Proteomic comparison between different tissue preservation methods for identification of promising biomarkers of urothelial bladder cancer.

Samples in biobanks are generally preserved by formalin-fixation and paraffin-embedding (FFPE) and/or optimal cutting temperature compound (OCT)-embedding and subsequently frozen. Mass spectrometry (MS)-based analysis of these samples is now available via developed protocols, however, the differences in results with respect to preservation methods needs further investigation. Here we use bladder urothelial carcinoma tissue of two different tumor stages (Ta/T1-non-muscle invasive bladder cancer (NMIBC), and T2/T3-muscle invasive bladder cancer (MIBC)) which, upon sampling, were divided and pres

External cervical resorption: part 2 - management.

Effective management of external cervical resorption (ECR) depends on accurate assessment of the true nature and accessibility of ECR; this has been discussed in part 1 of this 2 part article. This aim of this article was firstly, to review the literature in relation to the management of ECR and secondly, based on the available evidence, describe different strategies for the management of ECR. In cases where ECR is supracrestal, superficial and with limited circumferential spread, a surgical repair without root canal treatment is the preferred approach. With more extensive ECR lesions, vital p

Fetal sigmoid colon mesentery - In relevance in fetal ultrasound application. A pilot study.

INTRODUCTION: Ultrasound examinations during pregnancy are routine procedures used to detect fetal congenital malformations. Ultrasound monitoring of sigmoid colon mesenterial development could be useful for early detection of subjects at risk of sigmoid colon volvulus. OBJECTIVE: The aim of our paper was to assess the sigmoid colon length, and sigmoid colon mesentery width and height in the late fetal period, and, using the results, to estimate the surface area of the mesocolon (in mm2) in living fetuses. Moreover, we attempted to repeat some of these measurements in living fetuses using ult

Keratocystic Odontogenic Tumour with Extraosseal Spread: Evaluation of the Effect Carnoy's Solution.

Keratocystic odontogenic tumour is relatively rare benign tumour. It is characterized by its fast aggressive growth and high risk of recurrence. Treatment is always surgical: conservative (enucleation, marsupialization) or aggressive (enucleation followed by application of Carnoy's solution, cryotherapy; peripheral ostectomy or en block resection of the jaw). Authors analysed retrospectively 22 patients who fulfilled inclusion criteria, i.e. had odontogenic keratocystic tumour of mandible, wherein antero-posterior dimension was at least 30 mm, and the tumour penetrated into the surrounding sof

Elution of High-affinity (>10-9 KD) Antibodies from Tissue Sections: Clues to the Molecular Mechanism and Use in Sequential Immunostaining.

Inconsistent results obtained with published methods for the elution of antibodies from tissue sections prompted the assessment of both old and new methods in combination with monoclonal rabbit antibodies of known, increased affinity (above 1×10(-9) KD). We tested an acidic (pH 2) glycine buffer, a 6 M urea hot buffer and a 2-Mercaptoethanol, SDS buffer (2-ME/SDS). Some antibodies were not removed by the glycine pH 2 or 6 M urea hot buffers, indicating that antibodies survive much harsher conditions than previously believed. We found that the elution is dependent upon the antibody affinity and

Treatment planning challenges in the maxillary anterior region consequent to severe loss of buccal bone.

UNLABELLED: The burgeoning popularity of dental implant usage in the maxillary anterior region is well recognized. On the other hand, challenges involved in achieving optimal esthetics through implants have also been discussed extensively. Presence of labial/buccal bone in the maxillary anterior region is necessary for esthetic success of treatment, with or without the use of dental implants. This case report describes the multidisciplinary management of a rare case with severe loss of buccal bone confined to the maxillary central incisor region. A patient with a recent history of orthodontic

Provisional use of a natural tooth crown following failure of replantation: a case report.

Despite a wide range of treatment options available, traumatized teeth may be inevitably lost on certain occasions. This case report presents an alternative treatment option for the premature loss of a traumatized maxillary central incisor because of extensive root resorption and mobility. A fixed appliance-type provisional prosthesis was fabricated by using the patient's natural clinical crown from the extracted tooth. This measure proved to be a very adequate, esthetic treatment solution before a permanent restorative plan could be developed in the future.

Development of passive haemagglutination (PHA) and haemagglutination inhibition (HAI) technique for potency estimation of Cobra Antisnake Venom Serum (ASVS).

Antibodies against snake venom or antivenom potency are assayed quantitatively by in-vivo neutralization test in mice, which requires large number of laboratory animals. In potency assays of biological substances such as antivenoms, it is highly desirable to avoid suffering and death of animals by substituting in-vivo method with in-vitro methods, provided such methods measure life-saving capability with precision similar to that of in-vivo method. The in-vitro tests determine the neutralizing power of antivenom by permitting the evaluation of a particular biological activity of the venom and

A case of oculo-auriculo-vertebral spectrum with long-term and comprehensive management.

AIM: To show that a patient with severe oculo-auriculo-vertebral spectrum can achieve satisfactory occlusion and facial appearance through long-term and comprehensive management. METHODS: A patient, with oculo-auriculo-vertebral spectrum, had bilateral cleft lip and palate, macrostomia, malformed pinna, and unilateral hypoplasia of the zygomatic arch and mandible, which resulted in an asymmetric facial appearance characteristic of hemifacial microsomia. After bilateral lip repair and palatal closure, surgical repair of the pinna, nose, and cheiloplasty was performed. The patient was seen at t

Cleidocranial dysplasia: modern concepts of treatment and a report of an orthodontic resistant case requiring a restorative solution.

A case is presented of a young boy with cleidocranial dysplasia, whose multiple supernumerary teeth prevented the eruption of most of his permanent teeth. His maxillary incisor teeth failed to erupt following removal of anterior supernumerary elements and orthodontic traction. Lack of abutment teeth and a difficult maxillary base made prosthetic treatment almost impossible. A horseshoe acrylic denture retained by milled crowns bonded to the deciduous canines and a maxillary first molar proved a very successful restoration. The problems of treating this group of patients are discussed.

Immunocytochemical demonstration of mineralocorticoid receptors in rat and human kidney.

Using a polyclonal antiserum against the hinge region of the recently cloned human mineralocorticoid receptor (MR) and indirect peroxidase immunohistochemistry, we have shown MR-like immunoreactivity (LI) in superficial nephron segments, including distal convoluted tubule, connecting piece and initial cortical collecting duct. The absence of staining in cells tentatively identified as intercalated cells on light microscopy was confirmed by pre-embedding electron microscopy. Though the intracellular distribution of immunostaining varied with the fixative used, the cellular distribution of MR-LI

Dermal mast cells in mastocytosis: fixation, distribution and quantitation.

The mast cell distribution and number were studied in skin biopsies of 18 mastocytosis patients and 10 controls. The biopsies were stained for mast cells with toluidine blue at pH 0.5. The number in the upper dermis of lesional abdominal skin was at least twice as high as that of normal adjacent skin. Fixation in iso-osmotic 0.6% formaldehyde and 0.5% acetic acid, revealed more mast cells than conventional 4% formaldehyde fixation. Staining for 5 days, when compared to the normal for 30 min, increased the number of demonstrable mast cells just as did the change in fixation. Conventional formal

Dental visits, teeth remaining, and prosthetic appliances in a Norwegian urban population.

Of a random sample (177 persons) of 35-year-old Oslo citizens, 117 subjects (66.1%) attended a dental examination. Questionnaires were returned by mail from 28 of the non-respondents. The most common reason for not attending was "difficulty in finding time for an appointment." About 89% of the respondents had visited a dentist regularly. Regular dental care seemed to be more common among people with a high level of education than among those with a low level of education. The most common treatment performed at the last dental visit was filling of cavities. The mean number of remaining teeth wa

Handling of multicohort data in pooled analysis of environmental exposures: confounding by cohort of the relationship between perfluoroalkyl substances and birthweight in the ECHO Program.

The relationship between exposure to perfluoroalkyl substances (PFAS) and birthweight remains unclear. Pooling data across multiple cohorts can increase power, leading to more representative populations and exposure distributions, but confounding by cohort can be a major source of bias. To understand this potential bias, we assessed the relationship between exposure to 5 PFAS and birthweight utilizing data from 5480 mother-infant dyads across 17 Environmental influences on Child Health Outcomes (ECHO) Cohort sites. The relationship was assessed in several ways: covariate-adjusted models withou

Children's books about scientists convey demotivating messages.

Biographies of scientists are widely used to spark children's interest and broaden participation in science. In telling the stories of successful scientists, these books convey implicit "recipes for success"-yet no study has systematically examined whether these recipes align with what motivation research recommends. Here, we provide the first large-scale analysis of motivational messages in children's science biographies (422 best-selling books, 1,355 protagonists). Four main findings emerged. First, scientific ability was portrayed equally often as fixed and as malleable, a pattern unlikely

Efficacy of a Standardized Low-Dose Insulin Infusion Protocol in the Emergency Stabilization of Diabetic Dogs.

BACKGROUND: Continuous insulin infusion protocols are essential for managing decompensated diabetic dogs, but comparative data between variable and fixed infusion rates are limited. METHODS: This prospective observational study evaluated the glycemic response of 21 diabetic dogs treated with a fixed-dose continuous-rate infusion (CRI) of regular insulin at 0.05 IU/kg/h for 12 h. Capillary blood glucose was measured hourly. Statistical analyses included Wilcoxon signed-rank tests, Friedman test, Mann-Whitney U, and Kruskal-Wallis tests. RESULTS: A significant reduction in glucose concentratio

Individualized and targeted coagulation management in bleeding trauma patients.

PURPOSE OF REVIEW: This review aims to summarize current evidence on hemostatic management of bleeding trauma patients, with a focus on resuscitation strategies using either coagulation factor concentrates or fixed-ratio transfusion concepts. It discusses the potential benefits and limitations of both approaches. RECENT FINDINGS: Recent studies have shown that coagulopathy caused by massive traumatic hemorrhage often cannot be reversed by empiric treatment. During initial resuscitation, a fixed-ratio transfusion approach uses the allogeneic blood products red blood cells, plasma, and platelet
