Distinct thick ascending limb cell types mediate divalent and monovalent ion transport.

PURPOSE OF REVIEW: The kidney thick ascending limb (TAL) is typically viewed as comprising a single cell type that expresses a Na-K-2Cl transporter and potassium channel at the apical membrane and a basolateral chloride channel. Together, these generate a lumen-positive transepithelial voltage. This review summarizes recent work revising this view substantially. RECENT FINDINGS: Very early work suggested that not all TAL cells express potassium channels at the apical membrane. More recently, it became clear that paracellular transport pathways for monovalent and divalent cations exhibit a mos

The direct-current and alternating-current transport properties of hydrogenated bilayer β12- and χ3-borophenes.

Using density functional theory and the non-equilibrium Green's function method, we investigated the structures, electronic properties, and the direct-current (DC) and alternating-current (AC) transport properties of two-dimensional (2D) hydrogenated bilayer (BL) β12- and χ3-borophenes. We found that when the H coverage is less than 60% for BL β12-borophene and 62.5% for BL χ3-borophene; hydrogenation can enhance the stability of the two BL structures and change the interlayer interaction of BL χ3-borophene from the van der Waals bonds to the covalent bonds. The electronic properties of hydrog

SLC40A1 (iron transporter): mechanistic regulation, role in disease pathogenesis, and prospects for targeted therapy.

Solute carrier family 40 member 1 (SLC40A1) encodes ferroportin 1 (FPN1), the only known iron efflux transporter in mammalian cells, which is critical for maintaining systemic iron homeostasis and cellular iron balance. SLC40A1 plays a key role in regulating iron homeostasis and is involved in the pathogenesis of inflammatory, fibrotic, neurodegenerative diseases, and cancer, with dysfunction linked to mutations or epigenetic silencing. Its biological functions and regulatory mechanisms are complex and diverse. This article provides an overview of SLC40A1's molecular and biological properties,

Terahertz-induced Berry curvature control of spin-selective transport in chiral DNA molecules.

CONTEXT: Chirality-induced spin selectivity (CISS) represents a remarkable quantum phenomenon whereby electron transmission through chiral molecules, such as DNA, becomes intrinsically spin-polarized even in the absence of magnetic fields. Despite extensive experimental verification of static CISS effects, achieving dynamic control over spin polarization remains an open challenge. Terahertz (THz) radiation offers a promising route to externally modulate molecular electronic structure on sub-picosecond timescales. In this study, we develop a theoretical model that unifies THz-driven Floquet dyn

Bee pollen-derived peptide with dual DPP-IV Inhibition and glucose transport modulation.

This study investigates the potential of bee pollen protein hydrolysate (BPPH) as a natural source of bioactive peptides capable of inhibiting dipeptidyl peptidase IV (DPP-IV) for the management of type 2 diabetes mellitus (T2DM), a metabolic disorder characterized by insulin resistance and hyperglycemia. BPPH was generated through pepsin–pancreatin digestion, followed by ultrafiltration and RP-HPLC purification. LC-Q-TOF-MS/MS analysis identified Ala-Thr-His-Ala-Leu-Leu-Ala (ATHALLA, AA-7) as a predominant peptide associated with DPP-IV inhibitory activity. AA-7 exhibited strong DPP-IV inhibi

Inhibition of the serotonin transporter and risk of heart valve disease: A systematic review and meta-analysis.

BACKGROUND: The serotonin transporter (SERT) plays an essential role in regulating a wide range of physiological and psychological processes, including neurotransmitter homeostasis, circadian rhythm, sleep regulation, platelet function, immune modulation, mood and emotions. Although SERT was initially studied in the context of neurological and psychiatric disorders, growing evidence highlights how its off-target exogenous inhibition, and the resulting dysregulation of serotonin signaling, can promote valvular interstitial cell proliferation, extracellular matrix remodeling, and fibrosis, there

Experimental study on settling, transport, and packing mechanisms of proppants with different shapes in a physical model.

Hydraulic fracturing, as a critical technology for reservoir stimulation, has functioned as a central stimulation technique in the large-scale development of shale gas and coalbed methane. During hydraulic fracturing operations, the movement and distribution of proppants within fractures serve as the key determinant of both fracture conductivity created by fracturing and long-term sustained production performance. Meanwhile, the settling and packing mechanisms of proppants greatly affects proppant transportation efficiency and distribution within fractures, which in turn determines the level o

Accurate trajectory inference in time-series spatial transcriptomics with structurally-constrained optimal transport.

New experimental and computational methods use genetic or gene expression observations with single cell resolution to study the relationship between single-cell molecular profiles and developmental trajectories. Most tissues contain spatially contiguous regions that develop as a unit, such as follicles in the ovary, or tubules and glomeruli in the kidney. We find that existing approaches designed to use time series spatial transcriptomics (ST) data produce biologically incoherent trajectories that fail to maintain these structural units over time. We present Spatiotemporal Optimal transport wi

Source-Only Transportability of Engineered ECG Features for Healthy-Versus-Myocardial Infarction Classification.

Background/Objectives: Electrocardiogram (ECG)-based myocardial infarction (MI) classifiers may achieve high internal validation performance but show reduced performance when applied to data from another source. The task is a controlled binary healthy-versus-MI benchmark and is not intended to represent real-world chest-pain triage or autonomous clinical deployment. This study evaluated the source-only transportability of engineered 12-lead ECG feature families for binary healthy-versus-MI classification across a cardiologist-annotated hospital dataset and PTB-XL. Methods: The hospital dataset

Thermophysical and Transport Properties of Ethanolammonium Carboxylate Protic Ionic Liquids in Water and Dimethyl Sulfoxide.

Volumetric and transport properties of protic ionic liquids (PILs) in molecular solvents provide essential insights into solute-solute and solute-solvent interactions in solution. In this study, the density and electrical conductivity of binary mixtures of ethanolammonium carboxylate PILs, ethanolammonium acetate, propionate, butyrate, and pentanoate with water and dimethyl sulfoxide (DMSO) were measured across the entire mole fraction range at 298 K. Excess molar volumes (V E) were derived from density (ρ) data and correlated using the Redlich-Kister polynomial. The V E values were negative o

Beyond distance: quantifying point cloud dynamics with persistent homology and dynamic optimal transport.

We introduce a framework for analysing topological tipping in time evolutionary point clouds by extending the recently proposed topological optimal transport (TpOT) distance. While TpOT unifies geometrical, homological and higher-order relations into one metric, its global scalar distance can obscure transient, localized structural reorganizations during dynamic phase transitions. To overcome this limitation, we present a hierarchical dynamic evaluation framework driven by a novel topological and hypergraph reconstruction strategy. Instead of directly interpolating abstract network parameters,

Weak electron-phonon coupling is insufficient to generate significant CISS in two-terminal transport.

A central open question in chiral-induced spin selectivity is whether weak electron-phonon coupling in a helical molecular junction can generate a sizable spin polarization in two-terminal transport without invoking additional strong symmetry-breaking ingredients. We address this question by implementing a self-consistent nonequilibrium Green's function (NEGF) calculation for a helical tight-binding model with spin-orbit coupling and electron-phonon interactions. The electron-phonon self-energies are evaluated self-consistently, and the transport signal is extracted using the standard magnetiz

Nature-derived exosome-like nanoparticles (NELNs) for intracellular transport and targeted RNA delivery.

As exosomes serve as pivotal mediators of intercellular communication, research on exosomes has expanded markedly from its conventional focus on mammalian sources to the burgeoning field of nature-derived exosome-like nanoparticles (NELNs). Endowed with favorable biocompatibility, low immunogenicity, and wide, readily accessible sources, NELNs have emerged as promising delivery vehicles to address long-standing bottlenecks in RNA-based therapeutic delivery. However, their clinical translation is significantly impeded by the lack of a consensus definition and standardized manufacturing workflow

Gut-eye axis and transporter-mediated strategies: enhancing retinal uptake of dietary nutrients.

The blood-retinal barrier (BRB) stringently restricts the delivery of dietary nutrients to retinal tissues, presenting a major bottleneck for targeted nutritional intervention. This review systematically delineates three core pathways through which food-derived bioactive compounds overcome the limited delivery efficiency of the BRB: transporter-mediated active transport across the barrier, biotransformation by gut microbiota and the liver into bioactive metabolites with superior bioavailability, and gut-retina axis-mediated remote regulation of retinal homeostasis. An innovative focus is place

Stress-Induced Mechanical Memory in Respiratory Mucus: Anisotropy, Network Reorganization, and Directional Transport.

Mucus transport is essential for lung health, as ciliated cells constantly propel mucus outward to clear bacteria, viruses, and particles. This defense relies on a material that must be elastic enough to maintain ciliary traction, but capable of reorganizing under sustained directional loading. How the mucin network reconciles these demands, and whether it retains a memory of the stresses it experiences, remains poorly understood. Here we show that lung mucus develops a persistent, direction-dependent mechanical asymmetry under physiologically relevant stress-a mechanical memory encoded in the

In vivo monitoring of the impact of microplastic particle size on the absorption and transport of Pyrethroids in Malabar spinach.

Microplastics' (MPs) role as pesticide carriers in vegetables remains unclear, particularly regarding size-dependent transport mechanisms. Herein, the fate of eight pyrethroids (PYs) in Malabar spinach was differentially affected by polystyrene microplastics (PS-MPs), as revealed using a novel hollow covalent organic framework-based solid-phase microextraction (HCOF-SPME) technology. This method enabled precise in vivo tracking of pesticide fate while minimizing vegetable sacrifice. Experimental results revealed that PS-MPs enhanced PYs accumulation in roots via adsorption, with smaller MPs (1

Modeling chimeric antigen receptor response at the single-cell level with conditional optimal transport.

Chimeric antigen receptor (CAR) T cell therapy is a promising cancer immunotherapy; however, several challenges hamper its clinical efficacy. Although new computational models are attempting to explore the vast combinatorial design space of CAR components and suggest novel designs, challenges remain in capturing cellular heterogeneity as well as generalizing to unseen CAR variants. Here, we introduce an optimal transport (OT)-based framework designed to predict responses to CAR expression at the single-cell level, including variants that have not been experimentally tested. Our model accuratel

Tumor-on-chip platforms for transport phenotyping: decoding CAF-driven barriers to drug delivery.

Physical barriers within solid tumors constitute a fundamental but often overlooked mechanism of therapeutic resistance, contributing to the poor success rate of cancer drug translation. Therapeutic molecules often fail to reach their intended targets due to mass-transport limitations imposed by the remodeled, spatially heterogeneous tumor microenvironment (TME). Cancer-associated fibroblasts (CAFs) drive dynamic remodeling of the extracellular matrix (ECM), generating local variations in stiffness, porosity, and intrinsic permeability that, together, shape evolving transport phenotypes that g
