Aptamer-Engineered Liposomal Platform Enables in Situ cDC1 Vaccination to Potentiate Immunotherapy in Prostate Cancer.

Prostate cancer (PCa) is a prototypical "cold tumor" with low immunogenicity and a highly immunosuppressive tumor microenvironment, which severely limits the efficacy of immunotherapy. Notably, the number and functionality of conventional type 1 dendritic cells (cDC1), which are critical antigen-presenting cells, are markedly reduced in PCa, thereby impairing T cell priming. Here, we developed an aptamer-modified liposomal platform (Apt-Flt3L@Lipo) for the targeted co-delivery of Flt3 ligand (Flt3L) and chlorin e6 (Ce6) to elicit in situ cDC1 vaccination. Upon ultrasound activation, Ce6 induce

Methionine-depleting engineered probiotics promote PD-L1 antibody immunotherapy by activating the STING pathway.

The metabolic differences between tumor cells and normal tissue cells offer potential targets for therapeutic intervention. For instance, compared to normal tissues, tumor cells exhibit a significantly higher dependency on methionine. However, current strategies for methionine restriction have limited clinical applicability due to systemic toxicity, poor patient compliance, and insufficient tumor targeting. In this study, we developed an engineered probiotic, Met-EcN, which is able to achieve targeted consumption of methionine within the tumor microenvironment. In the B16-F10 melanoma mouse mo

Spatiotemporally engineered tumor-derived extracellular vesicle-based scaffold vaccine for personalized cancer immunotherapy.

Personalized vaccines demonstrate remarkable potential in leveraging tumor-specific adaptive immunity for cancer therapy. Nevertheless, current platforms face persistent challenges, including premature systemic clearance and imprecise antigen-presenting cell targeting, culminating in transient and inefficient antitumor immunity. Furthermore, the technical complexity, extended timelines, and prohibitive costs required for tumor-specific neoantigen identification continue to impede the clinical translation of personalized cancer vaccines. Here, we report a tumor-derived extracellular vesicle-bas

Precision-engineered Carrageenan Gels: Boosting the Efficacy, Selectivity, and Release of Celecoxib for Lung Cancer Therapy.

BACKGROUND: Lung cancer is one of the most widespread malignancies among all types of cancers. There is uncertainty in its treatment because of the selectivity. The investigation is aimed to enhance therapeutic efficacy through targeted improvements in drug selectivity and reduced toxicity by analyzing well-accepted cyclooxygenase (COX)-2, which is an enzyme target and a known therapeutic target for anti-inflammatory and antitumor agents. OBJECTIVE: The objective of the present research was to identify the most suitable counterpart for celecoxib, which would produce synergistic effects and im

Biomimetic catalase-engineered black phosphorus nanosheets enhance photo-/radio-therapy for hepatocellular carcinoma.

The hypoxic tumor microenvironment (TME) critically impairs the therapeutic efficacy of photo- and radiotherapy (RT) in hepatocellular carcinoma (HCC). Ameliorating tumor hypoxia thus presents a promising strategy to potentiate these treatment modalities. Herein, we developed a biomimetic nanoplatform by functionalizing black phosphorus nanosheets (BP) with catalase (CAT), forming a stable nanocomposite (BPCAT) via covalent bonding between carboxyl and hydroxyl groups. To enhance tumor-targeting capability and immune modulation, BPCATwas further camouflaged with macrophage membranes engineered

Sterically Engineered Mo─V Cluster Catalyst for Switchable Z/E-Selective Alkyne Hydrophosphorylation.

Stereodivergent alkyne hydrophosphorylation represents a formidable challenge in synthetic chemistry, particularly for selective and tunable synthesis of (Z)- and (E)-alkenylphosphine oxides. Herein, we report a steric hindrance-mediated strategy that achieves switchable Z/E selectivity via rational design of a metal-organic-functionalized phosphovanadomolybdate [PMo8V4O40(VO)2][(VO)(IM)4]3·5.5H2O (IM = 1-methylimidazole), which features a highly congested steric environment. Steric repulsion between the cluster framework and substrate enables highly Z-selective hydrophosphorylation, affording

Engineered ATP-loaded extracellular vesicles: a dual-functional strategy for improving myocardial infarction therapy.

Myocardial infarction (MI) represents a major component of cardiovascular disease, primarily due to severe energy depletion in ischemic tissue. Extracellular vesicles (EVs) have recently emerged as promising cell-free nanocarriers capable of targeted delivery and intercellular communication. Leveraging these advantages, engineered EVs were investigated in this study as a direct ATP-delivery platform to cardiomyocytes. EVs were functionalized with an anti-myosin antibody to form targeted extracellular vesicles (T-EVs) and subsequently loaded with ATP, generating T-ATP-EVs for selective energy t

Differential Modulation of Colonic Inflammaging by Limonin Loaded in Pectin Emulsion Gels with Engineered Fine Structures.

The emulsion gel (EG) system offers an effective strategy to improve limonin bioavailability, yet the differential intervention targets of limonin-loaded EGs constructed from pectins with distinct fine structures in colonic inflammaging remain unclear. In this study, three structurally distinct pectin-based EGs were engineered by modulating the degree of esterification and rhamnogalacturonan-I side-chain ratio. In d-galactose-induced aged mice, these EGs exhibited superior anti-inflammatory effects against colonic inflammaging compared to free limonin with distinct functional preferences. LMLR

Selective C18-Methyl Oxidation of Sclareol via an Engineered P450 Enables the Synthesis of Spiromyrrhene B.

While selective oxidation of unactivated C-H bonds has emerged as an effective tactic to reshape retrosynthetic strategies in the synthesis of natural products, the primary methyl C-H bonds remain inert and inaccessible to contemporary methods without preinstalled directing groups. Herein, we report a biocatalytic C-H oxidation strategy that leverages an engineered P450BM3 platform for the selective C18-methyl hydroxylation of sclareol. Through five rounds of directed evolution, variant Ox18M3-3 was developed, elevating the regioselectivity from 4% to >99% alongside a 2.2-fold increase in cata

Construction of an engineered Escherichia coli strain with enhanced intestinal colonization and anti-inflammatory efficacy in colitis.

Engineered probiotics are considered effective and safe therapeutic strategies for the treatment of various diseases. Escherichia coli Nissle 1917 (EcN) has been widely used as a chassis strain because of its safety and well-established genetic manipulation system. However, the limited intestinal colonization ability of EcN limits its potential as a chassis for the construction of synthetic probiotics. Here, an engineered EcN strain (EcN-CPM) with enhanced gastric acid and bile salt tolerance and improved intestinal adhesion was constructed. Oral administration of EcN-CPM to mice with colitis

Allogeneic CRISPR-Engineered CAR-T Cells Drive Potent Antitumor Activity in Solid Tumors.

Chimeric antigen receptor (CAR) T-cell therapy has shown limited efficacy in solid tumors, in part due to variability in autologous T cells derived from heavily pretreated patients with advanced disease. To address these constraints, we developed an off-the-shelf allogeneic CAR-T platform using CRISPR-Cas9-mediated genome editing in T cells from healthy donors to enable targeted CAR insertion at the TRAC locus with concurrent disruption of B2M. Using adeno-associated virus (AAV) delivery, we designed CAR-T cells targeting glypican-2 (GPC2) and glypican-3 (GPC3), emerging antigens expressed in

IL-12-engineered human PSMA-CAR T cells for the treatment of advanced prostate cancer.

Adoptive cell therapies used to treat advanced prostate cancer are being developed to target several tumor-associated antigens, including prostate-specific membrane antigen (PSMA). Chimeric antigen receptor (CAR) T cell therapy using the single-chain variable fragment (scFv) derived from the humanized murine mAb clone J591 as the antigen-binding domain has shown promising anti-tumor activity. However, it has also been associated with macrophage activation syndrome and other unwanted toxicities, highlighting the need for more specific and human-derived antigen-binders with optimized construct d

In situ engineered silicon-magnesium implants orchestrate sequential immunomodulation, angiogenesis, and osteogenesis for bone repair.

Magnesium (Mg) is a promising candidate for next-generation bone implants due to its favorable mechanical properties and biodegradability. However, its rapid corrosion causes local alkalization, hydrogen release, and inflammation, severely limiting clinical translation. Herein, we developed a multifunctional Mg-based implant, denoted as Mg/Mg2SiO4/PDA (MSP), by constructing an in situ Mg2SiO4 interlayer on the Mg substrate through a one-pot hydrothermal process, followed by polydopamine (PDA) functionalization. This multilayered design orchestrates sequential bone regeneration: early antiinfec
