Immune-sparing radiotherapy in solid tumors: radiation-induced lymphopenia, circulating immune-cell dose, and biomarker-guided optimization of radioimmunotherapy.

PURPOSE: Radiotherapy is a central component of solid tumor management and can enhance antitumor immunity through antigen release, immunogenic cell death, innate immune activation, and tumor microenvironment remodeling. However, radiotherapy can also induce systemic immune injury by depleting circulating lymphocytes and irradiating lymphoid organs. This review aims to examine radiation-induced lymphopenia (RIL) as a clinically relevant manifestation of systemic immune injury and its potential implications for radioimmunotherapy. METHODS: This narrative review discusses the biological basis of

Gastrin-Releasing Peptide Facilitates Cholinergically Mediated Contractions of the Mouse Gastric Fundus.

Gastrin-releasing peptide (GRP) and neuromedin B (NMB) are found in the stomach's myenteric plexus and muscular layers. When exogenously administered, they contract stomach muscle in several species, but their ability to modulate enteric nerve functions has rarely been studied. We investigated the effects of GRP on cholinergic-mediated contractions of the stomach, after first characterizing the contractile effects of GRP, NMB, and bombesin (BB). Circular muscle rings, cut from the mouse gastric fundus, were studied using tissue bath techniques. Contractions evoked by electrical field stimulati

Combined application of block and modulation factors to reduce the volume of the low dose area in helical tomotherapy plans for lung cancer.

BACKGROUND: The helical tomotherapy (HT) system can expose more normal lung tissue to low radiation doses, resulting in extensive low-dose distribution in both lungs which may induce radiation pneumonitis (RP). PURPOSE: This study aims to optimize dosimetric parameters and identify a clinically feasible treatment plan by analyzing the impact of different block settings and modulation factor (MF) combinations on HT plans for lung cancer to reduce the low-dose exposure volumes in normal lung tissue. METHODS: We retrospectively reviewed 14 lung cancer patients who received radiotherapy. These c

Lipopolysaccharide transport during long-chain fatty acid exposure is mediated by caveolin-1 dependent endocytosis in murine jejunum.

The entry of bacterial-derived lipopolysaccharides (LPS) from the intestinal lumen to the circulation induces low-grade systemic inflammation. We have found that LPS is transcellularly transported to the portal vein during luminal long-chain fatty acid (LCFA) exposure via CD36- and lipid raft-mediated pathways in rat jejunum, consistent with the involvement of caveolae-mediated endocytosis. We thus examined LPS transport in wild-type (WT) and caveolin-1 (Cav1) knockout (KO) murine jejunum. FITC-LPS was added to the mucosal bath of Ussing chambered muscle-stripped jejunal mucosa of WT and Cav1K

Effects of ketamine and propofol on muscarinic plateau potentials in rat neocortical pyramidal cells.

Propofol and ketamine are widely used general anaesthetics, but have different effects on consciousness: propofol gives a deeply unconscious state, with little or no dream reports, whereas vivid dreams are often reported after ketamine anaesthesia. Ketamine is an N-methyl-D-aspartate (NMDA) receptor antagonist, while propofol is a γ-aminobutyric-acid (GABAA) receptor positive allosteric modulator, but these mechanisms do not fully explain how these drugs alter consciousness. Most previous in vitro studies of cellular mechanisms of anaesthetics have used brain slices or neurons in a nearly "com

In vitro study of spontaneous motility and cholinergic responses in the human Hirschsprung's disease colon.

BACKGROUND: The current standard treatment of Hirschsprung's disease (HD) involves complete resection of the aganglionic intestinal segment with routine rectal irrigation pre- and postoperatively. To date, our understanding of the underlying pathomechanisms is not sufficient to warrant modifying this therapeutic approach. Here, we utilized resected tissue specimens from children with HD to investigate spontaneous motility and cholinergic responses in circular muscle from both ganglionic and aganglionic colon in vitro. METHODS: Full-thickness intestinal samples (from both aganglionic and gangl

Does Duty Cycle Modification Affect Temperature During Thulium Fiber Laser Lithotripsy?

OBJECTIVES: Although the thulium fiber laser (TFL, 1940 nm) efficiently fragments stones, concerns have been raised regarding heat production. Recently, a TFL with a modified duty cycle (MDC) was designed to reduce heat generation. The purpose of this study was to compare heat generation between the MDC and conventional TFL. METHODS: Ten millimeter BegoStone phantoms were placed in the proximal ureter of a kidney and ureter model. The model was submerged in a 35°C saline bath, with continuous saline irrigation (22°C) maintained at 15 mL/min through a ureteroscope. Temperature was measured usi

Virtual screening of FDA-approved drugs identifies amlodipine as a novel anti-Gyrodactylus kobayashii agent with therapeutic potential in aquaculture.

Parasitic infestations remain a major challenge in aquaculture, causing substantial economic losses. Current treatments for gyrodactylosis predominantly rely on unapproved chemicals, which often exhibit suboptimal efficacy. In this study, a molecular docking approach was employed to elucidate interactions between FDA-approved drugs and key residues within the active site of Gyrodactylus kobayashii proteins. Using AutoDock Vina, we screened a library of FDA-approved compounds and identified amlodipine and colistin sulfate salt as the most potent candidates against G. kobayashii. The minimum con
