacy of systemic and/or intrathecal immunotherapy. LMD was established by direct intrathecal injection of murine cell lines (B16-F10, BP, D4M, D4M-UV2, MC38-gp100, RMS, YUMM

Reconsidering palliative radiotherapy in addition to PD-1 blockade for non-small cell lung cancer: results from the FORCE phase II trial (AIO/YMO-TRK-0415).

Introduction of immune checkpoint inhibitors (ICI) has improved overall survival (OS) for advanced non-small cell lung cancer (NSCLC). However, responses differ greatly amongst patients. Additional radiotherapy (RT) may promote tumor-specific immunity and synergize with ICI to improve tumor control. The multicenter phase II FORCE trial evaluated safety and efficacy of nivolumab with additional palliative radiotherapy (5 × 4 Gy) as clinically indicated in pre-treated metastatic non-squamous NSCLC (group A, n = 41), including pretreated patients without an indication for radiotherapy in a parall

Piperlongumine enhances the antitumor efficacy of PD-1 inhibitors by inducing immunogenic cell death in prostate cancer cells.

BACKGROUND: In advanced prostate cancers, the immunologically "cold" tumor microenvironment inhibiting lymphocyte infiltration has been a significant barrier to the successful application of immune checkpoint inhibitor-based therapy. In other cancer types, immunogenic cell death inducers have been shown to restore sensitivity to checkpoint inhibitor-based therapy by promoting lymphocyte infiltration and activating naïve cytotoxic T lymphocytes through cross-presentation of neoantigens by dendritic cells. Here, we describe the ability of natural product and reactive oxygen species-inducer piper

Effects of PD-1 inhibitors on ovarian function-based on two-sample mendelian randomization and visualization experimental validation.

Although immune checkpoint inhibitors (ICIs) have transformed cancer treatment by improving survival, their ovarian safety remains uncertain. This study combined Mendelian randomization (MR) and experimental validation to assess the impact of PD-1 inhibitors on ovarian function. MR analysis used summary statistics from large European-ancestry genome-wide association studies (GWAS), applying the inverse-variance weighted (IVW) method, supported by sensitivity analyses. For in vitro experiments, mouse follicles were cultured with 10 µg/ml PD-1 inhibitor ch15mt (clinically relevant concentration)

Integrin β4 drives immune evasion and therapeutic resistance to PD-1 blockade in bladder cancer via MEK/ERK signaling.

Bladder cancer (BLCA) is one of the most prevalent malignancies of the urinary system and accounts for 90 to 95% of urothelial carcinomas. Muscle-invasive BLCA (MIBC) is a malignant type of BLCA, with a 5-years survival rate of only 50%. While immune checkpoint inhibitors (ICIs) have shown promise in advanced MIBC, their responsiveness is limited by the immune evasion mechanisms in tumor cells. Integrin β4 (ITGB4), a member of the intercellular adhesion molecule family, is reported to be significantly upregulated in many cancers and implicated in tumor progression. Our previous work establishe

Impact of immunotherapy on outcomes of cutaneous melanoma and concurrent brain metastasis: a surveillance, epidemiology, and end results analysis of 2010-2020.

INTRODUCTION: Melanoma has the greatest incidence of brain metastases among all cancers and once patients develop melanoma brain metastases (MBM) it is the primary contributor to patient mortality. Following the FDA approval of immune checkpoint inhibitors in 2011, there has been a significant reduction in melanoma patient mortality. Therefore, we investigated the impact of immunotherapy on overall survival (OS) pre and post implementation of immunotherapy in the management of MBM. METHODS: Data from patients with MBM upon diagnosis and seen between 2010-2012 and 2018-2020 were collected from

Assessing immunization coverage in Southern togo: implications for perennial malaria chemoprevention delivery.

BACKGROUND: Malaria remains a major health challenge in sub-Saharan Africa, especially for children under five. The World Health Organization (WHO) recommends perennial malaria chemoprevention (PMC) to children in areas of medium to high perennial transmission. In Togo, since 2022, a pilot project has leveraged the Expanded Program on Immunization (EPI) to deliver PMC to children under two years; however, the extent to which PMC achieves its desired outcome may depend on EPI performance. This study assessed EPI coverage and factors associated with zero-dose status in southern Togo. METHODS: W

Microbial dysbiosis with tryptophan metabolites alteration in lower respiratory tract is associated with clinical responses to anti-PD-1 immunotherapy in advanced non-small cell lung cancer.

Lower respiratory tract microbiome constitutes a unique immune microenvironment for advanced non-small cell lung cancer as one of dominant localized microbial components. However, there exists little knowledge on the associations between this regional microbiome and clinical responses to anti-PD-1 immunotherapy from clinical perspectives. Here, we equivalently collected bronchoalveolar lavage fluids from 56 advanced NSCLC participants treated with none (untreated, n = 28) or anti-PD-1 immunotherapy (treated, n = 28), which was further divided into responder (n = 17) and non-responder (n = 11)

Modelling the impact of a quadrivalent ACWY meningococcal vaccination and vaccination targeting serogroup B in France.

Invasive meningococcal disease (IMD) represents a significant global health burden, with high fatality rate and substantial risk of lifelong sequelae. Most cases worldwide are caused by six serogroups (A, B, C, W, X, and Y). Effective vaccines are available to prevent disease caused by serogroups A, B, C, W, and Y. In France, approximately 500 IMD cases occur each year. Since the introduction of mandatory vaccination against serogroup C, the majority of IMD now involve serogroups B, W, and Y. Given these changes in the epidemiology of IMD, French National Authority for Health decided to recons

Differences in checkpoint-inhibitor-induced hypophysitis: mono- versus combination therapy induced hypophysitis.

OBJECTIVE: Immune checkpoint inhibitors (ICIs) are revolutionary in oncology but may cause immune-related (IR) side effects, such as hypophysitis. Treatment with anti-PD-(L)1, anti-CTLA-4 or anti-CLTA-4/PD-1 may induce hypophysitis, but little is known about the differences in clinical presentation or need for different treatment. We analyzed the differences of anti-PD-(L)1, anti-CTLA-4 and anti-CTLA-4/PD-1 induced hypophysitis. METHODS: retrospective analysis of 67 patients (27 anti-PD-(L)1, 6 anti-CLTA-4 and 34 anti-CTLA-4/PD-1 induced hypophysitis). RESULTS: The median time between starti

Risk factors and outcomes of pericardial effusion in cancer patients receiving PD-1 inhibitors.

BACKGROUND: Programmed cell death 1 (PD-1) inhibitors can induce various adverse reactions associated with immunity, of which cardiotoxicity is a serious complication. Limited research exists on the link between PD-1 inhibitor use and pericardial effusion (PE) occurrence and outcomes. METHODS: We conducted a retrospective study at the First Affiliated Hospital of Xi'an Jiaotong University from 2017 to 2019, comparing cancer patients who developed PE within 2 years after PD-1 inhibitor therapy to those who did not. Our primary outcome was the all-cause mortality rate at one year. We applied th
