The Copper Chaperone CcsA, Coupled with Superoxide Dismutase SodA, Mediates the Oxidative Stress Response in Aspergillus fumigatus.

Superoxide dismutases (SODs) are important metalloenzymes that protect fungal pathogens against the toxic effects of reactive oxygen species (ROS) generated by host defense mechanisms during the infection process. The activation of Cu/Zn-SOD1 is found to be dependent on copper chaperone for SOD1 (Ccs1). However, the role of the Ccs1 ortholog in the human pathogen Aspergillus fumigatus and how these SODs coordinate to mediate oxidative stress response remain elusive. Here, we demonstrated that A. fumigatus CcsA, a Saccharomyces cerevisiae Ccs1 ortholog, is required for cells in response to oxid

Kinetics of neomycin release from polylactide spheres and its antimicrobial activity.

BACKGROUND: Neomycin is a natural aminoglycoside antibiotic produced by actinomycete Streptomyces fradiae. It exerts bacteriostatic and bactericidal activity against Gram-negative bacteria, certain Gram-positive bacteria and Mycobacterium tuberculosis. Neomycin inhibits the biosynthesis of bacterial proteins by impairing their life functions, leading to death of cells. OBJECTIVES: To examine the effect of molecular weight of polylactide (PLA), the applied stabilizer as well as mixing speed used in the encapsulation process on the size of obtained spheres. Examination of the kinetics of neomyc

Epidemiology and Diagnosis of Mucormycosis: An Update.

Mucormycosis is an angioinvasive fungal infection, due to fungi of the order Mucorales. Its incidence cannot be measured exactly, since there are few population-based studies, but multiple studies have shown that it is increasing. The prevalence of mucormycosis in India is about 80 times the prevalence in developed countries, being approximately 0.14 cases per 1000 population. Diabetes mellitus is the main underlying disease globally, especially in low and middle-income countries. In developed countries the most common underlying diseases are hematological malignancies and transplantation. Τhe

Copper in Wood Preservatives Delayed Wood Decomposition and Shifted Soil Fungal but Not Bacterial Community Composition.

Copper-based fungicides are routinely used for wood and plant protection, which can lead to an enrichment of copper-tolerant microbial communities in soil. To investigate the effect of such wood preservatives on the soil fungal and bacterial community compositions, five different vineyard and fruit-growing soil environments were evaluated using incubation studies over time. Pine sapwood specimens were impregnated with either water or different biocide treatment solutions containing a mixture of copper, triazoles, and quaternary ammonium compounds (CuTriQAC), a mixture of triazoles and quaterna

A Computational Modeling Approach Predicts Interaction of the Antifungal Protein AFP from Aspergillus giganteus with Fungal Membranes via Its γ-Core Motif.

Fungal pathogens kill more people per year globally than malaria or tuberculosis and threaten international food security through crop destruction. New sophisticated strategies to inhibit fungal growth are thus urgently needed. Among the potential candidate molecules that strongly inhibit fungal spore germination are small cationic, cysteine-stabilized proteins of the AFP family secreted by a group of filamentous Ascomycetes. Its founding member, AFP from Aspergillus giganteus, is of particular interest since it selectively inhibits the growth of filamentous fungi without affecting the viabili

Relationship between Molarity and Color in the Crystal ('Dramada') Produced by Scytalidium cuboideum, in Two Solvents.

Pigments from wood-decay fungi (specifically spalting fungi) have a long history of use in wood art, and have become relevant in modern science due to their longevity and colorfastness. They are presently under investigation as colorants for wood, bamboo, oils, paints and textiles. Major hurdles to their commercialization have been color repeatability (in that the same strain of the same species of fungus may produce different colors over time), and the binding of the pigments to glass storage containers. This is persistent as they do not naturally exist in a loose form. Due to these issues, t

Lights Off for Arbuscular Mycorrhiza: On Its Symbiotic Functioning under Light Deprivation.

Plants are often exposed to shade over different time scales and this may substantially affect not only their own growth, but also development and functioning of the energetically dependent organisms. Among those, the root symbionts such as arbuscular mycorrhizal (AM) fungi and rhizobia represent particularly important cases-on the one hand, they consume a significant share of plant carbon (C) budget and, on the other, they generate a number of important nutritional feedbacks on their plant hosts, often resulting in a net positive effect on their host growth and/or fitness. Here we discuss our

Proteomic analysis of the secretions of Pseudallescheria boydii, a human fungal pathogen with unknown genome.

Pseudallescheria boydii is a filamentous fungus that causes a wide array of infections that can affect practically all the organs of the human body. The treatment of pseudallescheriosis is difficult since P. boydii exhibits intrinsic resistance to the majority of antifungal drugs used in the clinic and the virulence attributes expressed by this fungus are unknown. The study of the secretion of molecules is an important approach for understanding the pathogenicity of fungi. With this task in mind, we have shown that mycelial cells of P. boydii were able to actively secrete proteins into the ext

The effect of alum addition on microbial communities in poultry litter.

Alum [Al(2)(SO(4))(3).14H(2)O] is a common poultry litter amendment used to decrease water-soluble phosphorus or reduce ammonia volatilization, or both. Although the physiochemical effects of alum addition have been well researched, little attention has been given to the poultry litter microbial communities. The goal of this study was to use molecular biological methods [denaturing gradient gel electrophoresis (DGGE), community cloning, and quantitative real-time PCR] to characterize general, group-specific and pathogenic microbial communities in alum (10% wt/wt) and non-alum-treated litter. Liposomal amphotericin B: what is its role in 2008?

Although our antifungal armamentarium has been enlarged recently with new azoles (voriconazole and posaconazole) and echinocandins (caspofungin, micafungin, anidulafungin), the polyenes still have an important role in antifungal strategies because of their extended antifungal spectrum and rarity of mycological resistance. The use of conventional amphotericin B deoxycholate is limited by substantial toxicity that is either infusion-related or associated with renal failure. Its lipid derivatives, particularly liposomal amphotericin B (LAmB), are less nephrotoxic while maintaining a broad antifun

Relevance of the selected cytokine release (TNF-alpha, IL-6, IFN-gamma, and IFN-alpha) to the exacerbation of bronchial asthma from airway mycotic infections. Predominant role of TFN-alpha?

Airway fungal infections are often associated in asthmatics with the exacerbation of asthma symptoms. However, the pathomechanism of this phenomenon has not been fully understood. The aim of our study was to assess whether antimycotic treatment can influence the capacity of bronchoalveolar (BAL) leukocytes to release proinflammatory cytokines, which could contribute to increase in asthma severity. Ten patients with bronchial asthma complicated by airway fungal infections (Candida albicans and/or Aspergillus fumigatus) were included in the study. Seven asthmatics were treated with systemic and

Diseases of mites.

An overview is given of studies on diseases of mites. Knowledge of diseases of mites is still fragmentary but in recent years more attention has been paid to acaropathogens, often because of the economic importance of many mite species. Most research on mite pathogens concerns studies on fungal pathogens of eriophyoids and spider mites especially. These fungi often play an important role in the regulation of natural mite populations and are sometimes able to decimate populations of phytophagous mites. Studies are being conducted to develop some of these fungi as commercial acaricides. Virus di

[Pyogenic infectious complications in recipients with allogenic kidneys: clinical and bacteriological aspects].

The bacteriological study of wound discharge indicated no changes in the structure of the microflora in the allogenic kidneys of recipients throughout the follow-up. Among microbes isolated there were prevalent gram-positive microbes whose proportion has slightly increased in the past year, with Staphylococcus, mainly epidermal Staphylococcus, which contributes to the etiology. At the same time, studies of wound discharge in the past years showed that the incidence of mixed infections had increased from 4.3 in 1989-1991 to 15.6% in 1994-1997. Bacteriological urinalysis found no great differenc

Experimental pulmonary candidiasis in modified rabbits. Histopathological, ultrastructural and enzyme cytochemical studies of tissue reactions.

The present experiment was performed in order to analyze and compare the histopathological features of Candida infection in various states of host defense capacity. The pulmonary lesions were induced by an intratracheal inoculation of 10(8) live cells of Candida albicans into each of the rabbits in the following 4 groups: 1) untreated controls, 2) animals sensitized non-specifically to bovine serum albumin (BSA), 3) those sensitized specifically to formalin-killed C. albicans cells, and 4) those treated with cyclophosphamide and mitomycin C. The animals were sacrificed at appropriate intervals

The pathogen's playbook in cancer: from oncogenesis to progression.

Globally, around 15-20% of cancers are linked to microbial infections, involving viruses, bacteria, and fungi, each with distinct pathogenic features. Advanced multi-omics technologies have confirmed tumor-specific microbial communities within tumor tissues. This review categorizes microbes by their action mechanisms in cancer biology: one group (e.g., Helicobacter pylori, HPV, Aspergillus species) directly induces tumorigenesis via DNA damage, repairing pathway disruption, oncoprotein expression, and carcinogenic metabolite production; the other group (e.g., Fusobacterium nucleatum, HIV, Cand

Design, Synthesis, and Biological Evaluation of 1,2,3-Benzothiadiazine-1,1-dioxide Derivatives as Potential Antifungal and Herbicidal Agents with Preliminary Mechanism Study.

Plant pathogenic fungi threaten crop health and yield by disrupting plant physiological and biochemical functions. In this study, we designed and synthesized a series of N-acetylhydrazine (amine)-1,2,3-benzothiadiazine-1,1-dioxide derivatives and evaluated their antifungal activity. Compound B2 showed strong activity against Rhizoctonia solani (EC50 = 0.320 μg/mL, superior to boscalid 1.25 μg/mL) and Fusarium graminearum (EC50 = 0.830 μg/mL, comparable to carbendazim 0.880 μg/mL), surpassing or comparable to commercial fungicides. Structure-activity relationship and DFT analyses highlighted th

Successful Medical Management of Pigmented Fungal Keratitis Following Vegetative Trauma.

Fungal keratitis remains a frequent cause of corneal morbidity in tropical regions, particularly after trivial trauma involving plant material. Dematiaceous fungi constitute a smaller subgroup of filamentous organisms but may present with characteristic pigmentation over the corneal lesion. A middle-aged woman with controlled diabetes who developed pain and visual blurring after minor ocular trauma came to the ophthalmology outpatient department. Slit-lamp examination showed a paracentral ulcer with feathery margins and a brownish plaque. Direct microscopy confirmed septate pigmented hyphae. F

Editorial: Accelerating antibiotic development from natural products: tackling antimicrobial resistance (AMR).

Accelerating antibiotic development from natural products. Natural products with antimicrobial activity can act through multiple mechanisms to overcome antibiotic resistance. These include directly inhibiting bacterial growth, reversing resistance by blocking efflux pumps and disrupting biofilms, and strengthening host defenses. Together, these strategies help restore antibiotic effectiveness, overcome resistance, and support more sustainable approaches to treating infections. Image created by Dr Ankita Nag in BioRender.
