Utility of Saliva for Detecting Influenza A Virus by Cost-Effective Extraction-Free SYBR Green-Based PCR.

Respiratory specimens collected via nasopharyngeal and throat swabs are the recommended method of choice for the molecular detection of the Influenza A virus. However, they often cause discomfort to patients and require trained healthcare workers. The aim of this study is to validate a cost-effective nucleic acid extraction-free PCR method using SYBR Green chemistry with saliva samples to diagnose respiratory illnesses caused by the Influenza A virus. For this, we enrolled symptomatic pediatric and adult patients with influenza-like illness. A SYBR Green-based nucleic acid amplification test w

Novel Airway Challenges in DEGCAGS Syndrome: Managing Infant Laryngeal Hamartomas.

BACKGROUND DEGCAGS syndrome is an exceptionally rare genetic disorder caused by mutations in the ZNF699 gene. It presents with a broad spectrum of clinical features, including neurodevelopmental delays and dysfunction or anomalies affecting the gastrointestinal, cardiovascular, genitourinary, and skeletal systems. Although these features have been described in the literature, reports of airway complications remain exceedingly rare. This report describes the case of a 1-year-old infant with a confirmed diagnosis of DEGCAGS syndrome who presented with progressive stridor and respiratory distress

Bridging basic science and applied diagnostics: Comprehensive viral diagnostics enabled by graphene-based electronic biosensor technology advancements.

This study presents a graphene field-effect transistor (gFET) biosensor with dual detection capabilities for SARS-CoV-2: one RNA detection assay to confirm viral positivity and the other for nucleocapsid (N-)protein detection as a proxy for infectiousness of the patient. This technology can be rapidly adapted to emerging infectious diseases, making an essential tool to contain future pandemics. To detect viral RNA, the highly conserved E-gene of the virus was targeted, allowing for the determination of SARS-CoV-2 presence or absence using nasopharyngeal swab samples. For N-protein detection, s

Comparison of DNA extraction methods for COVID-19 host genetics studies.

The coronavirus disease 2019 (COVID-19) pandemic has resulted in global shortages in supplies for diagnostic tests, especially in the developing world. Risk factors for COVID-19 severity include pre-existing comorbidities, older age and male sex, but other variables are likely play a role in disease outcome. There is indeed increasing evidence that supports the role of host genetics in the predisposition to COVID-19 outcomes. The identification of genetic factors associated with the course of SARS-CoV-2 infections relies on DNA extraction methods. This study compared three DNA extraction metho

An international, interlaboratory ring trial confirms the feasibility of an extraction-less "direct" RT-qPCR method for reliable detection of SARS-CoV-2 RNA in clinical samples.

Reverse transcription-quantitative polymerase chain reaction (RT-qPCR) is used worldwide to test and trace the spread of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). "Extraction-less" or "direct" real time-reverse transcription polymerase chain reaction (RT-PCR) is a transparent and accessible qualitative method for SARS-CoV-2 detection from nasopharyngeal or oral pharyngeal samples with the potential to generate actionable data more quickly, at a lower cost, and with fewer experimental resources than full RT-qPCR. This study engaged 10 global testing sites, including laborato
