Bilateral laparoscopic adrenalectomy for congenital adrenal hyperplasia with severe hypertension, resulting from two novel mutations in splice donor sites of CYP11B1.

We present an in vivo and in vitro study of congenital adrenal hyperplasia in a patient with 11beta-hydroxylase deficiency. Sequencing of the CYP11B1 gene showed two new base substitutions, a conservative 954 G-->C transversion at the last base of exon 5 (T318T), and a IVS8 + 4A-->G transition in intron 8. In addition, two polymorphisms were found in exons 1 and 2. The genetically female patient was raised as a male because of severe pseudohermaphroditism. Glucocorticoid-suppressive treatment encountered difficulties in equilibration and compliance, resulting in uncontrolled hypertension with

Small-for-dates babies at the age of four years: health, handicap and developmental status.

221 small-for-dates (SFD) and 244 average-for-dates (AFD) children were personally examined at birth, and seen thereafter at regular intervals up to 4 years, when a thorough assessment was made. There was an excess of SFD children with major congenital abnormalities; but no difference between the groups in the number of children who were handicapped without congenital abnormality. Their general health was good, and the prevalence of specific disease or major ill-health did not differ. SFD girls seemed to have less acute hearing than AFD girls; but there was not difference for boys. The prevale

[The role of transcranial electrostimulation in the treatment of psychosomatic disorders in industrial workers].

BACKGROUND: The prevalence of psychosomatic disorders (PD) is steadily increasing from year to year. Suffice it to mention that the appealability for the primary health care during the recent period increased from 20% to 40-49%. Various psychological, pharmacological and physical methods for the treatment of somatoform disorders have been developed. However, the physiotherapeutic methods for the management of psychosomatic disorders remain virtually unexplored. At the same time, the stress-limiting effects of transcranial electrical stimulation (TES) have been convincingly demonstrated. AIM:

Neonatal testosterone exposure modulates exploration and object avoidance without exacerbating autism-like behavior in Shank3b-deficient mice.

A rapid increase in autism spectrum disorder (ASD) prevalence, high heritability, and higher incidence in boys suggest a role of gene-environment interactions, likely involving sex hormones in its etiology. Prenatal exposure to high testosterone concentration has been linked to risk of ASD, however, experimental proof in genetically predisposed animals is lacking. Since neonatal development in mice mirrors late prenatal neurodevelopment in humans, we investigated the effects of neonatal testosterone administration on the behavior of female and male Shank3b(-/-) mice. On postnatal day 1, neonat

Sex-Specific Effects of Hypocretin Receptor Signaling in Corticotropin-Releasing Factor Neurons on Alcohol Drinking, Anxiety, and Extended Amygdala Neuronal Excitability.

BACKGROUND: Alcohol use disorder (AUD) is characterized by compulsive alcohol consumption and negative emotional states during withdrawal, often perpetuating a cycle of addiction through arousal dysfunction. The hypocretin/orexin (HCRT) neuropeptide system is a key regulator of arousal that is implicated in these processes, particularly in its interactions with corticotropin-releasing factor (CRF) neurons within the bed nucleus of the stria terminalis (BNST). METHODS: Using CRF-specific genetic deletion of Hcrt r 1 and/or Hcrt r 2 receptors in mice combined with behavioral and electrophysiolo

Plasma leptin levels are lower in females, but not males, with ketamine use disorder.

Background: Ketamine has emerged as a prominent substance of misuse. Leptin, an adipocyte-derived polypeptide hormone, has been implicated in the development of addiction. Sex-specific changes in leptin levels have been demonstrated following acute ketamine administration; the persistence of long-term ketamine use on leptin levels is uncertain. Objectives: To assess the sex-difference of leptin levels, and their persistence, in individuals with ketamine use disorder (KUD) compared to healthy controls. Methods: Plasma leptin levels were measured in 62 healthy controls (37 males, 25 females) and 6

Sex-dependent differences in vulnerability to early risk factors for posttraumatic stress disorder: results from the AURORA study.

BACKGROUND: Knowledge of sex differences in risk factors for posttraumatic stress disorder (PTSD) can contribute to the development of refined preventive interventions. Therefore, the aim of this study was to examine if women and men differ in their vulnerability to risk factors for PTSD. METHODS: As part of the longitudinal AURORA study, 2924 patients seeking emergency department (ED) treatment in the acute aftermath of trauma provided self-report assessments of pre- peri- and post-traumatic risk factors, as well as 3-month PTSD severity. We systematically examined sex-dependent effects of 1

Sex differences in recent first-onset depression in an epidemiological sample of adolescents.

Prior studies provide limited and contradictory evidence regarding sex differences in the incidence of depression during adolescence, a critical period for development of the disorder. Data from six consecutive years (2009-2014) of a national survey of US adolescents aged 12-17 (N=101 685) are used to characterize sex differences in the incidence of depression by age and to compare recent first-onset and persistent depression with respect to impairment, suicide attempts, conduct problems and academic functioning. Projecting from age-specific incidence proportions, the cumulative incidence of d

Factors Influencing the Phenotypic Expression of Hypertrophic Cardiomyopathy in Genetic Carriers.

INTRODUCTION AND OBJECTIVES: Hypertrophic cardiomyopathy (HCM) is a disorder with variable expression. It is mainly caused by mutations in sarcomeric genes but the phenotype could be modulated by other factors. The aim of this study was to determine whether factors such as sex, systemic hypertension, or physical activity are modifiers of disease severity and to establish their role in age-related penetrance of HCM. METHODS: We evaluated 272 individuals (mean age 49 ± 17 years, 57% males) from 72 families with causative mutations. The relationship between sex, hypertension, physical activity,

Integrated lipidome and miRNome analyses reveal sex-based differences in circulating extracellular vesicles of alcohol use disorder patients.

Integrated multi-omics and extracellular vesicle (EV) analysis are emerging as powerful, complementary strategies for biomarker discovery. These approaches offer promising tools to enhance early detection, diagnosis, and treatment of alcohol use disorder (AUD). Here we applied an integrated miRNomic and lipidomic approach to analyze plasma EVs from AUD patients and controls of both sexes to gain a comprehensive understanding of the underlying molecular mechanisms. We identified an AUD signature with predictive potential for diagnostic applications, representing an initial step in this directio

Sex differences in prevalence of subclinical hypothyroidism and its clinical correlates in overweight or obese, first-episode drug-naïve Chinese patients with major depressive disorder.

BACKGROUND: Sex differences in both major depressive disorder (MDD) and subclinical hypothyroidism (SCH) are established, but their interplay in overweight/obese, first-episode drug-naïve (FEDN) MDD patients with study-defined higher-TSH subgroup (TSH > 8 mIU/L with normal FT4) is unclear. We investigated sex-specific prevalence and clinical correlates of this elevated TSH phenotype in patients with FEDN-MDD. METHODS: Among 1026 FEDN-MDD outpatients with BMI ≥ 24 kg/m2 (348 males; 678 females), depression, anxiety, and psychotic symptoms were assessed using the HAMD-17, HAMA, PANSS-Positive s

Exploring the role of Cav3.2 calcium channels in autism-like cognitive behavior induced by prenatal valproic acid exposure.

Recent findings indicated that CACNA1H mutations may contribute to Autism Spectrum Disorder (ASD) by reducing Cav3.2 activity, disrupting neuronal function, and brain development. To explore how Cav3.2 deficiency affects autism-related cognition, we induced autism-like behaviors in wild-type (WT) and Cav3.2 knockout mice (KO) using the prenatal valproic acid model (pre-VPA). We analyzed how cognitive behavior (repetitive behavior, spatial working memory, sociability, social preference, and anxiety) in this model is differentially impacted in WT and Cav3.2 KO mice of different sexes and ages. Single prolonged stress in mice: interactions between alcohol drinking, negative affect, and fear learning.

RATIONALE & OBJECTIVE: Exposure to traumatic stressors can have detrimental effects on one's well-being. Alcohol is often used as a coping mechanism to alleviate stress, leading to the development of alcohol use disorder (AUD). Furthermore, sex dimorphisms in stress and AUD alter their progression and sustenance. To investigate these interactions for effective treatment strategies, validated and exhaustive preclinical models are necessary. Here, we designed a comprehensive study that examines how traumatic stress influences ethanol drinking patterns and PTSD-like phenotypes in male and female

An FSCV Study on the Effects of Targeted Typical and Atypical DAT Inhibition on Dopamine Dynamics in the Nucleus Accumbens Shell of Male and Female Mice.

Understanding the neurochemistry underlying sex differences in psychostimulant use disorders (PSUD) is essential for developing related therapeutics. Many psychostimulants, like cocaine, inhibit the dopamine transporter (DAT), which is largely thought to account for actions related to their misuse and dependence. Cocaine-like, typical DAT inhibitors preferentially bind DAT in an outward-facing conformation, while atypical DAT inhibitors, like modafinil, prefer a more inward-facing DAT conformation. Modafinil and R-modafinil have emerged as potential therapeutic options for selected populations
