Effects of early cigarette smoke exposure on early immune development and respiratory disease.

Exposure to tobacco constituents during early development remains a common but avoidable toxic exposure, which has been clearly linked with decreased lung growth and subsequent wheezing illness. There is also now emerging evidence that tobacco smoke can influence early immune function. This includes alterations in cytokine production by the fetoplacental unit, as detected ex vivo in cord blood, as well as in patterns of fetal mononuclear cell responses in vitro. Recent studies also suggest that the newborns of smoking mothers have altered signalling through Toll-like receptors (TLRs) that are

Phospholipases and their industrial applications.

Phospholipids are present in all living organisms. They are a major component of all biological membranes, along with glycolipids and cholesterol. Enzymes aimed at modifying phospholipids, namely, phospholipases, are consequently widespread in nature, playing very diverse roles from aggression in snake venom to signal transduction and digestion in humans. In this review, we give a general overview of phospholipases A1, A2, C and D from a sequence and structural perspective and their industrial application. The use of phospholipases in industrial processes has grown hand-in-hand with our abilit

Interactions among multiple genomes: tsetse, its symbionts and trypanosomes.

Insect-borne diseases exact a high public health burden and have a devastating impact on livestock and agriculture. To date, control has proved to be exceedingly difficult. One such disease that has plagued sub-Saharan Africa is caused by the protozoan African trypanosomes (Trypanosoma species) and transmitted by tsetse flies (Diptera: Glossinidae). This presentation describes the biology of the tsetse fly and its interactions with trypanosomes as well as its symbionts. Tsetse can harbor up to three distinct microbial symbionts, including two enterics (Wigglesworthia glossinidia and Sodalis gl

Role of an iron-dependent transcriptional regulator in the pathogenesis and host response to infection with Streptococcus pneumoniae.

Iron is a critical cofactor for many enzymes and is known to regulate gene expression in many bacterial pathogens. Streptococcus pneumoniae normally inhabits the upper respiratory mucosa but can also invade and replicate in lungs and blood. These anatomic sites vary considerably in both the quantity and form of available iron. The genome of serotype 4 pneumococcal strain TIGR4 encodes a putative iron-dependent transcriptional regulator (IDTR). A mutant deleted at idtr (Δidtr) exhibited growth kinetics similar to parent strain TIGR4 in vitro and in mouse blood for up to 48 hours following infec

Tigecycline displays in vivo bactericidal activity against extended-spectrum-β-lactamase-producing Enterobacteriaceae after 72-hour exposure period.

Progressively enhanced activity of a humanized tigecycline (TGC) regimen was noted over 3 days against an extended-spectrum-β-lactamase (ESBL)-producing Escherichia coli isolate and an ESBL-producing Klebsiella pneumoniae isolate. Bacterial density reduction approximated 3 log(10) approaching bactericidal activity at 72 h. This level of activity has not been previously noted for compounds such as tetracyclines, normally considered bacteriostatic antimicrobials. Extended regimen studies in vivo may aid in better delineation of antimicrobial effects, producing improved correlation with clinical

Oxidative stress as a trigger for growth phase-specific sigmaE-dependent cell lysis in Escherichia coli.

Sigma(E), encoded by rpoE, causes lysis of viable but nonculturable (VBNC) cells at the early stationary phase in Escherichia coli. However, the factors that trigger this lysis phenomenon remain unidentified. In this study, we found that the defective strain of katE, encoding catalase, exhibited a sigma(E)-dependent cell lysis phenotype, showing a decrease in turbidity of cell culture and an increase in accumulation of proteins in the culture medium as well as an increase in rpoE expression. The level of intracellular reactive oxygen species became maximal at the transition period from exponen

Antimicrobial susceptibility of clinical isolates of Enterobacteriaceae producing complex beta-lactamase patterns including extended-spectrum enzymes.

The antimicrobial susceptibility of 103 clinical isolates of Enterobacteriaceae to 11 antibiotics, was investigated, using a conventional inoculum size (5 x 10(5) CFU) and a higher inoculum size (5 x 10(8) CFU). All the isolates produced complex beta-lactamase patterns, including an extended-spectrum beta-lactamase (ESBL) of the TEM- or SHV-type plus other enzymes (a TEM-type or an SHV-type non-ESBL and/or a class C enzyme). The following repertoire of ESBLs was produced by the isolates: TEM-15, TEM-19, TEM-26, TEM-52, TEM-72, TEM-87, TEM-92, SHV-2a, SHV-5 and SHV-12, as assessed by sequencing

Understanding genetic determinants of bacterial adaptation to plant niches: Toward more robust bioinoculant design.

Plant-associated bacteria offer a sustainable alternative to chemical inputs in agriculture, yet the field performance of microbial inoculants remains inconsistent. A central but frequently overlooked cause is that bioprospecting pipelines still select strains on the basis of plant-growth-promoting activities, while the bacterial genetic traits that determine whether a strain can reach, colonise and persist in a given root compartment are rarely assessed. Previous reviews have largely catalogued growth-promoting functions or described root microbiome assembly from the plant's perspective, here

A Multiomic Approach Integrating Genomic and Metabolomic Data Highlights Colorectal Cancer Pathways.

Numerous genetic variants have been identified by genome-wide association studies as being associated with colorectal cancer (CRC) risk. Metabolome-wide association analysis was performed for 187 CRC-associated genetic variants using genomic data and untargeted 1H nuclear magnetic resonance urine metabolomics from 1951 Airwave Health Monitoring Study participants. We identified statistically significant associations between seven CRC single-nucleotide polymorphisms (SNPs) and urinary metabolites. This included SNPs within or close to RHPN2 with sucrose (P = 1.2 × 10-7), SLC6A18 with amino acid

Xylo-oligosaccharide supplementation exacerbates colitis susceptibility in mice via taurine depletion.

Xylo-oligosaccharides (XOS) are widely utilized as functional food ingredients and additives. However, the impact of long-term dietary supplementation with XOS on intestinal inflammation and its underlying mechanisms has yet to be fully elucidated. This study investigated the XOS supplementation on susceptibility to DSS-induced ulcerative colitis (UC) in C57BL/6 J mice and explored the underlying mechanisms, with a particular focus on the intestinal microenvironment and bile acids (BAs) metabolism. The experimental results demonstrated that a 12-week dietary intervention with XOS markedly aggr

Large-scale genomic analysis of jumbo phages: coevolution, genome architecture, and host interaction mechanisms.

BACKGROUND: Jumbo phages are phages with comparatively large genome sizes. Jumbo phages have been identified in various microbial communities. However, their diversity, genome structure, potential function, and their interactions with hosts and other phages are largely unknown due to insufficient genomic data. RESULTS: We collected 59,652,008 putative viral genomes from seven habitats by using 38 public metagenome datasets, an integrated public viral genome database (IGN), and pig gut viral genome databases. We obtained 10,754 jumbo phage genomes with sizes ranging from 200 to 831 kb. Most (94

Microplastic biofilms enhance environmental stress, disinfectant, and antibiotic tolerance of Salmonella enterica serovar typhimurium in simulated fresh vegetable processing systems.

Microplastics (MPs) are widely found in food supply chains. They act as a stable surface for microorganisms to stick to, which makes it more likely that pathogens will be present. This study aims to investigate the biofilm formation on MPs and evaluate the tolerance of foodborne pathogen Salmonella enterica serovar Typhimurium (ST) to environmental stresses, disinfectants, and antibiotics. The microplastics biofilms (MBs) were constructed on polyethylene MPs by the microbial samples from a vegetable washing sink. Microorganisms can form biofilms on MPs in the lettuce juice-based cultures. The

A Bibliometric Analysis of Global Research Hotspots and Progress on Microbial Extracellular Polymeric Substances in Bioremediation.

Extracellular polymeric substances (EPSs) are high-molecular-weight biopolymers secreted by microorganisms, showing great potential for bioremediation. However, comprehensive analyses of the development context and quantitative research on the overall trends of EPSs in bioremediation are lacking. This study conducted a systematic bibliometric analysis of microbial EPS research using VOSviewer and CiteSpace. Keyword burst and thematic evolution analysis indicate a distinct thematic shift: early research focused on "structural characterization and adsorption mechanisms of EPSs", whereas current

The vector effect of microplastics and nanoplastics: co-transport and ecological risks of chemical pollutants and antibiotic resistance genes in the soil-water continuum.

Microplastics (MPs) and nanoplastics (NPs) act as dynamic environmental vectors across the soil-water continuum, allowing them to enter organisms through direct ingestion, leading to potential tissue accumulation. Under specific exposure conditions, these vectors can undergo trophic transfer through food chains, contributing to combined toxicological risks. This paper reviews how the adsorption and co-transport behaviors of chemical pollutants by MPs and NPs are collectively regulated by the intrinsic physicochemical properties of the material and environmental weathering processes. The format

Suspended sludge-biofilm niche cooperation enables stable mainstream nitrogen removal via mixotrophic partial denitrification coupled with anammox.

The sustainable application of partial denitrification/anammox (PD/A) under mainstream wastewater conditions depends critically on relieving the strong competition between heterotrophic and autotrophic microorganisms for substrates and ecological space. In this study, ferrous sulfide (FeS) was employed as a biofilm carrier to establish a mixotrophic PD/A (MPD/A), which maintained a total nitrogen removal efficiency of 90.25% under fluctuating organic carbon conditions in mainstream wastewater. To elucidate how niche differentiation between suspended sludge and biofilms supports synergistic nit
