Brain-kidney cross-talk: A call for action!

The cascade of events initiated by traumatic brain injury (TBI) has been a major focus of research. Despite significant advances, a deeper understanding of both favorable and unfavorable factors is still needed in this special cohort, especially concerning non-neurological complications - such as respiratory dysfunction, cardiovascular issues, sepsis, and acute kidney injury (AKI). This underscores the need to look beyond the immediate "what kills first" approach and address other critical problems that are essential for both patient management and resource utilization. The brain-kidney cross-

Hypoxic development down-regulates endothelial Piezo1 via HIF1-α signaling and impairs Piezo1-mediated vasorelaxation.

Hypoxia plays a critical role in regulating vascular function, with endothelial mechanosensitive proteins, such as the piezo-type mechanosensitive ion channel component 1 (Piezo1), emerging as key players in maintaining vascular homeostasis. Piezo1 is essential for nitric oxide-mediated vasodilation and vascular tone regulation. However, the impact of hypoxia on endothelial Piezo1 expression and function remains poorly understood. The present study investigated the regulation of Piezo1 and mechanosensitive-related genes (MRGs) during hypoxic development and their role in fetal growth restricti

Multifunctional CoFcMOF Nanozyme Facilitates a Self-Validating Triple-Modal Immunosensor for CRP in Serum and Synovial Fluid.

Accurate quantification of C-reactive protein (CRP) in serum and synovial fluid is critical for diagnosing and monitoring localized inflammatory arthritis, but it remains challenging due to the complexity of the matrix and the lack of robust and sensitive assays. To overcome this critical challenge, a novel self-validating triple-modal immunosensing platform was developed based on a rationally designed multifunctional cobalt-ferrocene metal-organic framework (CoFcMOF) nanozyme, which ensures robust detection reliability through a synergistic signal fusion mechanism. The innovation lies in the

Regional wasteosome accumulation across neurodegenerative diseases points to a shared underlying mechanism potentially related to glymphatic insufficiency.

The glymphatic system plays a key role in clearing waste products from the brain and is essential for maintaining brain homeostasis. When dysfunctional, it appears to contribute to pathological changes that exacerbate brain disorders, including neurodegenerative diseases. Additionally, wasteosomes, also known as corpora amylacea, are structures that function as waste containers and are thought to increase in response to chronic glymphatic insufficiency. Hence, in this study, we evaluated whether the accumulation and distribution of wasteosomes are compatible with both the potential role of was

Murine models of neovascular AMD revisited: Mechanistic pathways, immune-metabolic crosstalk, and the impact of aging and sex.

Age-related macular degeneration (AMD) is a leading cause of irreversible central vision loss in older adults. Neovascular AMD (nAMD) is characterized by choroidal neovascularization (CNV), vascular leakage, and fibrotic remodeling. Although anti-vascular endothelial growth factor (VEGF) therapy has improved visual outcomes for some patients, emerging findings highlight growing concerns regarding treatment resistance, persistent disease activity, and progression to subretinal fibrosis, which remain major challenges. Growing evidence indicates that nAMD arises from the interplay among metabolic

The Role of Arterial Stiffness in the Prognosis, Assessment, and Treatment of Patients with Peripheral Artery Disease: A Comprehensive Review.

Peripheral artery disease (PAD) is a prevalent and disabling vascular condition characterized by progressive atherosclerotic narrowing and stiffening of peripheral arteries. Arterial stiffness, most accurately measured by pulse wave velocity (PWV), has gained prominence as a critical biomarker in the assessment and management of PAD. Unlike traditional diagnostic tools that focus primarily on anatomical lesions, PWV offers dynamic insights into vascular function, elasticity, and systemic cardiovascular risk. Elevated PWV has been consistently associated with adverse outcomes in PAD patients, i

The Complex Interplay of Malaria and EBV in Burkitt Lymphoma.

Burkitt lymphoma (BL) is an aggressive B-cell lymphoma endemic in children in regions of sub-Saharan Africa, where its incidence geographically overlaps holoendemic Plasmodium falciparum malaria and poorly controlled childhood Epstein-Barr virus (EBV) infection. Despite decades of research, the precise mechanistic synergy between these two pathogens remains incompletely defined. This review synthesizes current epidemiological, immunological, and molecular evidence to propose an integrated model for the etiology of endemic BL. We outline a paradoxical, dual-edged relationship wherein EBV infect

How Ocean Acidification Makes Marine Fish Reproduction Vulnerable: Physiological Mechanisms and Ecosystem Consequences.

Ocean acidification, driven by the absorption of anthropogenic carbon dioxide into seawater, poses a significant threat to marine environments and biodiversity. This comprehensive review examines the specific impacts of acidification on the reproductive physiology of fish, integrating recent advances in our understanding of gonadal development, gametogenesis, and early embryonic development. We synthesize current findings on how altered carbonate chemistry disturbs the reproductive system from gametogenesis and fertilization to larval development and beyond. Our analysis reveals that ocean aci

Genetic determinants of HIV-1 elite control.

PURPOSE OF REVIEW: Elite controllers, rare individuals who maintain undetectable HIV-1 viremia without antiretroviral therapy, represent a human model of a functional cure. Understanding the host genetic factors that enable this spontaneous viral control is a key goal of HIV-1 research. This review synthesizes recent findings that are challenging and expanding the classic, human leucocyte antigen (HLA)-centric view of elite control. RECENT FINDINGS: While the dominant association of specific HLA class I alleles (e.g., HLA-B*57, HLA-B*27) with low set-point viral load remains the cornerstone o

Unlocking the healing power of Berberine: A promising aid for multiple sclerosis.

Multiple sclerosis (MS) is a debilitating autoimmune disorder characterized by inflammatory demyelination and progressive neurodegeneration within the central nervous system (CNS). Despite advances in disease-modifying therapies (DMTs), current treatments primarily mitigate relapses and slow disease progression but fall short in comprehensively addressing cumulative disability or neurodegeneration. Berberine (BBR), a naturally occurring isoquinoline alkaloid, has emerged as a promising therapeutic candidate due to its potent immunomodulatory, anti-inflammatory, and neuroprotective properties.

KDELR3 is transcriptionally activated by FOXM1 and accelerates lung adenocarcinoma growth and metastasis via inhibiting endoplasmic reticulum stress-induced cell apoptosis.

Lung cancer is still considered to be the leading cause of cancer-related death worldwide, and lung adenocarcinoma (LUAD) is the most common kind. KDEL Endoplasmic Reticulum Protein Retention Receptor 3 (KDELR3) is a critical regulator of the endoplasmic reticulum (ER) stress and the followed unfolded protein response (UPR) process, which are critical in tumor development. However, the role of KDELR3 in LUAD tumor progression remains poorly understood. In this work, we demonstrated that KDELR3 is significantly upregulated in LUAD tumor tissues and cell lines. Suppression of KDELR3 promoted the

Intestinal oxygen and microbiota crosstalk: implications for pathogenesis of gastrointestinal diseases and emerging therapeutic strategies.

The gut microbiota and its hypoxic host environment play a critical role in human health. Despite its importance, the mechanisms maintaining homeostasis and the characteristics defining dysbiosis remain largely undefined. In particular, the regulation of intestinal oxygen (IO) levels emerges as a critical factor in maintaining microbial balance. Host-driven factors, including epithelial oxygen consumption, mucosal perfusion, and luminal gas diffusion, establish a hypoxic gradient essential for the stable colonization by obligate anaerobes. Disruptions to this gradient, leading to pathological

PDE5 Inhibitors as Modulators of Alzheimer's-Associated Inflammation and Oxidative Stress: A Meta-Analytical Assessment of Preclinical Studies.

Neuroinflammation and oxidative stress contribute significantly to cognitive decline, a hallmark of neurodegenerative diseases such as Alzheimer's disease (AD). Addressing cognitive decline is a critical medical need, and phosphodiesterase-5 inhibitors (PDE5Is) may offer a promising solution. This meta-analysis highlights the anti-inflammatory and antioxidant actions of PDE5Is, which may help counter neuroinflammation and oxidative stress in neurodegenerative diseases including AD. We reviewed over 1,258 studies and considered 34 preclinical rodent studies of inflammation or oxidative stress.

MIG-6 Plays a Critical Role as a PGR Mediator in Maintaining Epithelial and Stromal Cells for Uterine Receptivity.

Ovarian steroid hormones-estrogen and progesterone-play a central role in regulating epithelial-stromal interactions in the uterus. These interactions are critical for uterine function, including endometrial receptivity, implantation, and decidualization. These interactions involve complex signaling crosstalk between the uterine epithelium and the underlying stroma, with dynamic cell population-specific roles. Mitogen-inducible gene 6 (MIG-6) is a key mediator of progesterone signaling in the endometrium, and loss of MIG-6 results in implantation failure due to a non-receptive endometrium. To

The AI-2/LuxS quorum sensing system enhances stress tolerance and enological performance of Lactiplantibacillus plantarum during malolactic fermentation.

Although Lactiplantibacillus plantarum shows potential for malolactic fermentation (MLF), its stress sensitivity compared to Oenococcus oeni limits its enological applications. This study reveals the critical role of the AI-2/LuxS quorum sensing (QS) system in the wine stress adaptation of L. plantarum XJ25 through targeted luxS gene manipulation. Deletion of luxS did not impact growth or acid production of L. plantarum XJ25 in optimal conditions (no stress) but significantly decreased AI-2 signal molecule activity. However, under simulated wine stress conditions, including low pH and high eth

Impact of High Glucose on Bone Collagenous Matrix Composition, Structure, and Organization: An Integrative Analysis Using an Ex Vivo Model.

Diabetes mellitus is a widespread metabolic disorder linked to numerous systemic complications, including adverse effects on skeletal health, such as increased bone fragility and fracture risk. Emerging evidence suggests that high glucose may disrupt the extracellular matrix (ECM) of bone, potentially altering its composition and organization. Collagen, the primary organic component of the ECM, is critical for maintaining structural integrity and biomechanical properties. However, definitive evidence and a comprehensive understanding of the molecular mechanisms through which high glucose impac
