[Study of proliferation and apoptosis parameters of gastric epithelium cells in patients with precancerous pathologies of the stomach].

Changes in proliferation and apoptosis parameters of the gastric epithelium in patients with chronic Hp-associated gastritis were studied depending on the prevalence of the inflammatory process, presence of intestinal metaplasia and atrophic changes of the stomach glandular epithelium. The obtained results demonstrated a change in the cellular updating processes of the stomach epithelium depending on the localization, prevalence of the process, and more expressed - during the development of atrophic and dystrophic changes of the stomach mucous coat. Moderate amplification of apoptosis was reco

Investigation of cytokinin-deficient phenotypes in Arabidopsis by ectopic expression of orchid DSCKX1.

The plant hormone cytokinin plays a major role in regulating plant growth and development. Here we generated cytokinin-reduction Arabidopsis plants by overexpressing a heterologous cytokinin oxidase gene DSCKX1 from Dendrobium orchid. These transgenic plants exhibited reduced biomass, rapid root growth, decreased ability to form roots in vitro, and reduced response to cytokinin in growing calli and roots. Furthermore, the expression of KNAT1, STM, and CycD3 genes was significantly reduced in the transgenic plants, suggesting that cytokinin may function to control the cell cycles and shoot/root

Risk factors for the development of primary cutaneous malignant melanoma.

It is now possible to identify a small number of genes in which identifiable abnormalities are associated with an increased risk of melanoma, and a large number of phenotypic factors, which are associated with a lesser risk affecting a very much larger proportion of the melanoma population. As with colon cancer, it seems likely that there is a stepwise development of invasive malignant melanoma, and that interaction between genotype and environmental stimuli may be very different at each successive stage of melanoma progression. Unraveling these steps is the current challenge.

p16(INK4a) Promoter hypermethylation of non-tumorous tissue adjacent to gastric cancer is correlated with glandular atrophy and chronic inflammation.

The p16(INK4a) tumor suppressor gene can be inactivated by promoter region hypermethylation in many tumor types including gastric cancers. However, p16(INK4a) promoter hypermethylation in the surrounding non-tumorous tissues of gastric cancers has not been studied in detail. We therefore examined 46 gastric cancers, corresponding adjacent non-tumorous tissue samples and 8 gastric tissue samples of chronic gastritis by performing methylation-specific polymerase chain reaction, and we analyzed p16(INK4a) protein expression using immunohistochemistry and Western blot. p16(INK4a) promoter hypermet

A disrupted homologue of the human CLN3 or juvenile neuronal ceroid lipofuscinosis gene in Saccharomyces cerevisiae: a model to study Batten disease.

1. In order to investigate the biological function of the human CLN3 gene that is defective in Batten disease, we created a yeast strain by PCR-targeted disruption of the yeast gene (YHC3), which is a homologue of the human CLN3 gene. 2. The phenotypic characterization revealed that the yhc3 delta mutants are more sensitive to combined heat and alkaline stress than the wild-type strains as determined by inhibition of cell proliferation. 3. This suggests that the yhc3 delta mutant is a good model to investigate the biological function of human CLN3 gene in mammalian cells and to understand the

Impact of p16 expression on surgical management of malignant melanoma and pancreatic carcinoma.

BACKGROUND: Recent advances in molecular oncology have provided explanations at the DNA level for the malignant transformation and metastatic potential of various cancers. Malignant melanoma and pancreatic cancer may be classified together in both these cancers exhibit mutations in, or loss of, the cell-cycle inhibitory gene, p16. This paper reviews the current literature on p16 expression in melanoma and pancreatic cancer, explores factors that place patients with these cancers in categories of high risk for metastases or recurrence, and addresses whether aberrant gene expressions should infl

OVCA2 acts as an oncogene in pediatric AML by negatively regulating CDKN1A to drive cell cycle progression.

OBJECTIVES: Pediatric acute myeloid leukemia (AML) is biologically distinct from adult AML and carries a poor prognosis. OVCA2, a serine hydrolase with context-dependent roles, has unknown function in AML. We aimed to investigate its role, regulation, and clinical significance in pediatric AML. METHODS: OVCA2 was identified as a downstream target of a pediatric AML-specific core transcriptional regulatory circuit using CUT&Tag integrative analysis. Expression and prognostic associations were assessed in TARGET (pediatric) and TCGA (adult) datasets. Functional validation via lentiviral shRNA k

CRISPR/Cas9-Mediated Gene Knockout Reveals a Nonredundant Role for p16INK4A in Controlling TCR-Dependent and Independent CD8 T Cell Expansion.

The possibility of enhancing T cell function by deleting specific genes represents a long-sought goal in preclinical studies and ultimately for clinical applications. Using CRISPR/Cas9 genome editing, we report that, in human cytotoxic CD8 T cell clones, the cell cycle checkpoint gene CDKN2A, encoding p16INK4A, plays a nonredundant role in controlling T cell receptor (TCR)-dependent and independent cell expansion. Deletion of CDKN2A dramatically enhanced antigen-driven and homeostatic proliferation, while preserving effector functions. In contrast, the deletion of other cell cycle inhibitors (

CDK8 phosphorylates SOX2 to maintain stemness of pituitary adenoma.

Pituitary adenomas (PAs) are intracranial tumours with severe clinical complications and increased morbidity. Stem cell-like characteristics play a crucial role in the initiation and progression of PAs. In this study, we identified CDK8 as a critical regulator of stemness in PA tumorigenesis. Immunohistochemical analysis demonstrated that CDK8 expression is elevated in clinical PA samples and correlates significantly with Knosp grades, indicating its potential role in parasellar invasion. Inhibition of CDK8 significantly impaired the self-renewal capacity of patient-derived PA stem-like cells

Induced pluripotent stem cell-derived models of malignant nerve sheath tumor progression mimic glial to neuro-mesenchymal transition and uncover therapeutic opportunities.

Neurofibromatosis Type 1 (NF1) predisposes to peripheral nerve tumor development. The progression from a benign plexiform neurofibroma (PNF) towards a deadly malignant peripheral nerve sheath tumor (MPNST) is not completely understood but commonly involves the sequential loss of NF1, CDKN2A, and polycomb repressive complex 2 (PRC2). Here we use an iPSC-derived neural crest (NC) model to reproduce this malignant transformation through gene editing. NF1-CDKN2A double-knockout (2KO) NCs form neurofibroma-like tumors in vivo, requiring inactivation of p14ARF and p16INK4a. Additional PRC2 loss (3KO

Divergent neuropsychiatric and systemic toxicity profiles of abemaciclib and palbociclib: a triangulation study integrating pharmacovigilance, genetic epidemiology, and multi-omics profiling.

Cyclin-dependent kinase 4/6 inhibitors improve outcomes in hormone receptor-positive, human epidermal growth factor receptor 2-negative advanced breast cancer, but their toxicity profiles may differ in clinically meaningful ways. We aimed to compare the neuropsychiatric and systemic toxicity patterns of abemaciclib and palbociclib and to explore pharmacokinetic and molecular features that might contribute to these differences. We conducted a triangulation study integrating pharmacovigilance, Mendelian randomization, explainable machine learning, transcriptomics, and molecular docking. We analy

Exploring estrogen synthesis and metabolism related markers and immune dynamics in osteoporosis-insights from single-cell RNA sequencing and Mendelian randomization.

BACKGROUND: Osteoporosis (OP) is characterized by decreased bone mineral density and an increased fracture risk, often due to declining sex hormone levels that weaken bone strength. This study aims to explore estrogen related markers in OP to improve diagnosis and treatment. METHODS: Differentially expressed estrogen synthesis and metabolism-related genes (DE-ERGs) were identified through differential expression analysis and intersected with estrogen synthesis and metabolism-related genes (ERGs). Machine learning techniques and expression analysis were used to identify markers, followed by th

Abemaciclib in meningiomas with somatic NF2 or CDK pathway alterations: the phase 2 Alliance A071401 trial.

Systemic treatments are limited for patients with meningiomas that have progressed after surgery or radiation. Loss of NF2 and CDKN2A/CDKN2B is common in higher-grade meningiomas and promotes progression in preclinical models. We evaluated the efficacy of abemaciclib, a cyclin-dependent kinase 4/6 inhibitor, as one arm of the Alliance umbrella trial A071401, a genomically driven phase 2 study in recurrent and progressive meningiomas. Eligible patients with grade 2 or 3 tumors and NF2 mutations or CDK pathway alterations were treated with abemaciclib. Two co-primary endpoints were used: progres

Rare Germline Variants in CDKN2A-Negative Children and Adolescents With Cutaneous Melanoma.

Cutaneous melanoma is a complex disease influenced by both environmental and genetic factors. Inherited susceptibility plays a significant role, involving a combination of high-, intermediate- and low-penetrance genes. Melanoma in children and adolescents has been speculated to have a stronger genetic component due to the early onset. This study investigates germline variants in early-onset melanoma through exome sequencing of 154 patients in Australia diagnosed with cutaneous melanoma before the age of 20. Potentially pathogenic variants in shelterin complex genes were identified in 3% of the
