Brain structure abnormalities in neurodevelopmental synaptopathies.

Advanced brain imaging studies have been scarcely reported in neurodevelopmental encephalopathies. In this study we assess structural brain alterations in three rare neurogenetic disorders primarily affecting glutamatergic neurotransmission, aiming to identify shared and disease-specific neuroanatomical patterns and their clinical correlations. A cohort of patients with SYNGAP1 (n = 19), GRIN gene family (n = 19), and STXBP1 (n = 10) mutations underwent magnetic resonance imaging. Advanced segmentation tools were used to extract regional brain volumes. Volumetric differences between patient an

Neurodevelopmental Outcome in Isolated Vermian Hypoplasia: A Single-Center Retrospective Cohort Study and Literature Review.

Vermian hypoplasia (VH) is characterized by reduced cerebellar vermis volume without significant hemispheric involvement. To date, data on long-term neurodevelopmental outcomes in isolated VH cases are limited, complicating prenatal counselling. This is single-center retrospective cohort study analyzing fetuses with posterior fossa abnormalities (PFA) diagnosed between 2010 and 2021. VH cases were confirmed through ultrasonography and fetal MRI, excluding non-isolated anomalies or ischemic/hemorrhagic etiologies. We collected data on Neurodevelopmental outcomes through chart review and structu

Small for gestational age subgroups have differential morbidity, growth, and neurodevelopment at age 2: the INTERBIO-21st Newborn Study.

BACKGROUND: Small for gestational age is a complex perinatal syndrome associated with increased neonatal morbidity, mortality, and impaired childhood growth and neurodevelopment. Current classifications rely primarily on birthweight, which does not capture the heterogeneity of the condition nor predict long-term health outcomes. Here we aim to identify and characterize distinct small for gestational age subgroups and assess their neonatal and early childhood health trajectories. OBJECTIVE: To refine the classification of small for gestational age by identifying subgroups based on maternal, fe

Beyond the Extra X and Y Chromosome: The Contribution of Familial Risk for Psychopathology to the Neurodevelopmental Phenotype of Children With Sex Chromosome Trisomy.

Individuals with an extra X or Y chromosome (sex chromosome trisomy or SCT) have an increased risk for symptoms of psychopathology and neurocognitive dysfunction. In this study, we evaluated the contribution of family history (FH) of neuropsychiatric or neurocognitive disorders to the phenotype of SCT. One hundred and six children with SCT and 102 nonclinical controls, all aged 1-7 years, and their primary caregiver (parent) participated. Rates of neuropsychiatric and neurocognitive disorders were collected for all first-degree family members of the children. Neurocognitive tests and parental

Using melatonin to induce sleep as an alternative to general anaesthesia in auditory brainstem response testing.

OBJECTIVES: Auditory Brainstem Response (ABR) is an objective, non-invasive hearing test requiring the subject to remain still for accurate results. This poses challenges in children with neurodevelopmental disabilities. Traditionally, general anaesthesia (GA) was employed, but it carries risks, high costs, and long waiting times. Melatonin, with minimal side effects and no need for monitoring, offers a potential alternative. This study evaluates the feasibility and efficacy of melatonin-induced sleep as an alternative for GA in ABR testing. METHODS: A retrospective review was conducted on 52

Elevated cerebral perfusion in neonatal encephalopathy is associated with neurodevelopmental impairments.

BACKGROUND: Neonatal encephalopathy (NE) represents a primary cause of neonatal death and neurodevelopmental impairments. In newborns with NE, cerebral hyperperfusion is related to an increased risk of severe adverse outcomes, but less is known about the link between perfusion and mild to moderate developmental impairments or developmental delay. METHODS: Using arterial spin labelling perfusion MRI, we investigated the link between perfusion in 36 newborns with NE and developmental outcome at 2 years. RESULTS: 53% of the infants demonstrated a normal outcome at 24 months, while two had cereb

Identifying novel data-driven subgroups in congenital heart disease using multi-modal measures of brain structure.

Individuals with congenital heart disease (CHD) have an increased risk of neurodevelopmental impairments. Given the hypothesized complexity linking genomics, atypical brain structure, cardiac diagnoses and their management, and neurodevelopmental outcomes, unsupervised methods may provide unique insight into neurodevelopmental variability in CHD. Using data from the Pediatric Cardiac Genomics Consortium Brain and Genes study, we identified data-driven subgroups of individuals with CHD from measures of brain structure. Using structural magnetic resonance imaging (MRI; N = 93; cortical thickness

Comparing Healthcare Needs in Extremely Low Birth Weight Infants With NEC and Spontaneous Intestinal Perforation.

BACKGROUND: Necrotizing enterocolitis (NEC) and spontaneous intestinal perforation (SIP) affect 6-8% of extremely low birth weight (ELBW) infants. SIP has lower mortality than NEC, but with similar short-term morbidity in length of stay, growth failure, and supplemental oxygen requirements. Comparative long-term neurodevelopmental outcomes have not been clarified. METHODS: Data were prospectively collected from 59 North American neonatal units, regarding ELBW infants (401-1000 g or 22-27 weeks gestational age) born between 2011 and 2018 and evaluated again at 16-26 months corrected age. Transcriptomic, proteomic, and metabolomic analyses identify candidate pathways linking maternal cadmium exposure to altered neurodevelopment and behavior.

Cadmium (Cd) is a ubiquitous toxic heavy metal of major public concern. Despite inefficient placental transfer, maternal Cd exposure impairs fetal growth and development. Increasing evidence from animal models and humans suggests maternal Cd exposure negatively impacts neurodevelopment; however, the underlying molecular mechanisms are unclear. To address this, we utilized multiple -omics approaches in a mouse model of maternal Cd exposure to identify pathways altered in the developing brain. Offspring maternally exposed to Cd presented with enlarged brains proportional to body weights at birth

An Examination of the Long-Term Neurodevelopmental Impact of Prenatal Zika Virus Infection in a Rat Model Using a High Resolution, Longitudinal MRI Approach.

Since Zika virus (ZIKV) first emerged as a public health concern in 2015, our ability to identify and track the long-term neurological sequelae of prenatal Zika virus (ZIKV) infection in humans has been limited. Our lab has developed a rat model of maternal ZIKV infection with associated vertical transmission to the fetus that results in significant brain malformations in the neonatal offspring. Here, we use this model in conjunction with longitudinal magnetic resonance imaging (MRI) to expand our understanding of the long-term neurological consequences of prenatal ZIKV infection in order to i

A multi-task, multi-stage deep transfer learning model for early prediction of neurodevelopment in very preterm infants.

Survivors following very premature birth (i.e., ≤ 32 weeks gestational age) remain at high risk for neurodevelopmental impairments. Recent advances in deep learning techniques have made it possible to aid the early diagnosis and prognosis of neurodevelopmental deficits. Deep learning models typically require training on large datasets, and unfortunately, large neuroimaging datasets with clinical outcome annotations are typically limited, especially in neonates. Transfer learning represents an important step to solve the fundamental problem of insufficient training data in deep learning. In thi

Visual Sequences Drive Experience-Dependent Plasticity in Mouse Anterior Cingulate Cortex.

Mechanisms of experience-dependent plasticity have been well characterized in mouse primary visual cortex (V1), including a form of potentiation driven by repeated presentations of a familiar visual sequence ("sequence plasticity"). The prefrontal anterior cingulate cortex (ACC) responds to visual stimuli, yet little is known about if and how visual experience modifies ACC circuits. We find that mouse ACC exhibits sequence plasticity, but in contrast to V1, the plasticity expresses as a change in response timing, rather than a change in response magnitude. Sequence plasticity is absent in ACC,
