Analysis of the effects of cell spacing and liquid depth on nitric oxide and its oxidation products in cell cultures.

A series of reaction-diffusion models was developed to describe spatial and temporal variations in the concentrations of NO and related species (O(2)(-), peroxynitrite, and N(2)O(3)) in a cell culture configuration that is commonly used to study the toxicity and/or mutagenicity of NO. In the analysis, adherent cells that generate NO and O(2)(-) (e.g., macrophages) were assumed to be present in sub-monolayer quantities at the bottom of a culture dish, with a stagnant layer of culture medium separating them from the incubator gas. It was assumed also that the level of O(2)(-) production is lower

Promoter transgenics reveal multiple gonadotropin-releasing hormone-I-expressing cell populations of different embryological origin in mouse brain.

Gonadotropin-releasing hormone-I (GnRH-I) is thought to be expressed by a single, highly spatially restricted group of neurons, which originate in the olfactory placode and migrate through the nose into the medial septum and hypothalamus from where they control fertility. Transgenic mice bearing a 13.5 kb GnRH-I-lacZ reporter construct were derived and found to express high levels of beta-galactosidase mRNA and protein within the septohypothalamic GnRH neurons in a correct temporal and spatial manner. Unexpectedly, low levels of beta-galactosidase were also present in three further populations

The expression of the mouse Zic1, Zic2, and Zic3 gene suggests an essential role for Zic genes in body pattern formation.

We examined the expression of Zic1, Zic2, and Zic3 genes in the mouse embryo by means of in situ hybridization. Zic genes were found as a group of genes coding for zinc finger proteins that are expressed in a restricted manner in the adult mouse cerebellum. We showed that the genes are the vertebrate homologues of Drosophila odd-paired, which may play an essential role in parasegmental subdivision and in visceral mesoderm development. The expression of the three Zic genes was first detected at gastrulation in a spatially restricted manner. At neurulation, the expression became restricted to th

A voxel-based method for the statistical analysis of gray and white matter density applied to schizophrenia.

We describe a novel technique for characterizing regional cerebral gray and white matter differences in structural magnetic resonance images by the application of methods derived from functional imaging. The technique involves automatic scalp-editing of images followed by segmentation, smoothing, and spatial normalization to a symmetrical template brain in stereotactic Talairach space. The basic idea is (i) to convert structural magnetic resonance image data into spatially normalized images of gray (or white) matter density, effected by segmenting the images and smoothing, and then (ii) to use

Controlling the Arrangement of [Fe4L6]8+ Cages in the Solid State.

Metallosupramolecular cages are widely studied for their unique host-guest properties in solution yet translating these functionalities to the solid state remains challenging due to cavity inaccessibility and dense packing. Here, we demonstrate a hierarchical approach that employs halogen bonding to control the spatial arrangement of tetrahedral [Fe4L6]8+ cages in the solid state. By incorporating perfluorinated halogen bond donors of varying geometry, we demonstrate selective modulation of cage packing motifs without disrupting the cage architecture. Structural analysis reveals that incorpora

Toward Mapping Spatiotemporally Resolved Transcriptomes and RNA-Protein Interactions.

RNAs navigate highly regulated life cycles that include synthesis, processing, ribonucleoprotein complex formation, localization, and degradation. However, most transcriptome-wide methods capture static, cell-wide averages that obscure the spatiotemporal aspects-where and when-of RNA life. Here we review recent advances that bring spatial and temporal resolution to transcriptome and RNA-protein interaction analyses by labeling RNA either in defined subcellular locales (RNA proximity labeling) or during defined transcriptional time windows to capture the kinetics of RNA synthesis, degradation,

A Greener Approach toward Ynones via Visible-Light-Induced Oxidation of Propargyl Alcohols under Aqueous-Aerobic Conditions.

A simple and convenient approach involving visible-light-induced, TBHP-mediated oxidation of propargyl alcohols to ynone in aqueous media has been developed. This photoinduced protocol was conducted under an aerobic-aqueous medium using Eosin Y as a photosensitizer. A diverse range of ynone derivatives were prepared from propargyl alcohol in moderate to excellent yield. Furthermore, the successful gram-scale experiment of this method also indicates its flexibility and applicability for industrial applications. Steric mapping of the series 2a-2q, exemplified by 2j, revealed an optimized buried

Functional Specialization of Cellular and Extracellular Fractions of Bacteroides thetaiotaomicron Contributes to Glycan Utilization.

Bacteroides thetaiotaomicron (Bt) degrades dietary glycans via carbohydrate-active enzymes (CAZymes), but their spatial distribution and functional coordination remain unclear. In this study, we characterized the "labor division" during glycan degradation across four bacterial fractions: cytoplasm, outer membrane, extracellular vesicles, and supernatant. All fractions displayed hydrolytic activity. Proteomic analysis identified abundant glycoside hydrolases, mainly GH32 enzymes encoded by PUL22, and their components varied greatly. Untargeted metabolomics identified 45 shared differential meta

An opposing molecular gradient axis underlies primate cortical organization.

The principles organizing cellular diversity and connectivity in primate brains remain elusive. By integrating spatial transcriptomics, magnetic resonance imaging, and retrograde labeling in marmosets, we identified two opposing molecular gradients that undergo postnatal refinement, emanating from allocortices and primary sensory cortices, respectively. These gradients reconcile conflicting hypotheses on cortical expansion and characterize distinct cortical areas. Cortical gradients align with thalamic gene expression and thalamocortical projection patterns. At gradient intersections, the defa

Pan-Cancer Multi-Omics Analysis Reveals the Potential of ZDHHC16 in Tumor Progression and Immune Regulation.

S-palmitoylation is a crucial post-translational modification that regulates diverse cellular processes, particularly signalling pathways. The ZDHHC family of enzymes catalyzes this modification, however, the role of ZDHHC16 in cancer warrants in-depth investigation. This study presents a comprehensive pan-cancer analysis of ZDHHC16, examining its expression patterns, clinical relevance, associations with immune responses, genomic characteristics, and potential as a therapeutic target. We further analysed single-cell and spatial transcriptomic data to identify specific cell populations associa

Chrysin Cocrystals: Prediction, Preparation, Characterization, and In Vitro/In Vivo Evaluation.

In this study, crystal engineering was employed to enhance the solubility and druggability of Chrysin (CHR). Four nitrogen heterocyclic compounds, including piperazine (PIP), 4,4'-bipyridine (BIP), imidazole (IMI), and sophoridine (SOP), were investigated using computational screening methodologies. Screening experiments were conducted to validate the computational screening results, and four CHR crystals were successfully prepared, three of which were reported for the first time. The structures of these cocrystals were characterized by using single-crystal X-ray diffraction (SXRD), powder X-r

Cardiac Computed Tomography in Structural Heart Interventions: From Preprocedural Planning to Procedural Strategy.

Cardiac computed tomography (CT) has become an essential imaging modality in structural cardiac interventions, providing high-resolution anatomical and functional assessments. Moreover, the role of cardiac CT spans pre-procedural planning, intra-procedural guidance, and post-procedural follow-up in interventions such as transcatheter aortic valve implantation (TAVI), mitral, tricuspid, and pulmonary valve interventions, left atrial appendage occlusion (LAAO), atrial septal defect (ASD), and paravalvular leak (PVL) closures. Furthermore, compared to traditional imaging techniques, cardiac CT of

Spatial centrosome proteome of human neural cells uncovers disease-relevant heterogeneity.

The centrosome provides an intracellular anchor for the cytoskeleton, regulating cell division, cell migration, and cilia formation. We used spatial proteomics to elucidate protein interaction networks at the centrosome of human induced pluripotent stem cell-derived neural stem cells (NSCs) and neurons. Centrosome-associated proteins were largely cell type-specific, with protein hubs involved in RNA dynamics. Analysis of neurodevelopmental disease cohorts identified a significant overrepresentation of NSC centrosome proteins with variants in patients with periventricular heterotopia (PH). Expr

An evaluation of a case of agenesis of the corpus callosum with Rourke's nonverbal learning disorder model.

This article evaluates the case of an 11-year-old male with congenital agenesis of the corpus callosum, against Rourke's NLD models. A comprehensive neuropsychological assessment was completed, and impairments with attention, movement, fine motor and sensory perception, visuo-spatial organization, and verbal memory were identified. Additionally, a volumetric analysis of the magnetic resonance image (MRI) was completed. Although no gross structural abnormalities, beyond agenesis, were present, the possibility of unobservable abnormalities was considered. Many predicted NLD deficits were clearly

Visual field effects of classical migraine.

Although classical migraine is known to produce lateralized sensory disturbances (e.g., visual "fortification" structures), its effect on higher order processes is unknown. Here four lateralized visual tasks were repeatedly presented to a single subject (the author) over a 2 1/2-year span, during both classical migraine attacks and headache-free periods. Attacks varied as to inferred hemispheric locus. All four tasks ("words," "typing," "bar graphs," and "locations") produced significant or marginally significant visual field (VF) by condition interactions, results generally consistent with a

A New Unsupervised Machine Learning Method for Metabarcoding Analysis Based on Selected Mock Community Data.

DNA metabarcoding analysis has emerged as a powerful tool for biological monitoring, enabling the identification and assessment of biodiversity across ecosystems. Conventional analytical methods, including operational taxonomic unit (OTU) clustering and amplicon sequence variant (ASV) analysis, have demonstrated strengths and limitations, including bias in estimating biodiversity and challenges in detecting rare sequences. To address these challenges, we developed unsupervised machine learning (UML)-based methods utilizing hierarchical density-based spatial clustering of applications with nois

Decoding of Motor Coordination Imagery Involving the Lower Limbs by the EEG-Based Brain Network.

Compared with the efficacy of traditional physical therapy, a new therapy utilizing motor imagery can induce brain plasticity and allows partial recovery of motor ability in patients with hemiplegia after stroke. Here, we proposed an updated paradigm utilizing motor coordination imagery involving the lower limbs (normal gait imagery and hemiplegic gait imagery after stroke) and decoded such imagery via an electroencephalogram- (EEG-) based brain network. Thirty subjects were recruited to collect EEGs during motor coordination imagery involving the lower limbs. Time-domain analysis, power spect

Microstructural white matter abnormalities in obsessive-compulsive disorder: A coordinate-based meta-analysis of diffusion tensor imaging studies.

BACKGROUND: There are no conclusive diffusion tensor imaging (DTI) findings on obsessive-compulsive disorder (OCD) for now. We conducted a comprehensive meta-analysis of DTI studies to identify white matter (WM) microarchitecture changes in OCD, and also to compare the results differences between the two most frequently used methods (voxel-based analysis, VBA versus tract-based spatial statistics, TBSS) for DTI data. METHODS: A systematic search was performed on relevant studies that reported fractional anisotropy (FA) alterations between patients with OCD and healthy controls (HC). Seed-base

Disturbance of Immune Microenvironment in Androgenetic Alopecia through Spatial Transcriptomics.

Androgenetic alopecia (AGA) is characterized by microinflammation and abnormal immune responses, particularly in the upper segment of hair follicles (HFs). However, the precise patterns of immune dysregulation remain unclear, partly due to limitations in current analysis techniques to preserve tissue architecture. The infundibulum, a major part of the upper segment of HFs, is associated with significant clusters of immune cells. In this study, we investigated immune cells around the infundibulum, referred to as peri-infundibular immune infiltration (PII). We employed spatial transcriptome prof
