Should Metastatic Site Inform First-Line Therapy in Clear Cell Renal Cell Carcinoma?

Frontline systemic therapy for metastatic clear cell renal cell carcinoma (RCC) has evolved to include doublet immunotherapy (IO/IO) and immunotherapy-tyrosine kinase inhibitor combinations (IO/TKI), with selection typically guided by histology and risk stratification. However, optimal regimen selection remains undefined owing to limited head-to-head data. Subanalyses of landmark clinical trials and emerging real-world multicenter study data suggest differential treatment benefit derived by metastatic site, particularly for bone metastases. Evidence suggests that bone metastases promote an imm

[Trastuzumab-deruxtecan (T-DXd) in HER2+ metastatic breast cancer: efficacy, clinical management, and perspectives].

HER2-positive breast cancer accounts for approximately 15-20% of all breast tumors and is characterized by a more aggressive biology compared with other subtypes. However, the introduction of anti-HER2 agents has made this disease increasingly manageable, even in advanced stages. Among the most recent therapeutic advances, trastuzumab deruxtecan (T-DXd) has emerged as a new standard of care, thanks to its remarkable efficacy, ability to induce deep and durable responses, and significant activity in challenging sites such as the central nervous system and bone. Clinicians play a crucial role in

Advances in the Management of Mediator-Related Symptoms in Non-Advanced Systemic Mastocytosis.

Systemic mastocytosis (SM) is a rarely occurring clonal mast-cell disorder defined by aberrant mast-cell accumulation and episodic or chronic mediator release, giving rise to a broad range of manifestations from pruritus and flushing to gastrointestinal symptoms and anaphylaxis. In non-advanced SM, mediator-related symptoms are the major source of morbidity and substantially impair quality of life. Traditional symptom-directed therapies-including antihistamines, leukotriene modifiers, and mast-cell stabilizers-remain the foundation of care, but a subset of patients experience persistent, refra

Aryl hydrocarbon receptor signaling in osteosarcopenia: an integrative framework for musculoskeletal aging.

Aging is accompanied by a progressive decline in tissue integrity and functional capacity and is a major risk factor for chronic disease. Osteosarcopenia, defined as the concurrent loss of bone and skeletal muscle mass and function, is a major contributor to frailty, fractures, disability, and reduced quality of life in older adults. While musculoskeletal aging has traditionally been attributed to endocrine and mechanical factors, growing evidence highlights an important role for environmental, microbial, and dietary signals in influencing tissue homeostasis across the lifespan. The aryl hydro

Establishment of an Immortalized Canine Hippocampal Neural Stem Cell Line via SV40LT Retroviral Transduction.

Dogs represent a promising animal model for analyzing human neurodegenerative diseases, owing to their similarities to humans in nervous system architecture and behavioral phenotypes. Neural stem cells (NSCs) serve as a highly valuable in vitro experimental model for investigating neurogenesis, neurodegenerative disease pathogenesis, and neural molecular biology; however, studies on immortalized canine neural stem cell lines remain scarce. Herein, we successfully established an immortalized canine hippocampal neural stem cell line that can be continuously passaged in vitro via SV40 large T ant

Macrophage Extracellular Traps in the Oral Mucosa: Autoimmune Disease and Platelet-Derived Epithelial Modulation.

Extracellular traps (ETs) are immune-derived chromatin networks initially described as antimicrobial barriers but increasingly recognized as modulators of tissue homeostasis and autoimmunity. The oral mucosa, constantly exposed to inflammatory stimuli, is particularly sensitive to ET-mediated remodeling (extracellular traps-mediated remodeling). In this study, we investigated how platelet-rich plasma (PRP), platelet-poor plasma (PPP), and washed platelets (WPT), widely used in regenerative medicine, influence ETosis in monocytes and macrophages, and how these ETs modulate the responses of prim

Gross anatomy of the skeleton of neonates of the Orinoco Matamata turtle (Chelus orinocensis).

The Orinoco Matamata turtle (Chelus orinocensis) was described in 2020, representing the second evolutionary lineage within the genus Chelus. The anatomy of this species has not been described, and such information is essential for understanding its biology, ecology, and for its potential relevance in clinical diagnosis. This research provides the first description of the macroscopic morphology of the skeletal system in neonates of the Orinoco Matamata turtle. A group of confiscated turtles was donated from the Alexander von Humboldt institute to our university for academic purposes. From this

Brown Adipose Tissue: Molecular Mechanisms of Regulation, Physiological Functions, and Therapeutic Targets.

Brown adipose tissue (BAT) biology has undergone a profound evolution over the past two decades, expanding from a narrow focus on thermogenic capacity in rodents to the establishment of its pleiotropic roles in human metabolic regulation. While this transition has enabled major progress in defining underlying molecular mechanisms and strengthened the clinical rationale for targeting BAT, it has also exposed unresolved challenges and knowledge gaps. In this review, we synthesize current evidence across the field, integrating findings from molecular, physiological, and translational studies to d

Understanding Tryptase Physiology, Clinical Significance, and Diagnostic Applications: Where Do We Stand?

Tryptase is the principal biomarker of mast cell (MC) burden and activation, routinely used in the diagnosis and management of MC-related disorders. Beyond its established role in confirming anaphylaxis, serum tryptase has gained increasing relevance in the diagnosis of systemic mastocytosis, hereditary α-tryptasemia (HαT), cutaneous mastocytosis, and selected haematologic diseases. Recent advances in MC biology, tryptase genetics, and disease classification have substantially improved the interpretation of baseline and acute (event-related) tryptase measurements. In particular, validation of

In vitro induction of human germinal center B cells.

During antibody responses, germinal centers (GCs) act as a key site for somatic hypermutation (SHM) and affinity maturation of B cells. However, their lymphoid tissue localization has made experimental manipulation of human germinal center B (GCB) cells difficult. Here, we describe a simple in vitro method, modeling T cell-derived signaling, that converts human naive B cells into BCL6+ induced GCB-like (iGCB) cells. Through side-by-side proteomic and transcriptional single-cell analysis of iGCB and human tonsil GCB cells, we show that iGCB cells recapitulate the surface marker, transcription f

Levodopa in retinal disease: Dopamine pathways, neuroprotective mechanisms, and clinical evidence.

We review the role of levodopa (L-DOPA), a dopamine precursor used to treat Parkinson disease, as a candidate for retinal neuroprotection, focusing on retinal dopamine biology, mechanisms of action, and evidence from preclinical and clinical studies. Selected experimental and clinical studies indicate that L-DOPA and related dopaminergic interventions influence multiple retinal pathways relevant to degeneration. L-DOPA, synthesized in the pigmented retinal pigment epithelium, activates GPR143 signaling and promotes the release of neuroprotective factors. In animal studies, L-DOPA supplementati

Phosphatidylinositol-4-phosphate signaling regulates dense granule biogenesis and maturation in Toxoplasma gondii.

Phosphoinositide metabolism defines the foundation of a major signaling pathway that is conserved throughout the eukaryotic kingdom. Although the 4-OH phosphorylated phosphoinositides phosphatidylinositol-4-phosphate (PtdIns4P) and phosphatidylinositol-4,5-bisphosphate are each individually required for the viability of all eukaryotic cells studied thus far, their activities in parasite biology are less well understood. Using intracellular tachyzoites of the apicomplexan parasite Toxoplasma gondii as model for studying PtdIns4P signaling in a protozoan, we demonstrate the presence of PtdIns4P

The microbiota-gut-brain axis in Huntington's disease: Evidence, mechanisms and therapeutic opportunities.

Huntington's disease (HD) is increasingly recognized as a multisystem disorder in which perturbations of the gut microbiota may influence peripheral homeostasis and shape central neurodegeneration. Across human cohorts, HD is marked by reproducible β-diversity shifts, loss of butyrate-producing taxa, and disruptions in bile-acid and tryptophan-derived metabolites, indicating broad remodeling of microbiota-host metabolic interfaces. Integrating metabolomic evidence with mechanistic data from HD models, we delineate three convergent axes through which dysbiosis may modify disease biology: (i) sh

Recent advances in omics and genetic tools in schistosome research and control.

Schistosomiasis is a neglected tropical disease posing a persistent threat to global health, particularly in endemic areas. Mass drug administration with praziquantel has substantially reduced disease burden, yet its limited efficacy against juvenile worms, reinfection, and emerging resistance underscore the urgent need for innovative strategies. Recent advances in omics technologies including genomics, bulk and single-cell transcriptomics, non-coding RNA omics, and proteomics, together with functional genetic tools such as RNA interference, transgenesis and CRISPR/Cas9 genome editing, have pr

Smoking accelerates immunosenescence in multiple sclerosis.

Multiple sclerosis (MS) is a chronic immune-mediated disease of the central nervous system characterized by inflammation, neuroaxonal injury, and progressive neurologic disability. Although its etiology is multifactorial, cigarette smoking is among the most consistent modifiable environmental exposures associated with increased MS risk and worse clinical outcomes. Smoking has been linked to greater inflammatory activity, accelerated disability accumulation, and earlier progression, suggesting effects across multiple stages of disease biology. In this review, we examine the hypothesis that smok

CRISPRi-based screen of autism spectrum disorder risk genes in microglia uncovers roles of ADNP in microglia endocytosis and synaptic pruning.

Autism Spectrum Disorders (ASD) are a set of neurodevelopmental disorders with complex biology. The identification of ASD risk genes from exome-wide association studies and de novo variation analyses has enabled mechanistic investigations into how ASD-risk genes alter development. Most functional genomics studies have focused on the role of these genes in neurons and neural progenitor cells. However, roles for ASD risk genes in other cell types are largely uncharacterized. There is evidence from postmortem tissue that microglia, the resident immune cells of the brain, appear activated in ASD.

Crosstalk between the microbiota and intestinal dendritic cells in IBD.

Conventional dendritic cells (cDCs) play a pivotal role in orchestrating the delicate balance between immunity and tolerance within the gastrointestinal tract by interacting with other cell types, particularly T cells. Meanwhile, the microbiota is critical for the induction and modulation of the immune system in the gut and plays a key role in the function of cDCs. So far, the study of intestinal cDCs has been encumbered by their limited numbers and phenotypic overlap with other myeloid cells. Recent advancements in single-cell sequencing technology have helped define cDCs and their subsets, w

Ubiquitinome profiling identifies USP13-CMAS axis as critical regulator of hair follicular melanogenesis via K48-linked polyubiquitination.

The ubiquitin-proteasome system critically regulates melanogenesis through post-translational modifications. However, the specific deubiquitination substrates involved in this regulation remain poorly characterized. This study employed multi-omics integration and functional validation to decipher the role of USP13 in melanocyte (MC) biology. Through quantitative melanin assays and apoptosis/proliferation analyses, we established the dual functionality of USP13 in promoting MC pigmentation and survival. Systematic proteome-ubiquitinome analysis of USP13-overexpressing MCs identified 23 high-con

Small extracellular vesicles: connecting early life exposure outcomes to air pollution during pregnancy to early childhood health.

Exposure to air pollution poses a serious threat to maternal and child health, particularly during critical developmental periods. Extracellular vesicles (EVs), as key mediators of intercellular communication, have emerged as a novel mechanism through which environmental exposures, including air pollutants, exert systemic effects. This review synthesizes current evidence on the role of small EVs (sEVs) in mediating the biological impacts of prenatal air pollution exposure, with a focus on their molecular profile, including the presence of signaling molecules like miRNAs and proteins, and their
