Cerebellar granule cell membranes and glutamate mediates transactivation of glutamine synthetase promoter.

Potential region of glutamine synthetase promoter driving astrocyte-specific transactivation, mediated by cerebellar granule cell membrane and glutamate has been identified by deletion analysis of promoter and transient transfection. The promoter region from -420 to -765 was found to be potentially important for this transactivation. These results provided further evidence for importance of neuronal-glial and glutamate-glial interactions in regulation of glial gene expression.

Poly(lactic acid)-poly(ethylene glycol) nanoparticles provide sustained delivery of a Chlamydia trachomatis recombinant MOMP peptide and potentiate systemic adaptive immune responses in mice.

UNLABELLED: PLA-PEG [poly(lactic acid)-poly (ethylene glycol)], a biodegradable copolymer, is underexploited for vaccine delivery although it exhibits enhanced biocompatibility and slow release immune-potentiating properties. We document here successful encapsulation of M278, a Chlamydia trachomatis MOMP (major outer-membrane protein) peptide, within PLA-PEG nanoparticles by size (~73-100nm), zeta potential (-16 mV), smooth morphology, encapsulation efficiency (~60%), slow release pattern, and non-toxicity to macrophages. Immunization of mice with encapsulated M278 elicited higher M278-specifi

Modulation of SK channel trafficking by beta adrenoceptors enhances excitatory synaptic transmission and plasticity in the amygdala.

Emotionally arousing events are particularly well remembered. This effect is known to result from the release of stress hormones and activation of beta adrenoceptors in the amygdala. However, the underlying cellular mechanisms are not understood. Small conductance calcium-activated potassium (SK) channels are present at glutamatergic synapses where they limit synaptic transmission and plasticity. Here, we show that beta adrenoceptor activation regulates synaptic SK channels in lateral amygdala pyramidal neurons, through activation of protein kinase A. We show that SK channels are constitutivel

Equisetin targets membrane integrity and transcriptional networks to combat multidrug-resistant Enterococcus faecalis.

This study evaluates the natural product Equisetin as a potent agent against multidrug-resistant Enterococcus faecalis (MDR E. faecalis). Equisetin exhibited strong antibacterial activity (MIC = 5.84 μg/mL) by disrupting membrane integrity, inducing protein/DNA leakage, depolarizing membrane potential, elevating ROS, and depleting ATP. It also inhibited biofilm formation and showed minimal hemolytic activity without inducing resistance. Transcriptomics revealed upregulation of DNA repair/stress genes and downregulation of metabolic/protein synthesis genes, indicating a multi-target mechanism.

FIRS: Neonatal considerations.

Fetal Inflammatory Response Syndrome (FIRS) is the fetal counterpart of systemic inflammatory response syndrome (SIRS) described in adults. When the fetus is directly exposed to inflammation of the fetal membranes or the placental-fetal circulation, and organs are adversely affected, the disorder is known as FIRS. This syndrome can significantly affect multiple organs with significant short and long term implications for the newborn. In cases of neonatal encephalopathy when no obvious etiology is identified, FIRS needs to be considered. Based on the significant incidence of chorioamnionitis an

Mammalian aquaglyceroporin function in metabolism.

Aquaglyceroporins are integral membrane proteins that are permeable to glycerol as well as water. The movement of glycerol from a tissue/organ to the plasma and vice versa requires the presence of different aquaglyceroporins that can regulate the entrance or the exit of glycerol across the plasma membrane. Actually, different aquaglyceroporins have been discovered in the adipose tissue, small intestine, liver, kidney, heart, skeletal muscle, endocrine pancreas and capillary endothelium, and their differential expression could be related to obesity and the type 2 diabetes. Here we describe the

Peritoneal dialysis: a biological membrane with a nonbiological fluid.

During chronic peritoneal dialysis (PD), the peritoneal membrane undergoes structural and functional alterations that, in some patients, may eventually lead to loss of function as the 'dialysis membrane'. These alterations include loss of mesothelial cells, thickening of the submesothelial compact zone, and changes of vascularization. At least some of these effects are thought to be associated with chronic exposure to unphysiological PD fluids. On a cellular level, chronic PD is associated with the phenomenon of epithelial-to-mesenchymal transformation (EMT) as well as premature aging and sene

Immunoisolation cell therapy for CNS diseases.

Delivery of potentially therapeutic drugs to the brain is hindered by the blood-brain barrier (BBB), which restricts the diffusion of drugs from the vasculature to the brain parenchyma. One means of overcoming the BBB is with cellular implants that produce and deliver therapeutic molecules directly into the CNS region of interest. In this paper we describe the current status of one iteration of cell-based therapy that uses xenogeneic cells encased within a selectively permeable polymeric membrane; this is known as immunoisolation. For the purposes of this review, cell immunoisolation for treat

The glomerular basement membrane in lupus nephritis.

Systemic lupus erythematosus is a disease that is particularly suited for studies of glomerular basement membrane pathology. Classification of the renal pathology of lupus nephritis is usually based on light microscopic features, combined with immunofluorescence findings and electron microscopic alterations. Study of renal biopsy helps to distinguish potentially reversible and irreversible disease, and to estimate prognosis of patients with lupus nephritis. Moreover, studies of human disease, as well as the availability of animal models and in vitro cell culture systems employing biochemical a

[Post-traumatic ARDS--a potential indication for lung transplantation?].

Posttraumatic adult respiratory distress syndrome (ARDS) still involves significant mortality, despite progress in management concepts. Current therapeutic strategies are briefly described, including kinetic therapy, high-frequency jet ventilation and extracorporeal membrane oxygenation. In addition, a spectacular case of the first successful lung transplantation for posttraumatic ARDS after failed ECMO (extracorporeal membrane oxygenation) support is reported. This young man with severe posttraumatic ARDS developed a potentially lethal bilateral pulmonary hemorrhage under treatment with ECMO,

Immunoregulation through membrane proteins modified by reducing conditions induced by immune reactions.

Selected disulfide bonds in membrane proteins are labile and are thus susceptible to changes in redox potential and/or the presence of thiol isomerase enzymes. Modification of these disulfide bonds can lead to conformational changes of the protein that in turn may alter protein activity and function. This occurs in the entry of several enveloped viruses into their host cells, e.g. HIV, hepatitis C virus and Newcastle disease virus. Labile disulfide bonds are also important in platelet activation, cytokine signalling and in a variety of diseases including cancer and arthritis. In this review we

Burow's solution-induced acute sensorineural hearing loss: report of two cases.

This is the report of rare cases to show the potential ototoxicity of Burow's solution in clinical use. We encountered two cases of acute sensorineural hearing loss occurring after the administration of 8% Burow's solution (pH 2.93) into the tympanic cavities. Prior to treatment, both cases presented with tympanic membrane perforations in the posterior and inferior portions and a small amount of serous discharge. Fortunately, hearing loss recovered almost completely except for high tones within a few weeks. These two case reports demonstrate that the ototoxicity of Burow's solution should be c

Could a cervical occlusion suture be effective at improving perinatal outcome?

Cervical weakness and infection have long been regarded as major causes of preterm birth. Cervical cerclage has been used extensively to reduce the risk of preterm birth arising as a result of cervical weakness, but increasing evidence suggests that the cervix plays more than just a mechanical role. Immunological function of the cervix and mucus plug is thought to be important in minimising the ingress of microbes, which can lead to chorioamnionitis and rupture of the amniotic membranes. In this review, we examine the background of traditional cervical cerclage and introduce the concept of the

Amniotic membrane to the rescue of partially necrosed dermis fat graft.

A 25-year-old woman underwent dermis fat graft on the right side for persistent ocular pain with her polymethylmethacrylate (PMMA) spherical orbital implant. One week later she developed dermal necrosis of the dermis fat graft with potentially devastating fat atrophy if left uncovered. Amniotic membrane graft epithelial side up was sutured to the conjunctiva surrounding the fat. 2 weeks later the graft epithelialized with preservation of fat. Role of amniotic membrane graft as a routine lining over dermis fat grafts to promote conjunctival healing and prevent fat exposure needs to be studied i

[Potential-dependent Ca2+ channels and contractile function of the heart in toxic-infectious shock caused plague].

Influence of intravenous administration to rats of murine toxin of Y. pestis (1 mg/ml, LD100) on the number of potential-operated Ca(2+)-channels, alpha- and beta-adrenergic and M1-cholinergic receptors of plasma membranes and heart contractility function has been studied in rats. The number of Ca(2+)-channels in plasma membranes and contractility of heart decreased by 50% in 1 hour after the i.v. administration of toxin and further decreased up to the lethal end. During the agonal stage of the shock desensitization of beta-adrenergic receptors has been discovered, while alpha-adrenergic and M

On the Dynamics of Noncovalent Interactions Modulating Passive Diffusion of Cisplatin Across a Human Breast Cancer Cell Membrane: An amIGM + GLED View.

Passive permeation across biological membranes is governed by a delicate balance of transient noncovalent interactions that are difficult to resolve experimentally or through conventional molecular dynamics (MD) analyses alone. Here, we present a perspective on the integration of the averaged modified independent gradient model (amIGM) based on promolecular density approximation and the graphic representation of the local electron energy density (GLED) with classical MD simulations to investigate the passive translocation of the cisplatin (cisPt) anticancer drug across a model membrane represe

Fabrication of a high-performance MXene-ATP composite membrane: enhancing flux and retention for dye wastewater treatment.

In order to overcome the poor mechanical properties and low permeation flux of MXene nanosheets, as well as the easy agglomeration and limited adsorption capacity of attapulgite (ATP), a composite membrane material (Fe-Co/MXene-ATP) was prepared by vacuum filtration method, of which ATP was inserted into the stacking layers of MXene and bimetallic oxides were loaded as active substances. Through a series of characterization and experiments, it was proved that MXene-ATP composite membrane was achieved, and Fe and Co oxides were successfully loaded on it. The membrane material was used to remove

Biomimetic trident lipid anchoring enables stable neutrophil-based combination therapy.

Anchoring-conjugation strategy, characterized by benign hydrophobic membrane anchoring and swift covalent drug-coupling, has been extensively practiced to attach therapeutic agents to cell surfaces in cell-based combination therapies. However, this approach faces significant challenges in phagocytic cells due to their enhanced membrane fluidity, which destabilizes the hydrophobic interactions and causes premature payload dissociation from cells, ultimately impairing drug delivery. This research proposes a biomimetic trident anchoring lipid featuring three hydrophobic tail chains to amplify hyd

Regulating the membrane-fusion effect of liposomes via the protonation of tertiary amines.

Membrane fusion, a fundamental biological process enabling precise exchange of biomolecules within and between cells, has inspired the development of various fusogenic model systems for applications in drug delivery and beyond. However, conventional systems typically operate in an "always-on" state, lacking spatiotemporal control. Although cleavable polyethylene glycol coatings can confer temporal regulation through stimuli-responsive activation, such strategies are often unidirectional and synthetically challenging. Here, we report a simple yet reversible strategy to regulate liposomal membra
