Phosphate-Mediated Regulation of Intracellular Calcium Dynamics.

Phosphate (Pi) and calcium (Ca2+) are essential mineral ions that play coordinated roles in maintaining normal cellular functions. While various steps of calcium signaling are well characterized, emerging evidence suggests the critical role of both intracellular and extra cellular phosphate in regulating intracellular Ca2+. In the cytoplasm, phosphate influences ATP production and organelle calcium buffering and influences the activity of calcium pumps, such as sarcoplasmic/endoplasmic reticulum Ca2+-ATPase (SERCA) and the plasma membrane Ca2+-ATPase (PMCA). Extracellular phosphate, taken up v

Interconnection between mechanistic and kinetic investigations of organometallic complex formation in palladium detection.

The main goal of this work was to explain the mechanism of organometallic complex formation during the process of palladium determination. The azo dye tropaeolin OO and Pd(II) ions in various media (H2O, acetate buffer, ethanol) were used as a model system to predict the mechanism. The changes occurring during complex formation were monitored spectrophotometrically. Based on the obtained spectra evolution, at selected wavelengths kinetic curves were constructed. Rate constants for formation of the intermediate and final products were determined by fitting equations to the kinetic curves. The c

Interaction with CYP20-3 limits OPDA flux into jasmonate biosynthesis in wounded leaves of arabidopsis grown at eCO2.

Upon recognition of a stress, such as wounding, a rapid increase in jasmonates leads to plant resistance against necrotrophic pathogens and chewing insect herbivores. This jasmonate burst is weaker in Arabidopsis thaliana plants grown under future predicted carbon dioxide levels (eCO2) compared to todays' levels. Even though foliar levels of jasmonoyl-isoleucine are lower in wounded arabidopsis at eCO2, levels of their precursor, 12-oxo-phytodienoic acid (OPDA), are not affected by atmospheric CO2 levels. We focused on the role of the OPDA-binding protein CYP20-3 in regulating jasmonate levels

Bridging the accuracy-speed divide in reactive molecular dynamics with QuantaMind MD.

Simulating chemical reactivity remains a central challenge in molecular dynamics, historically constrained by the trade-off between ab initio accuracy and classical efficiency. We present QuantaMind, a machine learning force field (MLFF) framework that attains density functional theory (DFT)-level accuracy, enabling fully reactive simulations of complex molecular systems. QuantaMind achieves exceptional numerical stability over tens-of-nanosecond trajectories, maintaining accuracy throughout long-time reactive dynamics. It reproduces spontaneous bond formation and cleavage in key chemical proc

Armed conflict accelerates cropland loss and food insecurity worldwide.

Armed conflict is an escalating threat to global food security yet its impact on cropland dynamics is poorly understood at global scale. Here we combine annual high-resolution cropland maps (2000-2020) with more than 460,000 conflict events across 155 countries to assess how armed conflicts affect cropland loss and food security. By 2020, conflict-exposed areas within 5 km of conflict events covered only 3% of global land but contained 6% of global cropland, indicating disproportionate exposure of agricultural land to conflict. Difference-in-differences estimates show that conflict exposure ca

Gelatin-Stabilized Water-in-Water Emulsions for Biocatalysis: Interfacial Enzyme Confinement, In Situ Product Extraction, and Cryogenic Recyclability.

Water-in-water (W/W) emulsions offer high biocompatibility for enzymatic processes but are difficult to stabilize with conventional oil-water surfactants due to their ultralow interfacial tension. Here, we design a gelatin-stabilized PEG/dextran (PEG/Dex) emulsion platform enabling integrated "reaction-separation-recovery" biocatalysis. By tuning the pH and polymer concentration, programmable microarchitectures such as core-shell, Janus, or triple-phase droplets were achieved. In cascade oxidation of guaiacol, the compartmentalized system reached an 86% yield of tetraguaiacol─50% higher than i

Rainfall deficit reduces biodiversity and destabilizes a saline-soil plant community.

Rainfall deficits are reshaping plant communities worldwide, yet their impacts on salinity-stressed ecosystems remain unclear. In saline systems, rainfall is essential for flushing soil salts and sustaining biodiversity. Here, we tested the hypothesis that rainfall deficit undermines ecosystem stability by eroding biodiversity in these systems. We conducted a 7-year experiment in the Yellow River Delta, simulating summer-autumn rainfall loss under both ambient and elevated winter-spring temperatures. Rainfall loss increased soil salinity (+43.3% under ambient; +25.2% under warming), promoted s

Residential green space, air pollution, and related metabolites in association with depression among cancer survivors.

The association of natural environmental exposure and air pollution with depression incidence among cancer survivors, as well as the potential role of plasma metabolomics, remains unclear. Here, we analyze 21,507 cancer survivors from the UK Biobank over a median follow-up of 12.39 years and find that individuals exposed to higher levels of green space and natural environment (tertile 3 vs. tertile 1) within a 1000-m buffer have 15.8% (95% CI: 4.0%-26.1%) and 18.2% (95% CI: 7.0%-28.1%) lower risks of depression, respectively. The strongest protective association is observed among breast cancer

The Dorsal Root Ganglia: From a Neuronal Relay to a Sensory Organ.

Somatosensory ganglia are often cast as passive relays, yet growing evidence shows the dorsal root ganglion (DRG) is a specialized sensory-immune organ. In the DRG, perineuronal and perivascular units act as sentinels that detect danger and calibrate immune tone. A permeable, macrophage-guarded blood-DRG barrier admits systemic cues, while neuron-glia microdomains set sensory gain and help restore homeostasis. Throughout the organ, neurons, glia, and vascular-stromal cells share immune receptors, enabling coordinated responses to infection, inflammation, and autoimmunity. In turn, neuronal sig

Introgression across ploidies contributes to genetic diversity in introduced urban Capsella bursa-pastoris.

Successful establishment of a species in a new range is a useful way to understand the impact of demography and selection on the evolution of globally distributed species. In particular, introductions influence genetic diversity and population structure in the introduced range in unpredictable ways. Additionally, introgressive hybridization is often associated with successful establishment in new ranges. In this study, we explore the impact of introgressive hybridization on the polyploid Capsella bursa-pastoris in the New York City metropolitan area. We find Capsella bursa-pastoris in the New

Surgical treatment of a congenital rectal cystic hamartoma in a 20-day Kurdish foal.

Hamartomas are mostly benign tumor-like malformations characterized by a disorganized overgrowth of mature cells and tissues indigenous to the affected anatomical site. A 20-day-old male Kurdish foal was presented to the Veterinary Teaching Hospital of Urmia University, Urmia, Iran, with a history of hematochezia and tenesmus. Clinical examination identified a pedunculated tissue mass within the rectal lumen. The abnormal mass was surgically excised via polypectomy under local anesthesia. Tissue samples were obtained, fixed in 10.00% neutral buffered formalin, and processed routinely for histo

TXNL1 in redox and protein quality control: mechanisms and disease relevance.

TXNL1 (thioredoxin-like protein 1), a member of the thioredoxin superfamily, integrates thioredoxin oxidoreductase activity with molecular chaperone function. Through its N-terminal TRX domain and C-terminal PITH domain, TXNL1 not only helps maintain cellular redox homeostasis but also prevents toxic aggregation of oxidatively damaged proteins. Consequently, TXNL1 acts as a critical buffer in the cellular defense against oxidative stress and in protein quality control. TXNL1 is widely expressed across diverse tissues, and its dysregulation is closely associated with multiple pathological condi

Cerebrospinal fluid dynamics and brain function regulation: from homeostasis to neurological disorders.

Cerebrospinal fluid (CSF) is increasingly recognized as an active regulator of brain function rather than a passive mechanical buffer. Beyond its classical roles in cushioning the brain and removing metabolic waste, CSF participates in a tightly coupled system linking neural activity, vascular dynamics, molecular signaling, and tissue mechanics. Here, we present an integrated theoretical framework that unifies three major conceptual strategies in contemporary CSF research: metabolic clearance, neuromodulatory signaling, and bidirectional coupling between fluid dynamics and neural activity. We

Design, radiolabeling, and biological evaluation of a novel 177Lu-AAZTA5-Carfilzomib conjugate as a proteasome-targeted theranostic agent for multiple myeloma.

The development of proteasome-targeted radiopharmaceuticals represents a promising strategy for the treatment of multiple myeloma. In this work, a novel Carfilzomib-based radioconjugate was designed through conjugation with the hybrid chelator AAZTA5 and subsequently radiolabeled with Lutetium-177. The conjugate was labeled efficiently under mild conditions with high radiochemical yield and exhibited excellent in vitro stability in both phosphate-buffered saline and serum. Biological evaluation using myeloma cell lines demonstrated a pronounced cytotoxic effect of the 177Lu-AAZTA5-Carfilzomib

Fluorescence-colorimetric dual-mode detection of cysteine in artificial urine with potential relevance to cervical cancer.

As cervical cancer poses a serious threat to the health of women, non-invasive and convenient diagnostic methods are still in urgent need. With cysteine (Cys) in urine as the disease marker, a xanthylium-based colorimetric and fluorescent probe SH-OH-NBD for Cys detection was constructed and its response to Cys was explored in artificial urine rather than common buffer solutions. Upon the action of Cys, the nitrobenzoxadiazole ether in SH-OH-NBD was cleaved, generating a new fluorescence peak at 567 nm. Thus, the concentration of Cys could be accurately determined based on the turn-on fluoresc

Beyond the Tics: Experiences of Stigma and Psychosocial Impact in Tourette Syndrome.

Persistent tic disorders (PTD), including Tourette Syndrome (TS), impact individuals' lives well beyond motor and vocal tics, affecting self-beliefs, relationships, and daily functioning. This qualitative study examined stigma and discrimination experienced by youth and adults with TS, their caregivers, and healthcare providers through in-depth interviews with 35 participants. Thematic analysis revealed four major themes: limited resources and funding, widespread misconceptions and stereotypes, negative experiences shaping self-perceptions and behaviors, and the importance of supportive enviro

The relationship between perceived parental knowledge and adolescent gambling.

This study revisits and extends prior evidence on the role of adolescents' perceived parental knowledge-how much they believe their parents know about their whereabouts and companions-in reducing gambling through attitudes and peer norms. Using two waves of the nationally representative survey ESPAD® Italia (2022-23; N = 24,631), we re-examine this relationship after major societal and digital shifts, including the COVID-19 pandemic and the expansion of online gambling platforms. Our results confirm that higher perceived parental knowledge is associated with lower gambling frequency, both dire

Effective Reduction of Anti-PEG Antibody Recognition via PEG Copolymers with Ethyl Glycidyl Ether and Isopropyl Glycidyl Ether.

Anti-PEG antibodies (APAs) lead to accelerated blood clearance of PEGylated drugs and nanocarriers. Randomized PEG (rPEG) reduces APA recognition by randomly incorporating glycidyl methyl ether in PEG. Extending this concept, statistical copolymerization of ethylene oxide with ethyl glycidyl ether (EGE) or isopropyl glycidyl ether (iPGE) was investigated. Copolymers of EO with 13-28 mol % EGE or 6-20 mol % iPGE showed low dispersity (Đ ≤ 1.09). Via online-NMR monitoring, reactivity ratios in DMSO were rEO = 1.39 and rEGE = 0.72 for P(EO-co-EGE) and rEO = 1.35 and riPGE = 0.74 for P(EO-co-iPGE)

Not paper people: application forms as ethical stories at the wellbeing garden.

This paper reports on ethnographic observations about the filling in, and handling, of application forms for a garden-based wellbeing program. It describes the ethical challenges involved in determining the eligibility of applicants, particularly where application forms aren't completed clearly or don't present cases indicative of potential health benefit. The paper utilises a narrative approach to the anthropology of ethics to interpret application forms as gambits in self-narratives about worth and need. These narratives are diverse, sometimes unclear, and sometimes do not fit easily with in

Decoding heterogeneous single-cell perturbation responses.

Understanding how cells respond differently to perturbation is crucial in cell biology, but existing methods often fail to accurately quantify and interpret heterogeneous single-cell responses. Here we introduce the perturbation-response score (PS), a method to quantify diverse perturbation responses at a single-cell level. Applied to single-cell perturbation datasets such as Perturb-seq, PS outperforms existing methods in quantifying partial gene perturbations. PS further enables single-cell dosage analysis without needing to titrate perturbations, and identifies 'buffered' and 'sensitive' re
