Relationship between impulsivity, prefrontal anticipatory activation, and striatal dopamine release during rewarded task performance.

Impulsivity, and in particular the negative urgency aspect of this trait, is associated with poor inhibitory control when experiencing negative emotion. Individual differences in aspects of impulsivity have been correlated with striatal dopamine D2/D3 receptor availability and function. This multi-modal pilot study used both positron emission tomography (PET) and functional magnetic resonance imaging (fMRI) to evaluate dopaminergic and neural activity, respectively, using modified versions of the monetary incentive delay task. Twelve healthy female subjects underwent both scans and completed t

Default mode network connectivity and reciprocal social behavior in 22q11.2 deletion syndrome.

22q11.2 deletion syndrome (22q11DS) is a genetic mutation associated with disorders of cortical connectivity and social dysfunction. However, little is known about the functional connectivity (FC) of the resting brain in 22q11DS and its relationship with social behavior. A seed-based analysis of resting-state functional magnetic resonance imaging data was used to investigate FC associated with the posterior cingulate cortex (PCC), in (26) youth with 22qDS and (51) demographically matched controls. Subsequently, the relationship between PCC connectivity and Social Responsiveness Scale (SRS) sco

Neural correlates of disbalanced motor control in major depression.

BACKGROUND: Motor retardation is a common symptom of major depressive disorder (MDD). Despite the existence of various assessment methods, little is known on the pathobiology of motor retardation. We aimed to elucidate aspects of motor control investigating the association of objective motor activity and resting state cerebral blood flow (CBF). METHODS: Nineteen control subjects and 20 MDD patients were investigated using arterial spin labeling (ASL) at 3T in the morning to quantify resting state CBF. Afterwards wrist actigraphy was recorded for 24h. CBF, group and activity level (AL) were en

Prior auditory information shapes visual category-selectivity in ventral occipito-temporal cortex.

Objects in our natural environment generate signals in multiple sensory modalities. This fMRI study investigated the influence of prior task-irrelevant auditory information on visually-evoked category-selective activations in the ventral occipito-temporal cortex. Subjects categorized pictures as landmarks or animal faces, while ignoring the preceding congruent or incongruent sound. Behaviorally, subjects responded slower to incongruent than congruent stimuli. At the neural level, the lateral and medial prefrontal cortices showed increased activations for incongruent relative to congruent stimu

Alterations in cortical activity of male methamphetamine abusers performing an empathy task: fMRI study.

OBJECTIVES: We investigate possible differences in neural correlates of empathy processing between abstinent methamphetamine (MA) abusers and healthy subjects using functional magnetic resonance imaging (fMRI). METHODS: Nineteen abstinent MA abusers (mean age of 36.06 years, range 31-52 years) and 19 healthy subjects (mean age of 37.05 years, range 33-42 years) participated in this study. A visual fMRI activation paradigm was used, comprising a series of cartoons, each depicting a short story. There were two categories of stories: empathy (Empathy) and Physical causality (Physical). fMRI imag

Deficient activity in the neural systems that mediate self-regulatory control in bulimia nervosa.

CONTEXT: Disturbances in neural systems that mediate voluntary self-regulatory processes may contribute to bulimia nervosa (BN) by releasing feeding behaviors from regulatory control. OBJECTIVE: To study the functional activity in neural circuits that subserve self-regulatory control in women with BN. DESIGN: We compared functional magnetic resonance imaging blood oxygenation level-dependent responses in patients with BN with healthy controls during performance of the Simon Spatial Incompatibility task. SETTING: University research institute. PARTICIPANTS: Forty women: 20 patients with BN

Meta-analysis of the effects of repetitive transcranial magnetic stimulation (rTMS) on negative and positive symptoms in schizophrenia.

BACKGROUND: A growing body of evidence suggests that repetitive transcranial magnetic stimulation (rTMS) can alleviate negative and positive symptoms of refractory schizophrenia. However, trials to date have been small and results are mixed. METHODS: We performed meta-analyses of all prospective studies of the therapeutic application of rTMS in refractory schizophrenia assessing the effects of high-frequency rTMS to the left dorsolateral prefrontal cortex (DLPFC) to treat negative symptoms, and low-frequency rTMS to the left temporo-parietal cortex (TPC) to treat auditory hallucinations (AH)

Coding region paraoxonase polymorphisms dictate accentuated neuronal reactions in chronic, sub-threshold pesticide exposure.

Organophosphate pesticides (OPs), known inhibitors of acetylcholinesterase (AChE), are used extensively throughout the world. Recent studies have focused on the ACHE/PON1 locus as a determinant of inherited susceptibility to environmental OP exposure. To explore the relationship of the corresponding gene-environment interactions with brain activity, we integrated neurophysiologic, neuropsychological, biochemical, and genetic methods. Importantly, we found that subthreshold OP exposure leads to discernible physiological consequences that are significantly influenced by inherited factors. Cortic

Sleep-dependent motor memory plasticity in the human brain.

Growing evidence indicates a role for sleep in off-line memory processing, specifically in post-training consolidation. In humans, sleep has been shown to trigger overnight learning on a motor-sequence memory task, while equivalent waking periods produce no such improvement. But while the behavioral characteristics of sleep-dependent motor learning become increasingly well characterized, the underlying neural basis remains unknown. Here we present functional magnetic resonance imaging data demonstrating a change in the representation of a motor memory after a night of sleep. Subjects trained o

Abnormal object recall and anterior cingulate overactivation correlate with formal thought disorder in schizophrenia.

BACKGROUND: The neural basis of formal thought disorder (FTD) is unknown. An influential theory is that FTD results from impaired semantic memory processing. We explored the neural correlates of semantic memory retrieval in schizophrenia using an imaging task assessing semantic object recall. METHOD: Sixteen healthy control subjects and sixteen schizophrenia patients whose FTD symptoms were measured with the Thought Disorder Index completed a verbal object-recall task during functional magnetic resonance imaging. Participants viewed two words describing object features that either evoked (obj

Sumoylation profiles and mitochondrial dynamics in in vivo and in vitro models of Parkinson's disease.

Parkinson's disease (PD) is characterized by progressive dopaminergic neuron loss and motor and non-motor impairments. Mitochondrial dysfunction and altered post-translational modifications, including SUMOylation, have emerged as key mechanisms in PD pathogenesis. Here, we investigated global SUMOylation changes in in vivo and in vitro PD models induced by MPTP and MPP+, respectively. Intranasal MPTP administration in rats induced region- and time-dependent changes in Ubc9 and SENP3 across the olfactory bulb, prefrontal cortex, striatum, and hippocampus. In primary cortical neurons, MPP+ reduc

A prefrontal peptidergic circuit for psychogenic itch.

Psychological stress can evoke itch without a peripheral stimulus, yet the underlying mechanism remains undefined. Here, we demonstrate that a peptidergic circuit within the medial prefrontal cortex (mPFC) mediates this phenomenon. In mice under chronic restraint stress, stress-elicited scratching coincided with elevated gastrin-releasing peptide (GRP) release and heightened activity of GRP receptor (GRPR)-expressing neurons in the mPFC. Microinjection of GRP into the mPFC reproduced stress-induced itch, whereas ablation of GRPR neurons abolished it. Optogenetic activation of GRPR neurons or t

SpaDiff denoises sequence-based spatial transcriptomics via diffusion process.

Spot-swapping in sequence-based spatial transcriptomics redistributes RNA molecules across neighboring spots, distorting spatial gene expression and biasing downstream analysis. We introduce SpaDiff, a mass-conserving denoising framework that corrects this artifact through score-guided reverse diffusion. Across simulated and real spatial transcriptomics datasets, SpaDiff consistently improves the recovery of spatial gene-expression distributions. In particle-level simulations with known ground truth, SpaDiff robustly restores gene-level spatial patterns across diverse contamination regimes. In

Behavioral Decoding Reveals Cortical Endocannabinoid Potentiation during Δ9-THC Impairment.

How Δ9-tetrahydrocannabinol (THC) impairs natural behaviors in mice remains unknown. We developed a video-monitored behavioral platform with machine learning classifiers to unravel discrete changes in natural mouse behaviors. THC infusion into the medial prefrontal cortex (mPFC) disrupted walking kinematic features characteristic of impairment responses. THC predominantly increased mPFC GABAergic activity preceding walk initiation shifting the mPFC excitatory/inhibitory (E/I) balance. Pose-defined closed loop photo-stimulation of mPFC GABAergic neurons demonstrated that THC exacerbates selecte

Gestational diabetes mellitus, not obesity, triggers postpartum brain inflammation and premature aging in Sprague-Dawley rats.

Previous studies showed that women with gestational diabetes mellitus (GDM) are susceptible to cognitive dysfunction. We investigated the effects of GDM on brain pathologies and premature brain aging in rats. Seven-week-old female Sprague-Dawley rats were fed a normal diet (ND) or a high-fat diet (HFD) after two weeks of acclimatization. On pregnancy day 0, HFD-treated rats received streptozotocin (GDM group) or vehicle (Obese mothers). ND-treated rats received vehicle (ND-control mothers). On postpartum day 21, brains and blood were collected. The GDM group showed increased inflammatory and p

Isolation of psychedelic-responsive neurons underlying anxiolytic behavioral states.

Psychedelics hold promise as alternate treatments for neuropsychiatric disorders. However, the neural mechanisms by which they drive adaptive behavioral effects remain unclear. We isolated the specific neurons modulated by a psychedelic to determine their role in driving behavior. Using a light- and calcium-dependent activity integrator, we genetically tagged psychedelic-responsive neurons in the medial prefrontal cortex (mPFC) of mice. Single-nucleus RNA sequencing revealed that the psychedelic drove network-level activation of multiple cell types beyond just those expressing 5-hydroxytryptam

Cognitive impairments in type 1 diabetes mellitus model mice are associated with synaptic protein disorders.

An association between type 1 diabetes mellitus (T1DM) and cognitive impairment was recently reported. However, the mechanisms by which T1DM induces cognitive impairment are still unknown. Here, we confirmed that T1DM mice induced by streptozotocin (STZ) injection had impaired working memory and spatial memory. We observed long-term potentiation (LTP) induction defects and synaptic loss in mice 20 weeks after STZ injection. We also found decreased levels of synaptic proteins, including the N-methyl-D-aspartic acid receptor (NMDAR) subunit NR2A, synaptophysin (SYP), and postsynaptic density 95

What part of the brain is involved in graphic design thinking in landscape architecture?

Graphic design thinking is a key skill for landscape architects, but little is known about the links between the design process and brain activity. Based on Goel's frontal lobe lateralization hypothesis (FLLH), we used functional magnetic resonance imaging (fMRI) to scan the brain activity of 24 designers engaging in four design processes-viewing, copy drawing, preliminary ideas, and refinement-during graphic design thinking. The captured scans produced evidence of dramatic differences between brain activity when copying an existing graphic and when engaging in graphic design thinking. The res
