Effects of tamoxifen on autosomal genes regulating ovary maintenance in adult mice.

Environmental endocrine-disrupting chemicals (EDCs), known to bind to estrogen/androgen receptors and mimic native estrogens, have been implicated as a main source for increasing human reproductive and developmental deficiencies and diseases. Tamoxifen (TAM) is one of the most well-known antiestrogens with defined adverse effects on the female reproductive tract, but the mechanisms related to autosomal gene regulation governing ovary maintenance in mammals remain unclear. The expression pattern and levels of key genes and proteins involved in maintaining the ovarian phenotype in mice were anal

Fabry disease mimicking hypertrophic cardiomyopathy: genetic screening needed for establishing the diagnosis in women.

AIMS: Fabry disease, an X-linked storage disorder caused by defective lysosomal enzyme alpha-galactosidase A activity, may resemble sarcomere-gene-associated hypertrophic cardiomyopathy (HCM). The 'cardiac variant' of Fabry disease which only affects the heart may be missed unless specifically tested for. METHODS AND RESULTS: We evaluated 90 consecutively recruited HCM probands and their relatives. Probands without sarcomere-gene mutations were tested for alpha-galactosidase A gene (GLA) mutations. Of the 90 families, 31 (34%) had sarcomere gene mutations and were therefore excluded. In the r

DNA methylation of multiple promoter-associated CpG islands in meningiomas: relationship with the allelic status at 1p and 22q.

The purpose of this research was to examine the DNA methylation profile of meningiomas. Accordingly, we examined the DNA methylation status of ten tumor-related genes (RB1, p16(INK4a), p73, MGMT, ER, DAPK, TIMP-3, p14(ARF), THBS1, and Caspase-8) in 98 meningiomas (68 grade I; 27 grade II; and 3 grade III samples) using methylation-specific PCR and sequencing. The most frequently methylated genes were THBS1 (30%), TIMP-3 (24%), p16(INK4a) (17%), MGMT (16%), p73 (15%), ER (15%), and p14(ARF) (13%), whereas methylation was relatively rare in the other genes (<10%). Methylation occurred in at leas

Integration target site selection for retroviruses and transposable elements.

When a retrovirus infects a cell, its RNA genome is reverse transcribed into a double-stranded DNA, which is then permanently integrated into the host chromosome. Integration is one of the essential steps in the retroviral life cycle. Many transposable elements also move around and integrate into the host genome as part of their life cycle, some through RNA intermediates and some through 'cut and paste' mechanisms. Integration of retroviruses and transposable elements into 'sensitive areas' of the genome can cause irreparable damage. On the other hand, because of their ability to integrate per

Differential display of human marrow stromal cells reveals unique mRNA expression patterns in response to dexamethasone.

Human bone marrow stromal cells (hBMSC) are pluripotent cells that have the ability to differentiate into bone, cartilage, hematopoietic-supportive stroma, and adipocytes in a process modulated by dexamethasone (DEX). To characterize changes in hBMSC in response to DEX, we carried out differential display experiments using hBMSC cultured for 1 week in the presence or absence of 10(-8) M DEX. When RNA from these cells was used for differential display, numerous cDNA bands were identified that were up-regulated and down-regulated by DEX. The cDNA bands were reamplified by PCR and directly used t

Characterization of the Microbiome and Host's Metabolites of the Lower Respiratory Tract During Acute Community-Acquired Pneumonia Identifies Potential Novel Markers.

PURPOSE: Community-acquired pneumonia (CAP) is one of the most frequently encountered infectious diseases worldwide. Few studies have explored the microbial composition of the lower respiratory tract (LRT) and host metabolites of CAP. We analyzed the microbial composition of the LRT and levels of host metabolites to explore new biomarkers for CAP. PATIENTS AND METHODS: Bronchoalveolar lavage fluid (BALF) was collected from 28 CAP patients and 20 healthy individuals. Following centrifugation, BALF pellets were used for amplicon sequencing of a variable region of the bacterial 16S rDNA gene to

Hyperhomocysteinemia, diabetes mellitus, and carotid atherosclerosis independently increase atherosclerotic vascular disease outcome in Japanese patients with end-stage renal disease.

BACKGROUND: Patients with end-stage renal disease (ESRD) have high mortality from atherosclerotic/atherothrombotic vascular disease (AVD). However, the role of an elevated plasma total homocysteine (tHcy) level as a risk factor is uncertain in ESRD. METHODS: We enrolled 55 ESRD patients in a prospective follow-up study in order to evaluate the prognostic significance of their tHcy levels, common methylenetetrahydrofolate reductase (MTHFR) gene polymorphism, and other atherosclerotic risk factors, in combination with the results of B mode ultrasound for carotid arteries. RESULTS: Mean intima-

Genome-wide analysis of the soybean GmGRP gene family identifies putative cold-responsive genes.

Glycine-rich proteins (GRPs) play essential roles in plant growth, development, and stress responses, but systematic studies in soybean remain limited. In this study, using the soybean reference genome, 11 GmGRP gene members were identified with tools such as Pfam, SMART, CDD, and HMMER, and their physicochemical properties, phylogenetic relationships, gene structure, conserved motifs, and promoter cis-acting elements were systematically analyzed. Phylogenetic analysis showed that GmGRPs can be divided into multiple subgroups and share conserved biological functions with GRPs in Arabidopsis, r

Genome-Wide Identification of the BhKCS Gene Family in Wax Gourd (Benincasa hispida) and Stress-Responsive Expression of BhKCS8/9/14/15.

The β-ketoacyl-CoA synthase (KCS) family catalyzes the rate-limiting step in very-long-chain fatty acid (VLCFA) synthesis and plays critical roles in cuticular wax formation and abiotic stress responses. However, the KCS gene family in wax gourd (Benincasa hispida) has remained uncharacterized. In this study, 27 BhKCS genes were identified genome-wide and unevenly distributed across ten chromosomes, with a dense cluster on chromosome 1. Phylogenetic analysis classified the BhKCS proteins into five groups together with Arabidopsis KCSs. Conserved motif and exon-intron structure analyses reveale

Microbiome-linked transcriptomic signatures in NMIBC: Toward personalized uro-oncology.

BACKGROUND: Nonmuscle Invasive Bladder Cancer (NMIBC) is a prevalent malignancy marked by high recurrence and progression rates. Emerging evidence suggests that demographic and environmental factors may alter the bladder's native oncobiome, influencing tumor behavior. This exploratory pilot study examined whether paired tumor and adjacent normal bladder mucosa exhibit distinct host transcriptomic and microbial signatures that may illuminate early tumor-microbiome interactions in NMIBC. METHODS: A meta-transcriptomic analysis was conducted on paired tumor and adjacent normal bladder mucosa fro

Severe Hyperlipidemia and Multifocal Atherosclerosis With Negative Genetic Testing for Familial Hypercholesterolemia: A Case Report.

Familial hypercholesterolemia (FH) is a genetic disorder characterized by severely elevated cholesterol levels and increased cardiovascular risk. While typically associated with mutations in the low-density lipoprotein receptor (LDLR), apolipoprotein B (APOB), or proprotein convertase subtilisin/kexin type 9 (PCSK9) genes, many clinical cases remain genetically unexplained. Identifying novel variants in non-canonical genes is essential for understanding the complex genetic architecture of lipid disorders. A 75-year-old female patient presented with severe hyperlipidemia, multifocal atheroscler

Transcriptomic analysis of naturally occurring bovine respiratory disease in Australian feedlot cattle identifies bovine viral diarrhoea virus-induced immunosuppression and Mollicutes as the dominant drivers of lung inflammation.

Bovine respiratory disease (BRD) is the most economically significant syndrome of feedlot cattle worldwide, yet the relative contributions of individual BRD pathogens to host transcriptional responses remain poorly understood. We performed pathogen-stratified transcriptomic analysis of lung tissue obtained from naturally occurring BRD cases (n = 22) and non-BRD controls (n = 9) in feedlot cattle. BRD-affected tissue showed significant immune activation with 3148 differentially expressed genes (DEGs; 18.7% of the 16,873 genes tested), of which 70.4% were upregulated. Pathogen-stratified analysi

Enhanced antibiofilm and gene-suppressive effects of cinnamon nanoemulsion against multidrug-resistant Enterococcus faecalis.

BACKGROUND: Enterococcus faecalis is a major cause of persistent endodontic infections due to its strong biofilm-forming ability and resistance to conventional disinfectants. Cinnamon (Cinnamomum zeylanicum) essential oil (EO) has been recognized for its antimicrobial properties; however, its poor solubility and stability limit its clinical application. This study evaluated the antibacterial efficacy of cinnamon EO and its nanoemulsion (EM) against multidrug-resistant E. faecalis and its biofilm-associated gene expression. METHODS: Cinnamon EO was extracted via Soxhlet and emulsified using com

Transcriptional Heterogeneity Underlying Cancer Cell Migration Converges on Shared Regulatory Programs.

Background/Objectives: Cancer cell migration is a hallmark of cancer and is associated with metastasis. While large-scale functional screens have identified regulators of migration, less is known about how intrinsic transcriptional heterogeneity drives highly migratory phenotypes within individual cancer models or whether these transcriptional changes are conserved across different models of varying tissues of origin. This study aims to define shared and cell line-specific transcriptional programs associated with cancer cell migration and analyze their relevance to patient datasets. Methods: F
