In situ, antibody-independent, and multiplexed characterization of amyloid plaques by MALDI MS/MS imaging using iprm-PASEF.

Amyloidosis collectively describes a heterogeneous group of protein aggregation-based diseases involving the misfolding and extracellular accumulation of fibril-forming amyloid proteins. Diagnosing amyloidosis is difficult due to its many subtypes (e.g., AA, AL, ATTR), with varying symptoms. Current diagnosis often involves Congo red staining, but it has limitations in quantification and specificity. A novel method called iprm-PASEF exploits MALDI imaging and offers a faster, spatially resolved, antibody-independent technique for identifying peptides while preserving tissue structure. In this

Solid lipid nanoparticles as carriers for Ulva rigida peptide extract with demonstrated anti-β-amyloid and antioxidant properties.

INTRODUCTION: Alzheimer's disease (AD) remains one of the greatest challenges in neurodegenerative research, largely due to the toxic aggregation of the β-amyloid (Aβ) peptide and the difficulty of delivering therapeutic compounds across the blood-brain barrier (BBB). METHODS: This work explored Ulva rigida (U. rigida), a green macroalga rich in bioactive peptides, as a natural source of anti-amyloidogenic and antioxidant agents. RESULTS: First, the peptides extraction process was optimized by design of experiments (DoE). Peptides extracted and hydrolyzed under optimized conditions showed an

Amyloid Plaques Ameliorate Memory Deficits and Hippocampal Neuron Loss in an Aβ4-42-Driven Alzheimer's Disease Mouse Model.

Extracellular deposition of amyloid-β (Aβ) peptides in the form of plaques is the most prominent pathological hallmark of Alzheimer's disease (AD). The postulated central pathophysiological role of fibrillary Aβ plaques has, however, been questioned, and small, soluble, pre-fibrillar Aβ aggregates (oligomers) have been implicated as the crucial neurotoxic species in AD etiology. While the relationship between insoluble amyloid plaques and soluble Aβ oligomers remains unclear, it has been hypothesized that plaques may serve as reservoirs, sequestering toxic Aβ oligomers in the initial stages of

Vitamin D3 Attenuates Alzheimer's Disease-Like Pathology via Remodeling Amyloid Protein Production and Clearance Pathway in Wistar Rats.

Alzheimer's disease widely affects millions of people worldwide, accounting for 60% of dementia cases. Clinically classified by the presence of cognition impairment, pathophysiological representation includes deposited senile plaques, neurofibrillary tangles, and neuroinflammation. The pathogenesis of Alzheimer's disease (AD) remains multifaceted and is governed by multiple hypotheses. However, it undeniably involves amyloid-β (Aβ) accumulation and hyperphosphorylated tau (p-tau) pathology as the crucial events in disease initiation. Substantial evidence has correlated Vitamin D3 as a vital su

High-throughput screening approach identifies substrate-selective Hsp104 variants that counter amyloid seeding with diminished off-target effects.

Hsp104, a yeast protein-remodeling factor, can disaggregate misfolded proteins implicated in neurodegeneration. Although many potentiated Hsp104 variants have been generated, suboptimal properties have limited their application in mammalian systems. Here, we present the development of a high-throughput screening approach for identifying enhanced Hsp104 variants. To screen a large library of variants in parallel and with a quantitative output, we coupled a live-or-die yeast-based selection with next-generation sequencing. The identified Hsp104 variants solubilize preformed α-synuclein and TDP-4

Lactoferrin amyloid-templated gold nanozymes combat pathogenic Bacteria via a synergistic assault of oxidative stress and metabolic collapse.

The escalating crisis of antibacterial resistance necessitates the development of novel therapeutic agents that operate via unconventional mechanisms. Herein, we report the design of a potent antibacterial nanozyme by engineering a food-grade protein, lactoferrin (LF), into a dual-function biological macromolecule template. Lactoferrin was first transformed into robust amyloid fibrils (LFF), which then served as an active scaffold for the in situ synthesis of gold nanoclusters (AuNCs@LFF). The resulting hybrid material exhibited robust peroxidase-like (POD-like) activity, efficiently catalyzin

Amyloid beta levels and occludin depletion in low dose heavy metal mixture mediated cerebellar toxicity in Wistar rats.

Co-exposure to heavy metals can result in additive or synergistic toxicity in the brain, culminating in neurotoxicity. This study investigated the neurotoxic effects of a low-dose mixture of two toxic heavy metals-lead (Pb, 20 mg/kg) and aluminium (Al, 35 mg/kg)-and one essential metal, manganese (Mn, 0.564 mg/kg), on the cerebellum of rats. Animals were divided into five groups (n = 5) and orally treated for 90 days as follows: Group I received normal drinking water and served as the control; Group II received a heavy metal mixture of Pb (20 mg/kg), Al (35 mg/kg), and Mn (0.564 mg/kg) body we

Predicting future cognitive impairment in preclinical Alzheimer's disease using amyloid PET and MRI: A multisite machine learning study.

Predicting the likelihood of developing Alzheimer's disease (AD) dementia in at-risk individuals is important for the design of and optimal recruitment for clinical trials of disease-modifying therapies. Machine learning (ML) has been shown to excel in this task; however, there remains a lack of models developed specifically for the preclinical AD population, who display early signs of abnormal brain amyloidosis but remain cognitively unimpaired. Here, we trained and evaluated ML classifiers to predict whether individuals with preclinical AD will progress to mild cognitive impairment or dement

Traumatic Brain Injury Modulates Synuclein-Associated Transcription, Amyloid Plaque Morphology and Cognitive Performance in APPswe/PS1dE9/Blg Mice.

Background/Goals: Traumatic brain injury (TBI) is increasingly recognised as an important risk factor for delayed neurodegeneration and has been implicated in the modulation of Alzheimer's disease (AD)-related amyloid pathology. However, experimental evidence remains equivocal, suggesting that the effects of TBI on amyloidogenesis are context-dependent and influenced by factors including disease stage, injury severity, and the pre-existing neurodegenerative background. This study aimed to comprehensively assess the effects of TBI on cognitive function, synuclein-family gene expression, neuroin

Cortical gray-white matter contrast alterations precede amyloid-β positivity and macrostructural changes in older adults without dementia.

INTRODUCTION: Early Alzheimer's disease (AD) involves subtle cortical changes that may precede atrophy. Magnetic resonance imaging (MRI) microstructural markers may detect earlier pathology than classical morphometry. METHODS: We analyzed cross-sectional MRI and amyloid-β (Aβ) positron emission tomography (PET) data from 1323 non-demented AMYPAD participants. Cortical volume, thickness, gray-white matter contrast (GWC), and mean diffusivity (MD) were related to global Aβ burden and estimated time to Aβ-positivity using regression, correlation, and change-point analyses. RESULTS: Microstructu

[Research progress on vascular endothelial growth factor C in meningeal lymphatic vessel-mediated clearance of amyloid β-protein].

Alzheimer's disease (AD) is a neurodegenerative disorder characterized by the abnormal deposition of amyloid β-protein (Aβ) as a core pathological feature. Meningeal lymphatic vessels are crucial for Aβ clearance, and their dysfunction accelerates AD progression. Vascular endothelial growth factor C (VEGF-C), through activation of vascular endothelial growth factor receptor 3 and downstream pathways, synergistically promotes lymphangiogenesis, enhances lymphatic permeability, and regulates lymphatic fluid flow, thereby improving Aβ clearance efficiency. Genetic factors and aging-related declin

A C. elegans model of familial Alzheimer's disease shows age-dependent synaptic degeneration independent of amyloid β-peptide.

The membrane-embedded γ-secretase complex is involved in the intramembrane cleavage of ∼150 substrates. Cleavage of amyloid precursor protein (APP)-derived substrate C99 generates 38-43-residue secreted amyloid β-peptides (Aβ), with the aggregation-prone 42-residue form (Aβ42) particularly implicated in the pathogenesis of Alzheimer's Disease (AD). However, whether Aβ42 is the primary driver of neurodegeneration in AD remains unclear. Dominant mutations in APP or presenilin-the catalytic component of γ-secretase-cause early-onset familial AD (FAD) and reduce one or more steps in the multi-step
