In silico optimized cell-penetrating peptides achieve transdermal siRNA delivery and regulate inflammatory environment in psoriasis.

To enable Small Interfering RNA (siRNA) transdermal delivery, we use computational modeling to predict key properties of four cationic peptide carriers. These parameters can be utilized for the prediction of peptide carrier diffusion within the stratum corneum, thereby facilitating the screening of carriers with transdermal delivery capabilities. We take this opportunity to examine the discrepancy between computer-simulated transdermal vehicle functions and actual therapeutic efficacy. We validate the therapeutic efficacy of four peptide carriers by employing both human cell-derived 3D skin mo

Numerical simulations to determine the stimulation of the crista ampullaris during the Head Impulse Test.

The Head Impulse Test, the most widely accept test to assess the vestibular function, comprises rotations of the head based on idealized orientations of the semicircular canals, instead of their individual arrangement specific for each patient. In this study, we show how computational modelling can help personalize the diagnosis of vestibular diseases. Based on a micro-computed tomography reconstruction of the human membranous labyrinth and their simulation using Computational Fluid Dynamics and Fluid-Solid Interaction techniques, we evaluated the stimulus experienced by the six cristae ampull

Classification of Cracks in Composite Structures Subjected to Low-Velocity Impact Using Distribution-Based Segmentation and Wavelet Analysis of X-ray Tomograms.

The problem of characterizing the structural residual life is one of the most challenging issues of the damage tolerance concept currently applied in modern aviation. Considering the complexity of the internal architecture of composite structures widely applied for aircraft components nowadays, as well as the additional complexity related to the appearance of barely visible impact damage, prediction of the structural residual life is a demanding task. In this paper, the authors proposed a method based on detection of structural damage after low-velocity impact loading and its classification wi

Standard PK/PD concepts can be applied to determine a dosage regimen for a macrolide: the case of tulathromycin in the calf.

The pharmacokinetic (PK) profile of tulathromycin, administered to calves subcutaneously at the dosage of 2.5 mg/kg, was established in serum, inflamed (exudate), and noninflamed (transudate) fluids in a tissue cage model. The PK profile of tulathromycin was also established in pneumonic calves. For Mannheimia haemolytica and Pasteurella multocida, tulathromycin minimum inhibitory concentrations (MIC) were approximately 50 times lower in calf serum than in Mueller-Hinton broth. The breakpoint value of the PK/pharmacodynamic (PD) index (AUC(0-24 h) /MIC) to achieve a bactericidal effect was est

Computer modeling to predict effects of implant malpositioning during TKA.

Computer-assisted surgery has focused on alignment of implants and soft tissue balancing but the end results during surgery as they pertain to long-term function of the implants are not yet realized. One parameter that continues to be variable despite the use of computer navigation is the placement of the implants in the transverse plane. The objective of this study was to determine whether implant and anatomic registrations could be used in a computer model (LifeMOD/KneeSIM, LifeModeler, Inc., San Clemente, California) to show differences in the resulting contact patterns of the medial and la

Computational Modeling of Parkinson's Disease Across Scales: From Mechanisms to Biomarkers, Drug Discovery, and Personalized Therapies.

Parkinson's disease (PD) is a multifactorial neurodegenerative disorder characterized by complex interactions across molecular, cellular, circuit, and behavioral scales. While experimental and clinical studies have provided critical insights into PD pathology, integrating these heterogeneous data into coherent mechanistic frameworks and translational strategies remains a major challenge. Computational modeling offers a powerful approach to bridge these scales, enabling the systematic investigation of disease mechanisms, candidate biomarkers, and therapeutic strategies. In this review, we surve

Mixed reality-assisted orthognathic surgery with pre-bent titanium plates.

Orthognathic surgery using pre-bent titanium plates and cutting guides has been shown to enhance surgical precision in positioning the maxilla. This technical note introduces an innovative approach that utilizes mixed reality to pre-bend titanium plates and simulate surgical guides with osteotomy lines and drilling holes, thereby aiding in the localization of the maxilla. Postoperative computed tomography showed a discrepancy of <2 mm in comparison to the preoperative virtual design. The virtual surgical planning for orthognathic surgery was dynamically transferred to the surgical procedure in

Increasing midtarsal joint stiffness reduces triceps surae metabolic costs in walking simulations but has little effect on total stance limb metabolic cost.

The human foot's arch is thought to be beneficial for efficient gait. This study addresses the extent to which arch stiffness changes alter the metabolic energy requirements of human gait. Computational musculoskeletal simulations of steady state walking using direct collocation were performed. Across a range of foot arch stiffnesses, the metabolic cost of transport decreased by less than 1% with increasing foot arch stiffness. Increasing arch stiffness increased the metabolic efficiency of the triceps surae during push-off, but these changes were almost entirely offset by other muscle groups

Surgery Requiring Detailed Preoperative Simulation and Scar De-epithelialization to Repair Severe Postoperative Scarring from Gastroschisis.

We present the case of a patient with severe postoperative scarring from surgical treatment for gastroschisis, with the intestine located immediately under the dermal scar. Although many patients are unsatisfied with the results of scar repair treatment, few reports exist regarding severe or difficult cases involving the surgical repair of postoperative scar contracture. We achieved an excellent result via simulation involving graph paper drawings that were generated using computed tomography images as a reference, followed by dermal scar deepithelialization. The strategy described here may be

Rescaled Local Interaction Simulation Approach for Shear Wave Propagation Modelling in Magnetic Resonance Elastography.

Properties of soft biological tissues are increasingly used in medical diagnosis to detect various abnormalities, for example, in liver fibrosis or breast tumors. It is well known that mechanical stiffness of human organs can be obtained from organ responses to shear stress waves through Magnetic Resonance Elastography. The Local Interaction Simulation Approach is proposed for effective modelling of shear wave propagation in soft tissues. The results are validated using experimental data from Magnetic Resonance Elastography. These results show the potential of the method for shear wave propaga

Model of a predatory stealth behaviour camouflaging motion.

A computational model of a stealth strategy inspired by the apparent mating tactics of male hoverflies is presented. The stealth strategy (motion camouflage) paradoxically allows a predator to approach a moving prey in such a way that it appears to be a stationary object. In the model, the predators are controlled by neural sensorimotor systems that base their decisions on realistic levels of input information. They are shown to be able to employ motion camouflage to approach prey that move along both real hoverfly flight paths and artificially generated flight paths. The camouflaged approache

Biophysics of the cochlea: linear approximation.

Several deficiencies affecting previous "box" models of the cochlea are overcome in this paper. Both mechanical and hydrodynamical aspects are treated at a level adequate to the complexity of realistic cochlear structures. The dynamics of the cochlea as a passive physical system, in the linear approximation, is described by an integral equation. It is further shown that this equation describes the properties of the working cochlea, provided a force term that accounts for hair cell motility is included. Numerical solutions for different degrees of outer hair cells activity, obtained by matrix m

CT evaluation of coverage and congruency of the hip prior to osteotomy.

A computer-assisted model has been developed to improve the results of surgical techniques for reconstruction of hip dysplasia. This method assesses the coverage and congruency of the femoral head by evaluating multiple factors that may influence surgical planning. It achieves a more reliable image because the measurements are based on a three-dimensional representation, and attention is focused on the cartilaginous coverage of the femoral head. A method to simulate the operative correction helps the surgeon in planning osteotomies of the femur and pelvis. This technique clearly establishes bo

Compound action potential and single acoustic nerve fibres activity generation: an equivalent neuron approach.

The peripheral acoustic system can be easily subdivided into three distinct subsystems: a mechanical one (middle ear and basilar membrane), the set of mechanical-to-neural transducers (the inner and outer hair cells) and the neural part. The functional behaviour of the external and middle ear is well known, but the processes concerned with the transduction of mechanical motion into neural activity are still under investigation. Actually the mechanism of transmission of information concerning acoustic stimuli to the central nervous pathways is not clearly known. On the basis of physiological kn

Cortical tension links curvature to tissue growth in the cellular Potts model.

The growth of biological tissue is sensitive to the physical properties of the environment. For example, the growth rate of contractile tissue under geometric confinement is proportional to local curvature as a result of tissue surface tension. It is not known how local cell behavior is coordinated to give rise to such behavior. Here we use computer simulations based on the cellular Potts model to investigate the role of cell adhesion and cortical contractility for curvature-dependent tissue growth. Our results show that tension-dependent local proliferation leads to the observed macroscopic c

Design and validation of a minibeam treatment delivery system for use with a radiation therapy research platform.

Minibeam radiation therapy (MBRT) is a promising treatment technique in the early stages of clinical use. In this work, two hexagonal minibeam collimators were designed, fabricated, and validated using film measurements and Monte Carlo simulations. Film and Monte Carlo results demonstrated strong agreement, with the greatest agreement near the beam central axis. One of the previously validated MBRT collimators was then modified to allow for dose calculations on a mouse cone beam computed tomography (CBCT) dataset, and a MBRT dose distribution was preserved in the animal dataset. This validated

Midtarsal joint stiffness alters the metabolic cost of simulated running via mechanisms other than changes in foot energy storage and return.

The foot's arch has been proposed to aid in metabolically efficient running. Computational musculoskeletal simulations of steady state rearfoot and non-rearfoot strike running across a range of midtarsal joint (i.e. foot arch) stiffnesses indicated that with increasing stiffness the metabolic cost of transport decreased by ∼5% in rearfoot strike running but increased by ∼11% in non-rearfoot strike running. The magnitude of mechanical work performed about the midtarsal joint as its stiffness increased followed a similar decreasing pattern in both running foot strike conditions, suggesting that
