Identifying and reprogramming softness-driven cancer stem-like cells overcomes CAR-T cell resistance in solid tumours.

Solid tumours show substantial mechanical heterogeneity, yet how such cues influence the susceptibility of cancer cells to T cell-based therapies remains unclear. Here we discover that cancer cells grown on soft matrices are less sensitive to chimeric antigen receptor T cell cytotoxicity and exhibit elevated extracellular adenosine triphosphate and sustained calcium activity. To understand the mechanisms underlying this reduced killing, we sought to identify the cells that respond to mechanical softness. We engineered a doxycycline-gated calcium-activated transcriptional mechano-recorder that

The Integrative and Conjugative Element ICEPmiW2 in Proteus mirabilis W2 Facilitates the Dissemination of Antibiotic-Resistance Genes.

BACKGROUND: The extensive use of antibiotics for treating infectious diseases leads to their release into the environment, which in turn results in antibiotic pollution and thereby facilitates the dissemination of antibiotic-resistance genes (ARGs). Recently, despite the implementation of strict antibiotic usage restrictions, the accumulation of ARGs and multidrug-resistant bacteria in the aquaculture environment continues to show a trend of persistent spread. METHODS: The W2 strain was isolated in the presence of 32 μg/mL doxycycline. A broth microdilution assay was employed to determine the

Screening of Chemical Residues and Antimicrobial Resistance in Dehydrated Chicken Feet Intended as Dog Treats: A Pilot Study.

The pet population is steadily increasing worldwide, and treats, which account for approximately 15% of the pet food market, are the fastest-growing segment of the pet food industry. However, the widespread use of antimicrobials in poultry farms may pose a direct risk to pets' health, leading to the emergence of antimicrobial resistance. For this reason, we sought to assess the possible presence of drug residues and antimicrobial-resistant bacteria in a limited cohort of 11 dehydrated chicken feet intended as dog treats purchased from the European market. Residues of nicarbazin (64%), 4-4' din

Species Diversity, Antimicrobial Susceptibility, and Provisional Epidemiological Cut-Off Values of Vibrio spp. Isolated From Pacific White Shrimp (Litopenaeus vannamei) Farms in Southern Thailand.

Vibrio spp. are the most common opportunistic pathogens in shrimp farms, and drug-resistant Vibrio spp. pose a major threat to both shrimp and human health. The current study investigated species diversity and antimicrobial susceptibility of Vibrio spp. from Pacific white shrimp (Litopenaeus vannamei) in Southern Thailand. Shrimp were collected from ten intensive farms with a history of disease outbreaks. Vibrio spp. were isolated from the hepatopancreas, identified to species level via 16S rRNA sequencing, and tested for minimum inhibitory concentrations (MICs) against eight antibiotics. A to

Treponema pallidum suppressive microenvironments and central nervous system damage: from mechanisms to tools and prevention.

The global resurgence of syphilis, marked by a dramatic 150% increase in cases over the last decade and the rise of the dominant SS14-Ω strain cluster, represents a critical threat to public health. This review explores the complex molecular mechanisms by which Treponema pallidum functions as a "stealth pathogen," establishing suppressive chemotactic microenvironments that facilitate neuroinvasion and persistent central nervous system (CNS) damage. We delineate a biomechanical and molecular cascade wherein the spirochete utilizes its unique "naked" outer membrane and hypervariable Treponema pa

Pellino1-mTOR/S6K1 signaling axis is a key pathogenesis for the development of polycystic kidney disease.

Ubiquitination serves a critical role in regulating both inflammatory responses and kidney injury. Among inherited renal disorders, autosomal dominant polycystic kidney disease (ADPKD) has demonstrated associations with disrupted ubiquitin signaling that exacerbates inflammation and cyst progression. In this study, we demonstrate that the E3 ligase Pellino1 (Peli1) acts as an essential contributor to the pathogenesis of ADPKD amid inflammatory conditions. In individuals with clear cell renal cell carcinoma (ccRCC), Peli1 exhibits markedly elevated expression, and this upregulation is associate

Phenotypical-genetic virulence and resistance in atypical avian pathogenic Escherichia coli.

Avian pathogenic Escherichia coli is a threat to poultry industries, as it is associated with infections, poor production ratings, and economic losses. The spread of multidrug-resistant bacteria is a major factor aggravating the situation. Therefore, this study aimed to characterize 28 E. coli isolates from poultry farms in Brazil regarding virulence, as well as genetic and phenotypical resistance. Antimicrobial susceptibility test was carried out by the disk diffusion method, while the genomic information of the isolates was determined by whole-genome sequencing. Resistance was identified for

Host modulation therapy in periodontitis: from established therapies to emerging technologies.

Periodontitis is a biofilm-induced chronic inflammatory disease, characterized by gingival inflammation and alveolar bone loss. According to a national survey, approximately half of the U.S. adults are affected by periodontal disease. To effectively prevent and treat periodontitis, it is essential to address its underlying causes. The primary etiological factors include polymicrobial synergy and dysbiosis of the oral microbiota, and a dysregulated immune response. The standard therapeutic approach, mechanical removal of biofilm through debridement, sometimes demonstrates limited efficacy, part

miR126-mediated alteration of vascular integrity in Rett syndrome.

Rett syndrome (RTT) is a neurodevelopmental disorder caused by mutations in methyl-CpG binding protein 2 (MeCP2). MeCP2 is a non-cell type-specific DNA binding protein, and its mutation influences not only neural cells but also non-neural cells in the brain, including vasculature-associated endothelial cells. Vascular integrity is crucial for maintaining brain homeostasis, and its alteration may be linked to the pathology of neurodegenerative diseases, but a non-neurogenic effect, such as the relationship between vascular alteration and RTT pathogenesis, has not been shown. Here, we developed

Pleural effusion associated with Strongyloides stercoralis hyperinfection in a splenectomised dog.

Strongyloides stercoralis is a soil-transmitted helminth that is increasingly reported in dogs across Europe. While most infections are subclinical, hyperinfection syndrome, marked by excessive larval proliferation and systemic spread, can occur in immunocompromised hosts. An 11-year-old male Boston Terrier dog, originating from southern France with a history of chronic diarrhoea and recent splenectomy was presented with severe regenerative anaemia, thrombocytopenia, and a degenerative left shift. Haemotropic Mycoplasma haemocanis infection was diagnosed, and doxycycline therapy was initiated.

Prevalence, antimicrobial resistance and detection of resistance genes of Escherichia coli and Salmonella spp. isolated from captive European wild boars reared in Brazil.

The expansion of invasive European wild boar (Sus scrofa scrofa) populations in Brazil and the increasing commercialization of this species raise concerns about their role as reservoirs of zoonotic and antimicrobial-resistant bacteria. This study aimed to determine the prevalence, antimicrobial resistance profiles, and detection of virulence and resistance genes in Escherichia coli and Salmonella spp. isolated from captive wild boars. A total of 100 rectal swab samples were collected from animals raised on commercial farms in Goiás State, Brazil. One Salmonella isolate (1%) was detected and id

Generation and characterization of the TRE-TNFR2 transgenic mouse for cell-specific and timed (over)expression of tumor necrosis factor receptor 2.

Tumor necrosis factor receptor 2 (TNFR2), the signaling partner of the transmembrane form of tumor necrosis factor (tmTNF), is implicated in cell survival and tissue healing processes. Despite recent advances, our knowledge of the precise cellular mechanisms engaged by TNFR2 and, most importantly, its downstream targets is still limited. To address this gap, we generated tetracycline responsive element (TRE)-TNFR2 transgenic mice that, once crossed with tetracycline-controlled trans-activator (tTA) or reverse tetracycline-controlled trans-activator (rtTA) transgenics, allow for timed (over)exp
