Neuron-tumor crosstalk in cancer: molecular mechanisms and translational advances.

Tumor neuron hijack is a malignant adaptive program whereby tumor cells recruit, physically engage and functionally reprogram the peripheral and central nervous system within the local microenvironment and host macroenvironment; this process not only exploits neuro-immune regulatory machineries, neural endocrine signaling, and nutrient supply for sustaining tumor growth, invasion, metastasis, immune escape and treatment resistance, but also closely involves the induction and amplification of cancer-associated pain, a common and debilitating manifestation of the host's pathological response to

Inferring effective neuronal circuits via network flux counting.

Extracting circuit mechanisms from neuronal population activity is challenging due to the heterogeneous neuronal properties and diverse strengths in synaptic connections. Standard inference methods, such as Generalized Linear Models (GLMs), typically regress for parameters on all neuronal activity at once. Such a global fitting approach can face identifiability difficulties-for example, where the statistical estimation of strong, opposing weights becomes ill-conditioned in excitatory-inhibitory balanced networks. Here, we introduce FLux-based Effective Coupling (FLEC), a framework that maps sp

NEuRT: A Transformer-Based Model for Explainable Neuronal Activity Analysis.

The study of neuronal activity is essential for understanding brain function and its alterations in neurodegenerative diseases. Advances in in vivo imaging have enabled real-time observation of neuronal dynamics, but classical statistical methods struggle to capture the complex, time-dependent interactions within neuronal networks. Machine learning offers promising solutions for analyzing high-dimensional neuronal data, yet their application in neuroscience remains limited. Here, we introduce NEuRT, a Bidirectional Encoder Representations from Transformers (BERT)-based model adapted for neuron

Behavioral and neuronal dynamics in a rat model of obsessive-compulsive disorder.

While cognitive and behavioral manifestations of obsessive-compulsive disorder (OCD) are well known, the neuronal dynamics underlying these symptoms remain poorly understood. Theoretical work suggests that changes in the attractor dynamics of neuronal networks towards increased stability and decreased flexibility might cause behavioral and cognitive symptoms of OCD. We used chronic treatment with the D2 and D3 dopamine receptor agonist quinpirole as a rat model of the disease. In this model, we examined changes in behavioral dynamics and, in a parallel experiment, changes in organization of ne

Feeding decision-making by a single neuron via disparate neurotransmitters.

Animals use gustatory information to decide whether to ingest nutritious substances or avoid toxic ones. Although certain neurons in the gustatory circuits respond to both aversive and appetitive signals, how these neurons resolve inputs with opposing valences is unknown. Here, we examine how the Drosophila melanogaster neuropeptide leucokinin (LK) affects gustatory information processing to elicit the appropriate feeding behaviors. We identify the subesophageal LK neurons (SELKs) as downstream synaptic partners of gustatory receptor neurons and show that these two groups are functionally conn

Temporal drift of sleep-wake representations in hypothalamic neuronal ensembles.

Correlative and causal evidence implicates distinct genetically-defined and evolutionary-conserved hypothalamic neurons in regulating wakefulness, non-rapid eye movement (NREM), and rapid eye movement (REM) sleep. The prevailing view is that these circuits govern sleep-wake states by recruiting stable, invariant neuronal substrates, yet, this remains unknown. Here, we show that inhibitory, excitatory, hypocretins/orexins-, and melanin concentrating hormone-expressing neurons in hypothalamus did not exhibit stable state-specific activities using longitudinal single cell calcium imaging in freel

[From Silent Spring to neuronal silence : pesticides and neurodegenerative diseases].

Over sixty years ago, Silent Spring, the groundbreaking book by biologist Rachel Carson, raised global awareness of the dangers of uncontrolled pesticide use and contributed to the ban on the use of dichlorodiphenyltrichloroethane (DDT) in the USA. Many years later, it is clear that most of the issues raised in this book remain highly relevant. Today, almost our entire environment is contaminated, sometimes permanently, by pesticides. Yet the chronic toxicity of these substances is still poorly understood. In recent years, a growing body of data seems to link environmental exposure to pesticid

Huntingtin, signaling endosomes and neuronal survival in health and Huntington disease.

The axonal transport of organelles mediates several functions throughout the lifespan of neurons. Over the past years, the role of endosomes, a specific type of trafficking organelle, has been extensively investigated for their increasing relevance during development, maturation and maintenance of neuronal networks. Endosome formation results from membrane invagination events driven by endocytic pathways. Following their formation, endosomes undergo maturation steps, and their fate is determined by the recruitment of specific cytosolic proteins to their membrane. Early endosomes may be recycle

APOE4 disrupts the central dogma by arresting neuronal proteome dynamics.

Apolipoprotein E4 (APOE4) is the strongest genetic risk factor for late-onset Alzheimer's disease and promotes neuronal dysfunction through incompletely understood mechanisms. Here, we integrated transcriptomic, translatomic, and proteomic profiling of isogenic APOE3 and APOE4 human iPSC-derived neurons and found that APOE4 fundamentally impairs neuronal proteome renewal. Although transcriptional changes were modest, APOE4 disrupted ribosome occupancy, altered translational dynamics, and uncoupled protein abundance from transcript levels. Proteome-wide turnover measurements revealed a global e

MARK4 regulates GLUT3 surface expression and neuronal glucose uptake.

Brain neurons rely predominantly on glucose as an energy source and express glucose transporter 3 (GLUT3) as their principal glucose transporter. Unlike GLUT4, which undergoes insulin-stimulated membrane translocation in peripheral tissues, GLUT3 mediates largely constitutive and relatively insulin-independent glucose uptake in neurons. Although GLUT3 is essential for neuronal functions, its regulatory mechanisms are not fully understood. Microtubule affinity-regulating kinase 4 (MARK4), a member of the AMP-activated protein kinase-related kinase family, has been implicated in Alzheimer's dise

Regulatory role of neuronal guidance proteins in spinal cord injury.

Spinal cord injury is a severe neurological condition with limited neuronal regeneration and functional recovery. Currently, no effective treatments exist to improve spinal cord injury prognosis. Neuronal guidance proteins are a diverse group of molecules that play crucial roles in axon and dendrite growth during nervous system development. Increasing evidence highlights their regulatory functions in spinal cord injury. This review provides a brief overview of the modulation patterns of key neuronal guidance proteins in neuronal axon growth during nervous system formation and subsequently focu

CDK3 induces neuronal death and brain atrophy in Alzheimer's disease.

Progressive neuronal loss and brain atrophy are principal determinants of cognitive decline in Alzheimer's disease (AD), yet most mouse models fail to recapitulate these features. Here we identify cyclin-dependent kinase 3 (CDK3) as a key driver of neurodegeneration in AD. CDK3 is elevated in human AD brains and correlates with disease severity. As laboratory mice carry a nonfunctional Cdk3 mutation, we generated two models with restored CDK3 activity and then crossed to AD backgrounds. Both models exhibited substantial neuronal loss, hippocampal atrophy, and cognitive deficits. Single-nucleus

Molecular hallmarks of excitatory and inhibitory neuronal resilience to Alzheimer's disease.

BACKGROUND: A significant proportion of individuals maintain cognition despite extensive Alzheimer's disease (AD) pathology, known as cognitive resilience. Understanding the molecular mechanisms that protect these individuals could reveal therapeutic targets for AD. METHODS: This study defines molecular and cellular signatures of cognitive resilience by integrating bulk RNA and single-cell transcriptomic data with genetics across multiple brain regions. We analyzed data from the Religious Order Study and the Rush Memory and Aging Project (ROSMAP), including bulk RNA sequencing (n = 631 indivi

The protein interactome of the Neuron Specific Gene family (NSG1-3).

The Neuron-Specific Gene (NSG) family members (NSG1-3) play critical and diverse roles in neuronal protein trafficking, but their precise molecular functions remain poorly understood. Here, we employed proximity-labeling proteomics to map the interactomes of each NSG protein. Unlabeled mass spectrometry identified over 1,000 significantly enriched interactors compared with a cytoplasmic control, revealing substantial overlap between NSG1 and NSG2, and a more divergent profile for NSG3. Gene ontology and KEGG pathway analysis confirmed established associations with glutamatergic synapses and en

UCP2 mediates mitochondrial dynamics to induce AgRP neuronal activity.

OBJECTIVES: The hypothalamic agouti-related protein (AgRP)- expressing neurons regulate feeding and whole-body energy homeostasis. A growing body of evidence indicates that changes in mitochondrial dynamics, such as fission and fusion, play a crucial role in regulating AgRP neuronal activity. However, the mechanisms underlying this process remain to be elucidated. Here, we showed a role of mitochondrial UCP2-mediated mitochondrial dynamics in AgRP neurons in regulating AgRP neuronal activity and fasting-induced feeding behavior. METHODS: We analyzed mitochondrial morphology, expression of act

Single-Neuron Responses to Odor-Related Words in the Human Amygdala.

Human imaging studies suggest that visually presented words are processed by distributed networks beyond classical language areas, reflecting the properties related to their meanings. Based on human single-neuron recordings, we investigated whether and how the odor aspect of words is processed in mediotemporal lobe regions involved in olfactory perception. We analyzed ensemble activity in response to odor-related versus control words in the piriform cortex, amygdala, hippocampus, entorhinal cortex, and parahippocampal cortex and identified stimulus-responsive and odor-associated neurons.
