(p)ppGpp mediates persister formation in Escherichia coli during glucose to fatty acid shift.

Bacterial persistence contributes to antibiotic failure and recurrent infectious disease, yet the metabolic cues that promote persister formation remain poorly understood. Here we investigated Escherichia coli persistence after nutrient downshifts from glucose to various carbons. Compared to shifts to gluconeogenic carbons (pyruvate, malate, succinate, and fumarate), the glucose-to-fatty acid shift induced exceptionally high persister levels, with cells tolerating ampicillin (56%), carbenicillin (22%), and gentamicin (1%) after 24-h treatment. With an RNA-based biosensor and HPLC quantificatio

Bispecific antibody vesicles: A multifunctional bioactive drug delivery platform for the treatment of Pseudomonas aeruginosa infection.

Pseudomonas aeruginosa (P. aeruginosa) is a major pulmonary pathogen that establishes chronic infection through Psl polysaccharide-mediated adhesion and biofilm formation, while accelerating acute disease via toxin injection through the type III secretion system (T3SS) PcrV protein. Conventional small-molecule antibiotics show poor target specificity, limited biofilm penetration and insufficient toxin neutralization, thereby compromising therapeutic efficacy. Here, we developed a bioactive nano delivery system that integrates a bispecific antibody (BsAb) with a nano-antibiotic for P. aeruginos

Intravesical therapies for catheter-associated urinary tract infections: current approaches and future directions.

SUMMARYUrinary tract infections (UTIs) are among the most common bacterial infections worldwide, with catheter-associated UTIs (CAUTIs) representing a major subset in healthcare settings. CAUTIs significantly increase patient morbidity, mortality, hospital stays, and healthcare costs, while driving antibiotic overuse and antimicrobial resistance (AMR). Systemic antibiotics often fail due to poor biofilm penetration, localized infection sites, and rising multidrug resistance, highlighting the urgent need for alternative, targeted therapies. This review discusses the role of intravesical therapi

Emerging hypervirulent and extensively drug-resistant Klebsiella pneumoniae with potent biofilm formation as a new clinical challenge.

Hypervirulent Klebsiella pneumoniae strains (hvKp) can induce invasive community-acquired infections in healthy individuals. A total of 71 K. pneumoniae isolates were obtained from patients admitted to teaching hospitals in Arak, Iran. The hypermucoviscous phenotype was assessed using the string test. The presumptive hvKp strains were identified by the iucA gene and screened for capsular genes (K1, K2, K5, K20, K54, K57) and virulence factors (rmpA, rmpA2, iroB, peg-344). Antimicrobial susceptibility and colistin broth disk elution were also conducted. The biofilm formation ability was evaluat

Transferable IncX3-blaKPC-2 plasmid and chromosomal blaCTX-M-27 in a commensal Escherichia coli ST7854 isolated from a healthy companion dog.

OBJECTIVES: Carbapenemase-producing Enterobacterales have disseminated globally, largely via highly transmissible plasmids. Their emergence in companion animals is of particular concern, indicating that clinically important carbapenem-resistance determinants have spread beyond healthcare settings. This study characterized a multidrug-resistant Escherichia coli isolate from a healthy dog in South Korea. METHODS: Antimicrobial susceptibility was determined by standardized reference methods. Hybrid whole-genome assembly resolved chromosomal and plasmid architectures, and plasmid transferability

A novel diagnostic technology rapidly detects the presence of infection and defines antimicrobial sensitivities in organisms typical for joint infection.

AIMS: Infective arthritis is rare, but serious. Timely treatment with appropriate antimicrobial therapy is potentially life- and limb-saving, but our ability to deliver this is limited by current laboratory practices, which confer a delay of several days before antimicrobial sensitivities are available. We propose a novel diagnostic modality, the Scattered Light Integrating Collector (SLIC), which monitors bacterial growth in real time, allowing a diagnosis of infection to be made in minutes, and antimicrobial sensitivities to be delivered in under an hour. METHODS: We collated data previousl

Ligilactobacillus agilis LA-V4 Isolated from Vulture Fecal Isolate: A Novel Probiotic Candidate with Broad-Spectrum Antibacterial Activity.

Vultures are extraordinarily adapted to feed on carrion, providing them with a constant microbiologically hostile environment. This peculiar ecological position has influenced the evolution of their gut microbiota, potentially conferring its uncommon antimicrobial traits and resistance to stress. In this study, we report on the isolation and comprehensive characterization of a lactic acid bacterium strain, identified as Ligilactobacillus agilis, from vulture feces via 16S rRNA gene sequencing. This strain exhibited potent antagonistic activity against several clinically relevant bacterial path
