An NIR-responsive FeS-loaded alginate hydrogel promotes diabetic wound healing through Nrf2 activation and macrophage polarization.

Diabetic wounds are characterized by excessive oxidative stress, persistent inflammation, and impaired tissue regeneration, which collectively hinder normal wound healing. Here, we developed a near-infrared (NIR)-responsive sodium alginate hydrogel incorporating nano‑iron sulfide (nFeS) through mild gelation mediated by calcium carbonate and glucono-δ-lactone. The resulting SA-nFeS hydrogel exhibited a porous structure, favorable rheological properties, hydrogen peroxide-responsive degradation, radical-scavenging activity, and good cytocompatibility. NIR irradiation accelerated the release of

Spatiotemporal Dissection of the Infection Response to Bacterial Soft Rot in Non-heading Chinese cabbage.

Bacterial soft rot caused by Pectobacterium brasiliense (Pbr) leads to substantial yield losses in cruciferous vegetables, including non-heading Chinese cabbage (pak choi, Brassica rapa subsp. chinensis). This study employed an integrated physiological and multi-tissue transcriptomic approach to decipher the infection response strategies of pak choi against Pbr. Age-dependent resistance was detected, with susceptibility decreasing as plants matured. In susceptible four-week-old plants, Pbr infection induced oxidative stress, characterized by the accumulation of hydrogen peroxide (H₂O₂) and mal

FeCoNiZnPt high-entropy hierarchical nanosheet-assembled nanozyme with enhanced peroxidase-like activity for colorimetric glucose detection.

The poor stability of natural enzymes and the limited catalytic activity of conventional nanozymes remain major challenges in the field of colorimetric glucose sensing. In this work, a high-entropy hierarchical nanosheet-assembled nanozyme (FeCoNiZnPt-HEHN) composed of Fe, Co, Ni, Zn, and Pt was successfully constructed through a solvothermal strategy. The as-prepared nanozyme exhibits a three-dimensional hierarchical microrod architecture assembled from ultrathin nanosheets, with multiple metal elements uniformly distributed throughout the structure. The synergistic catalytic centers involvin

Catalase Defines Radiotherapy Resistance and a Therapeutic Vulnerability in Rhabdomyosarcoma.

Therapeutic resistance remains a critical obstacle in rhabdomyosarcoma (RMS), the most common pediatric soft tissue sarcoma. Increasing evidence implicates redox adaptation in tumor survival and treatment failure. Here, we investigated the functional role and clinical relevance of catalase, a primary hydrogen peroxide-detoxifying enzyme, in modulating RMS therapeutic response. Integrated transcriptomic analyses revealed that while catalase is overall downregulated in RMS compared to healthy skeletal muscle, elevated expression strongly correlates with high-risk, metastatic disease and poor ove

Targeted catalytic eradication of antibiotic-resistant Helicobacter pylori by a pH-activated Fe-doped nanozyme.

The increasing prevalence of antibiotic-resistant Helicobacter pylori (R-H. pylori) poses a significant clinical challenge, contributing to persistent gastritis, peptic ulcers, and gastric cancer. Conventional antibiotic therapies are increasingly limited by resistance, incomplete eradication, and disruption of the gut microbiota. To address these challenges, we developed pH-responsive iron-doped ammonium citrate carbon dots (Fe-CDs), a nanozyme with intrinsic specificity for targeting and eradicating R-H. pylori within the acidic gastric microenvironment. Fe-CDs exhibit dual oxidase (OXD)- an

Oxidative Damage Induced Disruption of Mitochondrial Quality Control Underlying Abnormal Energy Metabolism in Peripheral Leukocytes of Schizophrenia Patients.

BACKGROUND AND HYPOTHESIS: Mitochondrial dysfunction is increasingly implicated in schizophrenia (SZ) pathogenesis. To maintain mitochondrial homeostasis under cellular stress, a sophisticated mitochondrial quality control (MQC) mechanism has developed, encompassing mitochondrial biogenesis, dynamics, and mitophagy. STUDY DESIGN: This study systematically evaluated MQC in peripheral leukocytes of 42 SZ patients and 43 healthy controls through morphological analysis, MQC gene expressions, mitochondrial DNA (mtDNA) maintenance, and oxidative damage. Besides, we validated the regulatory effects

Berberrubine protects against noise-induced cochlear damage and hearing loss by inhibiting the p38 MAPK signaling pathway.

Cochlear hair cells are critical sensory receptors for auditory function, and exposure to high-intensity noise can lead to irreversible hearing loss. However, the mechanisms underlying noise-induced hearing loss (NIHL) are poorly understood, and effective therapeutic strategies have yet to be developed. In this study, berberrubine (BB) was found to protect HEI-OC1 cells and hair cells of cochlear explants from hydrogen peroxide (H2O2)-induced damage. Furthermore, BB pretreatment protected against cochlear damage and preserved hearing in a mouse model of NIHL. Structural analysis using hematoxy

Influence of the menstrual cycle and sex hormones on the response to genotoxic stress in human cells.

Although the menstrual cycle and sex hormones have been shown to influence the molecular response to drugs and chemicals, this phenomenon is still insufficiently examined. In this study, the effects of the menstrual cycle and sex hormones on the genotoxic stress response were investigated, with the focus on the activation of poly(ADP-ribose) polymerases (PARPs). Peripheral blood mononuclear cells (PBMCs) were isolated from blood collected from naturally cycling female donors (n = 10) at three phases of their menstrual cycle: (I) early follicular, (II) late follicular/ovulatory, and (III) mid-l

Hydroxyurea-coordinated copper-based metal-organic framework nanoreactor for glucose-gated nitric oxide release and infected wound treatment.

Nitric oxide (NO)-based therapy represents a promising strategy against multidrug-resistant infections owing to its broad-spectrum antibacterial activity and minimal propensity to induce resistance. Hydroxyurea (HyU), a clinically approved drug for sickle cell disease, has recently emerged as a hydrogen peroxide (H2O2)-responsive NO precursor. However, the hydroxamic acid moiety responsible for NO generation is highly susceptible to hydrolytic degradation in aqueous environments, severely limiting its therapeutic utility. Herein, we report a coordination-driven stabilization strategy that inco

Tobramycin enhances Mycobacterium abscessus fitness through whiB7 induction.

Nontuberculous mycobacteria (NTM) are opportunistic pathogens that cause pulmonary disease (PD) in people with bronchiectasis and other chronic airway diseases. Difficulty treating and eradicating NTM-PD highlights the need for improved understanding of bacterial mechanisms to establish chronic infections. People with the genetic disorder cystic fibrosis (CF) develop bronchiectasis and are the population at highest risk of NTM-PD, caused mainly by Mycobacterium avium or Mycobacterium abscessus (Mabsc). The majority of people with cystic fibrosis (pwCF) and bronchiectasis develop chronic Pseudo

Redox-mediated optochemical plasmonic sensing of H2O2-equivalent reducing capacity in exhaled breath condensate.

Oxidative-stress-related diseases generate elevated levels of hydrogen peroxide (H2O2) and other redox-active species in exhaled breath condensate (EBC), offering a promising avenue for non-invasive disease assessment. Despite its diagnostic potential, clinical translation of EBC redox species detection has been constrained by bulky analyzers, labor-intensive operation, and limited sensitivity. By harnessing the redox potential, we present a Redox-mediated Optochemical Plasmonic Sensing (ROPS) system, an ultrasensitive platform that enables real-time, easy-to-use, point-of-care quantitative an

Cascade-responsive biomimetic nanoparticles remodel tumor metabolism and potentiate ferroptosis for synergistic therapy of aggressive breast cancer.

Breast cancer presents a formidable global health challenge, with traditional monotherapies frequently hampered by limited specificity and off-target toxicity. Herein, we developed a highly integrated, biomimetic, and pH-responsive zeolitic imidazolate framework-8 (ZIF-8) nanoplatform, RBCm-coated nanoparticles (R-A/G@Z), tailored for cascaded starvation and potentiated ferroptosis therapy. Through a one-pot biomineralization approach, the hydrophobic ferroptosis inducer artesunate (ART) and the hydrophilic metabolic enzyme glucose oxidase (GOx) were co-encapsulated within a ZIF-8 framework, f

Plasma-catalytic remediation of pharmaceutical-contaminated wastewater: Catalyst design and mechanistic insights from DFT.

Contaminants of emerging concern (CECs), characterized by persistence, poor biodegradability, and high toxicity, pose significant risks to aquatic ecosystems and human health. Plasma-catalytic processes may eliminate CECs, particularly micropollutants, from wastewater. However, discharge-zone thermalization and heavy-metal leaching from metal-based catalysts pose challenges for industrial adoption of cold plasma technology. This study presents a novel coaxial plasma electrode and a heteroatom-doped carbonaceous catalyst that overcome these limitations. Catalysts with varying boron and oxygen c
