The widely accepted theory of endometriosis posits that endometriosis stems from the translocation of endometrial tissue through the fallopian tubes into the abdominal cavity. However, the exact pathogenesis and critical molecules of endometriosis remain unclear. Here, we find that alanyl-tRNA synthetase 1 (AARS1) is abundantly expressed in endometrial tissues and promotes the proliferation, migratory capability, and invasive potential in endometriotic stromal cells (EESC) and 11Z cells. Moreover, AARS1 enhances epithelial-to-mesenchymal transition in EESC and 11Z cells. In addition, AARS1 cou

Bartter syndrome type II is an autosomal recessive salt-losing tubulopathy caused by variants in the KCNJ1 gene, with a typical clinical phenotype of hypokalemia. In this study, we identified a 3-year-old child exhibiting hypokalemia and lower limb weakness. Genetic analysis identified a novel compound heterozygous mutation in the KCNJ1 gene (c.640A>G, p.Thr214Ala; c.1017C>A, p.Cys339Ter) in the patient, whereas each of the unaffected parents carried only one of these heterozygous mutations. Structural analysis indicated that the p.Thr214Ala variant impairs the stability of ROMK protein. Our f

Atherosclerotic cardiovascular disease (ASCVD) remains a leading cause of morbidity and mortality despite effective low-density lipoprotein cholesterol-targeted therapies. This review explores the crucial role of inflammation in the residual risk of ASCVD, emphasizing its impact on atherosclerosis progression and plaque stability. Evidence suggests that high-sensitivity C-reactive protein (hsCRP), and potentially other inflammatory biomarkers, can be used to identify the inflammatory residual ASCVD risk phenotype and may serve as future targets for the development of more efficacious therapeut

RNA modifications have recently emerged as critical posttranscriptional regulators of gene expression programs. They affect diverse eukaryotic biological processes, and the correct deposition of many of these modifications is required for normal development. Messenger RNA (mRNA) modifications regulate various aspects of mRNA metabolism. For example, N 6-methyladenosine (m6A) affects the translation and stability of the modified transcripts, thus providing a mechanism to coordinate the regulation of groups of transcripts during cell state maintenance and transition. Similarly, some modification

Copper complexes of a poly-His/poly-Gly peptide (EDDHHHHHHHHHGVGGGGGGGGGG-NH2), a natural component of a snake venom, were studied by means of both experimental (thermodynamic, spectroscopic and MS) techniques and molecular dynamics (MD) simulations and density functional theory (DFT) calculations. This peptide proved to be an exceptionally effective copper chelator, forming complexes which are thermodynamically more stable than those formed by both the albumin-like ATCUN motif and several other poly-histidine protein fragments. We show that, in a poly-histidine stretch, copper seems to prefer

We present the case of a 27-year-old female with subcortical osteonecrosis of the humeral capitulum. Percutaneous retrograde drilling of the lesion and application of recombinant human bone morphogenetic protein (BMP)-7 were combined with autologous bone grafting. At follow-up the patient was almost pain-free, had normalized her range of motion, and radiography showed consolidation of the lesion without any heterotopic bone formation. By timing surgery prior to subchondral collapse, biomechanical stability of the subchondral bone was maintained. To our knowledge, this is the first report on th

Mutations in NEK1 in mice are causal for cystic kidneys, and model the ciliopathy polycystic kidney disease caused by abnormal ciliary structure or signaling. NEK1 has previously been shown to localize near centrosomes and to play a role in centrosomal stability and ciliogenesis. Recent data suggest that the etiology of kidney cysts involves aberrant signaling from the primary cilium to the nucleus. Here we demonstrate that NEK1 contains functional nuclear localization signals, is exported from the nucleus via a nuclear export signal-dependent pathway and that the protein cycles through the nu

We examined autistic behavior in a cross-sectional sample of 179 children with fragile X syndrome (FXS) and a longitudinal subset of 116 children using the Childhood Autism Rating Scale (CARS) to (a) determine a prevalence of autistic behavior in FXS, (b) examine the stability of autistic ratings over time, and (c) assess the association between the fragile X mental retardation protein (FMRP) and autistic behavior. Approximately 21% of the sample of 129 children (25.9% of boys) scored at or above the cutoff for autism. CARS scores increased slowly, yet significantly, over time, and low levels

1. Site directed mutations were constructed in the yeast iso-1-cytochrome c gene adjacent to the lysine 77 (methylation site) codon. 2. These mutant genes were then cloned and transformed into the S. cerevisiae strain B-6642 which contains a deficiency in the iso-1-cytochrome c gene. 3. The resulting transformants were screened for cytochrome c production using gel electrophoresis. 4. Amino acid analysis of the mutated cytochromes c demonstrated varying levels of trimethyllysine formation, depending on the nature of the site directed mutation. 5. The resulting transformants were then used as t

Pyroptosis is a recently discovered inflammatory form of programmed cell death which is mostly triggered by infection with intracellular pathogens and critically contributes to inflammation. Mitigating pyroptosis may be a potential therapeutic target in inflammatory diseases. However, small chemicals to reduce pyroptosis is still elusive. In the present study, we screened 155 chemicals from a microbial natural product library and found Geldanamycin, an HSP90 inhibitor, profoundly rescued THP-1 cells from pyroptosis induced by LPS plus Nigericin treatment. Consistently, other HSP90 inhibitors,

Tuberous sclerosis complex (TSC) is a genetic condition characterized by the growth of benign tumours in multiple organs, including the brain and kidneys, alongside intellectual disability and seizures. Identification of a causative mutation in TSC1 or TSC2 is important for accurate genetic counselling in affected families, but it is not always clear from genetic data whether a sequence variant is pathogenic or not. In vitro functional analysis could provide support for determining whether an unclassified TSC1 or TSC2 variant is disease-causing. We have performed a detailed functional analysis

YTH N6-methyladenosine RNA binding protein F1 (YTHDF1) enables N6-methyladenosine-containing RNA binding activity, involved in mRNA destabilization and positive regulation of translational initiation. Here we aimed to investigate the main molecular events associated with YTHDF1 during the course of metabolic dysfunction-associated steatotic liver disease (MASLD). YTHDF1 expression was detected by western blotting and real-time PCR. Ythdf1 hepatocyte-specific knockout mice were generated in this study. RNA sequencing and proteomic analyses were performed to investigate potentially involved mole

Human Epidermal Growth Factor Receptor 2 (HER2) negative breast cancer is a type of breast cancer that does not overexpress the HER2 protein. In some breast cancers, overexpression of HER2 makes the cancer more aggressive. Computational-aided drug discovery (CADD) was taken as an efficient alternative to conventional methods of drug design and development. This study has also been carried out using the same drug design and development methods from Rhazya stricta; whose extracts are traditionally approved to possess anti-cancer activity while choosing Aurora Kinase A (AURKA) as our target of in

Rapidly progressive Alzheimer's disease (rpAD) is a rare but severe form of Alzheimer's disease characterized by accelerated cognitive decline and limited therapeutic options. Conventional anti-amyloid-β interventions have shown little success due to poor target specificity, neurotoxicity, and lack of efficacy, underscoring the need for novel therapeutic strategies. This study aimed to identify and prioritize molecular targets associated with rpAD by investigating the protein interactome of amyloid-β (Aβ42) using integrative computational approaches. This study developed a ratiometric electrochemical aptasensor to detect the H1N1 virus by targeting its hemagglutinin (HA) protein. In the experimental design, a Cu-MOF-NH2@Au nanocomposite was utilized as the sensing platform, where the Cu-MOF provided a stable internal reference signal at approximately − 0.2 V (vs. Ag/AgCl). Simultaneously, the ferrocene-L-cysteine complex (Fc-L-Cys) produced a target response signal at + 0.3 V, and the combination of these two signals enabled a ratiometric output mode. By immobilizing capture aptamers on the Cu-MOF-NH2@Au surface and enabling them to specif

Copper (Cu) is an essential mineral nutrient in animal, however, excess Cu causes mitochondrial homeostasis imbalance and multiple regulated cell death. This study explored how Cu overload disrupts the SAM50-MICOS-ATAD3-mtDNA axis and further activates cGAS-STING-NLRP3-mediated pyroptosis in pig liver via in vivo feeding model and in vitro hepatocyte with Cu chelator TTM, SAM50 overexpression and H₂O₂ positive control. We verified that excess dietary Cu triggers hepatic lipid metabolic disorder and impairs mitochondrial respiratory activity. Mechanistically, Cu weakens bidirectional protein in

Enterococcus faecium is a Gram-positive bacteria that infects the human gastrointestinal tract and it is a leading cause of hospital-acquired infections, due to its ability to cause various types of infections, such as endocarditis, bacteremia, urinary tract infections, and others, exacerbated by its multidrug resistance, notably to vancomycin. Because they are linked to major infections that are difficult to manage, and also due to the widespread acquisition of resistance genes, management remains difficult. This study employs pangenomic analyses and subtractive genomics to explore genetic di

Non-small cell lung cancer (NSCLC) remains a leading cause of cancer-related mortality worldwide, with limited therapeutic efficacy due to tumor heterogeneity in conventional treatments. In the present study, an integrative, network pharmacology approach was employed to elucidate the multi-target mechanism of action of phytochemicals derived from Glossocardia bosvallia against NSCLC. Among 38 phytocompounds identified, 31 compounds that satisfied pharmacokinetic properties were selected for subsequent analysis. Ligand-based target prediction identified 429 potential protein targets, which are

Brain somatic variants in the SLC35A2 gene, encoding for a Golgi galactose transporter, represent the major cause of mild malformation of cortical development with oligodendroglial hyperplasia in epilepsy (MOGHE). Clinical features associated with MOGHE include early-onset epileptic encephalopathy, drug-resistant focal epilepsy with developmental delay, and intellectual disability. Half of somatic SLC35A2 variants identified in MOGHE patients are predicted to encode full-length SLC35A2 protein or stable protein products. We investigated the pathophysiological basis of MOGHE by analyzing the fu
