Systematic Structure-Based Virtual Screening Approach to Antibody Selection and Design of a Humanized Antibody against Multiple Addictive Opioids without Affecting Treatment Agents Naloxone and Naltrexone.

Opioid drug use, especially heroin, is known as a growing national crisis in America. Heroin itself is a prodrug and is converted to the most active metabolite 6-monoacetylmorphine (6-MAM) responsible for the acute toxicity of heroin and then to a relatively less-active metabolite morphine responsible for the long-term toxicity of heroin. Monoclonal antibodies (mAbs) are recognized as a potentially promising therapeutic approach in the treatment of opioid use disorders (OUDs). Due to the intrinsic challenges of discovering an mAb against multiple ligands, here we describe a general, systematic

US Food and Drug Administration, Centers for Disease Control and Prevention, and National Institutes of Health Co-Sponsored Public Workshop Summary-Development Considerations of Antimicrobial Drugs for the Treatment of Gonorrhea.

There is an unmet need for developing drugs for the treatment of gonorrhea due to rapidly evolving resistance of Neisseria gonorrhoeae against antimicrobial drugs used for empiric therapy, an increase in globally reported multidrug-resistant cases, and the limited available therapeutic options. Furthermore, few drugs are under development. Development of antimicrobials is hampered by challenges in clinical trial design, limitations of available diagnostics, changes in and varying standards of care, lack of robust animal models, and clinically relevant pharmacodynamic targets. On 23 April 2021,

Synthesis and Antimalarial Activities of New Hybrid Atokel Molecules.

The currently spreading resistance of the malaria parasite Plasmodium falciparum to artemisinin-based combination therapies makes an urgent need for new efficient drugs. Aiming to kill artemisinin-resistant Plasmodium, a series of novel hybrid drugs named Atokels were synthesized and characterized. Atokels are based on an 8-amino- or 8-hydroxyquinoline entity covalently bound to a 1,4-naphthoquinone through a polyamine linker. These drugs have been designed to target the parasite mitochondrion by their naphthoquinone moiety reminiscent of the antimalarial drug atovaquone, and to trigger a dama

Linear-g-hyperbranched and cyclodextrin-based amphiphilic block copolymer as a multifunctional nanocarrier.

In this paper, a novel, multifunctional polymer nanocarrier was designed to provide adequate volume for high drug loading, to afford a multiregion encapsulation ability, and to achieve controlled drug release. An amphiphilic, triblock polymer (ABC) with hyperbranched polycarbonsilane (HBPCSi) and β-cyclodextrin (β-CD) moieties were first synthesized by the combination of a two-step reversible addition-fragmentation transfer polymerization into a pseudo-one-step hydrosilylation and quaternization reaction. The ABC then self-assembled into stable micelles with a core-shell structure in aqueous s

Medicinal chemistry strategies targeting viral proteases: From classical design to next-generation therapeutics.

Viral proteases are central targets in antiviral drug discovery and development because they play essential roles in viral replication and maturation. Although protease inhibitors have achieved major clinical success, traditional design strategies face challenges, including resistance development, poor oral exposure of early peptidomimetics, and off-target toxicity of highly reactive covalent warheads. Classical approaches, such as peptidomimetics, macrocyclization, and covalent warhead engineering, are discussed alongside contemporary strategies, including allosteric modulation and targeted p

Design, Synthesis, Biological Evaluation, and In Silico Analysis of Novel Antifungal Agents Targeting Trichophyton Species.

Terbinafine (Tb) is the primary commercial treatment for superficial nail infections; however, resistance is an emerging public health concern, largely driven by point mutations in the squalene epoxidase (SQLE) gene that reduce the drug's effectiveness. In this work, five novel synthetic compounds were developed and evaluated for their antifungal activity against 24 clinical isolates of Trichophyton rubrum and Trichophyton mentagrophytes. Among them, compound antifungal 2 demonstrated potent activity at the nanomolar level (ca. 73 nM), surpassing the efficacy of Tb (ca. 253 nM), and with less

Novel short-acting non-depolarizing muscle relaxants derived from galantamine: Design, synthesis, and pharmacological evaluation.

This study evaluated two novel galantamine-derived compounds, DR-976 and compound 15, as potential short-acting, non-depolarizing muscle relaxants. The aim was to introduce a long carbon chain bearing a quaternary ammonium group on the galantamine scaffold, in order to redirect its pharmacological profile from allosteric potentiating activity toward neuromuscular blocking effect. The compound effects were compared with those of d-tubocurarine (dTC), the classical competitive antagonist of nicotinic acetylcholine receptors (nAChRs). In anesthetized rats, both compounds produced dose-dependent i

Discovery of Novel Molecular Scaffolds to Overcome Pseudomonas aeruginosa Aminoglycoside Resistance: Insights for a Consensus Scoring Rational Design Approach.

The berberine derivative 13-(2-methylbenzyl)-berberine (BED) has been shown to inhibit the MexXY-OprM efflux system of Pseudomonas aeruginosa (PA), a key contributor to aminoglycoside resistance, by interacting with the inner membrane protein MexY at an allosteric pocket (ALP). To enhance binding efficacy, this study aims to identify novel chemical scaffolds that target the MexY allosteric pocket through an integrated computational strategy. In this work, a ligand-based virtual screening (LBVS) approach was employed using a 2D/3D pharmacophore model derived from BED to perform in silico screen

Design and structural optimization of lappaconitine derivatives as Nav1.7 inhibitors with improved analgesic efficacy and reduced lethality in mice.

Lappaconitine (LA), a diterpenoid alkaloid with analgesic properties, is limited clinically by its narrow safety margin. To overcome this, we synthesized novel LA derivatives modified at the 20-N position to enhance Nav1.7 potency and reduce lethality. Systematic SAR studies identified representative compound 39 for further characterization. Compound 39 inhibited human Nav1.7 (IC50 = 2.87 ± 0.19 μM), exhibiting an 8.6-fold increase in potency over LA (IC50 = 24.71 ± 1.64 μM). This translated to superior suppression of sodium currents and action potential firing in DRG neurons. In a mouse posto

Rigidity vs Activity: Design of Gramicidin S Analogs against Multidrug-Resistant Bacteria Based on Molecular Engineering.

Antimicrobial peptides are a promising class of therapeutics to address antibiotic resistance; yet, their clinical use is limited by toxicity and narrow-spectrum activity. To better understand how conformational rigidity influences efficacy and safety, a series of β-sheet antimicrobial peptide analogs based on gramicidin S were designed and synthesized. Two stapled derivatives (GSC-FB and GSC-SS) and a flexible linear analog (GS-L) were prepared and evaluated. GSC-FB retained potent activity against Gram-positive bacteria with a significantly reduced cytotoxicity. GS-L, characterized by increa
