Glucosylceramide synthase is required for C6-ceramide nanoliposome-induced organelle stress and cell death.

Sphingolipids are bioactive lipids that regulate key signaling pathways both directly as ligands and through membrane re-organization. Ceramide sits at the center of this network and is considered pro-death in many contexts, making ceramide accumulation an attractive therapeutic strategy. Given the wide-ranging regulation of cellular responses this network exerts, better understanding ceramide metabolism may promote therapeutic efficacy of sphingolipid-based therapeutics. Ceramide glycosylation, catalyzed by glucosylceramide synthase (GCS; UGCG), is in turn widely regarded as a detoxification

Plant-Derived Nanovesicles: From Biogenesis and Lipid Fingerprints to Gut-Organ Axis and Smart Biomaterial Platforms.

Plant-derived nanovesicles (PDNVs) are natural bioactive nanocarriers, yet existing reviews remain fragmented across immunology, drug delivery, and engineering. This review provides the first integrated framework bridging plant cell biology, immunology, materials science, and nanomedicine to present a complete biogenesis-composition-immunomodulation-engineering-translation continuum for PDNVs. We systematically compare three biogenesis pathways-multivesicular body (MVB), plasma membrane budding, and the plant-specific exocyst-positive organelle (EXPO) pathway-and delineate the unique lipid fin

Malonic acid promotes glycolytic activity in cumulus cells and improves the quality of bovine oocytes matured in vitro.

Malonic acid (MA) is a dicarboxylic acid that inhibits the mitochondrial electron transport chain. Cumulus cells surrounding human oocytes that achieve conception after in vitro fertilization (IVF) contain higher levels of MA than those that do not. Although MA has no effect on embryo developmental culture, the effects of MA on cumulus cells and oocyte quality remain unclear. In this study, the addition of 10 μM MA to the in vitro maturation (IVM) medium cultured cumulus cell-oocyte complexes improved embryo production efficiency, meeting multiple conception-related criteria after IVF (P < 0.0

Site-1 protease-mediated cholesterol metabolism is essential for lymphatic development in mice.

Recent evidence suggests that cellular metabolism, including glycolysis and fatty acid synthesis in lymphatic endothelial cells (LECs), plays essential roles in developing functional lymphatic systems. Site-1 protease (S1P) proteolytically activates membrane-bound latent transcription factor sterol regulatory element-binding proteins (SREBPs), which are required to induce lipid biosynthesis. In this study, we generated mice with pan-endothelial or LEC-specific deficiency of either S1P or SREBP2. Mouse embryos with pan-endothelial deletion of S1P showed defective lymphatic vessel migration in s

Evidence and unknowns for the relevancy of applying current parenteral nutrition support recommendations among infants born less than 750 g or younger than 25 weeks' gestation: A narrative review.

With advancements in neonatal care, the viability of extremely low-birth-weight (ELBW) infants, especially those born extremely preterm, is increasing. However, specific recommendations for managing parenteral nutrition (PN) support in nanopreterm infants (<750 g or <25 weeks' gestation) are lacking. We aim to evaluate current recommendations and highlight considerations for applying them to nanopreterm infants. The author team used English-language articles related to nutrition in ELBW with emphasis on nanopreterm infants, along with studies on fetal growth and metabolism. Current PN support

A comprehensive review of challenges and advances in exosome-based drug delivery systems.

Exosomes or so-called natural nanoparticles have recently shown enormous potential for targeted drug delivery systems. Several studies have reported that exosomes as advanced drug delivery platforms offer efficient targeting of chemotherapeutics compared to individual polymeric nanoparticles or liposomes. Taking structural constituents of exosomes, viz., proteins, nucleic acids, and lipids, into consideration, exosomes are the most promising carriers as genetic messengers and for treating genetic deficiencies or tumor progression. Unfortunately, very little attention has been paid to the facto

Beyond Mortality: Exploring the Influence of Plant Phenolics on Modulating Ferroptosis-A Systematic Review.

Ferroptosis is a recently discovered type of programmed cell death that is mechanistically different from other types of programmed cell death such as apoptosis, necroptosis, and autophagy. It is characterized by the accumulation of intracellular iron, overproduction of reactive oxygen species, depletion of glutathione, and extensive lipid peroxidation of lipids in the cell membrane. It was discovered that ferroptosis is interconnected with many diseases, such as neurodegenerative diseases, ischemia/reperfusion injury, cancer, and chronic kidney disease. Polyphenols, plant secondary metabolite

Coagulation protease-induced extracellular vesicles: their potential effects on coagulation and inflammation.

Coagulation proteases, in addition to playing an essential role in blood coagulation, often influence diverse cellular functions by inducing specific signaling pathways via the activation of protease-activated receptors (PARs). PAR activation-induced cellular effects are known to be cell-specific as PARs are expressed selectively in specific cell types. However, a growing body of evidence indicates that coagulation protease-induced PAR activation in a specific cell type could affect cellular responses in other cell types via communicating through extracellular vesicles (EVs) as coagulation pro

Advances in Extracellular-Vesicles-Based Diagnostic and Therapeutic Approaches for Ocular Diseases.

Extracellular vesicles (EVs) are nanoscale membrane vesicles of various sizes that can be secreted by most cells. EVs contain a diverse array of cargo, including RNAs, lipids, proteins, and other molecules with functions of intercellular communication, immune modulation, and regulation of physiological and pathological processes. The biofluids in the eye, including tears, aqueous humor, and vitreous humor, are important sources for EV-based diagnosis of ocular disease. Because the molecular cargos may reflect the biology of their parental cells, EVs in these biofluids, as well as in the blood,

Extracellular vesicles: effectors of transfusion-related acute lung injury.

Respiratory transfusion reactions represent some of the most severe adverse reactions related to receiving blood products. Of those, transfusion-related acute lung injury (TRALI) is associated with elevated morbidity and mortality. TRALI is characterized by severe lung injury associated with inflammation, pulmonary neutrophil infiltration, lung barrier leak, and increased interstitial and airspace edema that cause respiratory failure. Presently, there are few means of detecting TRALI beyond clinical definitions based on physical examination and vital signs or preventing/treating TRALI beyond s

Exosomes released by Brucella-infected macrophages inhibit the intracellular survival of Brucella by promoting the polarization of M1 macrophages.

Exosomes, membrane vesicles released extracellularly from cells, contain nucleic acids, proteins, lipids and other components, allowing the transfer of material information between cells. Recent studies reported the role of exosomes in pathogenic microbial infection and host immune mechanisms. Brucella-invasive bodies can survive in host cells for a long time and cause chronic infection, which causes tissue damage. Whether exosomes are involved in host anti-Brucella congenital immune responses has not been reported. Here, we extracted and identified exosomes secreted by Brucella melitensis M5
