Efflux pump modulation by Montelukast and its roles in restoring antibiotic susceptibility in multidrug-resistant Staphylococcus aureus.

BACKGROUND: Staphylococcus aureus and its drug-resistant mutants are mentioned among the WHO's high-priority list of pathogens. Antibiotics like fluoroquinolones and cephalosporins are used to treat multidrug-resistant S. aureus infections. However, a higher expression of efflux pumps (NorA, NorB, and AbcA) induces multidrug resistance. The master regulator, MgrA, regulates the expression of most of these efflux pumps in S. aureus. The phosphorylation status of MgrA is determined by the cellular PknB/RsbU ratio, where PknB, a serine-threonine kinase, and RsbU, a serine-threonine phosphatase, a

Native Valve Infective Endocarditis Caused by Dual Infection of Staphylococcus aureus and Pseudomonas monteilii: A Case Report.

Infective endocarditis (IE) is most commonly associated with Gram-positive bacteria such as Staphylococcus aureus. Gram-negative causes are less common, and are typically associated with pacemakers, prosthetic valves or intravenous drug use. Pseudomonas monteilii is an environmental organism that is rarely reported in human infection, with its presence in sputum often regarded as colonisation or contamination. This case describes a 30-year-old man with no underlying structural heart disease who was found to have a 6-8mm mitral valve vegetation on echocardiography. His blood cultures grew Staph

In Silico Lead Identification of Staphylococcus aureus LtaS Inhibitors: A High-Throughput Computational Pipeline Towards Prototype Development.

The emergence of multidrug-resistant Staphylococcus aureus underscores the urgent need for novel therapeutic agents targeting essential bacterial pathways. The lipoteichoic acid synthase (LtaS) is crucial for the synthesis of lipoteichoic acid in the cell wall of Gram-positive bacteria and represents a promising and vulnerable target for antimicrobial drug development. This study employed a comprehensive computational pipeline to identify potent inhibitors of the LtaS enzyme. A library of natural compounds was retrieved from the COCONUT database and screened against the crystal structure of th

Multifunctional antibacterial micelle with targeting ability and lipase-responsibility for therapy of methicillin-resistant Staphylococcus aureus infection.

Bacterial infections frequently trigger severe inflammatory cascades accompanied by oxidative stress responses, which collectively impair tissue regeneration and exacerbate disease progression. Consequently, there is a pressing clinical need to develop therapeutic strategies capable of simultaneously eradicating pathogenic bacteria and modulating the infection microenvironment. The infected tissue microenvironment is characterized by macrophage infiltration and upregulation of folate receptor expression on their cellular surfaces. Concurrently, pathogenic bacteria actively secrete abundant lip

Prevalence and demographic correlates of Methicillin-Resistant Staphylococcus aureus (MRSA) colonization in patients undergoing total knee replacement.

BACKGROUND: Methicillin-resistant Staphylococcus aureus (MRSA) remains a significant concern in orthopedic surgery, particularly in total knee replacement (TKR), where infection can lead to severe complications. In procedures like TKR, where implants act as a foreign body and potential surface for biofilm formation, infections can lead to severe complications, including delayed healing, and implant failure, and often need multiple revision surgeries. Screening for MRSA before surgery has become a standard practice in many hospitals to reduce the risk of infection. This study aims to evaluate t

Genome-wide screen reveals glycerol-induced aminoglycoside potentiation against Staphylococcus aureus via boosting GlpK-initiated energy metabolism.

Potentiation of existing antibiotics represents a promising strategy for combating the global health crisis of antibiotic resistance. Here we report that 1 min co-treatment with glycerol markedly enhances aminoglycoside efficacy against stationary-phase Staphylococcus aureus cells. This rapid combined treatment also effectively eliminates S. aureus biofilms and persister cells, methicillin-resistant S. aureus clinical isolates in vitro, and S. aureus in a mouse skin infection model. The potentiation is achieved through rapid enhancement of proton motive force-dependent aminoglycoside uptake. A

Microwave-Excited Thermoelectric Catalysis of Organic-Inorganic Nanohybrid for Highly Effective Treatment of Staphylococcus aureus-Infected Osteomyelitis.

Osteomyelitis caused by Staphylococcus aureus (S. aureus) is difficult to cure with antibiotics or phototherapy. Microwave (MW) has become a promising method for treating deep tissue infections due to its strong penetration ability, however, the effects of MW dynamics still need to be improved to achieve rapid and effective treatment. In this work, tin selenide/polypyrrole (SnSe/PPy) nanocomposites with MW thermoelectric catalytic performance are successfully prepared, and their ideal thermal production capability stems from increased dielectric loss, including interfacial polarization and con

Staphylococcus aureus vertebral osteomyelitis: a single-centre retrospective cohort study with focus on oral flucloxacillin follow-up.

BACKGROUND: International guidelines for Staphylococcus aureus vertebral osteomyelitis recommend 6 weeks of treatment, including oral follow-up using antibiotics with high bioavailability such as a fluoroquinolone/rifampicin combination. Oral flucloxacillin is not recommended due to low bioavailability and scarce evidence. However, flucloxacillin as oral follow-up treatment is common practice in Sweden based on favourable clinical experience, good tolerability, few interactions, and low ecological impact. Our aim was to review a single-centre experience of S. aureus vertebral osteomyelitis, wi

Staphylococcus aureus in Foodborne Diseases and Alternative Intervention Strategies to Overcome Antibiotic Resistance by Using Natural Antimicrobials.

Foodborne diseases are the most common causes of illness worldwide. Bacterial pathogens, including Staphylococcus aureus, are often involved in foodborne disease and pose a serious threat to human health. S. aureus is commonly found in humans and a variety of animal species. Staphylococcal enteric disease, specifically staphylococcal food poisoning (SFP), accounts for numerous gastrointestinal illnesses, through the contamination of food with its enterotoxins, and its major impact on human health imposes a heavy economic burden in society. Commonly, antibiotics and antimicrobials are used to t

A customizable and defined medium supporting culturing of Candida albicans, Staphylococcus aureus, and human oral epithelial cells.

UNLABELLED: Candida albicans, an opportunistic oral pathogen, synergizes with Staphylococcus aureus, allowing bacteria to co-invade and systemically disseminate within the host. Studying human-microbe interactions creates the need for a universal culture medium that supports fungal, bacterial, and human cell culturing, while allowing sensitive analytical approaches such as OMICs and chromatography techniques. In this study, we established a fully defined, customizable adaptation of Dulbecco's modified Eagle medium (DMEM), allowing multi-kingdom culturing of S. aureus, C. albicans, and human or
