Advanced assay design for characterizing anti-drug antibody responses in clinical serum samples.

Monitoring anti-drug antibody (ADA) responses is critical for evaluating the safety and efficacy of protein therapeutics. While traditional three-tiered testing (screening, confirmation, titration) reliably identifies ADA incidence, onset and magnitude, modern, complex biologics necessitate adapting our assessment strategies. Consequently, advanced assay designs are required to capture the full biological and clinical impact of these immune responses. Structured to guide bioanalytical scientists through this paradigm shift, this review first explores the transition toward integrated functional

Beyond prediction: Why experimental structural biology remains essential in plants.

Structural biology is undergoing a transformative era driven by advances in artificial intelligence (AI)-based protein structure prediction and cryo-electron microscopy. Predictive approaches have dramatically expanded structural coverage across proteomes and are increasingly integrated into experimental workflows. However, protein function frequently depends on dynamic molecular processes including ligand-dependent conformational remodeling, transient interactions, cooperative assembly, and transport-state transitions that remain difficult to define from static computational models alone. The

A comparative study of HSO-optimized PID and TID controllers with fuzzy adaptation for delta robot control.

This paper presents the development, tuning, and comparative evaluation of intelligent control strategies for a highly nonlinear Delta robot. The work includes the development and validation of a Simscape-based model derived from the experimental platform, followed by controller design using four approaches: Harmony-Search-optimized PID, Harmony-Search-optimized Tilt-Integral-Derivative (TID), self-tuning fuzzy PID, and self-tuning fuzzy TID control. System identification was used to obtain a reduced model suitable for efficient controller tuning, and the identified NLARX model achieved an MSE

Ask-Audit-Apply: A Framework for Clinical Reasoning with Artificial Intelligence.

Artificial intelligence (AI)-enabled clinical decision support (CDS) tools are increasingly embedded in clinical workflows, generating and prioritizing clinical information, including diagnostic suggestions, evidence summaries, and management recommendations. Yet guidance on how clinicians should reason with AI at the point of care remains limited, creating risks of over-reliance, inconsistent verification, and diffusion of responsibility. We propose Ask-Audit-Apply (AAA) as a clinician-facing framework for AI-supported clinical reasoning that specifies decision-level behavioral practices. AAA

Transforming tai ji quan into next-generation, evidence-based exercise medicine: A translational roadmap for falls prevention.

A substantial body of research provides compelling evidence that tai ji quan, a mind-body exercise rooted in traditional Chinese medicine, is effective in reducing falls and improving mobility and has gained increasing recognition as a form of evidence-based exercise medicine within mainstream healthcare. Despite this strong evidence base and growing policy support, the translation of tai ji quan from research into routine clinical practice remains limited. Addressing this evidence-to-practice gap will require innovative translational strategies that more effectively integrate evidence-based t

Heart Transplantation in 2076: From Scarcity to Cardiac Restoration.

Heart transplantation has historically been defined by the scarcity of donor organs and the uncertainties surrounding them. Over the past 50 years, advances in immunosuppression, surgical techniques, and posttransplant surveillance have improved outcomes, yet the fundamental paradigm remains unchanged: the replacement of a failing heart still depends on the death of another person. In this Perspective, we argue that this paradigm is beginning to shift, with early evidence emerging from molecular diagnostics that enable noninvasive detection of graft injury. Over the coming decades, regenerativ

High-performance polydopamine and silver nanoparticle-modified MXene-based hydrogel flexible strain sensors for transfer-learning-assisted handwriting recognition and wrist-movement monitoring.

Hydrogel-based flexible strain sensors show great potential for applications in wearable electronics, human-machine interfaces, and soft robotics. However, it remains challenging to prepare hydrogel sensors with high sensitivities and wide detection ranges. Herein, we report a polydopamine and silver nanoparticle-modified MXene-based hydrogel, namely, MXene/Ag@PDA/P(AA-AM)/SA-Zn (MAPS), which exhibits a high sensitivity (with a gauge factor of 16.64), high stretchability (1100%), and short response time (0.2 s). This hydrogel-based strain sensor can be adhered to the human body surface as a we

Piezo1-Fstl1 Axis in Fracture Healing: Modulation of the Chondrocyte Inflammation-ROS-Mitochondrial Damage Cascade and Application of Smart Delivery System.

This study investigated the regulatory role of an intelligent drug delivery system in promoting fracture healing via Piezo1-Fstl1 signaling axis. It also verified its modulation of chondrocyte inflammatory response, mitochondrial oxidative stress, and osteoblast differentiation. Inflammation triggers the accumulation of pro-inflammatory factors, and reactive oxygen species (ROS) in chondrocytes. This leads to oxidative damage in mitochondria, a decrease in mitochondrial membrane potential (MMP), and the induction of mitochondrial permeability transition pore (mPTP) opening, thereby hindering f

A Neuro-Symbolic Bioinformatics Framework for Unlocking Chordate Physiological Dark Data and Validating Allometric Scaling.

Animal functional trait data are essential for macroecology, but massive datasets remain locked in unstructured scientific literature. Traditional manual extraction is inefficient, and general-purpose artificial intelligence (AI) systems struggle with complex biological tables and numerical accuracy. To address this bioinformatics challenge, we propose a multimodal neuro-symbolic framework combining visual-language perception and code-based reasoning. This approach reconstructs complex document layouts and delegates biostatistical calculations, such as unit normalization and thermodynamic ener

Childhood-Onset Lupus Nephritis in the Era of Triple Therapy and Steroid Minimization.

Childhood-onset lupus nephritis (cLN) should no longer be framed as a smaller version of adult lupus nephritis. It is a high-stakes pediatric kidney disease in which immune injury, treatment toxicity, growth, puberty, fertility, adherence, and transition to adult care intersect over decades. Approximately 10-20% of systemic lupus erythematosus begins in childhood, and 40-60% of affected children develop lupus nephritis. Regional cohorts report even higher renal involvement in some populations, including 65-73% among Saudi children with SLE. Contemporary guidance has moved from prolonged high-d

Lumbar spinal stenosis: current concept of management.

Lumbar spinal stenosis (LSS) is a common degenerative spinal condition where spinal canal narrowing causes symptoms such as neurogenic claudication, radiculopathy, and lower back pain. While non-operative and surgical approaches yield similar long-term outcomes, surgical intervention-particularly decompression-can provide earlier symptom relief, functional recovery, and fall prevention in selected patients with refractory symptoms. Recent advancements in surgical technologies and image guidance have brought about a paradigm shift in LSS management. Biportal endoscopic spine surgery (BESS) has
