Engineering of Copper-Based Nitrogen-Rich Covalent Triazine Frameworks (Cu@CTFs) as Highly Efficient Electrocatalysts for Carbon Dioxide Reduction.

In this study, we employed a simple and efficient method to synthesize two porous covalent triazine frameworks (CTFs) using a cyanuric chloride core with both 1,4-diaminobenzene and 1,1'-biphenyl-4,4'-diamine, which were subsequently embedded with copper acetate, giving Cu@CTF electrocatalysts. The composite materials were comprehensively characterized, and their performance in the electrochemical CO2 reduction reaction (CO2RR) was systematically evaluated. The Cu@CTFs exhibited significant enhancements in CO2RR performance compared to their unmodified counterparts. The as-synthesized Cu@CC-BP

Interfacial engineering of carbon quantum dot/metal-organic framework heterostructures for boosted urea electrosynthesis from carbon dioxide and nitrate.

Electrocatalytic urea synthesis from CO2 and NO3- offers a sustainable alternative to conventional energy-intensive industrial processes. However, it remains hindered by sluggish CN coupling kinetics and mismatched CO2/NO3- reduction rates. Herein, a carbon quantum dots (CQDs)/metal-organic framework (MOF) heterostructure (Cu-MOF-CQD) was constructed by integrating amino-rich CQDs into the porous Cu-BTC framework (BTC = 1,3,5-benzenetricarboxylic acid) via a one-step ultrasonic-static strategy. The integration of CQDs markedly enhances the electrical conductivity of Cu-MOF and modulates the in

Structural and functional characterization of a metagenomically derived γ-type carbonic anhydrase and its engineering into a hyperthermostable esterase.

The 16S microbial community profiling of a metagenomics library from geothermal spring at Lisvori (Lesvos island, Greece) enabled the identification of a putative sequence exhibiting 95% identity to the γ-type carbonic anhydrase (γ-CA) from Caloramator australicus (γ-CaCA). The sequence of γ-CaCA was amplified by PCR, cloned, and expressed in E. coli. Activity assays showed that γ-CaCA possesses very low, but detectable, anhydrase activity, while exhibiting no measurable esterase activity. Differential scanning fluorimetry (DSF) revealed that the enzyme shows high thermal stability with a melt

Engineering Mono- vs Dinuclear Iron(II)-Active Sites to Steer Liquid Oxygenate Selectivity in Methane Oxidation Pathways Using O2.

The selective oxidation of methane to liquid oxygenates using O2 under mild conditions remains a grand challenge due to the chemical inertness of methane and the competing thermodynamically favored overoxidation. We report the development of well-defined mono- and dinuclear bipyridyl Fe(II) hydroxyl species within a porous metal-organic framework [MOF-253(Al)], which enable methane oxidation by O2 via distinct oxidation pathways with exceptionally high product selectivity. The MOF-supported dinuclear Fe(II) catalyst (MOF-253-Fe2) having a [(bpy)FeII(μ2-OH)(OH)]2 active site achieves 100% liqui

Synergistical Engineering of Vacancy and Doping Enables High-Rate and Ultrastable Na4Fe2Mn(PO4)2P2O7 Cathode for Sodium Ion Batteries.

Iron-manganese-based polyanionic compounds (Na4Fe3-xMnx(PO4)2P2O7) have attracted extensive interest as cathode for sodium ion batteries (SIBs) owing to higher working voltage, and higher energy density compared to Na4Fe3(PO4)2P2O7. However, such cathode suffers from sluggish Na+ diffusion, severe voltage hysteresis, and large structural strain caused by Jahn-Teller distortion of Mn3+. Taking Na4Fe2Mn(PO4)2P2O7 (NFMPP) as an example, herein, a synergistic engineering strategy of vacancy and doping for constructing Na4Fe2Mn1-5y/3Nb2y/3□y(PO4)2P2O7 cathodes (□: vacancy) is developed where vacanc

Engineering an organoid culture system for enhanced murine and human hematopoietic stem and progenitor cell self-renewal and expansion.

Developing a biomimetic culture system is crucial for the efficient maintenance and expansion of rare hematopoietic stem and progenitor cells (HSPCs)in vitro. This advancement can significantly enhance the application of HSPC-based transplantation therapies and support the manufacturing of bone marrow (BM) organoids. Traditional two-dimensional culture systems fall short in replicating the interactions between cultured cells and the hematopoietic niche, resulting in excessive reactive oxygen species (ROS) production and triggering HSPC differentiation. In response, we have developed an innovat

Enzyme and cofactor engineering to increase d-xylonate dehydratase activity for improved d-1,2,4-butanetriol production from d-xylose.

d-1,2,4-Butanetriol (BTO), a C4 platform compound, is widely used in fields such as military and pharmaceuticals. Biosynthesis of d-1,2,4-BTO from lignocellulose-derived d-xylose presents a promising production route. However, the low catalytic activity of d-xylonate dehydratase leading to the accumulation of d-xylonic acid remains a key bottleneck for the efficient production of d-1,2,4-BTO. In this study, we aimed to enhance the catalytic activity of d-xylonate dehydratase through an integrated enzyme and cofactor engineering approach. Firstly, we evolved the d-xylonate dehydratase YjhG by u

Beyond Pristine Layers: Engineering van der Waals MPX3 Materials (M: Transition Metal, X: Chalcogen) via Defects, Adsorption, and Intercalation.

Recent breakthroughs and achievements in the studies of 2D materials have led to the increased attention to the respective van der Waals parent compounds. Herein, the class of layered materials-so-called transition metal phosphorus trichalcogenides (MPX3; M: transition metal, X: chalcogen)-has been recently investigated extensively due to the diversity in their properties depending on the M/X combination. Moreover, further studies demonstrate the large tunability of the electronic, optical, and magnetic properties of these materials using different methods, like defects' engineering, alloying,

Engineering Multi-Functional Enzyme-Mimetic Polyphenol-Catalase Complex for Reversing Hypoxia and Redox Homeostasis in Vascular and Muscular Regeneration.

Hypoxia contributes to a wide range of pathological conditions, including muscle atrophy and ischemic diseases, yet effective therapeutic strategies remain limited. In this study, we developed an epigallocatechin-3-gallate (EGCG)-catalase complex (EC) that simultaneously provides oxygenation and reactive oxygen species (ROS) clearance through multienzyme mimicry. EC exhibits superoxide dismutase (SOD)-like activity by converting superoxide anion (O2•-) into hydrogen peroxide (H2O2), followed by catalase-mediated decomposition of H2O2 into oxygen (O2) and water (H2O), thereby transforming harmf

Intervertebral disc degeneration (IVDD) is a leading cause of discogenic low back pain, contributing significantly to global disability and economic burden. Current treatments provide only short-term pain relief without addressing the underlying pathogenesis. Herein we report engineering of biomimetic therapies for IVDD guided by single-cell RNA-sequencing data from human nucleus pulposus tissues, along with validation using animal models. In-depth analyses revealed the critical role of mitochondrial dysfunction in fibrotic phenotype polarization of nucleus pulposus cells (NPCs) during IVDD pr

Combined effect of strain and intrinsic spin-orbit coupling on band gap engineering of GNRs: a first-principles study.

By employing first-principles calculations, we theoretically investigate the impact of uniaxial strain and intrinsic spin-orbit coupling (SOC) on the electronic properties of zigzag and armchair edge hydrogen (H)-passivated graphene nanoribbons (GNRs). We find that band structure and density of states of 4-zigzag graphene nanoribbon (ZGNR) and 15-armchair graphene nanoribbon (AGNR) are highly sensitive to the combined effect of strain and intrinsic SOC. In the case of H-passivated 4-ZGNR, SOC with a strain>10% increases the energy band by increasing spin-polarized states at the opposite edges.

Engineering a CRISPRoff Platform to Modulate Expression of Myeloid Cell Leukemia (MCL-1) in Committed Oligodendrocyte Neural Precursor Cells.

In vitro differentiation of human pluripotent stem cell (hPSC) model systems has furthered our understanding of human development. Techniques used to elucidate gene function during early development have encountered technical challenges, especially when targeting embryonic lethal genes. The introduction of CRISPRoff by Nuñez and collaborators provides an opportunity to heritably silence genes during long-term differentiation. We modified CRISPRoff and sgRNA Sleeping Beauty transposon vectors that depend on tetracycline-controlled transcriptional activation to silence the expression of embryoni

A Novel Engineering Cell Therapy Platform Mimicking the Immune Thrombocytopenia-Derived Platelets to Inhibit Cytokine Storm in Hemophagocytic Lymphohistiocytosis.

Hemophagocytic lymphohistiocytosis (HLH) is a common and highly fatal hyperinflammatory syndrome characterized by the aberrant activation of macrophages. To date, there is a lack of targeted therapies for HLH. It is validated that macrophages in HLH efficiently phagocytose anti-CD41-platelets (anti-CD41-PLTs) from immune thrombocytopenia (ITP) patients in previous research. Hence, the pathological mechanisms of ITP are mimicked and anti-CD41-PLTs are utilized to load the macrophage-toxic drug VP16 to construct macrophage-targetable engineered platelets anti-CD41-PLT-VP16, which is a novel targ

Influence of Oligomeric Lactic Acid and Structural Design on Biodegradation and Absorption of PLA-PHB Blends for Tissue Engineering.

The advancing development in biomaterials and biology has enabled the extension of 3D printing technology to the bioadditive manufacturing of degradable hard tissue substitutes. One of the key advantages of bioadditive manufacturing is that it has much smaller design limitations than conventional manufacturing and is therefore capable of producing implants with complex geometries. In this study, three distinct blends of polylactic acid (PLA) and polyhydroxybutyrate (PHB) were produced using Fused Deposition Modeling (FDM) technology. Two of these blends were plasticized with oligomeric lactic

Planar Chlorination Engineering: A Strategy of Completely Breaking the Geometric Symmetry of Fe-N4 Site for Boosting Oxygen Electroreduction.

Introducing asymmetric elements and breaking the geometric symmetry of traditional metal-N4 site for boosting oxygen reduction reaction (ORR) are meaningful and challenging. Herein, the planar chlorination engineering of Fe-N4 site is first proposed for remarkably improving the ORR activity. The Fe-N4/CNCl catalyst with broken symmetry exhibits a half-wave potential (E1/2) of 0.917 V versus RHE, 49 and 72 mV higher than those of traditional Fe-N4/CN and commercial 20 wt% Pt/C catalysts. The Fe-N4/CNCl catalyst also has excellent stability for 25 000 cycles and good methanol tolerance ability.
