Pharmacokinetics of the potent hallucinogen, salvinorin A in primates parallels the rapid onset and short duration of effects in humans.

Salvia divinorum, a mint plant originally used by the Mazatecs of Oaxaca, Mexico in spiritual rituals has gained popularity, in smoked form, as a legal hallucinogen in the United States and Europe. Abuse results in rapid onset and short-lasting effects that include visual hallucinations and motor-function impairment. Salvinorin A, the psychoactive component of S. divinorum, is a uniquely potent agonist at kappa-opioid receptors, targets for new therapeutic drugs. We labeled salvinorin A with C-11 by acylation of salvinorin B with [11C]-acetyl chloride to study whether its kinetic behavior in t

Saccade control and eye-hand coordination in optic ataxia.

The aim of this work was to investigate ocular control in patients with optic ataxia (OA). Following a lesion in the posterior parietal cortex (PPC), these patients exhibit a deficit for fast visuo-motor control of reach-to-grasp movements. Here, we assessed the fast visuo-motor control of saccades as well as spontaneous eye-hand coordination in two bilateral OA patients and five neurologically intact controls in an ecological "look and point" paradigm. To test fast saccadic control, trials with unexpected target-jumps synchronised with saccade onset were randomly intermixed with stationary ta

Multiple circuits relaying primate parallel visual pathways to the middle temporal area.

Parallel pathways in the primate visual system parse the sensory signal into magnocellular (M), parvocellular (P), and koniocellular (K) streams. These pathways remain anatomically separate and distinct from their origination in different retinal ganglion cell types, through distinct layers of the lateral geniculate nucleus, and into primary visual cortex (V1), with the M pathway terminating primarily in layer 4Calpha, the P pathway in layer 4Cbeta, and the K pathway in the cytochrome oxidase blobs of layer 2/3. Recent studies indicate that outputs from V1 are less compartmental than previousl

Mapping multiple visual areas in the human brain with a short fMRI sequence.

It is a fundamental insight of neuroscience that the cerebral cortex is divided into spatially separated and functionally distinct areas. In this study, we tried to map a large number of visual areas in individual subjects passively viewing a simple stimulus sequence during functional magnetic resonance imaging (fMRI) at 1.5 T. The blocked stimulus sequence contrasted static object photographs with video takes of movement through natural indoor and outdoor scenes, alternated with a control fixation task. Two runs of the 5-min sequence sufficed to invoke 29 distinguishable activations, 16 (13 b

Visual attention deficits in Alzheimer's disease: an fMRI study.

Cognitive and neuroscience studies indicate that attentional operations are impaired in Alzheimer's disease (AD). Our goal was to define the anatomical areas of activation associated with visual attention processing and to define deficits or changes that may occur in AD patients compared with control group. Thirteen AD patients and 13 age- and education-matched normal controls were tested in two visual search tasks (one was a conjunction task, where feature binding is required. The other was a subset task, where group stimuli is needed without feature binding) using fMRI techniques. After ster

The space of senses: impaired crossmodal interactions in a patient with Balint syndrome after bilateral parietal damage.

Balint syndrome after bilateral parietal damage involves a severe disturbance of space representation including impaired oculomotor behaviour, optic ataxia, and simultanagnosia. Binding of object features into a unique spatial representation can also be impaired. We report a patient with bilateral parietal lesions and Balint syndrome, showing severe spatial deficits in several visual tasks predominantly affecting the left hemispace. In particular, we tested whether a loss of spatial representation would affect crossmodal interactions between simultaneous visual and tactile events occurring at

Executive and visuospatial deficits in patients with chronic progressive external ophthalmoplegia and Kearns-Sayre syndrome.

Although neuropsychological deficits have been reported in mitochondrial cytopathies, patients with chronic progressive external ophthalmoplegia (CPEO) or Kearns-Sayre syndrome (KSS) have not been studied systematically using a comprehensive test battery. The aim of our study was to assess the range and extent of putative cognitive dysfunction in 22 patients with CPEO or KSS, and to compare cognitive performance of patients with healthy control subjects matched for age, sex and years of education. Genetic analysis of skeletal muscle tissue from 22 patients with CPEO or KSS included screening f

Shank3 deletion in PV neurons is associated with abnormal behaviors and neuronal functions that are rescued by increasing GABAergic signaling.

BACKGROUND: Phelan-McDermid syndrome (PMS) is a neurodevelopmental disorder characterized by developmental delay, intellectual disability, and autistic-like behaviors and is primarily caused by haploinsufficiency of SHANK3 gene. Currently, there is no specific treatment for PMS, highlighting the need for a better understanding of SHANK3 functions and the underlying pathophysiological mechanisms in the brain. We hypothesize that SHANK3 haploinsufficiency may lead to alterations in the inhibitory system, which could be linked to the excitatory/inhibitory imbalance observed in models of autism sp

Visual cortical regions show sufficient test-retest reliability while salience regions are unreliable during emotional face processing.

Functional magnetic resonance imaging studies frequently use emotional face processing tasks to probe neural circuitry related to psychiatric disorders and treatments with an emphasis on regions within the salience network (e.g., amygdala). Findings across previous test-retest reliability studies of emotional face processing have shown high variability, potentially due to differences in data analytic approaches. The present study comprehensively examined the test-retest reliability of an emotional faces task utilizing multiple approaches to region of interest (ROI) analysis and by examining vo

The macaque IT cortex but not current artificial vision networks encode object position in perceptually aligned coordinates.

Efficient interaction with the visual world requires not only object identification but also localization of where objects are in space. While spatial ("where") processing has classically been attributed to dorsal stream pathways, recent work has shown that object position can also be decoded from ventral stream responses, including the inferior temporal (IT) cortex. However, because object position in these paradigms is coupled to pixel-based location, it has remained unclear whether ventral stream position signals are perceptually meaningful or instead reflect incidental inheritance from ret

Microstructural Integrity of Hippocampal-Posterior Cortical White Matter Is Associated With Phenomenological Properties of Trauma-Related Intrusive Memories.

BACKGROUND: Trauma-related intrusive memories (TR-IMs) are spontaneous and emotionally intense sensory recollections experienced as occurring in the here and now and a core feature of posttraumatic stress disorder (PTSD). Although central to symptom burden, the neurobiological substrates of specific phenomenological properties of TR-IMs remain poorly understood. Prior work has linked dynamic hippocampus (HPC) coactivation with default mode and visual networks to TR-IM properties, but whether structural connectivity tracks with these properties is unknown. METHODS: In 114 symptomatic trauma-ex

Inducing Long-Term Plastic Changes and Visual Attention Enhancement Via One-Week Cerebellar Crus II Intermittent Theta Burst Stimulation (iTBS): An EEG Study.

Intermittent theta burst stimulation (iTBS) is a non-invasive technique frequently employed to induce neural plastic changes and enhance visual attention. Currently, most studies utilized a single iTBS session on healthy subjects to induce short-term neural plastic changes within tens of minutes post-stimulation and investigate its single-session effect on attention performance. Few studies have conducted multiple iTBS sessions on the cerebellum to explore long-term effects on the cerebral cortex and daily effects on visual attention performance. In this study, 18 healthy subjects were involve

Noninvasive Biomarkers for Assessing the Excitatory/Inhibitory Imbalance in Children with Epilepsy.

Epileptic seizures involve a cortical excitation-inhibition imbalance driven by dysfunctional interneurons that contribute to gamma oscillation generation. While impaired gamma oscillations are commonly reported in epilepsy, the dynamics of broadband and narrowband gamma as well as beta oscillations remain underexplored. These oscillations may serve as noninvasive electrophysiological markers associated with altered cortical dynamics, potentially reflecting underlying excitation-inhibition imbalance in epilepsy. Here, we recorded high-density electroencephalography and magnetoencephalography d

Characterizing the discriminability of visual categorical information in strongly connected voxels.

Functional brain responses are strongly influenced by connectivity. Recently, we demonstrated a major example of this: category discriminability within occipitotemporal cortex (OTC) is enhanced for voxel sets that share strong functional connectivity to distal brain areas, relative to those that share lesser connectivity. That is, within OTC regions, sets of 'most-connected' voxels show improved multivoxel pattern discriminability for tool-, face-, and place stimuli relative to voxels with weaker connectivity to the wider brain. However, understanding whether these effects generalize to other

Longitudinal brain functional connectivity changes induced by neurosurgical thalamotomy for tremor in Parkinson's disease: a preliminary study.

The hypothesis that the effectiveness of neurosurgical procedures in Parkinson's disease (PD) would be related to connectivity dysfunctions between the site of the stimulation and other brain regions is growing. This study aimed to assess resting-state functional connectivity between thalamic ventral intermediate nucleus (Vim) and the rest of the brain before and after thalamotomy in PD. A 76-year-old right-handed woman with refractory tremor-dominant PD was selected as a candidate for left Vim radiosurgery thalamotomy. Ten-m4 plays a unique role in the establishment of binocular visual circuits.

The patterning of binocular vision requires distinct molecular pathways for inputs arising from each side of the nervous system. Recent studies have demonstrated important roles for members of the Ten-m/Odz/teneurin family in the development of ipsilateral retinal projections. Here, we further highlight the significance of this gene family in visual development by identifying a role for Ten-m4 during the formation of the ipsilateral projection in the mouse. Ten-m4 was found to be expressed in the retina, dorsal lateral geniculate nucleus (dLGN), superior colliculus (SC), and primary visual cor

Preferential signal pathways during the perception and imagery of familiar scenes: An effective connectivity study.

The perception and imagery of landmarks activate similar content-dependent brain areas, including occipital and temporo-medial brain regions. However, how these areas interact during visual perception and imagery of scenes, especially when recollecting their spatial location, remains unknown. Here, we combined functional magnetic resonance imaging (fMRI), resting-state functional connectivity (rs-fc), and effective connectivity to assess spontaneous fluctuations and task-induced modulation of signals among regions entailing scene-processing, the primary visual area and the hippocampus (HC), re

The hypothalamus may mediate migraine and ictal photophobia: evidence from Granger causality analysis.

BACKGROUND: The hypothalamus plays a central role in the pathophysiology of migraine and is considered to be the "migraine generator." It participates in initiating a migraine attack through its connectivity to regions of the brain involved in processing and modulating pain. However, the underlying mechanisms of hypothalamic effective functional connectivity that bring about migraines remain unclear. This study investigated the hypothalamus-based directional effective connectivity in migraine without aura patients and assessed associations among the clinical characteristics. METHODS: Seven pa

Ventromedial prefrontal cortex activity differentiates sick from healthy faces: Associations with inflammatory responses and disease avoidance motivation.

BACKGROUND: Humans are able to discern the health status of others using olfactory and visual cues, and subsequently shift behavior to make infection less likely. However, little is known about how this process occurs. The present study examined the neural regions involved in differentiating healthy from sick individuals using visual cues. METHODS: While undergoing a functional magnetic resonance imaging scan, participants (N = 42) viewed facial photos of 30 individuals (targets) who had been injected with an inflammatory challenge--low-dose endotoxin (i.e., sick) or placebo (i.e., healthy),

Salience, emotion, and attention: The neural networks underlying tinnitus distress revealed using music and rest.

In the present study, we used an innovative music-rest interleaved fMRI paradigm to investigate the neural correlates of tinnitus distress. Tinnitus is a poorly-understood hearing disorder where individuals perceive sounds, in the absence of an external source. Although the great majority of individuals habituate to chronic tinnitus and report few symptoms, a minority report debilitating distress and annoyance. Prior research suggests that a diverse set of brain regions, including the attention, the salience, and the limbic networks, play key roles in mediating both the perception of tinnitus
