Universal fluorescent multiplex PCR and capillary electrophoresis for evaluation of gene conversion between SMN1 and SMN2 in spinal muscular atrophy.

We have developed a capillary electrophoresis (CE) method with universal fluorescent multiplex PCR to simultaneously detect the SMN1 and SMN2 genes in exons 7 and 8. Spinal muscular atrophy (SMA) is a very frequent inherited disease caused by the absence of the SMN1 gene in approximately 94% of patients. Those patients have deletion of the SMN1 gene or gene conversion between SMN1 and SMN2. However, most methods only focus on the analysis of whole gene deletion, and ignore gene conversion. Simultaneous quantification of SMN1 and SMN2 in exons 7 and 8 is a good strategy for estimating SMN1 dele

Is RNA manipulation a viable therapy for spinal muscular atrophy?

Childhood spinal muscular atrophy (SMA) is an autosomal recessive disorder characterised by loss of the alpha motor neurones of the spinal cord. SMA is cause by mutations in the survival motor neuron (SMN) gene. There are two copies of the SMN gene: SMN1 and SMN2. The two genes differ by only 11 nucleotides at the genomic level. One of these is a C to T single nucleotide polymorphism (SNP) at position 6 in exon 7. This change alters an exon splicing enhancer in exon 7, meaning that while SMN1 expresses exclusively full-length protein containing exon 7, SMN2 is predominantly alternatively splic

Universal multiplex PCR and CE for quantification of SMN1/SMN2 genes in spinal muscular atrophy.

We established a universal multiplex PCR and CE to calculate the copy number of survival motor neuron (SMN1 and SMN2) genes for clinical screening of spinal muscular atrophy (SMA). In this study, one universal fluorescent primer was designed and applied for multiplex PCR of SMN1, SMN2 and two internal standards (CYBB and KRIT1). These amplicons were separated by conformation sensitive CE. Mixture of hydroxyethyl cellulose and hydroxypropyl cellulose were used in this CE system. Our method provided the potential to separate two 390-bp PCR products that differ in a single nucleotide. Differentia

Progesterone restores retrograde labeling of cervical motoneurons in Wobbler mouse motoneuron disease.

The Wobbler mouse, a mutant characterized by motoneuron degeneration in the cervical spinal cord, has been used to test the efficacy of novel treatments for human motoneuron diseases (HMD). Previous reports have shown that slow axonal transport is impaired in Wobblers and other models of HMD. Since progesterone (PROG) corrects some morphological, molecular, and functional abnormalities of Wobbler mice, we studied if steroid exposure for 8 weeks restored retrograde axonal transport by measuring motoneuron labeling after injection of fluorogold into the limb muscles. The dye was injected into fo

Persistence of vibrissal motor representation following vibrissal pad deafferentation in adult rats.

The effect of sensory vibrissal pad denervation on M1 organization was studied in adult rats 2 weeks after the infraorbital nerve was severed. Cortical motor output organization was assessed mapping the representation size and thresholds of vibrissa movements evoked by intracortical electrical microstimulation (ICMS). Motor cortex output patterns of control and sham groups of rats were compared with those of rats that had received unilateral or bilateral infraorbital nerve lesions. The mean size of the vibrissa representation in both unilateral and bilateral input-deprived hemispheres was not

Natural history of 46 patients with multifocal motor neuropathy with conduction block.

We studied 46 consecutive patients with multifocal motor neuropathy with conduction block (MMN-CB). Typically, asymmetric weakness and atrophy of the hands or arms developed insidiously, but spontaneous improvement (without treatment) or death from this disease did not occur and 94% remained employed. For 18 patients examined on multiple occasions using the weakness subscore of the neuropathy impairment score [NIS(W)] for a median time of 2.3 years, worsening of 1.3 points per year was observed; many patients, however, had received intensive immunomodulating therapy. Median worsening to our fi

Human midbrain organoids reveal the characteristics of axonal mitochondria specific to dopaminergic neurons.

Mitochondrial dysfunction and abnormalities in mitochondrial quality control contribute to the development of neurodegenerative diseases. Parkinson's disease is a neurodegenerative disease that causes motor problems mainly due to the loss of dopaminergic neurons in the substantia nigra pars compacta. Axonal mitochondria in neurons reportedly differ in properties and morphologies from mitochondria in somata or dendrites. However, the function and morphology of axonal mitochondria in human dopaminergic neurons remain poorly understood. To define the function and morphology of axonal mitochondria

Synergistic Effects of Constraint-Induced Movement Therapy and Mirror Therapy on Neuroplasticity and Functional Recovery in Rats With Hemiplegic Cerebral Palsy.

IMPORTANCE: The mechanisms underlying the combined effects of constraint-induced movement therapy (CIMT) and mirror therapy (MT) on neuroplasticity in hemiplegic cerebral palsy (HCP) remain unclear. OBJECTIVE: This study investigated whether combined CIMT and MT improves functional recovery more than either therapy alone and examined accompanying motor cortical neuroplastic changes. DESIGN: HCP rats were randomly assigned to HCP, HCP+CIMT, HCP+MT, and HCP+CIMT+MT groups (n = 13 per group), with a separate control group. SETTING: The study was conducted at the Experimental Animal Center, Gua

Convection enhanced drug delivery of BDNF through a microcannula in a rodent model to strengthen connectivity of a peripheral motor nerve bridge model to bypass spinal cord injury.

Models employing peripheral nerve to bypass spinal cord injury (SCI), although highly promising, may benefit from improved nerve regeneration and motor bridge connectivity. Recent studies have demonstrated that neuronal growth factor-induced enhancement of endogenous neurorestoration may improve neuronal connectivity after severe neurologic injury, particularly if delivered intraparenchymally with zero-order kinetics. We sought to investigate the effect of convection-enhanced delivery of brain-derived neurotrophic factor (BDNF), a neuronal growth factor, on the connectivity of a peripheral mot

Transcutaneous spinal random noise stimulation enhances motor memory consolidation and corticospinal transmission in humans.

Stochastic resonance sensory input modulates the central nervous system's excitability, thereby possibly influencing motor skill learning and retention. We investigated the effects of transcutaneous spinal random noise stimulation (tsRNS) at the cervical level on motor skill learning and corticospinal transmission in healthy humans. Participants performed a 20 min visuomotor tracking training task requiring rapid shifts in pinch force, with motor performance tests conducted before, immediately after, 1 day after and 7 days after the training to assess motor skill learning and retention. Long-term plasticity may be manifested as reduction or expansion of cortical representations of actively used muscles in motor skill specialists.

Our aim was to study long-term plasticity in the organization of cortical muscle representations due to extensive motor training for different skills. We were especially interested in whether skill-specific demands on independent hand muscle movements and synchronous leg muscle movements are reflected differently in the reorganization of muscle representations. We used navigated transcranial magnetic stimulation to estimate the size of cortical representations of opponens pollicis, abductor digiti minimi, and tibialis anterior muscles in five string instrument players, five figure skaters, and

Oscillations in sensorimotor cortex in movement disorders: an electrocorticography study.

Movement disorders of basal ganglia origin may arise from abnormalities in synchronized oscillatory activity in a network that includes the basal ganglia, thalamus and motor cortices. In humans, much has been learned from the study of basal ganglia local field potentials recorded from temporarily externalized deep brain stimulator electrodes. These studies have led to the theory that Parkinson's disease has characteristic alterations in the beta frequency band (13-30 Hz) in the basal ganglia-thalamocortical network. However, different disorders have rarely been compared using recordings in the

Temporal classification of multichannel near-infrared spectroscopy signals of motor imagery for developing a brain-computer interface.

There has been an increase in research interest for brain-computer interface (BCI) technology as an alternate mode of communication and environmental control for the disabled, such as patients suffering from amyotrophic lateral sclerosis (ALS), brainstem stroke and spinal cord injury. Disabled patients with appropriate physical care and cognitive ability to communicate with their social environment continue to live with a reasonable quality of life over extended periods of time. Near-infrared spectroscopy is a non-invasive technique which utilizes light in the near-infrared range (700 to 1000

A single session of brief electrical stimulation enhances axon regeneration through nerve autografts.

Nerve graft reconstruction of gap defects may result in poor clinical outcomes, particularly with long regeneration distances. Electrical stimulation (ES) of nerves may improve outcomes in such patients. A single session of ES at 20 Hz for 1 h significantly enhances axon regeneration in animals and human subjects after nerve crush or nerve transection and repair. The objectives of this study were to evaluate if ES enhances axon regeneration through nerve grafts and if there is added benefit of a second, delayed session of ES (serial ES) on axon regeneration as compared to a single session only

A new familial disease of saccadic oscillations and limb tremor provides clues to mechanisms of common tremor disorders.

Tremor disorders pose fundamental questions about disease mechanisms, and challenges to successful neurotherapeutics: What causes motor circuits to oscillate in disorders in which the central nervous system otherwise seems normal? How does inheritance 'determine' the clinical phenotype in familial tremor disorders? Here, we address these questions. Analogies between the neural circuits controlling rapid eye movements (saccades) and those controlling limb movements allow us to translate the interpretations from the saccadic systems to the limb movement system. Moreover, the relatively well unde

Inhibition of the LHb glutamatergic pathway ameliorates depressive-like behaviors in a 6-hydroxydopamine-induced Parkinson's disease mouse model.

BACKGROUND: Depression is one of the most common non-motor disorders and neuropsychiatric comorbidities in Parkinson's disease (PD). The pathophysiology of depression in PD patients remains unclear and has been largely unexplored. METHOD: In this study, we employed chemogenetics and pharmacology to modulate the lateral habenula (LHb) and its downstream brain regions, the rostromedial tegmental nucleus (RMTg) and the ventral tegmental area (VTA) in wild type (WT) and 6-hydroxydopamine (6-OHDA) mice, to investigate the potential mechanisms underlying the improvement of PD-related depression. Distributed control circuits across a brain-and-cord connectome.

Just as genomes revolutionized molecular genetics, connectomes (maps of neurons and synapses) are transforming neuroscience. To date, the only species with complete connectomes are worms1-3 and sea squirts4 (103-104 synapses). By contrast, the fruit fly is more complex (108 synaptic connections), with a brain that supports learning and spatial memory5,6 and an intricate ventral nerve cord analogous to the vertebrate spinal cord7-11. Here we report the first adult fly connectome that unites the brain and ventral nerve cord, and we leverage this resource to investigate principles of neural contr

Identification of a brainstem locus that inhibits tumor necrosis factor.

In the brain, compact clusters of neuron cell bodies, termed nuclei, are essential for maintaining parameters of host physiology within a narrow range optimal for health. Neurons residing in the brainstem dorsal motor nucleus (DMN) project in the vagus nerve to communicate with the lungs, liver, gastrointestinal tract, and other organs. Vagus nerve-mediated reflexes also control immune system responses to infection and injury by inhibiting the production of tumor necrosis factor (TNF) and other cytokines in the spleen, although the function of DMN neurons in regulating TNF release is not known

Trichloroethylene induces dopaminergic neurodegeneration in Fisher 344 rats.

Trichloroethylene, a chlorinated solvent widely used as a degreasing agent, is a common environmental contaminant. Emerging evidence suggests that chronic exposure to trichloroethylene may contribute to the development of Parkinson's disease. The purpose of this study was to determine if selective loss of nigrostriatal dopaminergic neurons could be reproduced by systemic exposure of adult Fisher 344 rats to trichloroethylene. In our experiments, oral administration of trichloroethylene induced a significant loss of dopaminergic neurons in the substantia nigra pars compacta in a dose-dependent
