Pathogenic characteristics of a novel intranuclear coccidia in Japanese black calves and its genetic identification as Eimeria subspherica.

Bovine intranuclear coccidiosis is caused by the protozoans Eimeria alabamensis and Cyclospora spp. Here, we characterized the disease and genetically identified the causative species in Japanese black calves with chronic and refractory watery diarrhea. Histologic examinations revealed atrophy of the jejunal villi and numerous parasites in the nucleus of epithelial cells in the jejunum. Based on molecular analyses using 18S ribosomal RNA gene-specific primers that we designed, the parasites were found to be formed in the same cluster as Eimeria subspherica in the phylogenetic tree, which was s

Striated muscle laminopathies.

Lamins A and C, encoded by LMNA, are constituent of the nuclear lamina, a meshwork of proteins underneath the nuclear envelope first described as scaffolding proteins of the nucleus. Since the discovery of LMNA mutations in highly heterogeneous human disorders (including cardiac and muscular dystrophies, lipodystrophies and progeria), the number of functions described for lamin A/C has expanded. Lamin A/C is notably involved in the regulation of chromatin structure and gene transcription, and in the resistance of cells to mechanical stress. This review focuses on studies performed on knock-out

Non-DNA binding, dominant-negative, human PPARgamma mutations cause lipodystrophic insulin resistance.

PPARgamma is essential for adipogenesis and metabolic homeostasis. We describe mutations in the DNA and ligand binding domains of human PPARgamma in lipodystrophic, severe insulin resistance. These receptor mutants lack DNA binding and transcriptional activity but can translocate to the nucleus, interact with PPARgamma coactivators and inhibit coexpressed wild-type receptor. Expression of PPARgamma target genes is markedly attenuated in mutation-containing versus receptor haploinsufficent primary cells, indicating that such dominant-negative inhibition operates in vivo. Our observations sugges

[Ultrastructural studies of neurons and synapses in the visual system of amblyopic cats].

14 kittens of 2-4 weeks were randomly divided into normal (N) and monocular deprivation (MD) groups. PVEP was tested before and after monocular deprivation by blepharorrhaphy for 12 weeks, when development of amblyopia was confirmed. Under TEM, the central retinal ganglion cells, the ipsilateral geniculate nucleus layers, the neurons of layer IV of visual cortex area 17, and the neural synapses of MD and N eyes were examined to find that atrophic and degenerative changes to various degrees existed in the MD eyes, supporting the authors belief that the functional changes in amblyopia were based

The action of aflatoxin B1 on the rat liver.

The administration of a single dose of aflatoxin B(1) to the rat (7mg./kg. body wt.) results in the slow development of a periportal necrosis. Hepatic enzymes are released into the serum in the second 24hr. of the poisoning, closely preceding the onset of the necrosis, which is followed by a rise in serum alkaline-phosphatase activity and bilirubin concentration. Aflatoxin B(1) has been detected in the nucleus of the poisoned liver cell and in vitro it has been shown to interact with DNA. The toxin inhibits the production of nuclear RNA, probably by preventing the transcription of DNA by the R

Clinical and pathological analysis of intravascular large B-cell lymphoma of the prostate.

BACKGROUND: Intravascular large B-cell lymphoma is a rare type of lymphoma. Although this tumor can occur in several parts of the body, primary intravascular large B-cell lymphoma of the prostate is extremely rare. Currently, there are only four case reports of primary large B-cell lymphoma in the prostate, and there is no comprehensive analysis of case series. Here, we report a case of intravascular large B-cell lymphoma originating from the prostate. Due to the unique morphology and atypical location of tumors under a microscope, pathologists often overlook them. CASE PRESENTATION: A 52-yea

Integrated analysis reveals the dysfunction of intercellular communication and metabolic signals in dilated cardiomyopathy.

AIMS: Dilated cardiomyopathy refers to a heart muscle condition characterized by structural and functional irregularities in the myocardium that are not related to ischemia. Due to diverse etiologies such as genetic mutations, infections, and exposure to toxins, dilated cardiomyopathy can lead to substantial morbidity and mortality despite advances in the management of heart failure in dilated cardiomyopathy patients. We sought to analyze the characteristics of cell-cell communication and the metabolic signaling pathways in dilated cardiomyopathy. METHODS AND RESULTS: The single-nucleus seque

In Silico Localization of Perilymph Proteins Enriched in Meńier̀e Disease Using Mammalian Cochlear Single-cell Transcriptomics.

HYPOTHESIS: Proteins enriched in the perilymph proteome of Meńier̀e disease (MD) patients may identify affected cell types. Utilizing single-cell transcriptome datasets from the mammalian cochlea, we hypothesize that these enriched perilymph proteins can be localized to specific cochlear cell types. BACKGROUND: The limited understanding of human inner ear pathologies and their associated biomolecular variations hinder efforts to develop disease-specific diagnostics and therapeutics. Perilymph sampling and analysis is now enabling further characterization of the cochlear microenvironment. Rece

A glaucoma- and ALS-associated mutant of OPTN induces neuronal cell death dependent on Tbk1 activity, autophagy and ER stress.

Mutations in OPTN are associated with glaucoma, an eye disease, and also with amyotrophic lateral sclerosis (ALS), a motor neuron disease. A 2-bp insertion in OPTN (691_692insAG or 2bpIns-OPTN) is associated with both glaucoma and ALS. This mutation results in frame shift after 127 amino acids, giving rise to a protein with C-terminal aberrant sequence. We have explored the mechanism of induction of cell death by this mutant in a motor neuron cell line, NSC-34, and also in a retinal cell line, 661W. Compared to wild-type OPTN, this mutant induced more cell death in NSC-34 and 661W cells. This

Functional Defects From Endocrine Disease-Associated Mutations in HLXB9 and Its Interacting Partner, NONO.

The insulin-secreting pancreatic neuroendocrine tumors, insulinomas, characterized by increased pancreatic islet β-cell proliferation, express the phosphorylated isoform of the β-cell differentiation factor HLXB9 that interacts with NONO/p54NRB, a survival factor. Interestingly, two different homozygous germline mutations in HLXB9, p.F248L and p.F272L, were reported in neonatal diabetes, a condition with functional β-cell deficiency. Also, two somatic heterozygous NONO mutations were found in endocrine-related tumors, p.H146R (parathyroid) and p.R293H (small intestine neuroendocrine tumor). Ho

Bax shuttling after rotenone treatment of neuronal primary cultures: effects on cell death phenotypes.

Neonatal (P7) brain hypoxia-ischemia (HI) induces intracellular Bax protein shifts to the nucleus, mitochondria, and endoplasmic reticulum (ER), where it triggers the activation of the respective cell death signaling cascades. When compared with HI-treated rat pups, 100% O(2) resuscitation of HI-treated rat pups increases HI-induced ER Bax levels, ER-mediated cell death signaling, and resultant lesion volume and inflammation due to increased necrotic-like cell death. To better characterize the role of Bax intracellular shuttling ER cell death signaling and necrotic-like cell death, we used rot

β-Catenin-Deficient Intervertebral Disc Cells Reduce Chemotactic Transcription and Myeloid Cell Recruitment to Injured Discs of Mice.

Aging exacerbates the incidence of discogenic chronic low back pain by excessive inflammation and immune cell presence. Aging reduces intervertebral disc (IVD) Wnt signaling, which is associated with IVD cell differentiation, IVD cell proliferation and anti-inflammatory signaling. However, it is unclear how in vivo Wnt signaling-absent IVD cells, which model aged IVD cells, regulate chemotactic signaling and immune cell recruitment. We hypothesized that Wnt signaling-absent IVD cells attenuate chemotactic transcription and immune cell recruitment to naive IVD and in response to IVD injury. Fir

Involvement of DNA-PK and ATM in radiation- and heat-induced DNA damage recognition and apoptotic cell death.

Exposure to ionizing radiation and hyperthermia results in important biological consequences, e.g. cell death, chromosomal aberrations, mutations, and DNA strand breaks. There is good evidence that the nucleus, specifically cellular DNA, is the principal target for radiation-induced cell lethality. DNA double-strand breaks (DSBs) are considered to be the most serious type of DNA damage induced by ionizing radiation. On the other hand, verifiable mechanisms which can lead to heat-induced cell death are damage to the plasma membrane and/or inactivation of heat-labile proteins caused by protein d

Dynamic association of MLL1, H3K4 trimethylation with chromatin and Hox gene expression during the cell cycle.

Mixed lineage leukemias (MLLs) are histone H3 at lysine 4 (H3K4)-specific methylases that play a critical role in regulating gene expression in humans. As chromatin condensation, relaxation and differential gene expression are keys to correct cell cycle progression, we analyzed the dynamic association of MLL and H3K4 trimethylation at different stages of the cell cycle. Interestingly, MLL1, which is normally associated with transcriptionally active chromatins (G1 phase), dissociates from condensed mitotic chromatin and returns at the end of telophase when the nucleus starts to relax. Single-Nuclear RNA Sequencing Reveals Regional Specialization and Cellular Interactions in Epicardial and Perivascular Adipose Tissue.

BACKGROUND: Adipose tissue surrounding the heart and vasculature plays critical roles in cardiovascular homeostasis and disease, yet the cellular and molecular milieu of these depots at single-cell resolution remains incompletely characterized. Understanding how regional adipocytes differ transcriptionally and communicate with neighboring cardiovascular cells is essential for developing targeted therapeutic strategies. METHODS: We performed single-nucleus RNA sequencing (snRNA-seq) on human adipose tissue from four anatomically distinct depots: ascending aorta, left atrium, right coronary art

Single-cell spatiotemporal dissection of the human maternal-fetal interface.

The human maternal-fetal interface is characterized by mosaic intermingling of maternal and fetal cells1. Yet the underlying cellular, molecular and spatial programmes remain incompletely defined. Here we generate a comprehensive atlas of the human maternal-fetal interface across normal pregnancies from early gestation to term by integrating large-scale paired single-nucleus transcriptomic and chromatin accessibility profiling with submicrometre-resolution spatial transcriptomics and CODEX multiplex protein imaging2, substantially boosting the spatiotemporal resolution of prior research3. Platelet-Derived Growth Factor Receptor α-Targeted Cell Membrane-Camouflaged Nanotherapy Disrupts Fibrosis-Inflammation Coupling in Intervertebral Disc Degeneration.

Intervertebral disc degeneration (IVDD) is a primary cause of chronic low back pain. Although inflammation is a prominent feature of degenerating discs, anti-inflammatory therapies often provide limited and transient benefit, suggesting that disc degeneration is maintained by a more stable tissue-level program. We therefore hypothesized that IVDD is sustained by a fibrosis-inflammation-coupled cell state, and that effective intervention requires both disrupting fibrotic signaling and overcoming the delivery barriers imposed by the disc's avascular, ECM-dense environment. To test this, we integ

Integrated Multi-Omics Analysis and Experimental Validation Identify UBE2Q2 as a Candidate Molecule Target in Intervertebral Disc Degeneration.

Intervertebral disc degeneration (IVDD) is a primary cause of low back pain, with its molecular mechanisms remaining incompletely understood. Ubiquitin-conjugating enzymes (E2 enzymes) play critical roles in protein degradation and maintenance of cellular homeostasis, yet their expression profiles and functional significance in IVDD remain largely unexplored. Using Gene Expression Omnibus (GEO) datasets (GSE70362 and GSE23130), we performed differential expression analysis, intersected with ubiquitination-related genes, and applied weighted gene co-expression network analysis (WGCNA) to identi

Lack of cytotoxic and genotoxic effects of mPEG-silane coated iron(III) oxide nanoparticles doped with magnesium despite cellular uptake in cancerous and noncancerous lung cells.

Cytotoxic and genotoxic effects of novel mPEG-silane coated iron(III) oxide nanoparticles doped with magnesium (Mg0.1-γ-Fe2O3(mPEG-silane)0.5) have been investigated on human adenocarcinomic alveolar basal epithelial (A549) and human normal bronchial epithelial (BEAS-2B) cells. In the studies several molecular and cellular targets addressing to cell membrane, cytoplasm organelles and nucleus components were served as toxicological endpoints. The as-synthesized nanoparticles were found to be stable in the cell culture media and were examined for different concentration and exposure times. No cy
