Trophoblastic pseudotumor of the uterus: case report and ultrastructure.

A trophoblastic pseudotumor occurring in a young woman 6 months after a normal pregnancy is reported. Ultrastructural investigation demonstrated a close structural relationship between the infiltrating cells and those of the trophoblastic components of the normal human placenta, especially those seen in the primary villi of the developing placenta.

Immunogenetic and oncogenic properties of rat trophoblast cell lines.

PROBLEM: The nature of major histocompatibility complex (MHC) antigen expression on rat placentas, trophoblast cell lines, and tumors derived from trophoblast cells was explored. METHOD OF STUDY: Cytohistochemical and flow cytometric analysis and molecular techniques. RESULTS: MHC antigen expression and genomic imprinting on the placenta and on trophoblast cells varies with the time of gestation and with the type of MHC antigen. CONCLUSIONS: There is no correlation in trophoblast cells between class I expression and cell ploidy, on the one hand, and malignant potential, on the other hand. G

[Growth factors and apoptosis rate in an unusual chorangioma].

We describe an unusual type of cellular chorangioma with a high rate of proliferating cells and mitosis and high expression of the growth factor BFGF. The tumor was observed in the placenta of a 26-year-old gravida 1 with help syndrome. The pregnancy was terminated at 35+5 weeks by elective caesarean section. Chorangiomas are hamartoma malformations of the placenta which in some cases may be large enough to influence the course of pregnancy and associated complications are hydramnion, gestosis or hemorrhage. Complications for the fetus are due to hemodynamic alterations caused by the formation

Observations on the pathogenesis of Chlamydia psittaci infection of pregnant sheep.

Susceptible pregnant sheep were infected with Chlamydia psittaci at different stages of early and mid-gestation. Placentas and foetuses were examined by microbiological, serological, histopathological and immunochemical methods at various intervals thereafter. While ewes were susceptible to infection from early gestation onwards and infections of placentas and foetuses occurred from about 60 days gestation (dg), pathological changes were not detected until after 90 dg. These changes consisted of initial rapid replication of C. psittaci in the trophoblast at the limbus of the placentomal hilus,

Polonium-210 levels in placental maternal-fetal barrier: A pilot study conducted in the city of Sao Paulo, Brazil.

The accumulation of detectable amounts of radon progeny in the maternal-fetal placental barrier may be a risk factor for fetal development and lifelong disabilities. In this preliminary study, we analyzed the levels of alpha-emitting radon progeny Polonium-210 (210Po) (in Becquerel/kg) in the maternal-fetal placental barrier from placentas in Sao Paulo, SP, Brazil. Overall, there were higher levels of 210Po at the maternal portion of the placenta 1.45 (0.34) Bq/kg when compared to the fetal placental portion [1.08 (0.35) Bq/kg]. The presence of 210Po in the maternal-fetal placenta may indicate

TIG1 triggers placental senescence in preeclampsia through LMNA/p53 axis activation.

INTRODUCTION: The mechanism behind abnormal placental aging in preeclampsia (PE) is unclear. Although TIG1 is widely expressed in the human placenta, its function hasn't been well understood. Our previous study found a significant elevation of TIG1 in the placentas of PE patients. In this study, we focused on the molecular mechanism by which TIG1 functions in PE. METHODS: TIG1 expression of placentas from PE patients and L-NAME mice were analyzed using qRT-PCR, Western blot, and immunohistochemistry. TIG1-overexpressed HTR-8/SVneo cells were constructed for transcriptomic sequencing. Senescen

Is there any effect on imprinted genes H19, PEG3, and SNRPN during AOA?

Assisted oocyte activation (AOA) has been proposed as an effective technique to overcome the problem of impaired fertilization after intracytoplasmic sperm injection (ICSI) but the safety of AOA remains a concern. We aimed to investigate if AOA induces imprinting effects on embryos. We used 13 cleavage embryos, nine blastocysts, and eight placentas from 15 patients. The subjects were divided into six groups by tissue type and with or without AOA. The methylation levels of imprinted genes (H19, paternally expressed gene [PEG3] and small nuclear ribonucleoprotein polypeptide N [SNRPN]) were test

Placental pathology in patients using cocaine: an observational study.

OBJECTIVE: Although retroplacental hemorrhage is a major cause of fetal death, its etiology often remains obscure. In some reports, cocaine use by pregnant women has been associated with retroplacental hemorrhage and clinical abruptio placentae. This study was designed to assess the occurrence of chorionic villus hemorrhage, an entity shown recently to be associated with retroplacental hemorrhage, in the placentas of cocaine users. METHODS: Twenty-nine placentas from cocaine users and 15 placentas from drug-free controls, as determined by questionnaire and urine toxicology screen, were examin

Placental Adaptations Under Severe Anemia: Neonatal Outcomes at the Crossroads.

Anemia is a major global health problem, particularly in developing countries such as India, due to the greater prevalence of dietary iron deficiency, hemoglobinopathies, macronutrient deficiencies, and infections such as malaria, human immunodeficiency virus (HIV), and hookworm infestation. Anemia-induced hypoxia triggers a range of vascular and structural changes in the placenta, potentially leading to complications for both the mother and the fetus. Studies have shown that placentae from anemic mothers often exhibit hypertrophy, characterized by increased placental weight relative to fetal

Adverse neonatal outcomes are associated with a sex-specific placental inflammatory profile†.

BACKGROUND AND OBJECTIVE: The placenta is crucial for fetal development; altered function is associated with complications and adverse neonatal outcomes. Our goal was to ascertain changes in the placental transcriptome in relation to neonatal outcome and fetal/placental sex. STUDY DESIGN: Seventy-two mother-baby dyads were included. Demographic, obstetrical, neonatal, and infant health data were obtained through medical charts. Adverse neonatal outcome was defined as the presence of a pulmonary, cardiac, neurological or other health complication. Bulk RNA-sequencing of placental biopsies was

Placental expression quantitative trait loci in an East Asian population.

Expression quantitative trait loci (eQTL) analysis measures the contribution of genetic variation in gene expression on complex traits. Although this methodology has been used to examine gene regulation in numerous human tissues, eQTL research in solid tissues is relatively lacking. We conducted eQTL analysis on placentas collected from an East Asian population in an effort to identify gene regulatory mechanisms in this tissue. Placentas (n = 102) were collected at the time of cesarean delivery. mRNA was extracted, sequenced with NGS, and compared with matched maternal and fetal DNA arrays per

Neutrophil infiltration leads to fetal growth restriction by impairing the placental vasculature in DENV-infected pregnant mice.

BACKGROUND: Dengue virus (DENV) infection during pregnancy increases the risk of adverse fetal outcomes, which has become a new clinical challenge. However, the underlying mechanism remains unknown. METHODS: The effect of DENV-2 infection on fetuses was investigated using pregnant interferon α/β receptor-deficient (Ifnar1-/-) mice. The histopathological changes in the placentas were analyzed by morphological techniques. A mouse inflammation array was used to detect the cytokine and chemokine profiles in the serum and placenta. The infiltration characteristics of inflammatory cells in the plac

AMPK Hyper-Activation Alters Fatty Acids Metabolism and Impairs Invasiveness of Trophoblasts in Preeclampsia.

BACKGROUND/AIMS: Preeclampsia (PE) has long been assumed to be an ischemic disease of the placenta, although there is limited evidence as to how the ischemia impacts on the placenta. AMP-activated protein kinase (AMPK) is a key regulator of cellular energy metabolism and plays an important role in a variety of ischemic diseases by enhancing energy production. The present study investigated placental metabolism in PE, and the role of AMPK in regulating trophoblast function. METHODS: placentas from normal and PE complicated pregnancies were subjected to GC-MS to identify fatty acids (FA) metabo

Integrative single-cell and cell-free plasma RNA transcriptomics elucidates placental cellular dynamics.

The human placenta is a dynamic and heterogeneous organ critical in the establishment of the fetomaternal interface and the maintenance of gestational well-being. It is also the major source of cell-free fetal nucleic acids in the maternal circulation. Placental dysfunction contributes to significant complications, such as preeclampsia, a potentially lethal hypertensive disorder during pregnancy. Previous studies have identified significant changes in the expression profiles of preeclamptic placentas using whole-tissue analysis. Moreover, studies have shown increased levels of targeted RNA tra

MiR 20a,-20b and -200c are involved in hydrogen sulfide stimulation of VEGF production in human placental trophoblasts.

UNLABELLED: Hydrogen sulfide (H2S) has been implicated to angiogenesis in various tissues. We sought to investigate the role of hydrogen sulfide (H2S) in regulating production of vascular endothelial growth factor (VEGF) proteins, the key factors of angiogenesis and vasculogenesis, in placenta. METHODS: Placental tissues were obtained from pregnant women with preeclampsia and healthy pregnant women who underwent elective cesarean section. Explants and trophoblasts were isolated from healthy placentas and treated with H2S donor and precursor. Western blotting was used to determine the levels o

Association of dysfunctional synapse defective 1 (SYDE1) with restricted fetal growth - SYDE1 regulates placental cell migration and invasion.

The transcription factor glial cells missing 1 (GCM1) regulates trophoblast differentiation and function during placentation. Decreased GCM1 expression is associated with pre-eclampsia, suggesting that abnormal expression of GCM1 target genes may contribute to the pathogenesis of pregnancy complications. Here we identified a novel GCM1 target gene, synapse defective 1 (SYDE1), which encodes a RhoGAP that is highly expressed in human placenta, and demonstrated that SYDE1 promotes cytoskeletal remodelling and cell migration and invasion. Importantly, genetic ablation of murine Syde1 results in s
