Protective effect of a T-cell-dependent immunosuppressive, B-cell-mitogenic protein (F3'EP-Si, or P90) produced by Streptococcus intermedius.

The role of a previously described bacterial protein (F3'EP-Si), now designated P90, in the survival of Streptococcus intermedius in the host was investigated, and the immunosuppressive and B-cell-mitogenic effects of this protein were further characterized. C57BL6 mice treated with P90 were about 50 times more susceptible to infection with this bacterium than untreated mice. One of seven splenocytes of C57BL/6 mice were activated by P90. Marked splenomegaly was observed in mice treated with P90, with increased numbers of splenic mononuclear cells and polyclonal immunoglobulin-secreting plaque

Identification of N-acetylneuraminyl alpha 2-->3 poly-N-acetyllactosamine glycans as the receptors of sialic acid-binding Streptococcus suis strains.

Streptococcus suis is a common cause of sepsis, meningitis, and other serious infections in young piglets and also causes meningitis in humans. The cell-binding specificity of sialic acid-recognizing strains of Streptococcus suis was investigated. Treatment of human erythrocytes with sialidase or mild periodate abolished hemagglutination. Hemagglutination inhibition experiments with sialyl oligosaccharides indicated that the adhesin preferred the sequence NeuNAc alpha 2-3Gal beta 1-4Glc(NAc). Resialylation of desialylated erythrocytes with Gal beta 1-3(4)GlcNAc alpha 2-3-sialyltransferase indu

Diagnosis of group A beta-hemolytic Streptococcus using clinical scoring criteria, Directigen 1-2-3 group A streptococcal test, and culture.

Cultures for group A beta-hemolytic Streptococcus were performed on 806 patients presenting with a sore throat to five urgent care centers. The accuracies of a clinical scoring system and of a liposomal in-office direct test for Streptococcus were compared with culture results. The Directigen 1-2-3 group A streptococcal test had a sensitivity of 67%, a specificity of 85%, a positive predictive value of 61%, and a negative predictive value of 89% compared with culture. The scoring system had a sensitivity of 26%, a specificity of 94%, a positive predictive value of 58%, and a negative predictiv

Maternal body mass index and the risk of early-onset Group B Streptococcus disease in newborns: A systematic review and meta-analysis.

BACKGROUND: Early-onset group B Streptococcus disease (EOGBS) remains a leading cause of neonatal morbidity and mortality. The incidence can be substantially reduced by intrapartum antibiotic prophylaxis in women with defined risk factors. However, the role of high prepregnancy body mass index (BMI) as a risk factor remains unclear. This systematic review and meta-analysis therefore aimed to evaluate the association between maternal BMI and the risk of EOGBS, as well as related proxy outcomes, including intrapartum vaginal GBS colonization and rectovaginal or urinary GBS colonization before te

Clinical and Economic Burden of Complicated Pneumonia and Invasive Disease due to Streptococcus pneumoniae Serotype 19A in an Ecuadorian Pediatric Reference Hospital.

Serotype 19A has emerged as an important cause of pneumococcal disease following the introduction of the 10-valent pneumococcal conjugate vaccine (PCV10). In this retrospective study of children hospitalized with culture-confirmed invasive pneumococcal disease at an Ecuadorian pediatric reference hospital, among 46 cases, 43 (93%) were serotype 19A, with most presenting as severe complicated pneumonia. The majority had received three PCV10 doses. Penicillin nonsusceptibility was identified in 88% of isolates. Serotype 19A infections were associated with longer hospitalization and higher medica

The Effect of a 5-Day Course of Azithromycin on Streptococcus Pneumoniae Carriage and Antimicrobial Resistance Among Kenyan Children Discharged From Hospital.

BACKGROUND: Mass azithromycin distribution reduces child mortality in some settings, potentially through reductions in nasopharyngeal carriage of Streptococcus pneumoniae, but has been associated with increased antimicrobial resistance. Individual-level data are lacking on the impact of azithromycin on antimicrobial resistance over time. METHODS: We analyzed data from a double-blind, randomized placebo-controlled trial (ClinicalTrials.gov; NCT02414399) which followed 1398 hospitalized Kenyan children to evaluate the impact of a 5-day course of oral azithromycin at discharge from hospital on p

Distribution of Streptococcus pneumoniae Serotypes in Nasopharyngeal Carriage Among Children in Indonesia and Estimated Coverage of Pneumococcal Conjugate Vaccines: A Systematic Review.

Background: Streptococcus pneumoniae may asymptomatically colonize the human nasopharynx and remains a leading cause of invasive and noninvasive disease in children, accounting for an estimated 294,000 global deaths in those aged under five years. Nationally representative serotype data from Indonesia remain limited despite national PCV13 rollout in 2022. This study aims to evaluate the distribution of Streptococcus pneumoniae serotypes and estimate the coverage of pneumococcal conjugate vaccines (PCVs) among children aged 0-18 years in Indonesia. Methods: Systematic search of PubMed, Scopus,

Trends in invasive and non-invasive Streptococcus pneumoniae disease in adults hospitalised in Bristol and Bath: a retrospective cohort study, 2006-2022.

BACKGROUND: Pneumococcal surveillance typically focuses on cases identified by positive microbiology and collects limited clinical data. The majority of pneumococcal studies in the literature focus on invasive respiratory disease. This study includes a cohort of all cause pneumococcal disease, including non-invasive disease and non-respiratory invasive disease with detailed clinical outcomes, allowing a more complete view of pneumococcal disease burden, and particularly meningitis cases. METHODS: A longitudinal adult cohort of invasive and non-invasive pneumococcal disease inpatients in three

Molecular Regulation of Virulence and Dental Biofilm Formation in Streptococcus mutans: Unravelling the Crosstalk Between the VicRK and LiaSR Two-Component Systems.

Streptococcus mutans is one of the primary causes of dental caries and is often found in persistent endodontic infections, primarily due to its ability to form acidogenic, aciduric and therapeutically recalcitrant biofilms. The VicRK and LiaSR two-component signal transduction systems (TCS) are particularly important regulators of virulence, envelope integrity and stress response. This paper will provide an updated synthesis of the evidence, at the molecular, genetic and systems levels, regarding the architecture, regulation and integration of VicRK and LiaSR in the control of S. mutans pathog

In Vitro Characterization and Safety Assessment of Streptococcus salivarius, Levilactobacillus brevis and Pediococcus pentosaceus Isolated from the Small Intestine of Broiler Breeders.

In poultry production, antibiotics have been excessively used as growth promoters to support well-being and decrease mortality caused by pathogenic microorganisms. The overuse of antibiotics has led to the emergence of antibiotic-resistant bacteria and the presence of antibiotic residues in poultry products. To counteract this problem, probiotics could be used as adjuncts or as substitutes for preserving a diverse and balanced microflora to prevent the colonization and multiplication of pathogenic bacteria in the GI tract. This study aimed to isolate and characterize the potential probiotic pr

A Case of Rectal Cancer Discovered Following the Occurrence of a Stent-Graft Infection with Streptococcus gallolyticus after Thoracic Endovascular Aortic Repair.

A 79-year-old man was admitted for transurethral resection of a bladder cancer. He had a history of thoracic endovascular aortic repair for Stanford type B acute aortic dissection and thoracic aortic aneurysm performed 2 years prior. During hospitalization, computed tomography scan findings raised suspicion of a stent-graft infection. Blood cultures confirmed the presence of Streptococcus gallolyticus ssp. pasteurianus. Gallium scintigraphy supported the diagnosis of a stent-graft infection. A subsequent lower gastrointestinal endoscopy revealed a colorectal cancer in the lower rectum. We then

Natural Carriage of Streptococcus pneumoniae Is Associated With Increased Experimental Pneumococcal Carriage but Reduced Conjugate Vaccine Efficacy in a Human Challenge Model.

BACKGROUND: In Malawi, the national 13-valent pneumococcal conjugate vaccine (PCV13) demonstrated less herd immunity than in the United States, likely due to higher natural pneumococcal carriage rates. We assessed PCV13 efficacy against experimental pneumococcal carriage in healthy Malawian adults. We explored how natural carriage (pneumococcal carriage of any serotype apart from 6B) influenced experimental carriage rates and vaccine efficacy. METHODS: Healthy adults aged 18 to 40 years were randomly assigned to PCV13 (n = 98) or saline (n = 106), followed by intranasal SPN 6B inoculation at

Protection acquired upon intraperitoneal group a Streptococcus immunization is independent of concurrent adaptive immune responses but relies on macrophages and IFN-γ.

Group A Streptococcus (GAS; Streptococcus pyogenes) is an important bacterial pathogen causing over 700 million superficial infections and around 500.000 deaths due to invasive disease or severe post-infection sequelae yearly. In spite of this major impact on society, there is currently no vaccine available against this bacterium. GAS strains can be separated into >250 distinct emm (M)-types, and protective immunity against GAS is believed to in part be dependent on type-specific antibodies. Here, we analyse the nature of protective immunity generated against GAS in a model of intraperitoneal

Brain Abscess Caused by Streptococcus intermedius and Aggregatibacter aphrophilus Secondary to Exfoliation of a Deciduous Tooth in a Previously Healthy Child: A Case Report.

This report describes the case of a previously healthy nine-year-old girl who developed a left temporal lobe brain abscess following spontaneous exfoliation of a healthy deciduous tooth. She presented in convulsive status epilepticus without fever or focal neurological symptoms, and the initial MRI revealed a ring-enhancing lesion with adjacent dural inflammation. Surgical drainage and intravenous antibiotic therapy led to a full recovery with no neurological sequelae after six weeks of treatment. Pus culture revealed Streptococcus intermedius and Aggregatibacter aphrophilus, both of which are

Differential expression profile of streptolysin S in the covR and covS mutants of group A Streptococcus is a mediator of keratinocyte death and aggressive local infection.

Group A Streptococcus (GAS) isolates with spontaneous mutations in covR and covS are frequently identified in patients with necrotizing fasciitis and toxic shock syndrome. CovR/CovS is a two-component regulator system, and the phosphorylation of CovR is solely modulated by CovS. Nonetheless, the phenotype of the ∆covS mutant and the ∆covR mutant is not entirely identical. It has been shown that the expression of SpeB protease is only detected in the ∆covR mutant, and the ∆covR mutant causes more severe local lesions than the ∆covS mutant in the mouse infection model. SpeB-mediated gasdermin A

First case of Streptococcus suis bacteremia caused by a serotype 14 strain without pig or pork contact in the Netherlands: A case report.

Streptococcus suis is a porcine pathogen that causes severe zoonotic infections in humans resulting in meningitis and sepsis. The main risk factors for S. suis zoonotic infections are consumption of raw pork products and direct contact with live pigs or pork, in particular in the presence of skin injuries. Most human infections are caused by clonal complex 1 (CC1) serotype 2 isolates. We describe an unusual case of S. suis bacteraemia caused by a CC1 strain carrying a serotype 14 capsule in a patient without a history of consumption of raw pork products or contact with either pig or pork.
