Microrobots for Precision Diagnosis and Treatment in the Digestive System: A Review of Actuation Mechanisms, Structural Design, Preclinical and Translational Applications.

Because lesions associated with digestive system diseases are often deeply seated, embedded within complex luminal milieus, and protected by substantial local delivery barriers, conventional diagnostic and therapeutic modalities remain constrained in their targeting capability, minimal invasiveness, and precision. Microrobots, leveraging micro-scale motion, active navigation, programmable controllability, and theranostic integration, open a new avenue for the precise diagnosis and treatment of digestive system diseases. Here, we review the major actuation modalities and structural designs of m

Carbohydrate polymer-based biomaterials for light-triggered Photo-Immunotherapy: Design strategies, immune mechanisms, and translational outlook.

Recent advances in biomaterials have highlighted the immense potential of carbohydrate polymers (CHPs) as biocompatible, multifunctional platforms for cancer therapy. Indeed, the structural diversity, tunable chemistry, and inherent biological affinity of these polymers enable the precise design of systems for immune modulation. Moreover, light- and laser-driven strategies have emerged as powerful tools for achieving spatiotemporal control over therapeutic activation. Therefore, integrating CHPs with photothermal or photodynamic nanocomponents enables these hybrid systems to induce localized t

Repetitive Transcranial Magnetic Stimulation for Peripheral Neuropathic Pain: From Cortical Analgesia to Phenotype-Stratified Clinical Translation.

Peripheral neuropathic pain (PNP) arises from lesions or diseases of the somatosensory system and remains difficult to treat because peripheral nerve injury is coupled to spinal, thalamic, cortical and affective network changes. Repetitive transcranial magnetic stimulation (rTMS) can engage cortical pain-modulation circuits without implantation or systemic drug exposure, but the field needs a clearer account of which peripheral neuropathic phenotypes, targets and trial designs can convert transient neuromodulation into clinically durable benefit. This narrative, non-systematic review synthesiz

Translational reprogramming of TGF-β signaling via TRMT61A-mediated tRNA m1A drives prostatic fibrosis and hyperplasia.

Dysregulation of the epitranscriptomic landscape is closely linked to pathological proliferation, but its specific role in benign prostatic hyperplasia (BPH) remains unclear. Here, we identify the tRNA methyltransferase TRMT61A as a critical driver of BPH progression. We found that TRMT61A and global N1-methyladenosine (m1A) levels are aberrantly upregulated in human BPH tissues. Functionally, TRMT61A knockdown potently suppresses prostate cell proliferation and reduces stromal fibrosis, inducing G1 cell cycle arrest and reversing pathological remodeling both in vitro and in vivo. Blueberry Bioactives as Adjunctive Nutritional Strategies for Pediatric Neurodevelopmental and Emotional-Behavioral Health: Mechanisms, Evidence, and Translational Challenges.

The rising prevalence of neurodevelopmental, emotional, and behavioral disorders in children has prompted interest in dietary strategies that target neuroinflammation, oxidative stress, and gut dysbiosis. Blueberries (Vaccinium spp.) contain substantial amounts of anthocyanins and other neuroactive polyphenols that may confer neuroprotective effects. We summarize the literature published between 2016 and 2025 to examine how the bioactives in blueberries affect symptoms relevant to children with diagnosed neurodevelopmental or emotional-behavioral disorders, including ADHD, mood problems, and c

Pyroptosis in triple-negative breast cancer: From inflammasome signaling to tumor immune remodeling and translational therapeutic strategies.

Triple-negative breast cancer (TNBC) remains an aggressive breast cancer subtype marked by molecular heterogeneity, immune evasion, limited actionable targets, and variable responses to immune-based therapy. Pyroptosis, a gasdermin-mediated inflammatory form of regulated cell death, may link tumor-cell injury to remodeling of the tumor immune microenvironment (TIME). However, its role in TNBC should not be interpreted as uniformly antitumor. Instead, pyroptotic signaling represents a context-dependent inflammatory process shaped by inflammasome activation, gasdermin cleavage, cellular source,

Preclinical translational screening of palladium(II)-porphyrin photosensitizers across human and Oncopig bladder cancer cell lines.

BACKGROUND: Bladder cancer remains a highly recurrent malignancy with limited long-term response to conventional therapies. The development of new treatments is also hindered by the lack of preclinical models that accurately reproduce human disease. In this context, Oncopig-derived models provide a translationally relevant platform for evaluating candidate therapies before progression to in vivo studies. METHODS: Here, we performed a preclinical in vitro screening of three palladium(II)-porphyrins, 3-Pd(PPh3), 3-Pd(dppf), and 3-Pd(PEPSI), in human (T24, RT4, 5637) and Oncopig-derived bladder

Discriminating single-amino acid differences and post-translational modifications in negatively charged peptides with α-hemolysin nanopores.

Effective and precise detection of peptides and their post-translational modifications (PTMs) is crucial for biomedical research but remains technically challenging. Here, we present a nanopore sensing strategy that enables label-free analysis of negatively charged peptides. By introducing arginine residues into the nanopore and using amino-cyclodextrin as a molecular adaptor, the system enables unambiguous discrimination of short negatively charged homopeptides differing by only a single residue. Building on this capability, the platform further demonstrates high sensitivity for detecting rep

Integrating Experimental Physicochemical Data with PBPK Modeling to Guide the Translational Development of the Antifungal 1,3,4-Oxadiazole Derivative LMM6.

The rising mortality of invasive fungal infections highlights an urgent need for novel antifungal agents with improved translational profiles. The 1,3,4-oxadiazole derivative LMM6 has emerged as a promising candidate. Therefore, this study aimed to build a comprehensive biopharmaceutical profile of LMM6 and apply PBPK modeling as a rational tool in early drug development to evaluate its pharmacokinetic feasibility in mice and humans. In vitro biopharmaceutical experiments assessed solubility (shake-flask method) in compendial and biorelevant media, alongside experimental logP determination. AD

Natural polysaccharides as multi-target therapeutic candidates for Alzheimer's disease: Mechanisms, structure-activity relationships, and translational perspectives.

BACKGROUND: Alzheimer's disease (AD) is increasingly recognized as a systems-level disorder involving amyloid-β (Aβ) deposition, tau pathology, oxidative stress, mitochondrial dysfunction, neuroinflammation, synaptic impairment, and microbiota-gut-brain axis dysregulation. Although previous reviews have summarized the anti-AD effects of natural polysaccharides according to individual pathological pathways, an integrated framework linking polysaccharide structure, gut microbial metabolism, peripheral immune regulation, and central AD pathology remains insufficiently developed. Stimuli-Responsive Biomimetic Nanomedicines for Targeted Therapy in Ischemic Stroke: Design Principles, Preclinical Evidence and Translational Challenges.

Ischemic stroke (IS) is a complex cerebrovascular disease with multifactorial etiology and pathological mechanisms, characterized by high morbidity, disability, and mortality rates. Although mechanical thrombectomy, intravenous thrombolysis, and neuroprotective interventions have improved acute management, effective brain-targeted delivery remains limited by the blood-brain barrier, short therapeutic windows, heterogeneous ischemic lesions, and secondary injury after reperfusion. Biomimetic nanomedicines have emerged as promising platforms for IS therapy because they can inherit biological fun

Phytochemical-driven angiogenesis and mucosal regeneration in gastric ulcer healing: molecular mechanisms, preclinical evidence, and translational perspectives.

Gastric ulcer remains a significant global gastrointestinal condition characterized by disruption of the gastric mucosal barrier, attributed to oxidative stress, inflammation, Helicobacter pylori infection, nonsteroidal anti-inflammatory drugs (aspirin, indomethacin, diclofenac), and compromised tissue repair. While traditional therapies effectively reduce acid secretion and eliminate H. pylori, they often do not actively promote mucosal regeneration and angiogenesis, both of which are crucial for the complete healing of ulcers and the prevention of recurrence. This review integrates molecular

Candida albicans in Oral Squamous Cell Carcinoma: From Microbial Dysbiosis to Tumor-Promoting Mechanisms and Translational Opportunities.

Oral squamous cell carcinoma (OSCC) remains a major global health burden with limited improvement in survival rates. While traditional risk factors such as tobacco and alcohol are well established, increasing evidence highlights the role of the oral microbiome in carcinogenesis. Among microbial species, Candida albicans (C. albicans) has emerged as a potential contributor to tumor-promoting processes. Clinical studies consistently report increased fungal colonization in oral potentially malignant disorders and OSCC, with associations to disease severity and recurrence. Mechanistically, C. albi

Macrophage-fibroblast communication driven by biomechanical stress in the pulmonary profibrotic niche: From signalling transduction to clinical translation.

Pulmonary (interstitial) fibrosis, the terminal pathological stage of various interstitial lung diseases, remains a major therapeutic challenge. The three approved antifibrotic agents - pirfenidone, nintedanib, and nerandomilast - receive only moderate endorsements in current guidelines, as they decelerate rather than arrest or reverse fibrotic remodelling, and none directly engages the mechanical dysregulation of the extracellular matrix (ECM) that underlies fibrogenesis. Macrophages and fibroblasts, two highly plastic cell populations, are the principal orchestrators of the pulmonary profibr

Copper Metabolism-Related Cell Death in Kidney Diseases: Molecular Mechanisms, Disease-Specific Evidence, and Translational Implications.

Copper is essential for mitochondrial respiration, antioxidant defense, extracellular matrix maturation, and cellular signaling, yet disturbances in its abundance or intracellular distribution can damage the kidney through mechanistically distinct pathways. Cuproptosis is a specific copper-dependent form of regulated cell death in which copper binds lipoylated mitochondrial proteins, promotes aggregation of tricarboxylic acid cycle components, destabilizes iron-sulfur cluster proteins, and elicits FDX1- and protein lipoylation-dependent proteotoxic stress. This mechanism should be distinguishe

A translational model of MASLD-associated HFpEF defines mitochondrial dysfunction and cardiac plasticity during disease progression and regression.

BACKGROUND: Metabolic dysfunction-associated steatotic liver disease (MASLD) and its advanced form, MASH, are closely linked to cardiac dysfunction, particularly heart failure with preserved ejection fraction (HFpEF). However, the mechanisms underlying MASLD-associated HFpEF and its reversibility remain poorly understood, largely due to the lack of robust preclinical models. Here, we established a translational model of MASLD-associated cardiac dysfunction that recapitulates the key features of human HFpEF. We applied functional and transcriptomic analyses of the left ventricle (LV) to define
