Activation of AMPKα2 attenuated doxorubicin-induced cardiotoxicity via inhibiting lipid peroxidation associated ferroptosis.

Ferroptosis has been suggested to involve in doxorubicin (DOX)-induced cardiotoxicity. However, the underlying mechanisms and regulatory targets of cardiomyocyte ferroptosis remains to be understood. This study demonstrated that the up-regulation of ferroptosis associated proteins genes were accompanied with the down-regulation of AMPKα2 phosphorylation in DOX treated mouse heart or neonatal rat cardiomyocytes (NRCMs). AMPKα2 knockout (AMPKα2-/-) significantly exacerbated mouse cardiac dysfunction, increased mortality, promoting ferroptosis associated mitochondrial injuries, enhanced ferroptos

Visible-light-assisted peroxydisulfate activation over Ag6Si2O7/Cu(II)-modified palygorskite composite for the effective degradation of organic pollutants by radical and nonradical pathways.

Photoassisted persulfate activation (PPA) is highly efficient oxidation process, but the eligible catalysts are scarce. Herein, a visible-light-responsive Ag6Si2O7 was anchored on Cu(II)-exchanged attapulgite (Cu-Pal) via a facile precipitation-deposition method to construct a novel PDS activator. The synthesized catalyst was systemically analyzed by a series of characterization techniques. The results revealed that Ag6Si2O7 nanoparticles were evenly dispersed on Cu-Pal to form heterojunction. 16%-Ag6Si2O7/Cu-Pal, an optimal catalyst exhibited excellent catalytic performance and stability in t

Interleukin-26 promotes the proliferation and activation of hepatic stellate cells to exacerbate liver fibrosis by the TGF-β1/Smad2 signaling pathway.

Liver fibrosis is a wound-healing process of liver featured by the activation of hepatic stellate cells (HSCs) and the deposition of extra cellular matrix (ECM). Accumulating facts have suggested that interleukin (IL) 26 is involved in the pathogenesis of liver fibrosis by the modulation of HSCs. However, the biological roles of IL-26 in liver fibrosis are still unclear. The present study aimed to determine the effect and mechanism of IL-26 on the proliferation and activation of HSCs in vitro. By cell counting kit (CCK)-8 assay, we observed that IL-26 significantly promoted the proliferation o

Novel Findings in Obstetric Brachial Plexus Palsy: A Study of Corpus Callosum Volumetry and Resting-State Functional Magnetic Resonance Imaging of Sensorimotor Network.

BACKGROUND: The response of the brain to obstetric brachial plexus palsy (OBPP) is not clearly understood. We propose that even a peripheral insult at the developmental stage may result in changes in the volume of white matter of the brain, which we studied using corpus callosum volumetry and resting-state functional magnetic resonance imaging (rsfMRI) of sensorimotor network. OBJECTIVE: To study the central neural effects in OBPP. METHODS: We performed an MRI study on a cohort of 14 children who had OBPP and 14 healthy controls. The mean age of the test subjects was 10.07 ± 1.22 yr (95% con

Activation of the hypothalamic paraventricular nucleus by forebrain hypertonicity selectively increases tonic vasomotor sympathetic nerve activity.

We recently reported that mean arterial pressure (MAP) is maintained in water-deprived rats by an irregular tonic component of vasomotor sympathetic nerve activity (SNA) that is driven by neuronal activity in the hypothalamic paraventricular nucleus (PVN). To establish whether generation of tonic SNA requires time-dependent (i.e., hours or days of dehydration) neuroadaptive responses or can be abruptly generated by even acute circuit activation, forebrain sympathoexcitatory osmosensory inputs to PVN were stimulated by infusion (0.1 ml/min, 10 min) of hypertonic saline (HTS; 1.5 M NaCl) through

Group-based trajectory modeling: a novel approach to examining symptom trajectories in acute bipolar episodes.

BACKGROUND: Pattern analysis can aid understanding of trajectories of symptom evolution. However, most studies focus on relatively homogeneous disorders with a restricted range of outcomes, prescribed a limited number of classes of medication. We explored the utility of pattern analysis in defining short-term outcomes in a heterogeneous clinical sample with acute bipolar disorders. METHOD: In a naturalistic observational study, we used Group-based trajectory modeling (GBTM) to define trajectories of symptom change in 118 bipolar cases recruited during an acute DSM IV episode: major depression

Thermal Degradation and Pyrolysis Kinetics of Alkaline-Pretreated Pinewood under Non-Isothermal Conditions.

Pinewood (PW) residues are abundant lignocellulosic wastes, but their elevated volatile and oxygenated fractions can complicate pyrolysis by promoting complex devolatilization behavior. This challenge motivates the use of NaOH pretreatment of PW to enhance thermal conversion efficiency. This study investigates the catalytic effect of sodium hydroxide (NaOH) on the pyrolysis kinetics of PW under non-isothermal conditions. Thermogravimetric analysis (TGA) was conducted on untreated PW and samples pretreated with varying concentrations of NaOH (0.2, 0.5, 1, and 2 wt %) at four different heating r

Lactate-induced metabolic reprogramming of TAMs impairs antigen presentation capacity via C/EBPα-CD74 axis in oral squamous cell carcinoma.

BACKGROUND: Throughout oral squamous cell carcinoma (OSCC) progression, tumor-associated macrophages (TAMs) lose their antigen-presenting capacity and anti-tumor function. The mechanisms that cause this dysfunction are not fully understood. CD74 is essential for antigen-presenting process, while little direct evidence describes its role in TAMs' immune function. METHODS: We integrated single-cell transcriptomic analysis, clinical cohort validation and CD74 conditional knockout mouse model to investigate the role of CD74 in TAMs during OSCC progression. Inhibition of replication factor C4-induced DNA damage repair reverses the chemoresistance of glioblastoma to temozolomide and reveals the synthetic lethal effect of combined targeting of checkpoint kinase 1.

Glioblastoma (GBM) is a highly aggressive malignant tumor of the central nervous system, and resistance to temozolomide (TMZ) remains a major cause of treatment failure and tumor recurrence. DNA damage repair (DDR) serves as a critical mechanism underlying chemoresistance, with replication factor C4 (RFC4) emerging as a key mediator. We aimed to investigate the mechanism by which RFC4 induces TMZ resistance in GBM through DDR activation. Analysis of clinical specimens revealed that RFC4 is overexpressed in glioma tissues and correlates with poor prognosis in patients with GBM. Functional assay

EXOC6B promotes cilial elongation via autophagy-dependent protein turnover.

Exocyst Complex Component 6b (EXOC6B), a constituent of highly conserved octameric exocyst complex, plays a pivotal role in tethering and spatial targeting of post-Golgi vesicles to the plasma membrane, thereby facilitating exocytosis. Mutations in the EXOC6B gene has been associated with severe neurological and skeletal disorders, such as spondyloepimetaphyseal dysplasia with joint laxity type 3 (SEMDJL3) and Laurin-Sandrow Syndrome, highlighting its critical role in the development. SEMDJL3 patient fibroblasts showed impaired ciliogenesis. Thus, in this study, we investigated the functional

EASIX as a moderately effective prognostic marker for mortality in severe acute pancreatitis: a retrospective study.

INTRODUCTION: The endothelial activation and stress index (EASIX) has been proposed as a surrogate laboratory index reflecting endothelial activation and stress. This study aimed to evaluate the prognostic value of EASIX in patients with severe acute pancreatitis (SAP). METHODS: This retrospective observational study analyzed 340 SAP patients hospitalized between December 2015 and December 2023. EASIX was calculated as LDH (U/L) × creatinine (mg/dL) / platelet count (109/L). Regression analyses identified mortality predictors, which were incorporated into a prognostic model.
